RESUMO

Objective:To evaluate the effect of thermocycling on nano-silver-incorporated silicone denture liner for Candida albicans (CA)adhesion.Methods:Specimens(1 0 mm ×1 0 mm ×3 mm)of silicone denture liner(control)and that incorporated with nano-sil-ver were prepared according to the manufacturer's direction.8 of the each kind of specimens were immersed in distilled water for 24 h at 37 ℃,and 8 were subjected to thermocycling for 4 000 cycles at temperatures ranging from (5 ±1 )℃to (55 ±1 )℃with a 60 s dwell time per temperature.In vitro CA adhesion assay was used to measure the levels of adherence to the specimens.Data were analysed sta-tistically.Results:Thermocycling increased CA adhesion to control specimens(P0.05 ).The adhesion of CA to nano-silver incorporated specimens was fewer than that to the controls with or wihout themocycling(P<0.01 ).Conclusion:Nano-silver incorporation may inhibit CA adhesion to silicone denture liner.Ther-mal cycling has no influence on C.albicans adhesion to silicone denture liner incorporated nano-sliver.

RESUMO

PURPOSE: The purpose of this study was to investigate the bonding properties of denture bases to silicone-based soft denture liners immersed in isobutyl methacrylate (iBMA) and 2-hydroxyethyl methacrylate (HEMA) for various lengths of time. MATERIALS AND METHODS: Polymethyl methacrylate (PMMA) test specimens were fabricated (75 mm in length, 12 mm in diameter at the thickest section, and 7 mm at the thinnest section) and then randomly assigned to five groups (n=15); untreated (Group 1), resilient liner immersed in iBMA for 1 minute (Group 2), resilient liner immersed in iBMA for 3 minutes (Group 3), resilient liner immersed in HEMA for 1 minute (Group 4), and resilient liner immersed in HEMA for 3 minutes (Group 5). The resilient liner specimens were processed between 2 PMMA blocks. Bonding strength of the liners to PMMA was compared by tensile test with a universal testing machine at a crosshead speed of 5 mm/min. Data were evaluated by 1-way ANOVA and post hoc Tukey-Kramer multiple comparisons tests (alpha=0.05). RESULTS: The highest mean value of force was observed in Group 3 specimens. The differences between groups were statistically significant (P<.05), except between Group 1 and Group 4 (P=.063). CONCLUSION: Immersion of silicone-based soft denture liners in iBMA for 3 minutes doubled the tensile bond strength between the silicone soft liner and PMMA denture base materials compared to the control group.

RESUMO

16 standered acrylic resin blocks with silicone resilient denture liner(10 mm ×8 mm ×3 mm)incorporated with nano-silver par-ticles at 5% (m/m,by dry wet)in-between were prepared.8 blocks were immersed in distilled water for 24 h at 37 ℃ as the controls (group 1).Another 8 were thermocycled for 4 000 cycles(group 2).16 acrylic resin bars were thermal cycled for 4 000 cycles.Then they were processed exactly as the control group (group 3).Tensile bond strength of the specimens was measured by a universal testing machine. The bond strength values(MPa)of group 1,2 and 3 were 2.683 ±0.435,2.179 ±0.633 and 1.460 ±0.566 respectively(group 1 vs 2,P>0.05;group 3 vs 1 or 2,P<0.05).The failure mode of the blocks in group 1 and 2 was mainly cohesive failure,in group 3 adhesion fail-ure.

O presente estudo teve como objetivo avaliar o efeito citotóxico dos monômeros isobutil metacrilato (IBMA) e 1,6 – Hexanediol dimetacrilato (1,6 – HDMA), do plastificante di-n-butil ftalato (DBP), e dos produtos de degradação ácido metacrílico (AM) e ácido benzóico (AB) sobre células L929. Esses compostos foram testados em faixas de concentrações liberadas, em um período de 30 dias, por materiais reembasadores rígidos, previamente quantificadas em estudos anteriores. O efeito citotóxico foi verificado por meio dos testes de MTT e 3H-timidina, após as células terem sido expostas às substâncias testadas nas concentrações estabelecidas. A classificação da citotoxicidade foi baseada na viabilidade celular em relação ao controle (células expostas ao meio sem as substâncias testadas). A atividade de síntese de DNA foi inibida por todos os compostos. Os resultados do presente estudo demonstraram que o teste de 3H-timidina foi mais sensível que o teste de MTT e que os compostos avaliados mostraram diferentes níveis de citotoxicidade in vitro. A atividade da desidrogenase mitocondrial diminuiu nas células tratadas com os monômeros, o plastificante e o produto de degradação AM; porém, para o AB, a maioria das concentrações testadas não apresentou efeito citotóxico.


The aim of this study was to evaluate the cytotoxic effect of the monomers 1,6 – hexanediol dimethacrylate (1,6 - HDMA) and isobutyl methacrylate (IBMA), the plasticizer di-n-butyl phthalate (DBP), and the degradation by-products methacrylic acid (MA) and benzoic acid (BA) on L929 cells. These compounds were tested in the range of concentrations released in 30 days from hard chairside reline resins that were quantified in previous investigations. Cytotoxic effects were assessed by using MTT and 3H - thymidine assays after the cells had been exposed to the test compounds at the given concentrations. Cytotoxicity was rated based on cell viability relative to controls (cells exposed to medium without test substances). The results presented in this study demonstrated that the 3H-thymidine assay was more sensitive than the MTT assay, and all compounds tested showed varying degrees of cytotoxic effect in vitro. DNA synthesis activity was inhibited by all compounds. Mitochondrial dehydrogenase activity decreased in cells treated with monomers, plasticizer and MA by-product, whereas no cytotoxic effect was observed on contact with BA at the majority of concentrations tested.
