RESUMO

Objective: The current work is intended towards the development of a novel, simple and precise high-performance thin layer chromatographic (HPTLC) method coupled with a densitometer for the estimation of teriflunomide (TEF) present in the marketed formulation. Methods: The chromatographic development was performed on aluminum plates coated with silica gel 60 F254 using toluene: ethyl acetate: glacial acetic acid (7.5:2: 0.5 v/v/v) as the mobile phase. Densitometric scanning was achieved at the absorbance maxima, UV 284 nm. Results: Well separated band was observed with Rf value 0.46. The calibration curve plotted in the concentration range 100-700ng/band exhibited an excellent linear relationship with the r2 value of 0.9928. The method was found to comply with all the validation parameters as per the ICH guidelines. Conclusion: The method ensures minimal use of mobile phase with minimal run time compared to other reported analytical methods. This validated method can be used by quality control laboratories for the routine quantitative analysis of tablets consisting of Teriflunomide.

Objective To analyze the metrological verification parameters and methods for dual-energy X-ray absorptiometry (DEXA) to facilitate magnitude traceability.Methods DEXA's principle was discussed from the aspects of dual-energy X-ray absorption method and hardware structure.The method combining corresponding theory and specifications was used to describe the measurement accuracy and repeatability as well as the verification method and data processing for radiation protection performance parameters.Theoretical analysis was executed on corresponding parameters.Results Theoretical analysis was carried out on the metrological verification parameters of DEXA from the aspects of principle structure and measuring method.Conclusion It's of great significance for enhancing DEXA metrological verification to master the metrological verification parameters and methods.

Osteoporosis is a disease in which the density of bone is decreased with consequent increase in bone fragility and susceptibility to fracture risk. Vitamin D deficiency may cause secondary hyperparathyroidism and low bone mineral density. Our study was aimed to assess relation between vitamin D status and parathyroid hormone and bone density in adults.Relatively healthy 369 participants aged between 20-60 were randomly selected from Ulaanbaatar city. Specially designed questionnaire was used in the survey. Bone mass density was diagnosed according to the WHO criteria by the T-score. Respondents serum calcium and phosphorus levels were described by the fully automated analyzer (Cobas Integra 2800, Germany). Serum parathyroid hormone and 25-hydroxivitamin D levels were described by using ELISA kit (Eucardio Laboratory, Inc. USA) at the Molecular biology laboratory of MNUMS.The statistical result was analyzed by SPSS 21 program.The subjects mean age was 47.61±12.92, with a range of 20-83. The mean T-score was -0.60±2.17. Our study showed that negative correlation between bone mass density values at parathyroid hormone (r=-0.58, p<0.05), 25-hydroxyvitamin D (r=-0.48, p<0.05) and serum calcium (r=-0.21, p<0.05).Our study showed increased serum parathyroid hormone led the releasing calcium from bone and decreasing bone mass density.

The cyanobacterium Oscillatoria agardhii was isolated from the fresh water Mawatha lake, Jaipur and was grown in Zarrouk's medium at 25 ± 2°C, illuminated with white fluorescent light at the intensity of 2 500 lux with 12:12 h light and dark photoperiod. The effects of photoperiod and temperature on the growth and protein expression by Oscillatoria agardhii were studied under different controlled culture conditions (ALR, ALC, CLR, CLC, and NDL), measuring optical density, cell count and dry weight. Protein content was measured quantitatively by Bradford assay and qualitatively by SDS-PAGE. The densitometric analysis was also carried out for the measurement of the expression level of different proteins/peptides under different culture conditions. Maximum growth and protein content were observed in ALR condition while minimum was in the CLC. Alternate light and dark periods proved efficient as contrasting banding patterns were observed with many new unique polypeptides such as 32, 36.3, 47.9, 60.8, and 67.0 kDa, whereas, expressions of three polypeptides of 57.2, 110.1, and 117.3 kDa were inhibited in constant light cultures.

Osteoporosis represents a reduced amount of bone tissue as compared with controls of same age and sex, and common feature of osteoporosis is a state of skeletal fragility leading to various fractures such as Colles' fracture, vertebral compression fracture, femur neck fracture, generally due to an inadequate amount of normally mineralized bone tissue, making the skeleton incapable of supporting normal mechanical demand. However it is difficult to quantify precisely its magnitude because of the variety of techniques used and the many bone sampling sites concerned. In 1970, Singh and associates(24.25) developed a method for grading the trabecular structure of the proximal end of the femur and reported that the femoral trabecular pattern index can clearly separate persons with osteoporosis from normal individuals. Cameron and its associstes developed an accurste method of determining bone mineral content by means of I-125 photon absorptiometry(5. 28). This method is consistently reproducible at the 98% level and has demonstrated agreement with ash measurement of cadaver bones at the 97% level. A comparative study of the usefulness of the femoral trabecular pattern index(Singh Index) and rsdial bone mineral content measurement by photon absorption was performed in 75 Kroean persons above forty years old. 25 of 75 persons were fracture patients due to osteoporosis. The results were as follows. 1. A significant difference in Singh index was noted between the fracture group snd control group. 2. A significant difference in bone mineral density was noted between the fracture group and control group. 3. A positive correlation was noted between the bone mineral denisty and Singh Index. 4. A significant difference in bone mineral density was noted between the fracture group and the control group which have same grade of Singh Index. So, we believe that bone mineral density is more objective and accurate than Singh Index. 5. We believe that both Singh Index and bone densitometer is useful in diagnosis and grading of osteoporosis.
