RESUMO

OBJECTIVE@#To investigate the effect of Heliotropium indicum L. (H. indicum L.) on uterine involution and its underlying mechanisms in both in vivo and in vitro study.@*METHODS@#For in vivo studies, postpartum rats were randomly divided into 2 groups (n=24 for each): control group and treated group which were orally and daily administered with ethanolic extract of H. indicum L. (250 mg/kg body weight) until day 5 of postpartum. Uteri were collected for analysis of weight, cross-sectional area, collagen cross-sectional area, and collagen content on postpartum day 1, 3, and 5 (n=8 for each) from both groups. Blood samples were collected for hepatotoxicity and 17β-estradiol (E2) measurement. For in vitro studies, the extract effects on uterine contraction at half maximum effective concentration of 2.50 mg/mL were studied in organ bath system for at least 20 min.@*RESULTS@#Uterine parameters were significantly decreased after treated with extract of H. indicum L. (P<0.05). H. indicum L. extract significantly accelerated the reduction of those parameters and significantly decreased E2 (P<0.05). The extract facilitated uterine involution with no hepatotoxicity. H. indicum L. extract significantly stimulated uterine contraction (P<0.05) and synergized with oxytocin, prostaglandin and its precursor, linoleic acid. By investigating the different sequencing of the extract with the additional stimulants (added before or after), the two showed antagonistic effects, but still showed potentiated force when compared with control (without the stimulants).@*CONCLUSIONS@#The underlying mechanisms by which H. indicum L. facilitated uterine involution might be due to reducing E2 which induces collagenase activity, leading to decreases in uterine weight and size and stimulating uterine contraction. Our study provides new findings for future drug development for facilitating uterine involution with H. indicum L.

RESUMO

Aim: Heliotropium (Boraginaceae) is a widely spread genus of plants found in the Central and Northern parts in the Sudan and used for the treatment of inflammations, and rheumatism from ancient times. The aim of this work is to provide answers to impending research questions on compounds present in petroleum ether and n-hexane extracts of the aerial parts of H. sudanicum. Methodology: Phytochemical analysis of plant extracts was performed by using Gas Chromatography/Mass Spectrometer. Various constituents were identified after matching their mass fragmentation pattern with data available in Gas Chromatography/Mass Spectrometer library of National Institute of Standards Technology (NIST). Results: Petroleum ether extract of H. sudanicum was characterized by abundance of 1,6,10,14,18,22- Tetracosahexaen-3-ol, 2,6,10,15,19,23-hexamethyl-, (all-E)- (33.99%), Squalene (20.94%) , Phytol (7.62%) and Hexatriacontane (7.49%). The constituents of the extract were fatty acid derivatives (15.24%), hydrocarbons (8.41%) and terpenoids (28.56%). H. sudanicum n-hexane extract was characterized by abundance of 9,12-Octadecadienoic acid (Z,Z)-,methyl ester (15.14%), 9,12,15-Octadecatrienoic acid, methyl ester (14.67%), Cholesterol (11.12%), Fucosterol (10.30%), gamma.-Sitosterol (9.59%) and1,6,10,14,18,22- Tetracosahexaen-3-ol, 2,6,10,15,19,23-hexamethyl-, (all-E)- (7.73%). Conclusion: The resulted compounds in this study provide evidence for the utilization of the plant as alternative natural resource of medicinal and industrial interest. To best of our knowledge this is the first report that directly compares the two extraction solvents for active compounds from Heliotropium sudanicum aerial parts shows that petroleum ether should be the solvent of choice.

RESUMO

The flavonoid 3,5-dihydroxy-7-methoxyflavanone ((-)-alpinone) isolated from sticky resinous exudate of Heliotropium huascoense was evaluated as immunostimulatory in mammalian cells . Preliminary observations had showed that (-)-alpinone had increased the expression levels of pro-inflammatory cytokine transcripts in salmonid. Due to high morbidity and mortality that infectious diseases cause in humans, we evaluate the effect of (-)-alpinone as an immunostimulant in mammalian cells. Reactive oxygen species (ROS) are produced by macrophages activators for the destruction of pathogens; we evaluated (-)-alpinone effect in ROS generation and the proliferation of macrophages. The results showed that proliferation in Raw 264.7 cells treated with 10 and 25 µg/mL of (-)-alpinone had a significant increase in macrophage proliferation. In relation to ROS formation, cells treated with 1 and 5 µg/mL of (-)-alpinone, induce ROS formation in macrophages.

Antioxidants are the substances that neutralize free radicals or their actions. The traditional antioxidant phytochemicals from plants have been reported to inhibit the propagation of free radical reactions and to protect the human body from disease. The use of synthetic antioxidants has always been questioned because of their toxicity therefore researches are being carried out on plants nowadays to bring out the potent natural antioxidants. With this intent three ethnomedicinal plants of Assam viz., Calamus leptospadix Griff., Heliotropium indicum Linn., Lasia spinosa Lour. were explored with a view to evaluate their in-vitro antioxidative potentials. The methanolic aerial extracts were studied for phytochemical analysis and antioxidative properties by different in-vitro assays: DPPH radical scavenging method, Nitric oxide radical scavenging activity, Hydrogen peroxide radical scavenging activity, Total phenolic content and Total flavonoid content. The percentage of inhibition was calculated by comparing the absorbance values of the control and test samples. The study conducted revealed that methanolic extract of Calamus leptospadix (CL) exhibited the highest DPPH radical scavenging activity (IC50 value of 29 ± 3.24 μg/ml), Nitric oxide radical scavenging activity (233.33 ± 3.54 μg/ml), Total phenolic content (8.02 ± 0.32 μg/ml expressed as gallic acid equivalents) and Total flavonoid contents (61.71 ± 1.16 μg/ml expressed as quercetin equivalents). Methanolic extract of Lasia spinosa (LS) showed highest hydrogen peroxide radical scavenging activity (IC50 value of 221.89 ± 2.14 μg/ml). The results obtained in the present study indicate that aerial parts of Calamus leptospadix have potent, Lasia spinosa have moderate and Heliotropium indicum (HI) have mild antioxidant and free radical scavenging activity.

RESUMO

The intracellular bacteria Piscirickettsia salmonis is the most prevalent pathogen in the Chilean salmon industry, responsible for 50 percent of losses in recent years. So far, there are no effective treatments to control infections by this pathogen due to the emergence of antibiotics resistance. Therefore, it is extremely important to conduct research to find successful antibacterial therapies. In this paper, we evaluated the in vitro bactericidal activity of flavonoids and aromatic geranyl derivatives isolated from the resinous exudate of species Heliotropium filifolium, H. sinuatum y H. huascoense. The results showed that the compounds Filifolinone, Naringenine and 3-O- methylgalangine cause different percentage of mortality of bacteria and therefore they are good candidates to continue its evaluation in vitro and in vivo.

Present study was designed to screen phytochemical constituents and antihyperglycemic activity of Heliotropium indicum (HI) in Streptozotocin (STZ) induced diabetic rats. Heliotropium indicum (Boraginaceae) whole plant is used as traditional medicine for a number of ailments including diabetes. The whole plant was collected, shade dried and extracted with different solvents in the increasing order of polarity. When different solvent extracts of HI each at a dose of 500 mg/kg bw were given to diabetic rats, the methanol and aqueous extracts produced significant (P<0.0001) antidiabetic activity. Phytochemical screening of various solvent extracts of HI whole plant revealed the presence of alkaloids, steroids, triterpenes, saponins and tannins. When methanol active fraction of Heliotropium indicum (MAFHI) was checked for its antidiabetic activity, the fraction at dose of 750 mg/kg bw produced marked antihyperglycemic activity. The antihyperglycemic activity was also exhibited during oral glucose tolerance test (OGTT) with the same dosage of MAFHI.

Introduction: leishmaniasis is a serious public health problem worldwide. In Colombia, its cutaneous clinical form has the greatest incidence, and is mainly caused by species of Leishmania (Viannia) panamensis. Given the ineffectiveness of the available treatments, it is necessary to find new therapeutic agents in plant species used in folk medicine. Objective: evaluate the phytochemical profile and the hemolytic, cytotoxic and antileishmanial in vitro activity of extracts and fractions from leaves of Cordia dentata and Heliotropium indicum. Methods: the total ethanolic extract and fractions obtained from leaves of C. dentata and H. indicum underwent phytochemical profiling and determination of their hemolytic activity, toxic effect on Artemia salina larvae and on human promonocytic cell line U937 (CRL-1593.2™), as well as their antileishmanial activity against intracellular amastigotes of L. (V.) panamensis (MHOM/CO/87/UA140) transfected with Green Fluorescence Protein. Results: hemolytic activity was not observed in the extracts and fractions from either species under the study conditions. H. indicum fractions were more toxic than C. dentata fractions against A. salina larvae and U937 cells. Activity of the H. indicum dichloromethane fraction (Hi-I-5B) on intracellular amastigotes is closely related to the presence of alkaloids. Conclusions: the ethanolic extract from H. indicum leaves has antileishmanial properties, unlike the extract from the species C. dentata. This suggests that the former plant species is a rich potential source of molecules useful for the treatment of cutaneous leishmaniasis.

RESUMO

The composition of the essential oil (EO) obtained by hydro distillation from dry leaves of Heliotropium stenophyllum (Heliotropiaceae) was analyzed using gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS). The insecticidal activity of the oil against the house fly Musca domestica was evaluated and the dose necessary to kill 50 percent of flies (LC50) in 2 h was determined at 25 +/- 1 °C. The essential oil from Heliotropium stenophyllum showed potent insecticidal properties (LC50 = 1.09 mg/dm3) in comparison with other essential oils, in which at shorter times, the same bio-assay was used. According to GC and GC/MS analysis, junenol (19.08 percent); longiborneol (9.34 percent); (E, Z)-geranyl linalool (6.81 percent); selina-3,11-dien-6-alpha-ol (6.70 percent); alpha-cedrene epoxide (6.60 percent); heliofolen-12-al D (6.23 percent) and beta-epi-bisabolol (4.83 percent were the principal components of the EO. The Heliotropium stenophyllum essential oil, made up exclusively of sesquiterpenes, showed a composition very different from the EOs of the other species of Heliotropium, studied, and present a great potential as a natural insecticide against houseflies.

The infectious salmon anemia virus (ISAV) of Orthomyxoviridae family, is responsible for heavy losses in industry aquaculture around the world, affecting several commercial aquatic organisms, mainly Salmo salar. Therefore, it is important to find effective antiviral therapies. In this work we evaluated in vitro and in vivo the antiviral activity of three natural flavonoids isolated from the resinous exudates of the plant Heliotropium sinuatum (Heliotropiaceae) against ISAV. The results show that 7-O-methyleriodictyol was able to inhibit the infectivity of ISAV in vitro assay with EC 50 of 0.20 ug/mL. Despite having a citotoxicity expressed as CC50 of 12.80 ug/mL, the in vivo study showed that this compound protected 100 percent to the fish infected with ISAV keeping 100 percent fish viability. These results allow the proposal of 7-O-methyleriodictyol as a good candidate to be used as antiviral therapy for ISAV in salmon industry.

RESUMO

A new compound E was isolated from the methanolic extract of the leaves of Heliotropium indicum by chromatographic fractionation. In the present study, the effect of the compound E on reproduction of Helopeltis theivora has been evaluated. The acute toxicity study (LD50) and sub-acute toxicity studies (haematological, biochemical and histopathological parameters) in albino Swiss mice were carried out to evaluate the safety aspect of the compound E. The compound showed significant inhibitory effect on the reproductive life of H. theivora. The oviposition period, fecundity and hatching percentage of H. theivora were found to be 15.67days, 39.33 and 28.00 % respectively after treatment with 2% compound E, whereas the control value were found to be 20.33 days, 77.67 and 77.33% respectively. The LD50 of the compound was found to be 780 mg kg-1 in Swiss albino female mice. The compound did not show any toxicity in mice at sub-lethal dose treatment (78 mg kg-1 b. wt., once daily) for 21 days as evident from different haematological, biochemical and histopathological parameters in compound E treated group when compared with control.

RESUMO

Heliotropium indicum (Boraginaceae) is used in the traditional Ivory Coast pharmacopeia to treat asthma. In the present study, wound healing effect of n- butanol fractions was evaluated in H292- cells. Fractions which possessed better wound healing activity were fractionated on Sephadex LH20 column chromatography. Two compounds have been isolated which were responsible for this wound healing effect. Their structures were established as Pestalamide B (1) and Glycinamide,N-(1-oxooctadecyl)glycyl-L-alanylglycyl-L-histidyl (2) on the basis of spectral analysis. Both compound 1 and 2 presented wound healing effect compared with the control (P<0.05).

RESUMO

The in vitro effect of the resinous exudate of Heliotropium filifolium, of the 3 H-spiro[1-benzofuran-2,1 '-cyclohexane] derivative called filifolinol 1, isolated from the resin and the semi-synthetic compounds filifolinone 2 and filifolinoic acid 3, obtained from filifolinol 1, were evaluated on the proliferation of an immortalized cell line, UCHT1, derived from rat thyroid. We evaluated the effect of these compounds on UCHT1 cell growth parameters by calculating doubling time; and toxicity using the LIVE/DEAD™ in vitro test. The results showed that the resin is not active, while filifolinone 2, filifolinoic acid 3 and filifolinol 1 produced a significant inhibition of cell doubling time, in concentrations equal or greater than 50, 25 and 75 uM, respectively. The LIVE/DEAD test showed no significant toxicity at these concentrations, compared to cultures kept in absence of compounds. These results suggest a possible cytostatic effect of these compounds, and could therefore constitute potential alternatives for antineoplasic therapy.

The different solvent extracts (viz Petroleum ether, Ethyl acetate and Methanol) obtained from leaves and flowers of Heliotropium indicum and Spilanthes calva were screened for antifeedant activity against Helopeltis theivora. All the six different extracts showed antifeedant activity at four different concentrations. The methanolic extracts of leaves of Heliotropium indicum and Spilanthes calva exhibited significant activity at 4% concentration. The numbers of spots produced were only 18.67 and 22.67 respectively, which are significantly less than the number of spots produced in control (104.00 and 93.33 respectively). The treatment with methanolic extracts of flowers of both the plants significantly reduced the number of feeding spots to 22.33 and 23.67 respectively in comparison to the control values of 101.33. All the activities are dose dependent. The mean results with SEM (mean±SE) were statistically significant at 1% level (p<0.01) for three observations.

RESUMO

Heliotropium indicum of the family Boraginaceae is used locally in Nigeria to treat ailments such as ulcer and fever. In this study, ulceration of the gastric mucosa in Wistar rats was induced via the oral administration of 80mg/kg/bodyweight of Indomethacin. Histological analyses of the stomach body wall in the rats of Groups 2 and 4 (which received 100mg/kg/bodyweight of extract before oral administration of 80mg/kg/bodyweight Indomethacin and 80mg/kg/bodyweight Indomethacin only respectively) showed erosion of the mucus-secreting cells, gastric pit, upper and middle parts of gastric glands and some of the parietal cells. Histological observations of the stomach body wall in rats of Group 5 (which received 200mg/ kg/bodyweight of extract before oral administration of 80mg/kg/bodyweight of Indomethacin) showed erosion of the mucus-secreting cells, gastric pit and the upper most part of the gastric gland. Histological observations of the stomach body wall in rats of Groups 1, 6 and 3 (which received 50mg/kg/bodyweight of Ranitidine and 400mg/kg/bodyweight of extract before oral administration of 80mg/ kg/bodyweight Indomethacin; and only 80mg/kg/bodyweight of Normal Saline respectively) showed normal morphological appearance of the different components of the mucosa layer. Thus, the aqueous extracts of the dried leaves of Heliotropium indicum have dose dependent histo-gastroprotective effects.

The Boraginaceae are represented in Peru by 16 genera and 136 species (Brako & Zarucchi, 1993), mostly shrubs and herbs. Here we recognize 33 endemic species in nine genera, Heliotropium being the most important. Endemic Boraginaceae grow mainly in the Desert Shrubland, Subtropical Costal Desert and Mesoandean regions, between 300 and 4000 m elevation. We applied IUCN categories and criteria to nine endemic species. None of the species have been collected within Peru's protected area system.
