RESUMO

ObjectiveTo observe the influence of basic fibroblast growth factor (bFGF),vascular endothelial growth factor (VEGF),epidermal growth factor (EGF),insulin and self-corium granule on the fat granule under cultivating and after transplantation.MethodsThe adipocytes of mouse were treated with bFGF,VEGF,EGF,insulin and self-corium granule in different concentration gradient post inoculation,respectively,to determine their different cytoactivity with MTT method,and then draw their activity curve with time.The various grafts were implanted into 32 SD rats,which contained bFGF,VEGF,EGF,insulin,self-corium granule or physiologic saline,and then the general condition of animals,quality determination,pathological changes and immunohistochemical staining were observed.At last,the effects of the different implants on the fat granule were compared in different methods.ResultsThe best cytoactive saturation of every experiental group was the same at different time points,except self-corium granule group,the cytoactivity of which was higher than that of control group (P<0.05).In animal experiment,focal necrosis and fat cistern were seen in comparatively larger area in the control group.The adipocyte of the experiental groups differed in size,and considerable small adipocytes were present.The capillary vessel density of all the experiental groups except the insuline group were higher than those of the control group (P<0.05).But the difference between the experiental groups and the control group became illegible gradually with time.ConclusionsThe activity of adipocyte being cultivated is differently increased by bFGF,VEGF,EGF and Insulin.The quantity of fat graft could be significantly raised by bFGF,VEGF,EGF and self-corium granule,which improve the revascularization of the fat graft after transplantation.
