@#AIM: To study serum soluble intercellular adhesion molecules -1(sICAM-1), soluble vascular cell adhesion molecule -1(sVCAM-1)and expression of miRNA-146a in peripheral blood mononuclear cells(PBMC)and its significance in patients with thyroid associated ophthalmopathy(TAO). <p>METHODS: From June 2014 to December 2015 in our hospital, 37 patients with TAO(TAO group), 40 patients with hyperthyroidism without TAO(non eye disease group)and 30 healthy people(control group)were enrolled and the serum concentrations of sICAM-1, sVCAM-1 and expression of miRNA-146a in PBMC were detected. <p>RESULTS: The serum sICAM-1 and sVCAM-1 of TAO group were 366.14±67.28g/L, 211.07±27.45g/L level was significantly higher than those of non eye disease group(286.62±51.09μg/L, 179.83±25.09μg/L)and healthy group(234.51±38.969μg/L, 164.51±22.57μg/L)(<i>P</i><0.05). In TAO group, miRNA-146a(0.071±0.016)in PBMC was lower than that in non eye disease group(0.381±0.084)and healthy group(1.105±0.216)(<i>P</i><0.05). The serum levels of sICAM-1 and sVCAM-1 were significantly higher in non eye disease group than in healthy group(<i>P</i><0.05), and the expression of RNA-146a in PBMC was lower in the non eye disease group than in the healthy group(<i>P</i><0.05). The serum levels of sICAM-1 and sVCAM-1 in the mild TAO group were significantly lower than those in the moderate-severe groups and extremely severe group(<i>P</i><0.05), and the miRNA-146a expression in the mild TAO group was higher than those in the moderate-severe group and extremely severe group(<i>P</i><0.05). The serum levels of sICAM-1 and sVCAM-1 were significantly lower in the moderate-severe group than those in the extremely severe group(<i>P</i><0.05), and the expression of miRNA-146a in the moderate -severe group was significantly higher than that in the extremely severe group(<i>P</i><0.05). <p>CONCLUSION: Serum sICAM-1, sVCAM-1 in TAO patients is with high expression, miRNA-146a in PBMC in TAO patients with low expression, and related to the degree of patient's condition.

Objective To investigate the clinical significance of soluble intercellular adhesion molecule‐1 and inflammatory factors (CRP ,IL‐17) in elderly patients with colorectal carcinoma .Methods The expression of sICAM‐1 ,CRP and IL‐17 in 76 ca‐ses of elderly patients and 32 cases of youg patients with colorectal cancer were detected by enzyme linked immunosorbent assay be‐fore and after surgery ,and to analyze its clinical significance correlated with pathological parameters .Meanwhile ,60 cases of healthy were controls .Results The serum levels of IL‐17 and sICAM‐1 were higher in patients with different ages of colorectal cancer than those of the normal control group (P0 .05) .The level of CRP before operation in the young group was higher than that in the normal control group (P0 .05) .The serum levels of sICAM‐1 and IL‐17 in colorectal cancer patients were associated with the degree of differentiation ,depth of invasion ,lymph node metastasis and TNM staging (P<0 .05) .The level of CRP was significantly correlated with the depth of invasion ,lymph node metastasis and TNM staging (P<0 .05) .Conclusion The serum levels of sICAM‐1 ,CRP and IL‐17 reflect the invasion and metastasis of colorectal cancer in a certain extent ,which play important roles in predicting the development and prognosis of colorectal cancer .

RESUMO

Objective To investigate the protective effects of thalidomide on rats with acute necrotizing pancreatitis (ANP) and its mechanism. Methods Fifty four SD rats were randomly divided into the three groups; ANP group, thalidomide group and control group with 18 rats in each group. The model of ANP was induced by retrograde injection of 5% sodium taurocholate into the bili-pancreatic duct. Rats in thalidomide group received thalidomide 200 mg/kg body weight gastric lavage 1 h after ANP induction. The rats were sacrificed 3 h, 6 h, and 12 h after ANP induction, and the amount of intraperitoneal ascites was quantified. The serum levels of TNF-α, IL-6, IL-18 were measured by EUSA. The proportion of CD4 + T cell, CD8 + T cell in peripheral blood was determined by flow cytometry. The expression of TNF-α mRNA in pancreatic tissue were measured by RT-PCR. The expression of ICAM-1 protein in pancreatic tissue was measured by immunohistochemistry. Pancreatic tissue underwent pathologic examination. Results Six hours after surgery, the amount of ascite, serum levels of TNF-α, IL-6, IL-18, CD4 + T cell, CD,+ T cell, pancreatic TNF-α mRNA and ICAM-1 protein expression, pathologic score in control group was (1.03 ±0.31)ml,(57.17±11.29)pg/ml, (24.45 ±4.14)pg/ml, (64.23 ±21.85)pg/ml, (47.58 ±9.21)% , (40.88 ± 2.96)%, 0.07 ±0.02, 0.57 ±0.30, 0.67 ±0.81, respectively, and the corresponding values were (3.63 ± 0.38)ml, (107.54 ±33.05) pg/ml, (47.30 ± 11.40) pg/ml, (367.76 ± 108.43 ) pg/ml, (54.90 ± 7.15)%, (17.17 ±3.12)%, 0.65 ±0.26, 3.20 ±0.57, 11.50 ±1.87 in ANP group; and (1.45 ±0.53)ml, (80.60 ±20.48) pg/ml, (26.61 ±10.85) pg/ml, (321.82 ±85.20) pg/ml, (29.80 ±2.19)% , (15.52± 1.96)%, 0.35 ±0.23, 2.37 ±0.67, 8.00 ±3.03. Besides the value of CD8 + T cell was significantly decreased, all other values were significantly increased when compared with control group (P <0.05). Conclusions Thalidomide can decrease the release of inflammatory mediator, and reduce the pathological damage of pancreas of ANP rats by inhibiting TNF-α mRNA expression.

OBJECTIVE: To determine the utility of intercellular adhesion molecule (ICAM)-1 antibody-conjugated gadolinium diethylenetriaminepentaacetic acid (Gd-DTPA-anti-ICAM-1) as a targeted contrast agent for the molecular magnetic resonance imaging (MRI) in collagen-induced arthritis (CIA). MATERIALS AND METHODS: Three groups of mice were used: non-arthritic normal, CIA mice in both the early inflammatory and chronic destructive phases. The MR images of knee joints were obtained before and after injection of Gd-DTPA-anti-ICAM-1, Gd-DTPA, and Gd-DTPA-Immunoglobulin G (Ig G) and were analyzed quantitatively. The patterns of enhancement on the MR images were compared with the histological and immunohistochemical ICAM-1 staining. RESULTS: The images obtained after injection of Gd-DTPA-anti-ICAM-1 displayed gradually increasing signal enhancement from the moment following injection (mean +/- standard deviation [SD]: 424.3 +/- 35.2, n = 3) to 24 hours (532 +/- 11.3), rather than on pre-enhanced images (293 +/- 37.6) in the early inflammatory phase of CIA mice. However, signal enhancement by Gd-DTPA and Gd-DTPA-IgG disappeared after 80 minutes and 24 hours, respectively. In addition, no significant enhancement was seen in the chronic destructive phase of CIA mice, even though they also showed inflammatory changes on T2-weighted MR images. ICAM-1 expression was demonstrated in the endothelium and proliferating synovium of the early inflammatory phase of CIA mice, but not in the chronic destructive phase. CONCLUSION: Molecular MRI with Gd-DTPA-anti-ICAM-1 displays specific images targeted to ICAM-1 that is expressed in the inflamed synovium of CIA. This novel tool may be useful for the early diagnosis and differentiation of the various stages of rheumatoid arthritis.

RESUMO

Intercellular adhesion molecules 1(ICAM-1) and vascular cell adhesion molecule 1(VCAM-1) play important roles in the inflammatory process of cerebral ischemic injury.The expression of ICAM-1 and VCAM-1 increases following cerebral ischemia;ICAM-1 and VCAM-1 promote ischemic inflammation through mediating leukocytes adhesion to the endothelial cells and eventually migration into brain tissue;the inhibition of the overexpression and effect of ICAM-1 and VCAM-1 can reduce cerebral ischemic injury.

Objective:To explore the levels of adhesion molecules in peripheral blood leucocyte from patients with multiple sclerosis(MS) and after intravenous methylprednisolone(MP) treatment.Methods:The positive percentag of intercellular adhesion molecule 1(CD54)?integrin LFA 1 ? subunit(CD18)?very late antigen 4 ??? subunit(CD49d?CD29) and L selectin(CD62L) were determined on lymphocytes and monocytes by flow cytometry from 28 patients with relapsing remitting MS (relapses n=20,remission n=8) and 20 controls.Results:The positive percentage of CD49d?CD29 on lymphocytes and monocytes,CD54 and CD62L on monocytes in relapses MS group were higher than in remission MS group and controls(P0.05).Conclusion:The adhesion molecules expression on peripheral blood leucocyte were elevated in MS patients and it may be suitable to be as a marker of MS activity.
