Objective To investigate the effect of phlorofucofuroeckol A(PFFE-A)on the proliferation and invasion of colorectal carcinoma cells and its regulation of transforming growth factor-β1(TGF-β1)and mothers against decapentaplegic hom-olog 2/3(Smad2/3)signaling pathway.Methods The cells were processed as follows:the cells were intervened with low,medium and high doses of 50,100,and 150 μmol·L-1 of PFFE-A,respectively and cells in the normal control group were also established.5-Ethynyl-2'-deoxyuridine(EdU)staining was used to detect the cell proliferation.The transwell chamber was used to detect the invasion ability.A xenograft colon cancer nude mice model was used to detect the growth and metastasis ability of the cells in vivo.Real-time quantitative polymerase chain reaction(RT-qPCR)was used to detect the expression of epithelial-to-mes-enchymal transition(EMT)related genes.Western blotting was used to detect the expression levels of TGF-β1 and p-Smad2/3 in cells.Results Compared with normal control group,the proliferation rate,the number of invaded cells,the tumor mass,the pro-portion of tumor metastasis,the expression of N-cadherin mRNA,the expression of TGF-β1 and p-Smad2/3 were significantly de-creased(P<0.05),and the mRNA expression of E-cadherin was significantly increased(P<0.05).All were presented with a sig-nificant dose-dependent(P<0.05).Conclusion PFFE-A could inhibit the EMT process of tumor cells,inhibit the prolifera-tion and invasion of HT29 cells in vitro,and down-regulate the growth and metastasis of HT29 cells in vivo,which may be achieved by down-regulating TGF-β1/Smad2/3 signaling pathway.

Introduction : L'interprétation de la spirométrie se base sur des équations de référence, prenant en compte l'âge, le sexe, la taille et la race. Au CNHU-PPC, deux équations ont été utilisées ces dernières années : celles de l'American Thoracic Society/European Respiratory Society "ATS/ERS" (publiées en 1983, actualisées en 1993), et celles du Global Lung Initiative (GLI) 2012, désormais recommandées par plusieurs sociétés savantes. Notre étude visait à comparer les interprétations de spirométries basées sur ces équations. Matériel et méthodes : Nous avons mené une étude rétrospective descriptive, avec recrutement exhaustif des spirométries réalisées au CNHU-PPC du 1er Janvier 2018 au 31 Mars 2020. Les données recueillies furent analysées avec le logiciel R. Le coefficient Kappa a été calculé pour apprécier la performance des équations ATS/ERS par rapport au GLI 2012. Pour tous les tests statistiques, la différence était statistiquement significative pour une p-value inférieure à 0,05. Résultats : Les 955 spirométries recensées concernaient une population majoritairement féminine (sex-ratio=0,7) et jeune (âge moyen=44±20 ans). Il y avait plus de spirométries normales selon les équations ATS/ERS (53,6%, versus 53,0% selon GLI 2012 ; Kappa=0,71). Un TVO était objectivé dans 18,6% des cas selon l'ATS/ERS (versus 18,0% selon GLI 2012, Kappa=0,90). Il y avait moins de TVR selon l'ATS/ERS (21,3%, versus 29 % selon GLI 2012 ; Kappa=0,72), et moins de TVM selon l'ATS/ERS (5,3%, versus 6,6% selon GLI 2012 ; Kappa=0,79). Les proportions d'asthmatiques étaient identiques (12,3%). Les équations ATS/ERS ont objectivé moins de BPCO et de maladies restrictives (respectivement 4,6% et 21,3%) que le GLI 2012 (respectivement 5,8% avec Kappa=0,74, et 29% avec Kappa =0,72). Conclusion : Les équations ATS/ERS objectivent moins d'anomalies spirométriques que celles du GLI 2012 au CNHU-PPC. Des études ultérieures s'imposent pour intégrer les valeurs de référence béninoises au GLI 2012, actuellement récommandées pour l'interprétation de la spirométrie
Introduction: Spirometry's interpretation is based on reference equations, taking into account age, sex, height and race. At the CNHU-PPC, two equations have been used in recent years: those of the American Thoracic Society / European Respiratory Society "ATS/ERS"(published in 1983, updated in 1993), and more recently, those of the Global Lung Initiative (GLI) 2012, now recommended by several learned societies. Objectives: Our study aimed to compare interpretations of spirometry based on these equations. Material and methods: We carried out a descriptive retrospective study, with exhaustive recruitment of the spirometry done at the CNHU-PPC from January 1, 2018, to March 31, 2020. The data collected were analyzed with the software R. The Kappa coefficient was calculated to assess the performance of the ATS/ERS equations compared to GLI 2012. Results: The 955 spirometries recorded concerned a predominantly female (sex ratio=0.7) and young (mean age=44±20 years) population. There was more normal spirometry according to the ATS/ERS (53.6%, vs 53.0% according to GLI 2012; Kappa=0.71). An obstructive ventilatory disorder was objectified in 18.6% of cases according to ATS/ERS (vs 18.0% for GLI 2012, Kappa=0.90). There was less restrictive ventilation disorder according to ATS/ERS (21.3%, vs 29% for GLI 2012; Kappa=0.72), and less mixed ventilatory disorder according to ATS/ERS (5.3%, vs 6.6% for GLI 2012; Kappa=0.79). The proportions of asthma patients were identical (12.3%). ATS/ERS objectified less COPD and restrictive diseases (respectively 4.6% and 21.3%) than GLI 2012 (respectively 5.8% with Kappa=0.74, and 29% with Kappa=0.72).

Endoplasmic reticulum is one of the most important organelles in maintaining cellular homeo⁃ stasis, mainly involved in intracellular lipid synthesis, protein folding and calcium ion homeostasis. Trau⁃ ma, ischemia and hypoxia and other pathological changes can cause protein folding dysfunction in the en⁃ doplasmic reticulum, triggering endoplasmic reticulum stress (ERS). Spinal cord injury (SCI) is a com⁃ mon traumatic disease with anextremely high disability rate, which seriously affects the quality of life. There is no safe and effective clinical methods so far. Investigations have shown that ERS is one of the important pathological changes leading to cell death and neuronal dysfunction after SCI, and is closely as⁃ sociated with signaling pathways such as apoptosis, autophagy, and inflammation in neurons after SCI; however, the molecular mechanisms between ERS and SCI have not yet been thoroughly investigated. A deeper understanding and exploration of the potential molecular mechanisms associated with ERS and SCI may be the future SCI therapeutics. In this paper, we first summarized the relationship between the chan⁃ ges of ERS⁃related genes and the pathological process of SCI, Then the interrelationships between ERS and the apoptosis, inflammation, and autophagy signaling pathways after SCI were analyzed, starting from the three main modes of regulation, including unfolded protein response (UPR), endoplasmic reticulum⁃ associated degradation (ERAD), and endoplasmic reticulophagy (ER⁃phagy),Finally, we summarize the relevant drugs and application prospects of targeting ERS for SCI in recent years, which may provide a theoretical basis for targeting the ERS pathway for SCI and provide some ideas and insights for the de⁃ velopment of future therapeutic strategies after SCI.

Endoplasmic reticulum stress (ERS) is a cellular defensive response to restore homeostasisand reduce the protein load. Over-activation of ERS can induce cell differentiation, proliferation, apoptosis, and autophagy. MicroRNAs (miRNAs) are endogenous non-coding RNAs (ncRNAs) thatregulate the expression of key proteins and genes in the ERS signaling pathway through post-transcriptional action. Meanwhile, activated ERS signaling pathway can indirectly regulate the expressionand function of target genes by decreasing miRNA stability. Based on a brief introduction of ERS classicalsignaling pathways, this paper further elaborated how microRNAs regulate ERS signaling pathways topromote apoptosis and proliferation, and what effect they would have on the expression profile of diseasesbased on this association. We also summarize the regulation of ERS on miRNAs expression and thecurrent research status. The mutual regulation between the two could provide a new idea for the follow-upresearch on the therapeutic targets of diseases.

Objective:To investigate the inhibitory effect of Fuzheng Jiedu prescription(FZJDP) combined with 5-fluorouracil (5-Fu) against postoperative recurrence and metastasis in gastric cancer-bearing mice and explore the possible mechanism based on changes in CD4<sup>+</sup> T cells, CD8<sup>+</sup> T cells, and regulatory T (Treg) cells of tumor microenvironment, endoplasmic reticulum stress (ERS) and phosphatidylinositol-3-kinase (PI3K)/protein kinase B(Akt)/mammalian target of rapamycin(mTOR) pathway. Method:Forty 615 mice were randomly divided into the model group,FZJDP (25 g·kg<sup>-1</sup>) group,5-Fu(25 mg·kg<sup>-1</sup>)group,and combined (25 g·kg<sup>-1</sup> FZJDP + 25 mg·kg<sup>-1</sup> 5-Fu)group, with 10 mice in each group. Mouse forestomach carcinoma (MFC) cells were implanted into the inner side of footpad of the left hind paw and the transplanted tumor was then surgically excised to establish a postoperative recurrence model. Hematoxylin-eosin(HE) staining was conducted to observe the pathological changes in mice with gastric cancer recurrence and lung metastasis. The CD4<sup>+</sup>/CD8<sup>+</sup> T cell ratio in recurrent tumor and the percentages of Treg cells [CD4<sup>+</sup>,CD25<sup>+</sup>, and forkhead box protein P3 (FOXP3)<sup>+</sup> cells] in spleen were detected by flow cytometry. The contents of ERS-related proteins [78-kDa glucose-regulated protein(GRP78),inositol-requiring enzyme 1 alpha (IRE1<italic>α</italic>),activating transcription factor 6(ATF6),and protein kinase R-like endoplasmic reticulum kinase(PERK)] and the expression of related proteins in the PI3K/Akt/mTOR signaling pathway were determined by Western blot and immunohistochemistry (IHC). Result:Compared with the model group, the combined group significantly increased the recurrent inhibition rate (<italic>P</italic><0.05). The recurrent tumor weight was significantly decreased in each treatment group (<italic>P</italic><0.05). The number of lung metastases and metastasis rate declined in each treatment group, and the lowest values were observed in the combined group, without any statistical significance. The CD4<sup>+</sup>/CD8<sup>+</sup> T cell ratios in the FZJDP group and combined group were significantly elevated (<italic>P</italic><0.05), while the percentages of Treg cells were reduced (<italic>P</italic><0.05). However, 5-Fu resulted in a significant increase in Treg cell percentage (<italic>P</italic><0.05). IHC results showed that the protein expression levels of ATF6 (<italic>P</italic><0.05,<italic>P</italic><0.01), IRE1<italic>α</italic>(<italic>P</italic><0.05),and Akt (<italic>P</italic><0.01) in each treatment group were significantly down-regulated as compared with those in the model group. As revealed by Western blot, the GRP78 expression level in the 5-Fu group was lower than that in the model group (<italic>P</italic><0.05), and the expression levels of PI3K, phosphorylated Akt (p-Akt), and mTOR were significantly decreased in the 5-Fu group and the combined group (<italic>P</italic><0.05). Conclusion:FZJDP combined with 5-Fu reduces postoperative recurrence and metastasis in tumor-bearing mice possibly by inhibiting PI3K/Akt/mTOR signaling pathway, diminishing ERS,and improving tumor immune microenvironment.

Objective:To investigate the regulatory mechanism of Bushen Zhuyun prescription(BSZYP) on endoplasmic reticulum stress (ERS) in rats with luteal phase defect (LPD) induced by mifepristone. Method:Fifty SD rats were randomly divided into a blank group, a model group, a positive control group (dydrogesterone,0.02 g·kg<sup>-1</sup>), and low-(0.08 g·kg<sup>-1</sup>)and high-dose (0.24 g·kg<sup>-1</sup>) BSZYP groups. Western blot and Real-time fluorescence-based quantitative polymerase chain reaction (Real-time PCR) were used to detect the mRNA and protein expression levels of immunoglobulin binding protein (BIP), protein kinase R-like endoplasmic reticulum kinase (PERK), inositol-requiring enzyme 1 (IRE-1), activating transcription factor 6 (ATF6), and C/EBP homologous protein (CHOP). The enzyme-linked immunosorbent assay (ELISA) was used to detect the serum progesterone (P) and estradiol (E<sub>2</sub>) levels. Result:Compared with the blank group, the model group showed the elevated protein expression of BIP, PERK, and CHOP (<italic>P</italic><0.01) and the dwindled mRNA expression of PERK and CHOP (<italic>P</italic><0.05), while no significant difference was observed in the protein expression of IRE-1 and ATF6, mRNA expression of IRE-1, BIP, and ATF6, and serum E<sub>2</sub> and P levels. Compared with the model group, the positive control group displayed diminished protein expression of CHOP (<italic>P</italic><0.01), while no significant difference was observed in the protein expression of PERK, IRE-1, BIP, and ATF6, mRNA expression of PERK, IRE-1, BIP, ATF6, and CHOP, and serum levels of E<sub>2</sub> and P. The protein expression of CHOP decreased (<italic>P</italic><0.01) and the mRNA expression of CHOP increased (<italic>P</italic><0.05) in the low-dose BSZYP group, while no significant difference was observed in the mRNA and protein expression of PERK, IRE-1, BIP, and ATF6, and serum E<sub>2</sub> and P levels. In the high-dose BSZYP group, the protein expression of PERK, BIP, and CHOP was down-regulated (<italic>P</italic><0.01), and the mRNA expression of CHOP was up-regulated (<italic>P</italic><0.01), while no significant difference was observed in the protein expression of IRE-1 and ATF6, mRNA expression of PERK, IRE-1, BIP, and ATF6, and serum E<sub>2</sub> and P levels. Conclusion:BSZYP can treat LPD by relieving ERS.

Aim To investigate the effect of ellagic acid on human cervical cancer cells and its correlation with endoplasmic reticulum stress ( ERS) - mitochondrial apoptosis pathway. Methods Hela cells were treated with ellagic acid, and cell survival rate, apoptosis and expression of ERS mitochondrial apoptosis pathway protein were detected. After pretreatment with ERS inhibitor 4-PBA, the expression of mitochondrial apoptosis pathway protein was detected. Results Ellagic acid showed concentration-dependent and time- dependent killing effect on Hela cells, and could significantly increase the apoptosis of Hela cells. Ellagic acid significantly increased the expression levels of ERS related proteins (GRP78, CHOP, p-PERK, p-IREl) in Hela cells, and significantly increased the expression levels of mitochondrial proapoptotic proteins . Bax and cleaved caspase-3, while significantly decreased the expression level of mitochondrial anti apop- totic protein Bcl-2. Another group of experiments showed that pretreatment with ERS inhibitor 4-PBA could significantly reduce the expression levels of Bax and cleaved caspase-3, and significantly increase the expression level of Bcl-2, which was close to the normal group. Conclusions Ellagic acid can significantly promote the apoptosis of human cervical cancer cells, and its mechanism is related to ERS mitochondrial apoptosis pathway.

Objective:To observe the effect of blood stasis syndrome on renal damage and endoplasmic reticulum stress (ERS) in diabetic kidney disease (DKD) rats, and to explore the relationship between renal syndrome of blood stasis damage and ERS in DKD rats. Method:The 50 Male SD rats of SPF level were selected to establish DKD rat model by high fat and high sugar diet combined with intra-abdominal injection of streptozotocin(STZ). They were randomly divided into normal group, diabetes mellitus group and diabetes mellitus and blood stasis syndrome group(0.05 mg·kg-1), among which diabetes mellitus and blood stasis syndrome group was prepared by dextran method. The general conditions, hemorheology indexes, 24 h urine protein, serum creatinine and renal pathology of the rats in each group were observed. Immunohistochemical analysis, Western blot and Real-time fluorescent quantitative polymerase chain reaction(Real-time PCR)were used to detect mRNA and protein expressions of endoplasmic reticulum stress-related proteins glucose regulated protein 78(GRP78), activating transcription factor-6(ATF6) and renal fibrosis index fibronectin(FN), and alpha smooth muscle actin(α-SMA). Result:Compared with normal group, the rats in diabetes mellitus group showed polyphagia, polyuria and weight loss, increased hemorheology index of whole blood viscosity and plasma viscosity(P<0.05,P<0.01), increased 24 h urine protein, serum creatinine and urea nitrogen(P<0.01), and increased renal pathology, and increased mRNA and protein expression of GRP78, ATF6, FN and α-SMA(P<0.05,P<0.01). After dextran preparation of blood stasis model. Diabetes mellitus and blood stasis syndrome group increased mortality, signs of change is more obvious in the diabetic group, whole blood viscosity, plasma viscosity, 24 h urine protein ration, serum creatinine and urea nitrogen were significantly higher than those in diabetic rats(P<0.01), pathological changes aggravated in the diabetes group. At the same time, mRNA and protein expressions of GRP78, ATF6, FN, and α-SMA in renal tissue were significantly higher than those in diabetic mellitus group(P<0.05,P<0.01). Conclusion:Under the combined disease and syndrome model, the blood stasis syndrome may further aggravate the pathological damage of the kidney of DKD rats, and is related to the enhancement of ERS in the kidney of DKD rats.

This study aimed to investigate the effect and possible mechanism of Bidens pilosa decoction on non-alcoholic fatty liver disease(NAFLD) induced by high fat and high glucose in mice. Bald/c mice were randomly divided into normal group, model group, metformin(200 mg·kg~(-1)) treatment group, Bidens pilosa decoction(10 g·kg~(-1)) treatment group, metformin and B. pilosa decoction(100 mg·kg~(-1)+5 g·kg~(-1)) treatment group. Except for the normal group, mice in the other four groups were fed with high-fat and high-glucose diet for 8 weeks to establish the non-alcoholic fatty liver model. After 4 weeks of treatment, blood was collected from the eyeballs, the mice were sacrificed, and relevant indicators were detected. The results showed that compared with the model group, blood lipid and blood glucose levels of each treatment group were significantly lower(P<0.05); HE staining results showed that liver pathological damage in each treatment group was significantly improved; oil red O staining results showed fat distribution in each treatment group significantly reduced(P<0.01); immunohistochemical staining showed that glucose regulated the protein expression of protein 78(GRP78) in liver tissues of each treatment group was also significantly reduced(P<0.01); Western blot results showed that endoplasmic reticulum stress signal pathway-related factors GRP78, phosphorylated-protein kinase R-like ER kinase(p-PERK), eukaryotic translation-initiation factor 2α(eIF2α), activating transcription factor 4(ATF4), C/EBP homologous protein(Chop), inositol requiring 1α(IRE1α), and cleaved-cysteinyl aspartate specific proteinase 12(cleaved-caspase-12) were significantly reduced(P<0.01). The results of the combined drug treatment group were better than those of the single drug treatment group. These results showed that B. pilosa decoction had the effect in improving non-alcoholic fatty liver, and its mechanism may be related to the down-regulation of the expression of endoplasmic reticulum stress(ERS)-related factors, and the reduction of the apoptosis of hepatocytes caused by ERS and the down-regulation of blood lipid and blood glucose levels.

Background: The hospital Emergency department (ED) is one of the most important components of the health delivery system. Objectives: To investigate the public awareness of the ERs in KSA, what the public knows about the provided services, and if they know the difference between the outpatient clinic and ERs. Methods: It is a cross-sectional descriptive community-based study carried out on 977 male and female, young and adult participants from all age groups, in different areas of the Kingdom of Saudi Arabia, during the period from 1st January to 31st July 2019. Data was collected through filling the pre-designed online questionnaire which guided us to the needed data. We utilized the SPSS program version 16. The X2 test was used as a test of significance, and differences considered significant at p-value less than 0.05. Results: Most of the participants (87.5%) reported that they know the difference between the outpatient clinic and ER. The majority (68.1%) of subjects said that ERs is meaning rapid and unplanned medical care, 17.3% said any needed health care is available there, 12.2% said that it means insufficient medical care and only 2.5% said it means availability of physician at any time for any purpose. As regards evaluation to the provided services in ERs; 32.5% of cases said it was very good followed by 28.5% good, 19.8% excellent, 10.2% accepted and 10% reported it was bad services. There were significant relations between the awareness and age (p=0.03) and education level (p=0.003), but no relation was found with the gender of the participant (p>0.5). Conclusion: In our study, Most of the participants reported that they know the difference between the outpatient clinic and ERs. The majority of subjects said that ERs is meaning rapid and unplanned medical care and/or availability of physician at any time for any purpose. There were significant relations between the awareness and age and education level, but insignificant relation was found with the gender of the participant.

Exercise-Induced Bronchoconstriction (EIB) is a transient acute narrowing of the airway which occurs as a result of exercise. EIB is common in children. The prevalence of EIB has been estimated to be 20% in general individuals without a known diagnosis of asthma. Eighty percent of all asthma individuals experience exercise-related symptoms (ERS). The purpose of this study was to investigate the prevalence of EIB in medical students using an exercise challenge test. Aims And Objectives – To study the prevalence of prevalence of EIB in medical student and to investigate the relationship between EIB and the history of asthma, history of ERS, allergy including rhinitis, atopic dermatitis, hay fever, allergies to food and animals. Methods: This study was done on medical student of NMCH, Jamuhar between January 2019 and March 2019. Medical students underwent exercise challenge test on bicycle -ergometry. Spirometry was performed a few minutes before and immediately after performance of the exercise. The criterion for a positive test was a greater than 10% decrease in FEV1 from the baseline measurement. Results: Total 188 students completed the study, out of which 24 came to be EIB positive. 17 had history of asthma of which 15 came to be EIB positive. 13 had history of ERS of which 11 came to be EIB positive. Conclusion: In our study prevalence of EIB came to be 12.7%. EIB in student having history of asthma came to be 88.2%. EIB in student having history of ERS came to be 84.6%.

With the advances in technology and research methods, the metabolomics of rheumatoid arthritis has made some progress. This article reviews the application of metabolomics in early biomarker screen-ing and pathogenesis of rheumatoid arthritis, so as lo provide reference for the prevention, diagnosis and treatment of the disease.

Objective: To explore the effect of endoplasmic reticulum stress (ERS) on neutrophil elastase (NE) induced mucin 5AC (MUC5AC) production in human airway epithelial cells. Methods: HBE16 airway epithelial cells were cultured and pretreated with reactive oxygen species (ROS) inhibitor N-acetylcysteine (NAC) or ERS inhibitor 4-phenylbutyrate (4-PBA), or transfected with small interfering RNA (siRNA) against inositolrequiring kinase 1α (IRE-1α) or X-box binding protein 1 (XBP-1), respectively before incubation with NE. NE group and blank control group were also set up. ROS production was assayed by detection kit; expression of glucose-regulated protein 78 (GRP78), phosphorylated protein kinase R-like endoplasmic reticulum kinase (pPERK), activating transcription factor 6 (ATF6), phosphorylated IRE-1α (pIRE-1α), and XBP-1 protein was detected by Western blotting; spliced XBP-1 (XBP-1s) mRNA was measured by real-time PCR; levels of MUC5AC protein in culture supernatant and cytoplasm were assayed by ELISA and immunofluorescence. Results: There was an obvious increase of ROS production with strong elevation of GRP78, ATF6, pPERK, and pIRE-1α protein in NE group cells after 24 h, compared with blank control group (P<0.05). The protein and mRNA of XBP-1s, and MUC5AC production also increased obviously (P<0.05). NAC and 4-PBA reduced ERS-related protein expression and MUC5AC production and secretion (P<0.05). Further studies showed that MUC5AC secretion was also blunted by IRE-1α siRNA or XBP-1 siRNA, accompanied with decreased expression of XBP-1s mRNA and protein (P<0.05). Conclusion: NE induces ERS by producing ROS, and increases MUC5AC protein production and secretion; IRE-1α/XBP-1 play a certain role in this process.

The aim of this study was to investigate the preventive and therapeutic effects of Erzhi Pills on 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine( MPTP)-induced Parkinson's disease( PD) in mice,and explore its possible mechanism of action. Mice were intraperitoneally injected with MPTP( 30 mg·kg-1,0. 01 m L·g-1) once daily to induce PD for 8 days. In the treatment group,Erzhi Pills were given by intragastric administration( 2. 5 g·kg-1,once daily for 30 days). The normal group received an equal volume of normalsaline. In terms of behavior,the limb movement coordination ability of the mice was detected by climbing,hanging and swimming experiments. The spatial learning and memory ability of the mice was detected by Morris water maze test. The content of MDA,as well as the activity of GSH-PX and SOD were determined in mice serum. Western blot was used to detect the protein expression levels of TH,MAOB and apoptosis-related factors CHOP and caspase-12 in brain tissues. Immunohistochemistry was used to detect the expression of TH in section of brain tissues in mice. The results showed that in behavioral aspects,as compared with the model group,the scores of limb movement ability as well as scores of spatial learning and memory ability were significantly improved in the treatment groups( P<0. 05). In terms of serological indicators,as compared with the model group,the activities of SOD and GSH-PX were significantly increased in the serum of treatment groups,and the content of MDA was significantly decreased( P<0. 05). The results of Western blot showed that as compared with the model group,the protein levels of TH in the brain tissues of the mice in treatment group were significantly up-regulated,while the protein levels of MAOB and apoptosis-related factors CHOP and caspase-12 were significantly down-regulated( P<0. 05). The results of immunohistochemistry showed that the number of TH positive cells in the brain tissues of the mice in the treatment group was significantly increased as compared with the model group( P<0. 05). In summary,Erzhi Pills have a certain preventive and therapeutic effect on MPTP-induced PD mice,which can significantly improve the limb motor coordination ability and spatial learning and memory ability of PD mice. Its mechanism may be related to down-regulating the expression of apoptosis-related factors CHOP and caspase-12,reducing the dopaminergic neuron damage and inhibiting dopaminergic neuronal apoptosis.

OBJECTIVE: To observe the effect of electroacupuncture (EA) and EA combined with intracerebral injection of vascular endothelial growth factor (VEGF) on endoplasmic reticulum stress (ERS) related proteins and genes as activating transcription factor (ATF 6), inositol requiring enzyme-1 (IRE 1), CCAAT/enhancer binding protein homologous protein (CHOP), X box-binding protein-1 (XBP 1) of cerebral ischemia reperfusion injury (CIRI) rats, so as to study its repair effect for CIRI. METHODS: Forty male SD rats were equally and randomly divided into 5 groups: sham operation, model, EA, VEGF and EA+VEGF groups (n=8). The CIRI model was established by occlusion of the middle cerebral artery (MCAO) with thread embolism method. For rats of the sham operation group, the right common carotid artery was isolated without MCAO. EA (2 Hz/100 Hz, 1-3 mA) was applied to "Baihui" (GV 20), left "Quchi" (LI 11) and left "Zusanli" (ST 36) for 30 min, once a day for 14 days. For rats of the VEGF and EA+VEGF groups, 10 µL VEGF (0.025 µg/µL) was injected into the lateral ventricle 24 h after successful modeling. The rats' neurological function was assessed by using the modified neurological severity score (mNSS), and the histopathological changes of cerebral tissue were observed by Nissl staining method. The expression levels of ERS related proteins and genes ATF 6, IRE 1, XBP 1 and CHOP were determined by western blot (WB) and fluorescent quantitative PCR, separately. RESULTS: After modeling, the level of mNSS was significantly higher in the model group than in the sham operation group (P<0.05), and the number of Nissl bodies was markedly lower in the model group than in the sham operation group (P<0.05). Following the treatment, the mNSS was significantly lower in the EA, VEGF and EA+VEGF groups than in the model group (P<0.05), and the numbers of Nissl bodies were obviously higher in the EA, VEGF and EA+VEGF groups than in the model group (P<0.05), suggesting an improvement of neurological dysfunction and a repair of the injured cerebral tissue after the treatment. The levels of CIRI-induced increase of mNSS and CIRI-induced decrease of the number of Nissl bodies in the EA+VEGF group were respectively remarkably lower or higher than those of the simple EA and simple VEGF groups (P<0.05). WB and PCR showed that the expression levels of ATF 6, IRE 1, XBP1 and CHOP proteins and genes were notably higher in the model group than in the sham operation group (P<0.05), and considerably lower in the EA, VEGF and EA+VEGF groups than in the model group (P<0.05). Comparison among the three treatment groups showed that after the treatment, the expression levels of ATF 6, IRE 1, XBP1 and CHOP proteins and genes were obviously lower in the EA+VEGF group than in the EA and VEGF groups (P<0.05). CONCLUSION: EA and EA plus intracerebral microinjection of VEGF can improve neurological function and promote cerebral tissue repair in CIRI rats, which is associated with their effects in down-regulating the expression of ERS related proteins ATF 6, IRE 1, XBP1 and CHOP. The effect of EA+VEGF is superior to that of simple EA and simple VEGF.

Objective To study the effect of formononetin on the expression of ASK1 and JNK on the protein level in rat after unilateral ureteral obstruction (UUO). Methods Rats were then randomly divided into control group, model group, the enalapril group, and the high, medium, and low dose groups of formononetin (100, 50, and 25 mg/kg). Renal interstitial fibrosis (RIF) rats model was established by unilateral ureteral obstruction except the control group. The rats were sacrificed 14 d after surgery, and blood samples were collected to detect serum creatinine (Scr) and blood urea nitrogen (BUN) levels. HE staining was used to observe renal pathologic change and determine renal tubular damage index. The area percentage of RIF was detected by Masson staining. Expressions of ASK1 and JNK protein in kidney were determined by Western blotting. Tubular epithelial cell apoptosis was detected by TUNEL assay. Results Serum Scr, BUN level, tubulointerstitial injury index, RIF, the expressions of ASK1 and JNK protein, and apoptotic index were significantly decreased in the treatment groups when compared with the model group (P < 0.05, 0.01). The high dose group of formononetin was more effective than enalapril group. Conclusion Formononetin inhibited the expressions of ASK1 and JNK protein in UUO rats model. Ultimately renal tubular epithelial cell apoptosis was suppressed and the progression of obstructive nephropathy pathologic process was retarded.

Objective To assess the effect of resveratrol on the expression of endoplasmic reticulum stress molecular partners150-KD oxygen-regulated protein (ORP150) in renal tissues of rats with unilateral ureteral obstruction (UUO). Methods The UUO rat model of renal interstitial fibrosis was established. The rats were randomly divided into sham operation group, model group, enalapril group, the high-dose, medium-dose and low-dose groups of resveratrol, each group of 10. After 14 d of surgery, rats were sacrificed to collect serum and kidney tissue to detect the level of serum Scratinine (Scr) and blood urea nitrogen (BUN); Pathological changes were stained by HE to evaluate of renal tubular damage index, renal interstitial collagen deposition area was detected by Masson staining, apoptosis of in situ cell in renal tissue was determined by TUNEL assay, and using the western blot for the detection of protein expression of ORP150, GRP78, and GRP94 in renal tissue. Results After comparison of the treatment groups with model group, BUN and Scr levels in serum were significantly reduced in high-dose resveratrol group, the damage degree of renal tubules was significantly reduced, the relative area of the renal interstitial collagen decreased, In addition, the expression of ORP150 was increased (P < 0.05, 0.01), GRP78 and GRP94 proteins expression were significantly reduced, (P < 0.05, 0.01). Moreover, the resveratrol high-dose group seems more effective than enalapril groups in reversing the phenotype (P < 0.05). Conclusion Resveratrol was able to protect renal function, alleviate hydronephrosis, reduce interstitial injury of renal tubular, induce the expression of the key endoplasmic reticulum stress signal molecule ORP150 and the down-regulated molecular partner GRP78 and GRP94, delay the apoptosis of renal tubular epithelial cell of rats after UUO and inhibit renal interstitial fibrosis caused by apoptosis deposition.

During growth,invasion and metastasis,tumor cells undergo endoplasmic reticulum stress(ERS)under hypoxia,hy-poglycemia and other environmental stresses.In response to ERS,tumor cells induce unfolded protein response(UPR).PERK signa-ling pathway as a key pathway to activate UPR can promote survival,proliferation,invasion and protection of tumor cells by increasing tumor tolerance to adverse microenvironment,inducting angiogenesis,inducing autophagosome formation and activating apoptotic signal molecules.Tumor cells are induced apoptotic and autophagic death when UPR reaches a certain extent.

Objective To compare the effects of different local intervention temperatures of pressure ulcer on endoplasmic reticulum stress (ERS) and apoptosis in rats, and to provide an experimental evidence for clinical prevention and treatment of pressure ulcer. Methods The rat model of pressure ulcer was established by ischemia reperfusion, and a total of 40 SPF adult, male SD rats were divided into 4 groups: the sham group (anesthesia only, without other treatment), model group (ischemia at 22℃ for 1 h and reperfusion at 22℃ as one cycle, repeated for 5 cycles), high-temperature intervention group (ischemia at 22℃ for 1 h and reperfusion at 32℃ as one cycle, repeated for 5 cycles) and low-temperature intervention group (ischemia at 22℃ for 1 h and reperfusion at 12℃ as one cycle, repeated for 5 cycles). At the end of the experiment, muscle tissues at the sites under pressure of the rats were taken on ice. The pathological changes of skeletal muscle tissues were observed by HE staining. The expression levels of ERS-related proteins GRP78, caspase-12 and CHOP were detected by Western blot, and the expression of caspase-12 and CHOP was also observed by immunofluorescence. Moreover, apoptosis in the skeletal muscle cells was examined by TUNEL staining. Results Compared with the model group, skeletal muscle cell damages became more severe and apoptotic cells were increased in the high-temperature intervention group. Besides, the results of the immunofluorescence assay showed an increased positive expression of caspase-12 and CHOP, and the results of Western blot showed that the expression levels of GRP78, caspase-12 and CHOP were all higher than those of the model group (P< 0. 05). In contrast, skeletal muscle cell damages were alliviated and apoptotic cells were reduced in the low-temperature intervention group. Meanwhile, the positive expression of caspase-12 and CHOP was decreased, as shown by immunofluorescence, and all the expression levels of GRP78, caspase-12 and CHOP detected by Western blot were lower than the control group (P < 0. 05). Conclusions Local low-temperature intervention can alleviate the pressure ulcer damages in rats through inhibition of the ERS-mediated apoptotic pathway. Local high-temperature intervention may exacerbate the pressure ulcer damages in rats by activating the ERS-mediated apoptotic pathway and promoting cell apoptosis. Local low-temperature intervention may be promising in clinical prevention and treatment of pressure ulcer.

With the development of whole genome and transcrip-tome sequencing technologies, a growing body of long noncoding RNAs (lncRNAs) and circular RNAs (circRNAs) has been i-dentified and is receiving increasing attention. LncRNAs and circRNAs are non-protein encoding transcripts whose functions are crucial for advancing our comprehensive understanding of bi-ological processes in human health and diseases, specifically tumor. And there is a close relationship between circRNAs and lncRNAs in their origin and function. In this paper, we have re-viewed the recent advances of lncRNAs and circRNAs in tumor by focusing on their relationships, which may help to distinguish them and identify tumor biomarkers further.
