RESUMO

Objective To study the cell proliferative effects of fungal immunomodulatory proteins from Ganoderma spp . on 26 gastric cancer cell lines in vitro .Method 26 human gastric cancer cell lines were treated with FIPs by MTS assay .The average optical density (OD) in 490 nm and inhibition rate (GI50 )was counted by Universal Microplate Spectrophotometer . Results Three FIPs showed similar profiling in 26 human gastric cancer cell lines after 72 h treatment in cell proliferation as-say ,which except for NUGC-4 and OCUM-1 did not showed obvious anti-proliferative effect ,the other 24 human gastric cell lines showed some anti-proliferative effects ,especially for 7 cell lines(NUGC-3 ,GTL-16 ,HGC-27 ,IM95m ,SNU-638 ,SNU-216 and SNU-5) showing strong potency ,with their GI50 less than 50 μg/ml .Conclusion FIPs showed strong anti-prolifera-tive effects in some human gastric cancer cell lines in vitro ,which had potential to be further developed as anti-gastric cancer drugs .

Uno de los fenómenos que abre más interrogantes en la Inmunología es ¿Por qué el embrión, comportándose como un injerto semialogénico, no es rechazado por la madre? Se sabe que la madre produce una activa respuesta inmunológica frente al feto y sin embargo, en condiciones normales, el rechazo inmunológico no se produce. En el presente trabajo de revisión, se describen algunos mecanismos por medio de los cuales se genera la tolerancia inmunológica específica de la madre hacia el embrión. Todos estos mecanismos son interdependientes, y en conjunto constituyen una trama para evitar el rechazo fetal. Aquí se detalla la acción de las hormonas sexuales femeninas sobre el sistema inmunológico, el cambio del perfil de citoquinas, la generación de proteínas inmunomoduladoras y de anticuerpos bloqueantes, el efecto de la expresión de los HLA-G y el papel de algunas células inmunocompetentes como los linfocitos T reguladores, las células dendríticas y las células asesinas naturales o Natural Killer. Asimismo se detallan otras vías por las cuales el embrión se defiende del ataque inmunológico materno como la inducción de la apoptosis en el endometrio y en los leucocitos, el metabolismo de hierro y triptofano, la inhibición del sistema del complemento y la expresión de anexinas.


One of the phenomena that offers more questions in Immunology is: Why the embryo, behaving like a semialogenic graft, is not rejected by the mother? It is known that the mother produces an active immunological response against the fetus and, nevertheless, in normal conditions, the immunological rejection does not take place. In the present work of revision, some mechanisms are described by means of which the specific immunological tolerance of the mother is generated towards the embryo. All these mechanisms are interdependent and altogether they constitute a safety network to avoid the fetal rejection. Here the effects of female sex hormones on the immunological system, the change of the profile of cytokines, the generation of immunomodulating proteins and blocking antibodies, the effect of the expression of the HLA-G and the paper of some cells, like Regulatory T lymphocytes, dendritic and Natural Killer cells, are detailed. Also other routes by which the embryo defends itself of the maternal immunological attack are described, like the induction of apoptosis in the endometrium and leukocytes, the tryptophan and iron metabolisms, the inhibition of the complement system and the expression of annexins.

RESUMO

The protein profile of the outer membrane of Leptospira interrogans serovar Hardjo subtype hardjoprajitno associated with the bovine natural immune response was investigated. The outer membrane proteins were extracted utilizing Triton X114 and precipitated with acetone. The protein sample was then resolved by SDS-PAGE and reacted in western blot against sera from a hyperimmune rabbit and from naturally infected bovines. In silver stained gels, 14 protein bands were observed, among which four proteins, with 22, 29, 47 and 63kDa, appeared as major constituents. Western blot tests with hyperimmune rabbit antiserum detected bands corresponding to proteins with 35; 27; 24; 21; 17 and 14kDa, while 32kDa and 45kDa proteins were the most immunoreactive with sera from naturally infected bovines.


Estudou-se o perfil proteico da membrana externa da Leptospira interrogans sorovariedade Hardjo, amostra hardjoprajitno, associado à resposta imune natural de bovinos infectados. Foram utilizados Triton X114 para a extração das proteínas de membrana externa e acetona para precipitá-las. As proteínas extraídas foram analisadas por SDS-PAGE e western blot contra soro de coelhos hiperimunes e de bovinos naturalmente infectados. Em géis corados com nitrato de prata, 14 bandas proteicas foram identificadas, e quatro dessas bandas, com 22, 29, 47 e 63kDa, foram as mais proeminentes. Os western blots com soro hiperimune de coelho detectaram bandas correspondentes a proteínas com pesos moleculares de 35, 27, 24, 21, 17 e 14kDa, e bandas de 32 e 45kDa destacaram-se nos testes com soros de bovinos naturalmente infectados.
