RESUMO

@#AIM: To evaluate the expression level of transforming growth factor β2(TGF-β2)in the aqueous humor and trabecular tissue of patients with primary angle-closure glaucoma(PACG)of Han and Kazakh ethnic groups and the formation of filter bleb after trabeculectomy.<p>METHODS: Prospective research. Between July 2018 to April 2019, 46 PAGG patients(49 eyes)underwent trabeculectomy in our hospital, including 25 Han nationality(26 eyes)and 21 Kazak nationality(23 eyes). Aqueous humor and trabecular tissue were obtained through trabeculectomy. ELISA method was used to detect the content of total aqueous TGF-β2(tTGF-β2)and activated TGF-β2(aTGF-β2). And the expression of TGF-β2 in trabecular tissue was detected by immunohistochemistry and immunofluorescence.<p>RESULTS:Immunohistochemical staining and immunofluorescence staining showed that the expression of TGF-β2 in the trabecular meshwork of Han patients was significantly higher than that of Kazakh patients. ELISA quantitative analysis showed that the aTGF-β2 content of Han and Kazak patients were 172.015±79.367pg/mL and 83.436±41.743pg/mL, respectively, the difference was statistically significant(<i>t</i>=4.794, <i>P</i><0.001). In patients ≥70 years old, the content of tTGF-β2 in the aqueous humor of patients of the two nationalities was 480.124±152.997 and 338.858±72.497pg/mL, respectively, the difference was statistically significant(<i>t</i>=2.421, <i>P</i>=0.026). In the comparison between preoperative and postoperative, there were time differences and interaction effects in intraocular pressure between Han and Kazak patients. At 6mo postoperatively, the formation of type Ⅰ and Ⅱ filter blebs in Han and Kazak patients was different, and the difference was statistically significant(50% <i>vs</i> 78%; χ2=4.841, <i>P</i>=0.028).<p>CONCLUSION: The expression of TGF-β2 in the aqueous humor and trabecular meshwork of patients with PACG in the two ethnic groups is different. The expression of TGF-β2 in the aqueous humor and trabecular meshwork of Kazakh patients is significantly lower, which reduces the promotion of postoperative filtering scar Functional filtration filtration bleb.

RESUMO

Objective To compare metabolic syndrome(MS)with Framingham risk score as predictors of cardiovascular disease( CVD) among Kazakhs. Methods The participants were the residents who had been followed up for more than 5 years in representative areas of Kazakhs in Xinjiang. We assigned MS a continuous risk score for predicting the development of CVD based on the weights of MS components. MS and Framingham risk score were compared in terms of their ability in predicting years in representative areas of Kazakhs in Xinjiang. We assigned MS a continuous risk score for predicting the development of CVD based on the weights of MS components. MS and Framingham risk score were compared in terms of their ability in predicting development of CVD using Cox regression and receiver operating characteristic curve. Results The incidence of CVD was 13.87%. The incidence of CVD was higher in the MS group than it in the non-MS group(21.59% vs 11.10%, P<0.001). The area under the receiver operating characteristic(ROC)curve of MS risk score was significantly larger than that of MS classification(0.727 vs 0.585, P<0.001);the area under the curve of MS risk score was close to that of Framingham risk score ( 0. 732 vs 0.727, P=0.673). The association between CVD and each quintile of MS risk score was more significant than that between Framingham risk score and CVD under the same exposed condition(4.61、9.33、14.15、22.29对3.69、6.36、8.47、16.99) . Conclusion MS risk score that included age may be a better predictor of CVD among Kazakhs.

RESUMO

Objective To investigate the correlation between the plasm homocysteine (Hcy) levels and cystathionine β synthase (CBS) T833C gene polymorphism in Xinjiang Kazakh with essential hypertension. Methods 239 Kazak patients with hypertension (hypertension group) and 206 with normotensive (control group) were selected for the study. Amplification Refractory Mutation System (ARMS) was used to analyze the polymorphism of CBS gene T833C, TT, TC and CC genotypes and the various sites of T, C allele frequencies in research group and the control group. Results Plasma of Hcy level was higher in the hypertensive group than those of control group, the difference was significant (P<0.05), and the individual plasma of Hcy with TC gene was higher than that with TT gene. The C allele frequencies was significantly higher in EH group than that in controls in Xinjiang Kazakh population, and the difference was significant (P<0.05). The risk of EH group in individuals carrying TC genetype was 2.39 times higher than in individuals carrying TT genetype (OR = 2.39, 95%CI:1.125 ~5.076, P = 0.02). Conclusion Elevated Hcy level may be a risk factor of Kazakhs hypertension in Xinjiang. The Cystathionine β synthase gene of T833C polymorphism may be associated with essential hypertension in Kazak people in Xinjiang.

RESUMO

Objective To investigate whether G1155942T polymorphism in WNK4 gene is associated with essential hypertension in a population with Kazakhs ethnicity,in Xinjiang. Methods This study covered 563 hypertension patients and 346 normotensive controls. The variant of G1155942T was determined by the TaqMan probe real-time PCR method. Some biochemical indices such as glucose (GLU),triglyeeride (TG) and total cholesterol (TC) were also measured. All of these results were under logistic regression analysis.Addictive model was applied to assess the interactive effects between WNK4 gene G1155942T mutation and environmental factors on hypertension. Results The G1155942T polymorphism was consistent with Hardy-Weinberg expectations in both case and control groups.Genotype and allele frequencies of G1155942T were observed (P=0.004,P=0.003).Data through logistic regression analysis showed that factors as age,BMI,total cholesterol as well as the GT + TT genotype frequencies of Exon 8 G1155942T polymorphism in WNK4 were responsible for the increased risks for hypertension.Positive interactions between G1155942T mutation and gender,BMI,GLU,the OR were 3.75 (95% CI:1.19-11.80),5.77 (95% CI:1.93-17.21 ) and 8.67 (95% CI:1.03-72.99),respectively.Conclusion Our result suggested that the Exon 8 G1155942T polymorphism in WNK4 gene was associated with hypertension in the studied Kazakhs ethnic group in Xinjiang and the T allele might be the risk factor for essential hypertension. There were interactive effects between WNK4 gene G1155942T mutation, gender,BMI,and GLU.
