RESUMO

With the rapid development and wide application of forensic DNA technology, strict management and guidance are required. Conclusion could be gained the public and judge recognition, would be more scientiifc, accurate and convincing by using the standard operation. In recent years, forensic DNA standard technology has been rapid development, but there are still insuffcient. This paper will summarize the present situation and the existing problems of forensic DNA technology standards and prospects.

Objective To explore the clinical value of examining HPV E6/E7 mRNA level in assessing cervical le?sions infected with high-risk human papillomavirus (HR-HPV). Methods The cervical epithelial cells were collected from 265 patients with HR-HPV infection, including 100 cases of neoplasia free/inflammation group (control group), 88 cas?es of cervical intraepithelial neoplasia (CIN)Ⅰ, 33 cases of CINⅡ, 28 cases of CINⅢand 16 cases of cervical carcinoma and the transcription of HPV E6/E7 mRNA level was examined using branched DNA (b-DNA) technology. Results The positive rate HPV E6/E7 mRNA were higher in CIN Ⅱ(81.82%), CINⅢ(89.29%) and cervical cancer group (100.00%) than tthat in control group (20.00%) and CINⅠ(35.23%) with significant difference, and there were no significant differences between other groups;The positive rate and transcription level of HPV E6/E7 mRNA in HSIL (high grade squamous intraepi?thelial lesion)and cancer group were significantly higher than normal, ASC(atypical squamous cell carcinoma) and LSIL(low grade squamous intraepithelial lesion) group (P<0.05). Conclusion The transcription level of HPV E6/E7 mRNA may re?flect the activity of the virus and the progression of disease, and could be use as an effective indicator to screen high grade cervical pathological changes and a complementary method of cervical lesion screening.

RESUMO

DNA topoisomerases are essential enzymes that control the topological state of DNA replication during mitosis. These enzymes are classified based on their mechanisms and physical properties. During mitosis, superhelical DNA must be unwound or relaxed by DNA topoisomerases prior to a decoding step by DNA processing enzymes, such as DNA polymerase and RNA polymerase. By blocking the reaction of resealing the breaks in the DNA ultimately can result in cellular death. Compounds that inhibit the catalytic function of these enzymes can serve as potential anticancer agents. DNA topoisomerases are found in nature and used as high quality and well-validated targets for the screening of potential anticancer agents. Our current work focuses on determining potential anticancer agents from natural resources using DNA topoisomerases as the screening targets. Large scale production of these enzymes using recombinant DNA technology in our academic laboratory is utilised to avoid dependence on expensive commercially available enzymes. The in-house produced enzymes can also be used to enhance our research in the field of molecular medicine by providing an enzyme source that can be used to screen potential anticancer agents, and for other newly developed diagnostic and medical research projects in the near future as well as a step in moving our efforts into the industrial sector.

RESUMO

Biotechnology is the knowledge and techniques of developing and using biological systems for deriving special products and services. The age-old technology took a new turn with the advent of recombinant DNA techniques, and boosted by the development of other molecular biological techniques, cell culture techniques and bioinformatics. Medical biotechnology is the major thrust area of biotechnology. It has brought revolutions in medicine – quick methods for diagnosing diseases, generation of new drugs and vaccines, completely novel approach of treatment are only a few to mention. The industrial and financial bulk of the industry mushroomed very rapidly in the last three decades, led by the USA and western advanced nations. Asian countries like China, India, South Korea, Taiwan and Singapore joined late, but advancing forward in a big way. In all the Asian countries governments supported the initiatives of the expert and entrepreneur community, and invested heavily in its development. Bangladesh has got great potential in developing biotechnology and reaping its fruits. However, lack of commitment and patriotism, and too much corruption and irresponsibility in political and bureaucratic establishment are the major hindrance to the development of biotechnology in Bangladesh.

RESUMO

La reciente utilización de la tecnología del ADN para la identificación individual a traído consigo una revolución en las ciencias forenses, que ha alcanzado también a la America Latina. El análisis histórico muestra que en Costa Rica sehan logrado importantes avances y en la actualidad se encuentra consolidado el trabajo con los STRs, y se están en proceso de implementación los marcadores de ADNmt y del cromosoma Y. Sin embargo, la incorporación delas innovaciones de la genética forense se ha venido realizando, cíclicamente, de 5 a 10 años tarde respecto a lospaises desarrollados en este campo. Se espera un cambio de actitud en el futuro, al estar disponibles nuevas generacionesde marcadores de ADN, que permitan explotar a corto plazo todo el potencial de esta útil herramienta al servicio de la justicia.
