RESUMO

Objective:To study the effect of tannins from Pericarpium Granati on hepatic microsomal enzyme in rats. Methods:Thirty Wistar rats were randomly divided into five groups:the blank control group, high, medium and low dose groups of tannins from Pericarpium Granati, the positive control group of phenobarbital sodium. The blank control group was given physiological saline. The three different doses groups were respectively given tannins from Pericarpium Granati orally at the dose of 150,100 and 75 mg·kg-1 · d-1 for 7 days. The positive control group was given phenobarbital sodium 80 mg·kg-1 ·d-1 with intramuscular injection for 5 days. At the end of the experiment, the activity ofⅠandⅡphase metabolic enzymes in liver microsomes of each group was determined by UV. Results:Compared with the blank control group, the high, medium and low dose groups of tannins from Pericarpium Granati could sig-nificantly decrease the content of CYP 450 and CYPb5, and inhabit the activity of ADM (P<0. 01);the high and medium dose group could significantly inhibit ERD enzyme activity (P<0. 01);the high dose group could significantly reduce GST enzyme activity (P<0. 01). Conclusion:Tannins from Pericarpium Granati has notably inhibitory effect on hepatic microsomal enzyme in rats, which can reduce the expression of CYP3A and CYP2E1 in a dose-dependent manner.

abstract The aim of this study was to evaluate the effects of caffeine, tea polyphenol and daidzein on the pharmacokinetics of lansoprazole and its metabolites. Rats were intragastrically administered caffeine (30 mg·kg-1, once per day), tea polyphenol (400 mg·kg-1, once per day) or daidzein (13.5 mg·kg-1, once per day) for 14 days, followed by an intragastric administration of lansoprazole (8 mg·kg-1) on the 15th day. The plasma concentrations of lansoprazole and its two primary metabolites, 5-hydroxylansoprazole and lansoprazole sulfone, were determined by high-performance liquid chromatography coupled with tandem mass spectrometry (HPLC-MS/MS). Tea polyphenol significantly elevated the Area Under the Curve (AUC) of lansoprazole from 680.29 ± 285.99 to 949.76 ± 155.18 μg/L.h and reduced that of lansoprazole sulfone from 268.82 ± 82.37 to 177.72 ± 29.73 μg/L.h. Daidzein increased the AUC of lansoprazole from 680.29 ± 285.99 to 1130.44 ± 97.6 μg/L.h and decreased that of lansoprazole sulfone from 268.82 ± 82.37 to 116.23 ± 40.14 μg/L.h. The pharmacokinetics of 5-hydroxylansoprazole remained intact in the presence of tea polyphenol or daidzein. Caffeine did not affect the pharmacokinetics of lansoprazole and its metabolites. The results imply that tea polyphenol and daidzein may inhibit the in vivo metabolism of lansoprazole by suppressing CYP3A.


resumo O objetivo deste estudo foi avaliar os efeitos da cafeína, do polifenol do chá e da daidzeína na farmacocinética do lansoprazol e de seus metabólitos. Administraram-se, intragastricamente, aos ratos cafeína (30 mg·kg-1, uma vez ao dia), polifenol do chá(400 mg·kg-1, uma vez ao dia) ou daidzeína (13,5 mg·kg-1, uma vez ao dia), por 14 dias, seguindo-se a administração de lansoprazol (8 mg·kg-1) no 15º. dia. As concentrações plasmáticas do lansoprazol e de seus dois metabólitos primários, 5-hidroxilansoprazol e sulfona de lansoprazol, foram determinadas por cromatografia líquida de alta eficiência acoplada com espectrometria de massas (CLAE-EM/EM). O polifenol do chá elevou, significativamente, a Área Sob a Curva (ASC) do lansoprazol de 680,29 ± 285,99 para 949,76 ± 155,18 μg/L.h e reduziu a da sulfona de lansoprazol de 268,82 ± 82,37 para 177,72 ± 29,73 μg/L.h. A daidzeína aumentou a ASC do lansoprazol de 680,29 ± 285,99 para 1130,44 ± 97,6 μg/L.h e reduziu a da sulfona de lansoprazol de 268,82 ± 82,37 para 177,72 ± 29,73 μg/L.h. A farmacocinética do 5-hidroxilansoprazol permaneceu intacta na presença de polifenol do chá ou daidzeína. A cafeína não afetou a farmacocinética do lansoprazol e de seus metabólitos. Os resultados sugerem que o polifenol do chá e a daidzeína podem inibir o metabolismo in vivo do lansoprazol por supressão da CYP3A.

RESUMO

Objective To investigate the degradation kinetics of paeoniflorin-6-O’- benzenesulfonate ( CP-25 ) in vitro. Methods The homogenates of liver and intestine were prepared in vitro, and concentrations of CP-25 in ho-mogenates were detected by HPLC. Results CP-25 was obviously degradable in liver and intestine homogenates, and half life of degradation was decreased when levels of homogenates increased;the metabolisms of CP-25 in dif-ferent homogenates of intestine were diverse, the metabolic actions in duodenum and colon were weaker than those of jejunum and ileum. Conclusion Oral administration of CP-25 suffers first pass elimination from intestine and liver, which suggests the absorption of CP-25 could be further improved by appropriate pharmaceutical preparations.

RESUMO

The pharmacokinetic study of Rifapentine by daily oral administration for 10d and followed by twice weekly for another 3 weeks in 9 volunteers and in comparism with Rifandin were reported. The results showed that the absorption and excretion of Rifapentine were markedly slower than that of Rifandin, its T1/2 was 15h. The Peak plasma level and T1/2 decreased after repeated administration of the drug. This suggested that Rifapentine and Rifandin in similar to Ri-fampin were hepatic microsomal enzyme inducers.

Sixty Wistar rats were divided into 5 groups, 12 in each:1)Casein; 2) Fish powder (FP) ; 3) Fibrin; 4)Liver residue(LR) , 5)Nitrogen free(NF). The feeds for all groups of rats were at the level of 10% protein and at the same calories except NF. Growth test, nitrogen balance and recovery test were used and the levels of serum proteins and Hb were determined.The protein efficiency ratio (PER) of casein, FP, fibrin and LR were 1.90, 3.07, 2.61 and 1.28 respectively. Net protein ratio (NPR), apparent digestibility, true digestibility and net nitrogen ratio (NNR) were same as PER suggesting the protein quality of LR was low, FP was high and fibrin in the medium. The values of serum proteins also showed that 10% protein of LR could not maintain rats in health, but compared with casein and FP the nutritional quality of fibrin was high and could be used in feed industry or other products.The effects of proteins on hepatic microsomal enzyme were observed. The influence factors were discussed.
