La administración crónica de cafeína evita la alteración de la glucosa postprandial en ratas. El aumento en el consumo de la cafeína alrededor del mundo no es discutible, es así como su investigación se ha vuelto extensa en sus diferentes campos. Objetivo. Analizar los efectos de la administración crónica de cafeína en ratas alimentadas con dieta de cafetería, a través de evaluar índices de consumo, antropométricos y bioquímicos. Materiales y métodos. La dieta de cafetería es un modelo dietético equivalente a las características de la dieta occidental típica que origina síndrome metabólico en humanos. En esta investigación se realizó la administración crónica vía intraperitoneal de cafeína por ocho semanas a ratas adultas macho Wistar alimentadas con dieta de cafetería. Dada la poca evidencia acerca de los efectos biológicos y comportamentales de la administración crónica de dicha sustancia frente a un modelo de dieta de cafetería se evaluaron parámetros de consumo, antropométricos y bioquímicos. Resultados. La dieta de cafetería ocasionó anomalías asociadas al síndrome metabólico; no obstante, la administración de cafeína en las ratas alimentadas con esa dieta resultó ser un factor protector en la glucosa postprandial, más no en la alteración de la tolerancia a la glucosa o perfil lipídico. Conclusiones. La cafeína permitió proteger los niveles de glucosa postprandial al término del experimento y un descenso en el peso corporal y consumo de alimento solo en la primera semana. Sin embargo, no se observaron mejoras significativas en el perfil de lípidos, adiposidad, tolerancia a la glucosa y glucosa plasmática(AU)


Chronic caffeine administration prevents postprandial glucose disturbance in rats. The increase in caffeine consumption is not debatable, this is how his research has become extensive in his different fields. Objective. To analyze the effects of chronic administration of caffeine in rats fed a cafeteria diet, by evaluating consumption, anthropometric and biochemical indices. Previous studies refer to administering caffeine in diets high in carbohydrates and / or in fat that induce obesity or symptoms of metabolic syndrome. Material and methods. The cafeteria diet is a dietary model equivalent to the characteristics of the typical western diet that causes metabolic syndrome in humans. In this research, chronic intraperitoneal administration of caffeine was performed for 8 weeks to adult male Wistar rats fed a cafeteria diet. Given the little evidence about the biological and behavioral effects of the chronic administration of this substance against a cafeteria diet model, consumption, anthropometric and biochemical parameters were evaluated. Results. After eight weeks it was found that the cafeteria diet given to the controls caused abnormalities associated with the metabolic syndrome; regarding the administration of caffeine in the rats fed this diet, the treatment turned out to be a protective factor in postprandial glucose, but not in the alteration of glucose tolerance or lipid profile. Conclusions. Caffeine allowed to protect postprandial glucose levels at the end of the experiment and a decrease in body weight and food consumption only in the first week. However, no significant improvements were seen in lipid profile, adiposity, glucose tolerance, and plasma glucose(AU)

The aim of this study was to evaluate the nutritional and behavioral effects of a cafeteria diet in dams during the breastfeeding period and in their offspring from weaning until early adulthood (70 days old). Pregnant Wistar rats were fed a chow diet until delivery. Postnatally (D0), litters were culled to 8 pups and lactating dams received control (CTRL n= 6) or cafeteria (CAF n= 6) diets and water ad libitum. At the end of the breastfeeding period, male offspring were placed in individual boxes receiving the same treatment from their respective dams (CTRL or CAF) until adulthood (70 days). All nutritional and behavioral evaluations were performed with the dams (n= 12) during the breastfeeding phase and with the male offspring (n= 24) after weaning to adulthood. CAF dams demonstrated a lower caloric and protein intake; higher intake of fats; loss of weight; greater accumulation of adipose tissue; and an anxiolytic effect. CAF male offspring showed lower caloric intake; higher intake of fats; and accumulation of adipose tissue. In addition, these animals continued to have decreased body weight, body length and tibia-femur length in relation to CTRL. In dams, a cafeteria diet promoted alterations in body composition and anxiety, and in offspring the diet resulted in adequate development.


El objetivo de este estudio fue evaluar los efectos nutricionales y de comportamiento de la dieta de la cafetería en las madres durante el período de lactancia materna y en su descendencia desde el destete hasta la edad adulta temprana (70 días de edad). Ratas Wistar embarazadas fueron alimentadas con una dieta estándar hasta el parto. Postnatalmente (D0), las camadas se ajustaron en 8 crías y las madres lactantes recibieron las dietas control (CTRL n= 6) o cafetería (CAF n= 6) además de agua ad libitum. Al final del período de lactancia materna, las proles machos fueron colocados en cajas individuales recibiendo el mismo tratamiento de sus respectivas madres (CTRL o CAF) hasta la edad adulta (70 días). Todas las evaluaciones nutricionales y comportamentales se realizaron con las madres (n= 12) durante la fase de lactancia y con la prole masculina (n= 24) después del destete hasta la edad adulta. Las madres CAF demostraron una menor ingesta calórica y proteica; mayor ingestión de grasas; pérdida de peso; mayor acumulación de tejido adiposo; y un efecto ansiolítico. La prole masculina CAF presentó menor consumo calórico; mayor ingestión de grasas; y la acumulación de tejido adiposo. Además, estos animales presentaron menor peso corporal, longitud corporal, y longitud de la tibia-fémur, en relación a CTRL. En las madres, la dieta de cafetería promovió cambios en la composición corporal y ansiedad, y en la prole la dieta comprometió el desarrollo adecuado.

High-fat diets affect male reproduction and sexual function. Therefore, we evaluated the effects of prolonged resveratrol administration on the metabolic, sperm, and testicular parameters of rats fed a cafeteria diet. Male Wistar rats were divided at weaning into control (C, n = 20) and cafeteria (CAF, n = 16) groups. At 3 months, half of them were given daily supplementations of resveratrol (C-R, n = 10; CAF-R, n = 8) at a dosage of 30 mg kg-1 body mass for 2 months. Animals were killed at 5 months of age, and blood, spermatozoa, and testes were collected for further analysis. Data were analyzed by one-way ANOVA, and P < 0.05 was considered statistically significant. The CAF diet promoted hyperglycemia (P < 0.0001), and treatment with resveratrol reversed this condition (P < 0.0001). The CAF diet reduced sperm viability and motility, while resveratrol improved these parameters (P < 0.05). Regarding testicular morphology, the height of the seminiferous epithelium was reduced in the CAF group compared with that of the C group (P = 0.0007). Spermatogenic cell proliferation was also reduced in the CAF group compared with that of the C group. However, the CAF-R showed an increase in cell proliferation rate compared with that of the untreated CAF group (P = 0.0024). Although it did not modify body mass, the consumption of a CAF diet promoted hyperglycemia, adverse testicular morphology remodeling, and abnormal sperm, which were attenuated by treatment with resveratrol, thus suggesting a protective effect of this antioxidant on spermatogenesis.

Antiobesity effect of phenolic rich fractions of fresh fruits of Solanum aethiopicum and Solanum macrocapon on cafeteria diet (CD)-induced obesity was investigated using Wistar rats to explore the potential of S. macrocapon and S. aethiopicum in the management of obesity. Phenolic-rich extracts (free and bound phenolics) were obtained from S. macrocapon and S. aethiopicum by extraction with 80% (v/v) acetone and ethyl acetate respectively. Obesity was induced in Wistar rats with CD for 42 days after which they were treated with phenolic-rich extracts of S. aethiopicum and S. macrocapon at 200 mg/kg BW and 400 mg/kg BW for six weeks. Body weights and blood glucose were monitored weekly. Lipid profiles (Total Cholesterol (TC), Triglyceride (TG), High Density Lipoprotein -cholesterol (HDL-C), Very Low Density Lipoprotein – cholesterol (VLDL-C) and Low Density Lipoprotein – cholesterol (LDL-C)), Aspartate Transaminase (AST), Alanine Transaminase (ALT), Alkaline Phosphatase (ALP), urea, creatinine, total protein, albumin, atherogenic coefficient (AC) and cardiac risk ratio (CRR) were estimated in the plasma. Treatment with phenolic-rich extracts of S. macrocapon and S. aethiopicum caused decrease in lee index, atherogenic coefficient, blood glucose level, plasma TC, TG, LDL-c, VLDL-c, ALT, AST, AC, CRR and increase in HDL-c, urea, creatinine, total protein and albumin concentrations relative to the obese rats. Conclusively, the phenolic-rich extracts of S. macrocapon and S. aethiopicum possess antiobesity potential and could be employed in the management of obesity.

ABSTRACT Lychee fruit has been studied owing to the presence of several bioactive compounds that can contribute to weight loss in obese individuals. However, the anti-obese potential of the fruit has not been explored yet. The aim of this study was to investigate the effect of different parts of lychee in reducing the adipose tissue mass of cafeteria diet-fed rats. Phenolic compounds and scavenging capacities were quantified. The food intake, apparent digestibility, weight of the body and liver, body mass, Lee Index, and the mass of epididymal and visceral adipose tissues were evaluated. The results were estimated by Tukey's Test at 5% probability. A higher amount of phenolic compounds and scavenging capacity were observed in the peel of lychee as compared to the other parts of the fruit. The hypercaloric diet with lychee flour resulted in a higher apparent digestibility. There was no difference between groups control (C), hypercaloric (H), hypercaloric with lychee flour - 50.00% peel and 50.00% seeds (H2F), and hypercaloric with lychee flour - 33.33% peel, 33.33% pulp and 33.34% seeds (H3F) with respect to body and liver weight, corporal mass, and Lee Index. The hypercaloric diet-fed group exhibited an increase in visceral and epididymal adipose tissue mass, whereas the group fed with hypercaloric diets and flour made from the peel and seed of lychee presented a lower visceral adipose tissue mass. In conclusion, the use of lychee flour was considered viable because it decreased visceral adipose tissue mass in rats.

The present study investigated the effects of short-chain fructooligosaccharides (scFOS) feeding on body weight, fat accumulation, glucose homeostasis and lipid profile in cafeteria (CAF) obese rats. Male Wistar rats were divided randomly into two groups: control group (CTL, n = 10), which received a chow diet and water and CAF (n = 20), which received the cafeteria diet, standard chow and soda. After 30 weeks of diet, 10 animals of CAF group received scFOS in the diet (50 g kg-1 of diet) over a period of 50 days, forming the CAF FOS group. Were evaluated the body weight, fat pad as well as, quantity of feces, glucose tolerance, insulin resistance (IR) and serum lipids levels. Animals submitted to the CAF diet displayed obesity, hyperglycemia, glucose intolerance, hyperinsulinemia and IR. The scFOS feeding not altered obesity, glucose intolerance, hyperinsulinemia and IR. CAF rats also presented hypertriglyceridemia and lower levels of HDL-cholesterol. The CAF FOS animals had reduced serum triglycerides (TG) and increased HDL-cholesterol. Thus, the use of scFOS in the diet can be considered as a hypolipidemic agent in the obese state.


O presente estudo investigou os efeitos da adição de frutooligossacarídeos de cadeia curta (scFOS) sobre o peso corporal, acúmulo de gordura, homeostase glicêmica e perfil lipídico em ratos obesos pela dieta de cafeteria (CAF). Ratos Wistar foram divididos em dois grupos: controle (CTL, n = 10), que receberam dieta padrão e água e CAF, que receberam dieta de CAF, ração padrão e refrigerante (n = 20). Após 30 semanas, dez animais do grupo CAF receberam 50 g kg-1 de dieta de scFOS na ração padrão durante 50 dias, formando o grupo CAF FOS. Foram avaliados o peso corporal e o peso das gorduras, bem como, quantidade de fezes, homeostase glicêmica e concentração de lipídios séricos. Animais do grupo CAF apresentaram obesidade, hiperglicemia, intolerância à glicose, hiperinsulinemia e RI. A adição scFOS não alterou a obesidade, intolerância à glicose, hiperinsulinemia e RI no grupo CAF FOS comparado ao grupo CAF. Animais CAF também apresentaram hipertrigliceridemia e redução na concentração de HDL-colesterol. Os animais CAF FOS apresentaram redução na concentração sérica de triglicerídeos (TG) e aumento no HDL-colesterol. Desta forma, a utilização de scFOS na dieta pode ser considerado como um agente hipolipidêmico nos estados de obesidade.

Obesity is reaching epidemic proportions all over the world yet it lacks adequate treatment. Most of the drugs have failed either due to ineffectiveness or adverse effects. Complementary and alternative system of medicine is being used since ancient times. However, many of them have not been tested for efficacy and safety using modern scientific methods. Therefore, the antiobesity effect of Safoof Mohazzil, a polyherbal formulation, was evaluated in cafeteria diet induced obesity in female Sprague Dawley rats. Animals weighing 100–150 g were divided into four groups (n=8) i.e. standard pellet diet, cafeteria diet control, cafeteria diet + Safoof Mohazzil and standard pellet diet plus Safoof Mohazzil. The formulation was administered orally at a dose of 1 g/kg/day for 14 weeks. At the end of study, cafeteria diet significantly increased body weight, Lee’s index, lipid profile (cholesterol and triglycerides), insulin and leptin levels as compared to standard pellet diet control group. Fourteen week treatment with Safoof Mohazzil significantly prevented the increase in body weight, Lee’s index, lipid profile, insulin and leptin levels as compared to cafeteria diet control group without affecting food and water intake. Safoof Mohazzil had no adverse effect on hepatic transaminases, locomotor activity and motor coordination. The study provides evidence for antiobesity effect of Safoof Mohazzil.

To study the efficacy of ethanolic extract of B. monosperma bark in cafeteria and atherogenic diet fed rats and monosodium glutamate (MSG) obese rats, different doses (200, 400 and 800 mg/kg) of ethanolic extract of B. monosperma bark showed dose dependent decrease in body weight, daily food intake, glucose, lipids, internal organs’ weight and fat pad weight in cafeteria and atherogenic diet fed rats and monosodium glutamate obese rats. The results suggested that B. monosperma has significant anti-obese activity.

OBJECTIVE: Obesity is a strong predictor of some kinds of diseases. High intake of high-fat foods contributes significantly to the growth of the obese population globally. The aim of this study was to verify if consumption of a cafeteria diet for fourteen weeks could increase white fat mass, body weight and skeletal muscle mass and promote insulin resistance in male Wistar rats. METHODS: Twenty animals were divided into two groups: control and obese. Both were fed standard chow and water ad libitum. Additionally, a cafeteria diet consisting of bacon, bologna sausage, sandwich cookies and soft drink was given to the obese group. RESULTS: The obese group was significantly heavier (p<0.0001) than controls from the second week until the end of the cafeteria-diet intervention. Absolute and relative fat mass, liver weight and Lee Index increased significantly (p<0.05) in the obese group. Furthermore, the obese group had lower (p<0.05) insulin sensitivity than the control group. CONCLUSION: In conclusion, fourteen weeks of cafeteria diet promoted a progressive increase of fat mass and insulin resistance. Therefore, this is a great and inexpensive diet-induced insulin resistance model.


OBJETIVO: A obesidade é um forte preditor de alguns tipos de doenças. A grande oferta de comida e a predominância de ácidos graxos presentes na maioria dos alimentos contribuem diretamente para o aumento da população obesa no mundo. O objetivo do estudo foi verificar se uma dieta de cafeteria durante um período de catorze semanas pode causar aumento dos pesos de tecido adiposo branco, corporal e muscular e provocar um quadro de resistência insulínica em ratos machos Wistar. MÉTODOS: Vinte ratos machos Wistar foram divididos em dois grupos: controle e obeso. Os dois grupos foram alimentados com ração padrão e água ad libitum. Ao grupo obeso foi ofertada dieta ocidental, composta por salsicha, mortadela, bolacha recheada, bacon e refrigerante. RESULTADOS: Os animais do grupo obeso estavam significantemente mais pesados a partir da segunda semana de tratamento e essa diferença permaneceu até o final do estudo (p<0,0001). O peso absoluto e relativo do tecido adiposo branco e do fígado, e o Índice de Lee foram maiores no grupo obeso (p<0,05), que apresentou uma menor sensibilidade à insulina no final do estudo quando comparado ao grupo controle (p<0,05). Podemos observar que a dieta cafeteria promoveu um aumento progressivo e significativo da massa de gordura corporal associada à disfunção da ação da insulina. CONCLUSÃO: Conclui-se que uma dieta de cafeteria por um período de catorze semanas é capaz de promover aumento progressivo da massa adiposa associada à disfunção da insulina, sendo ótimo e fácil modelo de para induzir resistência à insulina.

OBJECTIVES: Olanzapine, an atypical antipsychotic drug with affinities for dopamine, serotonin, and histamine binding sites appears to be associated with substantial weight gain and metabolic alterations. The aim of this study was to evaluate weight gain and metabolic alterations in rats treated with olanzapine on a hypercaloric diet. METHODS: We used 40 rats divided into 4 groups: Group 1, standard food and water conditions (control); Group 2, standard diet plus olanzapine; Group 3, cafeteria diet (hypercaloric); and Group 4, olanzapine plus cafeteria diet. Olanzapine was administered by gavage at a dose of 3 mg/kg for 9 weeks. RESULTS There were no significant changes in the cholesterol levels in any group. Glucose levels increased in Group 3 by the fourth week. Triglyceride levels were altered in group 2 toward the end of the experiment. Leptin levels decreased in Groups 2 and 4. Complex II activity in the muscles and liver was altered in Group 2 (muscle), and Groups 2, 3, and 4 (liver). Complex IV activity was altered only in the liver in Group 2, without significant alterations within the muscles. CONCLUSION: These results suggest that olanzapine is correlated with weight gain and the risks associated with obesity.


OBJETIVOS: A olanzapina, uma droga antipsicótica atípica com afinidade por locais de ligação de dopamina, serotonina e histamina, parece se associar a um ganho de peso e a alterações metabólicas consideráveis. O objetivo desse estudo foi avaliar o ganho de peso e as alterações metabólicas em ratos tratados com olanzapina numa dieta hipercalórica. MÉTODOS: Usamos 40 ratos divididos em 4 grupos: Grupo 1, condições padrão de alimento e água (controle); Grupo 2, dieta padrão mais olanzapina; Grupo 3, dieta hipercalórica; e Grupo 4, olanzapina mais dieta hipercalórica. Olanzapina foi administrada por gavagem a uma dose de 3 mg/kg por 9 semanas. RESULTADOS: Não houve alterações significativas nos níveis de colesterol em qualquer um dos grupos. Os níveis de glicose aumentaram no Grupo 3 por volta da quarta semana. Os níveis de triglicerídeos estavam alterados no Grupo 2 ao final do experimento. Os níveis de leptina diminuíram nos Grupos 2 e 4. A atividade do complexo II nos músculos e no fígado se alterou no Grupo 2 (músculos) e nos Grupos 2, 3 e 4 (fígado). A atividade do complexo IV se alterou apenas no fígado no Grupo 2, sem alterações significativas nos músculos. CONCLUSÃO: Esses resultados sugerem que olanzapina se correlaciona ao ganho de peso e aos riscos associados à obesidade.

The objective of this work was to study the effect of the aerobic physical training and the cafeteria diet introduced after weaning of Wistar rats and on the morphology of the main salivary glands (parotid, submandibular, sublingual). Male rats after weaning were subjected to the cafeteria diet or the standard rodent chow, and either performed aerobic physical training in a treadmill for 100 days, or did not performed any physical activity. Analyses were done considering the response in body weight, adipose tissues and salivary glands, and the data were submitted to statistical treatment (p < 0.05). The morphological and morphometric analyses of the salivary glands were performed through histological sections stained with hematoxylin and eosin. Despite the normophagic behavior, the rodents fed with the cafeteria diet became obese, with repercussions on parotid gland weight. However, this obesity and/or physical training did not influence the histological organization of the salivary glands. The morphometric analysis of the submandibular glands pointed out a reduction in the levels of serous acinar cells as an effect of the diet and physical training. In conclusion, the parotid and the submandibular glands alter themselves due to the nature and consistency of food present in the cafeteria diet as well as due to the aerobic physical training.


O objetivo deste trabalho foi avaliar as características morfológicas das glândulas salivares (parótida, submandibular, sublingual) apresentadas por ratos submetidos a treinamento físico aeróbio e dieta de cafeteria após o período de lactação. Ratos machos após a lactação consumiram dieta de cafeteria ou ração-padrão para roedores e realizaram treinamento físico aeróbio em esteira rolante por um período de 100 dias, ou não realizaram nenhuma atividade física. Foram feitas análises sobre a resposta do peso corporal, dos tecidos adiposos e das glândulas salivares e os dados submetidos a tratamento estatístico (p < 0,05). A análise morfológica e morfométrica das glândulas salivares foi realizada a partir de cortes histológicos corados com Hematoxilina e Eosina. Apesar do comportamento normofágico, os roedores alimentados com dieta de cafeteria apresentaram maior quantidade de gordura corporal, com repercussão sobre o peso da glândula parótida. A análise morfométrica das glândulas submandibulares indicou redução na proporção dos ácinos serosos como efeito da dieta de cafeteria e do treinamento físico. No entanto, excesso de gordura corporal, dieta de cafeteria e/ou treinamento físico não influenciou a organização histológica das glândulas salivares. Concluiu-se que as glândulas parótidas e submandibulares são mais influenciáveis em função da natureza e consistência dos alimentos presentes na dieta de cafeteria, assim como do treinamento físico aeróbio.
