RESUMO

Objective To observe the effects of Bushen Yizhi Decociton (BYD) on somatostatin-like (SS-like)neurons in hippocampal gyrus of rat models with Alzheimer's disease(AD). Methods Fifteen-month_old AD rat models were established by intraperitoneral injection of D-galactose for 4 weeks combined with ibotenic acid injection into bilateral nucleus basalis of Meynert. AD model rats were randomly allocated to AD model group(Group C), Hup-A treatment group(Group D) and BYD treatment groups (Group E and Group F,treated with high dosage and low dosage respectively), and 10 normal aged rats (Group B)and 10 normal youth rats (Group A)served as the normal control groups. The methods of immunohistochemistry and dig_labeling c-DNA probe in situ hibridization were used to detect the number of SS-like immunoreactive positive and SS mRNA expressed positive neurons in hippocampal CA1 and CA3 fields and the dentate gyrus. RESUMO

[Objective] To observe the influence of Xiaoyao Powder on the concentration of Ca2+ in hippocampal synaptosome of multi - stress model rats.[Methods] Wistar rats were randomly allocated to five groups: normal control (Group A) , model 1 (Group B), Xiaoyao Powder 1 (Group C), model 2 (Group D), and Xiaoyao Powder 2 (Group E) . All the rats except those of Group A were replicated to chronic multi-stress models. At the meantime, Groups C andE were given the decoction of Xiaoyao Powder, 1.06g each for 21 days and 42 days respectively. Groups A, B and D were administered by gavage the equal amount of normal saline. After the administration, the hippocampal homogenate was taken to detect the concentration of hippocampal synaptic Ca2+ by fluorimetry. RESUMO

After introcerebroventricular injection of kainic acid(KA) in the awake rat, three types of hippocampal unit responses, i. e- positive units, negative units and indifferent units, were recorded- Most positive units were hippocampal complex spike cells which correspond histologically to hippocampal pyramidal cells, and most hippocampal complex spike cells fired in positive bursts after KA treatment. In the early period after KA injection, the positive units were concentrated in CA3 area. It was suggested that:(1)the activities of positive units may be considered as the typical epileptiform hippocampal unit activity induced by KA, and the firing characteristics of the negative units may be the result of influence of hippocampal inhibitory interneurons or that of the excessive cellular depolarization;(2) the hippocampal pyramidal cells were more sensitive to the epileptogenic effect of KA than the hippocampal interneurons, and some pyramidal cells in CA3 area may serve as "pacemaker neurons" in KA induced epileptogenesis.
