RESUMO

Objective To investigate the effects of δ-opioid receptor agonist DADLE (D-Ala2-D-Leu5-enkephalin) on the hemodynamics and oxygen metabolism in rats with sepsis. Methods Eighty healthy male SD rats were randomly divided into 4 groups ( n = 20 each) : sham operation group (group S), sepsis group (group SEP) ,DADLE, group and DADLE2, group. Sepsis was induced by cecum ligation and puncture (CLP) in SEP, DADLE,and DADLE2 groups. In DADLE1 and DADLE2 groups, 0.5 mg/ml DADLE 10 mg/kg was injected intraperitoneally (IP) 0.5 h before CLP and immediately after CLP respectively. Mean arterial pressure (MAP), left ventricular systolic pressure (LVSP) and ± dp/dtmax were recorded at 0, 2, 4 and 6 h after CLP (T1-4). Blood samples from left common carotid artery and right external jugular vein were collected at T4 for blood gas analysis. The cardiac index (CI), O2 delivery (DO2), O2 consumption (VO2) and O2 extraction rate (ERO2) were calculated.Results Compared with group S, MAP and LVSP were significantly increased at T2, while decreased at T3,4, and ± dp/dtmax was significantly increased in group SEP, MAP was significantly increased at T2, while decreased at T3,4, LVSP was significantly increased at T2,3, while decreased at T4 , and ± dp/dtmax was significantly increased in DADLE, and DADLE2 groups, and CI, DO2 and VO2 were significantly decreased and ERO2 was increased in SEP, DADLE, and DADLE2 groups (P<0.05). Compared with group SEP, MAP, LVSP and ± dp/dtmax at T3,4 and CI, DO2 and VO2 were significantly increased, while ERO2 was significantly decreased in DADLE1 and DADLE2 groups (P < 0.05). There was no significant difference in the parameters mentioned above between DADLE1 and DADLE2 groups ( P > 0.05). Conclusion δ-opioid receptor agonist DADLE can obviously improve the hemodynamics and oxygen metabolism in septic rats.

RESUMO

Intravenous injection of milk in dose of 0.6ml/kg into rabbits produced fever, putting an acupuncture needle into "Baihui (Hundred Flower) point" and gave intermittent electric stimulations. The thermal response peak (△T) of fever fell, the thermal response index (TRI) reduced and body temperature recovered fast from fever. The evidence showed that electric acupuncture can inhibit this type of fever. The contents of cAMP and L-enkephalin (LEK) in cerebrospinal fluid before fever, at the intervals of 2 and 3.5 hours in fever were determined by radioimmunoassay (RIA), and the resuIts indicated the central concentrations of cAMP in fever increased significantly, as well as the central concentrations of LEK increased at interval of 2 hours, but recovered at interval of 3.5 hours. While electric acupuncture inhibited fever of rabbits, it reduced central concentrations of cAMP and increased the central concentrations of LEK. The results suggested the role of electric acupuncture on the type of fever may associate with the changes of the concentrations of these central substances.

RESUMO

C_(57)BL/G male mice were immunized with SRBC 9 hr after 75 mGy whole body X-irradiation. The hypothalamic M-Enk and L-Enk contents were decreased. Serum TS level kept on fluctuating over the control level. The change of testicular cAMP content was not in the same direction as that of the TS. The change of serum CS level was opposite to that of serum TS, These results suggest that the low dose radiation probably cause the changes of neuroendocrine function, leading to the decrement of hypothalamic EOP contents, the supression of CRF-ACTH-CS axis activity and the enhancement of GnRH-LH-TS axis activity.

RESUMO

By means of radioimmunoassay, leucine-enkephalin (L-ENK) in the uteri of women and rats was studied and its responses to ovarian steroids were observed. The histological localization of L-ENK and the influence of naloxone (NAL) on estrogen regulation of the rat uterus in terms of cAMP and eGMP determination were also studied. Results of the study demonstrated that presence of L-ENK like substance in the uteri of women and rats, and the concentrations varied in the various phases of the sexual cycle. indicating a positive correlation with levels of estrogen in the body. L-ENK was found localized in the interstetial cells of the human endometrium, and in the rat uterus. Effect of extrogen on nucleotide was influenced by NAL. The findings suggested that L-ENK may play an important role on physiologic function of utetus.

In presnt experiment, animal models for hyperthyroidism and hypothyroidism were made, and using radioimmunoassay, the immunoeactive leucine-enkephalin (Ir-LEK) levels of such discrete brain areas as hypothalamus, striatum, cerebral cortex(frontal lobe) and whole pituitary were determined in different groups. The results obtained indicated that both hyperthyroidism and hypothyroidism induced significant changes of concentration in CNS Ir-LEK.
