Gynerium sagittatum is a graminaceous plant widely used in the Caribbean coast of Colombia as a natural fiber source for the elaboration of handicrafts, particularly by the Zenú community. In the present investigation, the effect of different concentrations of cellulase and macerozyme enzymes at different incubation times and their interaction in the isolation of protoplasts was evaluated. Protoplasts were obtained from leaf mesophyll of G. sagittatum vitroplants exposed to enzymatic combinations of cellulase (1.5 and 2.0%), with macerozyme (0.3, 0.6 and 0.9%), for 3, 6 and 9 hours of incubation, for a total of 18 treatments with 5 replicates each. The highest numbers of isolated protoplasts corresponded to T18 (2.0% cellulase, 0.9% macerozyme), T12 (2.0% cellulase, 0.3% macerozyme), T3 (1.5% cellulase, 0.3% macerozyme) and T6 (1.5% cellulase, 0.6% macerozyme); at 9 hours incubation. The protoplast number for these treatments were: 88.625, 83.000, 75.000 and 53.375 protoplasts/mL respectively. Incubation time was significant in the isolation of protoplasts (p<0.05). The predictions between the factors showed that with an interaction of 2.0% cellulase and 0.9% macerozyme it is possible to obtain 44.302 protoplasts/mL, likewise, the incubation time-cellulase and incubation time-macerozyme interactions showed that it is possible to obtain 72.073 and 71.212 protoplasts/mL with 2.0% cellulase and 0.9% macerozyme for 9 hours of incubation respectively. The results indicate that the use of these enzymes and time, allows the isolation of of protoplasts from G. sagittatum in vitro plants.

Abstract The expansion of the use of machines in the field has promoted changes in the world of sugarcane work. Faced with this scenario, the question arises: How do work accidents and deaths behave in the mechanized reality of Brazilian sugarcane plantations? Predominantly quantitative, the research that supports this text uses secondary data collected from the Smartlab Platform and official government databases, linked to the formal labor market (RAIS). The results indicate that the set of agricultural activities (manual and mechanized) experienced a decrease in the incidence of work accidents, but did not show a reduction in the rate of work-related deaths. As a result, lethality increased in the period, demonstrating the preservation of the overexploitation of workers. This text is expected to shed light on the consequences generated by the agricultural mechanization process regarding the health and safety of sugarcane workers.

RESUMO

Abstract Xylan degradation is an important step in different industries, such as in biorefinery for biomass hydrolysis. Talaromyces wortmannii is a known fungus due to second metabolite production but only few works showed the xylanolytic potential of this fungus. In this way, the aim of this study was to evaluate the production of xylanolytic enzymes from T. wortmannii DR49 on industrial agro wastes. Cultivation in shake flask showed highest xylanase titration (10.3 U/mL; 9.5 U/mL) for wheat bran (WB) and hydrothermal pretreated sugar cane bagasse (HB); in β-xylosidase production WB and xylose were the best carbon sources (0.57 U/mL; 0.34 U/mL) respectively. STR cultivation revealed that 29°C and pH 6.0 were the best conditions for xylanase (14.5 U/mL) and β-xylosidase (1.7 U/mL) production. T. wortmannii DR49 showed to be a potential candidate for xylanolytic enzymes production using agro wastes in bioreactors, which has never been previously reported in this fungus.

ABSTRACT: The aim of this study was to evaluate a program for training basic probing cane techniques in the school environment. A 13-year-old female student with congenital blindness enrolled in the 1st grade of Middle School participated in the study. The instruments used were the evaluation protocols of the basic orientation and mobility techniques. The evaluation took place in three environments: corridors, classroom and schoolyard, and the quasi-experimental design consisted of pretest, intervention and posttest. The analysis was quantitative for the student's performance and, in the intervention, it was qualitative for the choice of the teaching strategies. The results of the pretest indicated that the student had no difficulty in performing the basic behaviors of the proposed techniques, but failed to perform those that are specific, which were the focus of the training. The teaching strategies were explanations, instructions and presentation of kinesthetic models. Posttest results pointed out that the student was able to perform most behaviors of all techniques in all environments. It was concluded that the training program was successful due to: the quasi-experimental study type that objectively allowed the evaluation of behaviors; registration and filming forms, which ensured control of the teaching process; the teaching strategies employed; teaching and training focused on the blind student's natural environment. Two points were identified as necessary to improve the use of the techniques: the type of clothing, the accessories, the student's shoes and the training time.

RESUMO

@#Introduction: The use of natural plant extracts to reduce blood glucose response has been practised from ancient times, although their industrial applications are rare. For the convenience of typical cane sugar consumers, selected tropical plant extracts were incorporated with cane sugar to make a low glycaemic sugar product. Methods: This study compared the glycaemic responses of a commercially available product (Product 1) containing seven herbal extracts; fenugreek (Trigonella foenumgraecum), turmeric (Curcuma longa), black pepper (Piper nigrum), ginger (Zingiber officinale), cinnamon (Cinnamomum verum), gooseberry (Phyllanthus emblica) and pomegranate (Punica granatum) against a novel product (Product 2) that was made by incorporating only two plant extracts that were cheaper and readily available in the local market - gooseberry (Phyllanthus emblica) and ginger (Zingiber officinale). Extracts were incorporated with cane sugar to make a series of crystallised solid sugar products and the formulae with the best sensory attributes was selected (Product 2). The glycaemic indices of both products were determined by standardised methodology using 12 healthy volunteers in a randomised crossover study. Results: The mean glycaemic index (GI) value for sugar in Product 1 was 49±9 and in Product 2 was 38±9. Both results elicited significantly (p<0.05) lower GI values than normal cane sugar (GI=65). The novel cane sugar product (Product 2) was far more superior compared to the commercially available product (Product 1) in reducing blood glucose response. Conclusion: Incorporation of suitable herbal extracts to cane sugar and foods like rice and wheat flour may be a suitable option to reduce their glycaemic impact.

RESUMO

Introduction: The fiber of the Gynerium sagittatum Aubl. P. Beauv is raw material for the elaboration of several handcrafts, which are symbols of Colombian cultural identity. In the manufacture process, different genotypes are used according to the fiber quality and the type of craftsmanship, but it is believed that Gynerium is a complex species, and to date, there is no agreement on whether these genotypes belong to the same species or to different species. Objective: The aim of this study was to quickly and accurately identify wild cane plants using the nuclear ribosomal internal transcribed spacer (ITS1+ITS2), three chloroplast regions (matK, rbcL, ycf1), and their combinations. Methods: Different tests were used for discrimination: (1) inter and intraspecific distances, (2) Best Match (BM), Best Close Match (BCM), and tree-based method (3) Neighbor Joining (NJ) and (4) maximum likelihood and bayesian inference in molecular data. Results: The results showed that BM and BCM approaches revealed the low rate of correct species identification for ITS+matK (33.3 %) and ITS (28.6 %) loci, showing similarity among sequences. These results were further supported by tree-based analyses, where all individual regions and the different gene combinations had a zero discrimination rate. Conclusions: all genotypes belong to the same species of wild cane, therefore existing morphological differences can be related to phenotypic plasticity.

INTRODUCTION Knee osteoarthritis is a relevant health problem given its high prevalence and associated disability. Within the non-pharmacological management alternatives, the use of canes has been proposed, however, there is no consensus in the literature regarding its indication. METHODS We searched in Epistemonikos, the largest database of systematic reviews in health, which is maintained by screening multiple information sources, including MEDLINE, EMBASE, Cochrane, among others. We extracted data from the systematic reviews, reanalyzed data of primary studies, conducted a meta-analysis and generated a summary of findings table using the GRADE approach. RESULTS AND CONCLUSIONS We identified three systematic reviews including four studies overall, of which one was randomized trials. We conclude that the use of a contralateral cane in patients with knee osteoarthritis probably reduces pain. In addition, it could slightly increase function, but the certainty of the evidence is low.

RESUMO

Forty eight crossbred calves, 24 males and 24 females, with weight and age of 203.6±3.7kg and 11.8±1.0 months were allotted to the following treatments: MT- mineral salt ad libitum and sugar cane mixed with 10g kg-1 mixture of urea and ammonium sulfate in a 9:1 proportion; MA - sugar cane mixed with 16g kg-1 of a concentrate of urea (550g kg-1), ammonium sulfate (60g kg-1) and minerals (390g kg-1); SU - sugar cane mixed with 50g kg-1 of a concentrate of soybean meal (830g kg-1), urea (52g kg-1) and minerals (118g kg-1) and MC - corn silage mixed with 10g kg-1 of a concentrate of urea (500g kg-1), ammonium sulfate (50g kg-1) and minerals (450g kg-1). Minerals mixed with sugar cane (MA) resulted in greater weight gain (0.258 x 0.188kg d-1) for similar intakes of DM (3.29 x 3.30kg d-1). Substitution of part of urea nitrogen in the MA by soybean meal (SU) resulted in greater ADG (0.538 x 0.258kg d-1), due to higher sugar cane intake (4.10 x 3.13kg d-1). ADG of calves receiving corn silage was greater (0.406 x 0.258kg d-1) than for calves receiving sugar cane.(AU)

ABSTRACT Soil erosion in earth roads of sugar cane is a problem that cannot be ignored, given the increase in planted area to this crop in Brazil. In this article soil losses are estimated using the Universal Soil Loss Equation (USLE), at four stretches of sugar cane earth roads of a farm at Itirapina (São Paulo State). Results showed that loss estimates ranged from 60.6 to 90.1 t.ha-1 for typic Quartzipsamment soil.

ABSTRACT The present work has the objective of investigating the possible recycling route for ether-amines, used for improving the efficiency of the process of iron ore's reverse flotation. A physical-chemical separation method has been studied using TANFLOC as flocculant. The wastewater contains, mainly, amine and silica, and the objective of this method was to separate the silica from the waste by flocculation. However, this method was not effective, as flocculation was not able to flocculate only silica, but also part of the ether-amine. Furthermore, the process of adsorption was assessed, and for this it was used as materials adsorbents Angelim's sawdust and sugar cane bagasse, both in natura, with which it was possible to remove approximately 95 and 90% of ether-amines, with adsorptive capacity of 4,2 and 2,7 mg.g-1, respectively, by using a mass of 1.5 g and a settling time of 30 minutes. It was verified that the removal efficiency is greater at a pH around 10. Thermodynamic studies were carried out and they were able to state, for sawdust, that this process is thermodynamically favorable and exothermic. In the case of sugar cane bagasse, it was not possible to analyze its thermodynamic behavior. In addition, the use of desorption revealed the possibility of regaining around 80% of the ether-amines. This proved that it is possible to recycle ether-amines by the adsorption mechanism.

RESUMO

Aim: Sugarcane (Saccharum officinarum L.) which is mostly cultivated in tropical and subtropical regions of the world is an important source of sucrose. This crop which was originally grown in South Eastern Asia and the Pacific for the sole purpose of chewing is mainly used nowadays for the production of sugar, bio-fuels, medicine etc. In spite of its numerous uses, the production of sugarcane in Buea, South West region of Cameroon is hampered by many factors including the incidence of disease. It is possible that some varieties are no longer cultivated because of their susceptibility to fungal diseases. It was for these reasons that we decided to survey the sugarcane farms in this area, to evaluate the presence of diseased symptoms due to activity of fungi. Study Design: Random sampling. Place and Duration of Study: Buea and its environs, Rubber Pathology Laboratory IRAD Ekona, and Life Sciences Laboratory, University of Buea, between June 2016 and July 2017. Methodology: During the survey, sugarcane farms in five villages in Buea and its environments were visited. The leaves and stems of the plants were observed for any symptoms of fungal activity. The incidence, severity and prevalence of the symptoms were noted. Leaf samples were collected from diseased sugarcane plants and transported to the laboratory for the isolation of fungi on Potato Dextrose Agar growth medium. The isolates were identified based on their cultural and micro-morphological characteristics. Results: Sugarcane plants with fungal disease symptoms were seen in all the farms visited during the survey, thus a prevalence of 100%. The disease incidence varied from 54- 100%, while the severity ranged from 20- 55%. Two genera of fungi were isolated from the diseased plants viz: Chaetomium sp. and Bipolaris sp. Conclusion: Chaetomium and Bipolaris are associated with fungal symptoms in diseased sugarcanes hence any management plans geared towards the improvement of sugarcane production in this area must include control measures for these fungi.

RESUMO

@#Background: Decreased rehabilitation time may increase the need for walking devices at the time of discharge to promote levels of independence among ambulatory individuals with spinal cord injury (SCI). However, using walking devices could create adverse effects on patients. This study explores the proportion of walking devices used, potential for walking progression, and associated factors among ambulatory individuals with SCI. Methods: Fifty-seven participants were assessed for their demographics and functional ability relating to the requirement for walking devices, including the Timed Up and Go Test (TUGT) and lower limb loading during sit-to-stand (LLL-STS). Results: Thirty-five participants (61%) used a walking device, particularly a standard walker, for daily walking. More than half of them (n = 23, 66%) had potential of walking progression (i.e., safely walk with a less-support device than the usual one). The ability of walking progression was significantly associated with a mild severity of injury, increased lower-limb muscle strength, decreased time to complete the TUGT, and, in particular, increased LLL-STS. Conclusion: A large proportion of ambulatory individuals with SCI have the potential for walking progression, which may increase their level of independence and minimise the appearance of disability. Strategies to promote LLL-STS are important for this progression.

ABSTRACT A culture medium for the multiplication of a coal solubilizing bacterial strain was designed (BSC3); were used three agroindustrial wastes: cane molasses, whey and crushed rice. Through Box-Behnken surfaces responses designs, two concentration ranks (2-10% y 0-6%) were evaluated, so the adequate combination to BSC3 biomass production was found. Bacterial growth curves were constructed to determine some kinetics parameters (growth specific rate [u], duplication time [Dt] and biomass final production), that were compared with the strain growth in a control culture medium; the optimized culture medium was also elementally caracterizated. The optimal concentrations to BSC3 biomass obtaining were: 6% molasses cane, 2,5% whey plus a salts minimum medium, with pH of 6,5. The kinetics parameters in this medium were: final biomass = 3,2 g.L-1, u=0,0206 h-1, Dt=33,64 h, and in the control medium: final biomass = 3,4 g.L-1, u=0,0139 h-1, Dt=49,85 h, this shows that the optimized medium allows the increase of the specific growth rate and a less duplication time of BSC3, so the optimized medium allowed to conserve its coal solubilizing activity.

RESUMO

Background: In this work, the xylanase production by Penicillium chrysogenum F-15 strain was investigated using agroindustrial biomass as substrate. The xylanase was purified, characterized and applied in hemicellulose hydrolysis. Results: The highest xylanase production was obtained when cultivation was carried out with sugar cane bagasse as carbon source, at pH 6.0 and 20°C, under static condition for 8 d. The enzyme was purified by a sequence of ion exchange and size exclusion chromatography, presenting final specific activity of 834.2 U·mg·prot-1. T he molecular mass of the purified enzyme estimated by SDS-PAGE was 22.1 kDa. The optimum activity was at pH 6.5 and 45°C. The enzyme was stable at 40°C with half-life of 35 min, and in the pH range from 4.5 to 10.0. The activity was increased in the presence of Mg+2 and Mn+2 and reducing agents such as DTT and ßmercaptoethanol, but it was reduced by Cu+2 and Pb+2 . The xylanase presented Km of 2.3 mM and Vmax of 731.8 U·mg·prot-1 with birchwood xylan as substrate. This xylanase presented differences in its properties when it was compared to the xylanases from other P. chrysogenum strains. Conclusion: The xylanase from P. chrysogenum F-15 showed lower enzymatic activity on commercial xylan than on hemicellulose from agroindustry biomass and its biochemistry characteristics, such as stability at 40°C and pH from 4.0 to 10.0, shows the potential of this enzyme for application in food, feed, pulp and paper industries and for bioethanol production.

RESUMO

ABSTRACT Saccharum spontaneum has been used for the development of energy cane a crop aimed to be used for the production of second-generation ethanol, or lignocellulosic ethanol. Lignin is a main challenge in the conversion of cell wall sugars into ethanol. In our studies to isolate the genes the lignin biosynthesis in S. spontaneum we have had great difficulty in RT-PCR reactions. Thus, we evaluated the effectiveness of different additives in the amplification of these genes. While COMT and CCoAOMT genes did not need any additives for other genes there was no amplification (HCT, F5H, 4CL and CCR) or the yield was very low (CAD and C4H). The application of supplementary cDNA was enough to overcome the non-specificity and low yield for C4H and C3H, while the addition of 0.04% BSA + 2% formamide was effective to amplify 4CL, CCR, F5H and CCR. HCT was amplified only by addition of 0.04% BSA + 2% formamide + 0.1 M trehalose and amplification of PAL was possible with addition of 2% of DMSO. Besides optimization of expression assays, the results show that additives can act independently or synergistically.

The effect of inclusion of cassava root silage on ruminal parameters and nitrogenous compounds in dairy cattle was evaluated. Five cattle cannulated in the rumen and with average body weight of 389 kg were distributed into a Latin square 5x5, with five levels of inclusion (0.00, 3.62, 7.23, 10.84 and 14.54% of the diet dry matter) of cassava root silage and five periods (10 days for adaptation and five days for data collection). The nitrogen excretion in urine and feces were not affected (P>0.05) by inclusion of cassava root silage in the diet. The total nitrogen intake (CNT) and nitrogen balance (NB) showed an increasing quadratic and linear behavior, respectively. The lowest CNT (142.14g/day) was obtained with the inclusion of 5.82% silage cassava root. The concentration of urea (U) and urea nitrogen (UN) in plasma and urine did not change (P>0.05). There was a quadratic behavior (P<0.05) in excreta of U and NU in the urine, with minimum point of 131.13 and 61.20g/day respectively, at the level of 7.27% inclusion of the cassava root silage. No interaction (P>0.05) was between the levels of cassava root silage and the collection time of rumen contents for the short-chain fatty acids, pH and ammonia nitrogen (NH3-N). Only the N-NH3 protein showed quadratic behavior, according to the inclusion levels, with the minimum point (12.62mg/100ml of rumen contents) at the level of 5.98%. The level of 7.23% of cassava root silage in the diet was more efficient with the use of nitrogen compounds. The diets contribute similarly to the production of short chain fatty acids, without disturbing the rumen pH.(AU)
