Background Cnidarian venoms and extracts have shown a broad variety of biological activities including cytotoxic, antibacterial and antitumoral effects. Most of these studied extracts were obtained from sea anemones or jellyfish. The present study aimed to determine the toxic activity and assess the antitumor and antiparasitic potential of Palythoa caribaeorum venom by evaluating its in vitro toxicity on several models including human tumor cell lines and against the parasite Giardia intestinalis. Methods The presence of cytolysins and vasoconstrictor activity of P. caribaeorum venom were determined by hemolysis, PLA2 and isolated rat aortic ring assays, respectively. The cytotoxic effect was tested on HCT-15 (human colorectal adenocarcinoma), MCF-7 (human mammary adenocarcinoma), K562 (human chronic myelogenous leukemia), U251 (human glyoblastoma), PC-3 (human prostatic adenocarcinoma) and SKLU-1 (human lung adenocarcinoma). An in vivo toxicity assay was performed with crickets and the antiparasitic assay was performed against G. intestinalis at 24 h of incubation. Results P. caribaeorum venom produced hemolytic and PLA2 activity and showed specific cytotoxicity against U251 and SKLU-1 cell lines, with approximately 50% growing inhibition. The venom was toxic to insects and showed activity against G. intestinalis in a dose-dependent manner by possibly altering its membrane osmotic equilibrium. Conclusion These results suggest that P. caribaeorum venom contains compounds with potential therapeutic value against microorganisms and cancer.

The immune system maintains the integrity of the organisms through a complex network of molecules, cells, and tissues that recognize internal or external antigenic substances to neutralized and eliminate them. The mechanisms of immune response have evolved in a modular fashion, where members of a given module interact strongly among them, but weakly with members of other modules, providing robustness and evolvability to the immune system. Ancestral modules are the raw material for the generation of new modules through evolution. Thus, the study of immune systems in basal metazoans such as cnidarians seeks to determine the basic tool kit from which the metazoans started to construct their immune systems. In addition, understanding the immune mechanisms in cnidarians contributes to decipher the etiopathology of coral diseases of infectious nature that are affecting coral reefs worldwide.


El sistema inmune mantiene la integridad de los organismos vivos por medio de una red compleja de moléculas, células y tejidos que reconocen sustancias antigénicas internas o externas para neutralizarlas y eliminarlas. Los mecanismos de respuesta inmune han evolucionado de una manera modular, en donde miembros de un módulo dado interactúan fuertemente entre sí, pero débilmente con componentes de otros módulos, otorgando así robustez y potencial evolutivo al sistema inmune. Módulos ancestrales representan el material básico para la generación de nuevos módulos durante el proceso evolutivo. Así, el estudio de sistemas inmunes en metazoarios basales como los cnidarios busca determinar cuales son los módulos ancestrales a partir de los cuales se constituyen los sistemas inmunes de animales derivados. Adicionalmente, el entendimiento de los mecanismos de respuesta inmune en cnidarios eventualmente contribuirá a descifrar la etiopatología de las enfermedades de corales de carácter infeccioso que está afectando los corales en el mundo.

La picadura de la medusa o agua viva Olindia sambaquiensis es de ocurrencia frecuente en las playas de la costa de la provincia de Buenos Aires. Se notifican entre quinientas y mil picaduras de medusas por temporada en la localidad balnearia de Monte Hermoso, al sur de la provincia de Buenos Aires. El daño cutáneo que provoca por su elevada acción urticante plantea un serio inconveniente para el desarrollo del turismo. Durante el mes de enero de 1998, en la localidad de Monte Hermoso se reunieron un total de 49 casos que fueron examinados dentro de la hora posterior a la picadura. Veintiocho eran de sexo masculino (57.1%). La edad promedio fue de 16 ± 4.1 (rango 5-80). El 54% presentó lesiones eritemato-edematosas de configuración lineal, el 28% presentó lesiones predominantemente eritematosas y el 18% presentó placas eritemato-edematosas. En el 73% de los casos las lesiones se localizaron en miembros inferiores. Hemos redefinido la lesión cutánea producida por la medusa O. sambaquiensis, su evolución, sus dimensiones y localizaciones más frecuentes. Además, se ha tipificado y cuantificado el dolor que produce, y otros signos y síntomas que acompañan a la picadura en la hora posterior, a las primeras 24 horas y a los 30 días. Describimos las conductas terapéuticas utilizadas en nuestra costa, evaluamos la efectividad de algunas de ellas en el control del dolor y finalmente proponemos un esquema terapéutico para dicha picadura.


Olindias sambaquiensis jellyfish sting occurs frequently in Buenos Aires province coast beaches. Among five hundred and one thousand stings by jellyfish are reported each season at Monte Hermoso, a beach village in the South of Buenos Aires province. The skin damage provoked because of its highly irritant effect poses a serious issue related to tourism development. A total number of 49 cases that were examined during the first hour after the sting were enrolled in Monte Hermoso during January 1998. Twenty eight were males (57.1%). The average age was 16 ± 4.1 (range 5-80). Of them, 54% showed linear erythema-edematous lesions, 28% showed predominantly erythematous lesions, and in 18% the injuries were erythema-edematous plaques. In 73% of the cases the lesions were located in lower limbs. We had hereby redefined cutaneous lesions produced by O. sambaquiensis, its evolution, its dimensions and most frequent localizations. Besides, it has been typified and quantified the pain it provokes and other signs and symptoms that go with the sting during the posterior hour, during the first 24 hours, and after 30 days. We described the therapeutic conducts used in our coasts and we assessed the effectiveness of some of them in pain control, and finally we propose a therapeutic scheme for this sting.

RESUMO

Seabather's eruption is characterized by the occurrence of intensely itchy erythematous papules observed mainly in the region covered by swimwear. The dermatitis occurs due to the contact of planula larvae of scyphomedusae Linuche unguiculata with the skin. The swimsuit pressure triggers the action of the poisonous stinging structures carried by the larvae. The case described occurred in a child who, while bathing in the ocean waters of the Northeast coast of Bessa's Beach located in the city of Joao Pessoa, state of Paraiba, showed typical clinical signs of the disease. It was concluded that the report of the case showed clinical and therapeutic implications for doctors working in all of the Brazilian coastal areas.


O prurido do traje de banho é uma erupção caracterizada por pápulas eritematosas, intensamente pruriginosas, localizadas principalmente sob os trajes dos banhistas de mar. A dermatite ocorre pelo contato com as larvas plânulas da cifomedusa Linuche unguiculata que disparam seus nematocistos na pele do acidentado a partir das suas células de defesa, os cnidócitos. O caso ocorreu em uma criança que ao se banhar nas águas oceânicas do litoral da Região Nordeste na Praia do Bessa localizada na cidade de João Pessoa, Estado da Paraíba, apresentou quadro clínico típico da enfermidade. Concluiu-se que o relato do caso evidenciou implicações clínicas e terapêuticas para os médicos que atuam em todas as áreas litorâneas brasileiras.

The unifying characteristic of cnidarians is the production of protein and polypeptide toxins. The present study describes the identification of a hemolytic toxin from the soft coral Sarcophyton trocheliophorum. The crude extract was highly cytotoxic (EC50 = 50 ng/mL) against human erythrocytes. It was also tested for hemolytic activity by the blood agar plate method, resulting in a hemolytic halo of 12 mm with 50 µg of protein. The stability of the venom under different physiological conditions was analyzed. The venom hemolytic activity was augmented by alkaline and neutral pH whereas it was reduced in acidic pH. The activity was stable up to 60º C. The hemolytic activity was completely abolished by the addition of serum and reduced significantly during frequent freezing-thawing cycles. Toxin purification was performed by ammonium sulfate precipitation and subsequently desalted by dialysis against 10 mM sodium phosphate buffer (pH 7.2), followed by anion exchange chromatography on DEAE cellulose column and gel filtration chromatography using Sephadex G-50 matrix. The purified active fractions possessed a prominent protein of approximately 45 kDa, as revealed by SDS-PAGE.(AU)

This study aimed to collect information on the diversity of corals and calcified hydroids of the Manuel Luiz Marine State Park (state of Maranhão, Northeast Brazil) to aid in the development of a management plan for the park. A total of 21 cnidarian species were identified, of which 16 were corals and calcified hydroids. The bathymetric and geographic distribution of each of these species was extended. This area has a rich coral fauna in relation to other parts of Brazil and is comparable in diversity to the Abrolhos Islands (Bahia State) - an unexpected result considering the region's proximity to the Amazon River Basin. Most of the specimens of corals and calcified hydroids observed were bleached.


O objetivo deste estudo foi obter informação sobre a diversidade dos corais e dos hidróides calcários do Parque Estadual Marinho do Parcel do Manuel Luiz (Estado do Maranhão, Nordeste do Brasil) para dar subsídios no desenvolvimento do plano de manejo do parque. Um total de 21 espécies de cnidários foi identificado, dos quais 16 eram corais e hidróides calcários. A distribuição batimétrica e geográfica de cada uma das espécies foram ampliadas. Esta área tem uma fauna coralínea rica com relação a outras partes do Brasil e é comparável em diversidade ao Arquipélago de Abrolhos (Estado de Bahia), um resultado inesperado considerando a proximidade da região à Bacia do Rio Amazonas. A maioria dos espécimes dos corais e dos hidróides calcários observados estava branqueada.
