RESUMO

Background: Four empirical studies have measured the impact of head-loading on female African porters posture using expensive radiography and manual kin anthropometry and goniometry. The reliability of cheaper, pragmatic smartphone goniometric technology as an alternate clinical tool to assess posture is needed. Objectives: This study was designed to test the inter-rater reliability of smartphone goniometry technology against manual goniometry in measuring selected sagittal postural angles in South African female youth who habitually head-load. Methods: Female South African youth who habitually headload voluntarily participated in the study (N = 100) and were randomly allocated into experimental (n = 50) and control (n = 50) groups. An observational randomized control design involving a pre-test post-test crossover was used, after which the control group crossed over into the experimental group and vice versa. The control group stood in the unloaded phase without a head load, while the experimental group carried the head load. The daily head loads and body mass were measured on an electronic scale. Demographic characteristics (age, body mass, and stature) were recorded and selected biomechanical angles were measured on the right side. Results: The mean age of the study participants was 12.3 ± 2.5 years; average body mass was 44.5 ± 13.7 kg. The average head load habitually carried was 8.0 ± 2.5 kg. The inter-rater reliability between the smartphone goniometry technology and manual goniometry was 0.9. Conclusion: The findings support the use of smartphone goniometry as a pragmatic method for assessing sagittal plane postural changes among rural South African youth who habitually head-load. Further studies are needed to validate these findings.

RESUMO

Objective:To report 3 cases of rare subtypes of hereditary epidermolysis bullosa. Methods:Clinical data were collected from the probands and their relatives, whole-exome sequencing was performed to screen disease-causing mutations in the probands, and Sanger sequencing or qPCR was conducted to verify the mutations in patients and their relatives. Results:Case 1 mainly presented with linear red scars on the back, and the proband, her mother with similar clinical manifestations and her asymptomatic daughter all carried a mutation c.4573G>A (p.Gly1525Arg) in the COL7A1 gene. Case 2 presented with generalized reticular pigmentation all over the body and occasional blisters restricted to the hand and foot, and carried a de novo mutation c.74C>T (p.Pro25Leu) in the KRT5 gene. Case 3 presented with pigmentation abnormalities mainly located at the sun-exposed sites and incomplete syndactyly of the left hand, and carried homozygous deletion mutations in exons 2-6 of the FERMT1 gene, which were inherited from her asymptomatic parents. Case 1 was diagnosed with dominant dystrophic epidermolysis bullosa pruriginosa, case 2 was diagnosed with epidermolysis bullosa simplex with mottled pigmentation, and case 3 was diagnosed with Kindler epidermolysis bullosa. Conclusion:The clinical manifestations of epidermolysis bullosa vary greatly, and gene detection is very important for confirmation of diagnosis of its rare types.

RESUMO

It has been proposed that age reprogramming enables old cells to be rejuvenated without passage through an embryonicstage (Singh and Zacouto in J. Biosci. 35 315–319, 2010). As such, age reprogramming stands apart from the inducedpluripotent stem (iPS) and nuclear transfer-embryonic stem (NT-ES) cell therapies where histo-compatible cells are produced only after passage through an embryonic stage. It avoids many of the disadvantages associated with iPS and NT-EScell therapies. Experimental evidence in support of age reprogramming is burgeoning. Here, we discuss possible newapproaches to enhance age reprogramming, which will have considerable benefits for regenerative therapies.

RESUMO

I have stories to tell. More than a decade ago and ironically almost a decade after my graduation from medical school, I trained and got certified in basic life support (BLS) skills because I did NOT want to fail my family, my friends and my patients as I had desired to NOT fail my conscience as a physician. I will tell you this that you will never regret honing BLS skills whenever you will see the smiles on the faces of revived and you will never forget when not forgiven – by else and/or by self – for your reluctance in knowing what’s expected from you especially as a physician. Even though my natural desire to hone BLS skills was born in the times when it was NOT mandated, my free will to care for my family, my friends and my patients had always brought me back to BLS training centers for revisiting my skills again and again, years after years. Essentially, I have recognized Brain-Heart-Lungs as the Trinity of Life and hereafter I plead to all my peers irrespective of which stage of career they are in: “Please hone your BLS skills, at least be a Heartsaver®, for your kin so that when your kin, your beloved, needs your help, you don’t delay providing help compared to leaving “To go find one”

RESUMO

The kinin-kallikrein system (KKS) is a system of endogenous multiprotein cascades. Its activation leads to activate the intrinsic coagulation cascade pathway and promote the enzymatic hydrolysis of kininogens with the consequent release of bradykinin and pancreatic peptides. KKS is involved in various pathophysiological processes such as inflammation, angiogenesis, cardioprotection, vascular permeability, blood pressure control, coagulation and pain. And KKS is closely related to the etiologic and pathologic progression of liver cancer, prostatic carcinoma and ovarian cancer. This review focuses on the role of KKS in the occurrence and development of hepatocellular carcinoma and the progress in the intervention of KKS in the treatment of hepatocellular carcinoma.

RESUMO

@#Objective To compare the effect of impulse electroacupuncture (impulse-EA) and music electroacupuncture (music-EA) for Tongdu Qishen on spatial learning and memory, and expression of cytokine in the hippocampus of APP/PS1 mice. Methods A total of 32 APP/PS1 double transgenic male mice were randomly divided into model group (n=8), drug group (n=8), impulse-EA group (n=8) and music-EA group (n=8), the same background and age male C57BL/6 mice were observed as normal group (n=8). The impulse-EA group and music-EA group accepted EA at Baihui (GV20) and Yintang (GV29), connected with their own electroacupuncture stimulators, for 20 minuts, then, they were pricked Renzhong (GV26) for a while. The drug group accepted donepezil hydrochloride 0.92 mg/kg intra-gastrically. The normal group, model group and drug group were grabbed and bounded in the same way. After 15 days of treatment, they were assessed with Morris water maze. The expression of high mobility group box pro-tein-1 (HMGB1) and interleukin-10 (IL-10) in hippocampus were measured with immunohistochemistry and Western blotting. Results The escape latency shortened inEA groups compared with that of the model group since the fourth day of Morris water maze training (P<0.05), and it was the least in the music-EA group; and the ratio of swimming across the target quadrant increased (P<0.05), but there was no significant difference between EA groups (P>0.05). The ex-pression of HMGB1 decreased (P<0.05) and the expression of IL-10 increased (P<0.05) in EA groups com-pared with that of the model group, and HMGB1 decreased more and IL-10 increased more in the music-EA group than in the impulse-EA group (P<0.05), according to the measurement in immunohistochemistry, not in Western blotting. Conclusion Both impulse-EA and music-EA based on Tongdu Qishen can promote the recovery of the learning and memory in APP/PS1 mice, and music-EA may do more effectively in inhibition of pro-inflammatory factors and promotion of the anti-inflammatory factors.

RESUMO

AIM:To investigate the roles of Notch signaling in lipopolysaccharide(LPS)-induced proliferation and secretion of interleukin-6(IL-6)and chemokine CXCL1 in bone marrow mesenchymal stem cells(BMSCs). METHODS:BMSCs were isolated by whole bone marrow culture. The expression levels of Notch signaling pathway recep-tors and ligands in the BMSCs treated with LPS were measured by qPCR and Western blot. The proliferation of BMSCs was analyzed by MTT assay and viable cell counting. The secretion levels of IL-6 and CXCL1 induced by LPS were measured by ELISA.RESULTS:Treatment with LPS at 1 mg/L effectively induced the proliferation of BMSCs and the secretion of IL-6.Obvious expression of Notch receptors and ligands in the BMSCs was observed, and LPS had little effect on the mRNA and protein levels of Notch receptors and ligands, but LPS increased the protein levels of Hes1 and Hey1,the target genes of Notch signaling.LPS at 1 mg/L increased the proliferation of BMSCs,whereas DAPT(Notch signal inhibitor)reduced the basal and LPS-induced proliferation of BMSCs(P<0.01).LPS treatment robustly increased the secretion of IL-6 and CXCL1 as assessed by ELISA.However, inhibition of Notch signaling almost completely abolished LPS-induced secretion of IL-6 and CXCL1(P <0.05).CONCLUSION: Inhibition of Notch signaling reduced not only the proliferation of BMSCs but also IL-6 and CXCL1 secretion induced by LPS.

RESUMO

Objective To observe the effect of Saccharomyces boulardii(SB) on interleukin-17, interleukin-10 and transforming growth factor-β1 of inflammatory bowel disease ( IBD ) rats which were induced by trinitro-benzene-sulfonic acid( TNBS) . Methods Balb/c female rats were randomly divided into three groups, normal group,TNBS group and TNBS+SB group. TNBS method was used to set up IBD rat models. TNBS group:the mice were injected by 5% TNBS 0. 1 ml+50% ethanol 0. 1 ml per mouse from rectum for 5 days, and then were filled in stomach by normal saline 0. 1 ml on the 2nd day for 2 weeks;TNBS+SB group:the mice were injected by 5% TNBS 0. 10 ml+50% ethanol 0. 10 ml per mouse from rectum for 5 days, and then were filled in stomach by SB (SB no less than 109 CFU/ml) 0. 1 ml on the 2nd day for 2 weeks;the normal group:the mice were injected by 50% ethanol 0. 2 ml per mouse from rectum for 5 days, and then were filled in stomach by normal saline 0. 1 ml on the 2nd day for 2 weeks. At the 15th day,the general situation of mice were observed, serum and colon tissue was collected. Alterations of colon inflamma-tion were observed by means of haematoxylin-eosin ( HE);the level of IL-17,IL-10 and TGF-β1 in serum were evaluated by enzyme linked immunosorbent assay (ELISA). The level of IL-17,IL-10 and TGF-β1 in colon tissue was evaluated by immunohistochemistry. Results The IL-17 level of serum and colon tissue in-creased(P<0. 05),but IL-10(P<0. 05) and TGF-β1(P<0. 05)decreased in TNBS group compared with those in the normal group. The level of IL-17 in TNBS+SB group was lower than that in TNBS group( P<0. 05),but the levels of IL-10(P<0. 05) and TGF-β1(P<0. 05)were higher than those in TNBS group. Compared with the normal group, the level of IL-17 in TNBS+SB group increased ( P<0. 05 ) , but IL-10 (P<0.05) and TGF-β1(P <0.05)decreased. Conclusion The expression of cytokine IL-17 increase, IL-10 and TGF-β1 decrease in peripheral blood and colon tissue of IBD. SB may ameliorate the intestinal inflammatory response of IBD by balancing the expression of IL-17,IL-10 and TGF-β1.

RESUMO

Abstract Newly hatched caterpillars of the butterfly Heliconius erato phyllis routinely cannibalize eggs. In a manifestation of kin recognition they cannibalize sibling eggs less frequently than unrelated eggs. Previous work has estimated the heritability of kin recognition in H. erato phyllis to lie between 14 and 48%. It has furthermore been shown that the inheritance of kin recognition is compatible with a quantitative model with a threshold. Here we present the results of a preliminary study, in which we tested for associations between behavioral kin recognition phenotypes and AFLP and SSR markers. We implemented two experimental approaches: (1) a cannibalism test using sibling eggs only, which allowed for only two behavioral outcomes (cannibal and non-cannibal), and (2) a cannibalism test using two sibling eggs and one unrelated egg, which allowed four outcomes [cannibal who does not recognize siblings, cannibal who recognizes siblings, "super-cannibal" (cannibal of both eggs), and "super non-cannibal" (does not cannibalize eggs at all)]. Single-marker analyses were performed using χ2 tests and logistic regression with null markers as covariates. Results of the χ2 tests identified 72 associations for experimental design 1 and 73 associations for design 2. Logistic regression analysis of the markers found to be significant in the χ2 test resulted in 20 associations for design 1 and 11 associations for design 2. Experiment 2 identified markers that were more frequently present or absent in cannibals who recognize siblings and super non-cannibals; i.e. in both phenotypes capable of kin recognition.

RESUMO

Objective To detect the damage of hippocampal neurons and the changes in inflammatory cytokines in rats after cerebral ischemia-reperfusion(I/R)and compare the expressions of IL-1β,IL-6 and TNFαin hippocampal DG,CA1 and CA3 subregions. Methods The focal cerebral I/R model was induced by an intraluminal filament embolism. The SD rats were randomly divided into the sham-operated group(SHAM group)and the middle cerebral artery occlusion-reperfusion group(MCAO group).HE staining was employed to detect the damage to hippocampal DG, CA1 and CA3 subregions. The expression levels of IL-1β, IL-6 and TNFα were detected by immunofluorescence assay. Results Compared with SHAM group, hippocampal DG,CA1 and CA3 subregion neurons in MCAO group were severely damaged, with occurred inflammatory cell infiltration, and a large amount of neurons apoptosis, and the expressions of IL-1β, IL-6 and TNFαin each subregion increased significantly. At the same time, in MCAO group, the expression of inflammatory cytokines in CA1 subregion was more significant than that in DG and CA 3 subregions(P<0.05).Conclusion Cerebral I/R could cause neuronal damage, inflammatory cell infiltration, and neuronal apoptosis in the DG, CA1 and CA3 subregions of the hippocampus and increase the release of inflammatory cytokines. In MCAO group, the expression of inflammatory cytokines in CA1 subregion of hippocampus is significantly higher than that in DG and CA 3 subregions, suggesting that CA1 region is more sensitive to I/R injury.

RESUMO

Objective To observe the effect of transcutaneous electrical stimulationon on Jiaji acupoint (Ex-B 05) on the expression of glial fibrillary acidic protein (GFAP), nuclear factor kappa B (NF-κB) and interleukin-6 (IL-6) in rats with spinal cord injury (SCI). Meth-ods Ninety healthy adult Sprague-Dawley rats were randomly divided into normal group (group A, n=30), transcutaneous electrical stimula-tion treatment group (group B, n=30) and control group (group C, n=30). Allen's method was used to establish the model of acute SCI in T9. All of them were assessed with Basso-Beattie-Bresnahan (BBB) scale and Slanting Board Test, and the expression of GFAP, NF-κB and IL-6 of the spinal cord were detected with immunohistochemistry one, three and seven days after operation. Results The results of BBB scale and Slanting Board Test were better in group B than in group C three and seven days after SCI (t>3.349, P20.815, P<0.001). Conclusion Transcutane-ous electrical stimulation can inhibit the inflammation and the expression of GFAP in spinal cord after SCI, and improve the motor function in rats after SCI.

RESUMO

Objective To explore the protective effcets of Schisandra chinensis extract (SCE) in paraquat (PQ)-induced pulmonary fibrosis in mice ,and its intrinsic molecular mechanisms thereof. Methods A total of 108 mice were randomly allocated into six groups (n=18):control group, model group, low concentration of SCE group (200 mg/kg), medium concentration of SCE group (400 mg/kg), high concentration of SCE group (800 mg/kg) and vitamin C group (100 mg/kg). Except control group, mice were given by intragastric administration with PQ (100 mg/kg) and administered with SCE and Vitamin C once per 24 h after PQ modeling. Mice were sacrificed at 7, 14 and 21 d after modeling. Six mice were executed at different time points. The degree of lung tissue inflammation and fibrosis were observed by HE staining and Masson staining. The mRNA and protein expression levels of transforming growth (TGF)-β1, interleukin (IL)-6 and IL-17 in lung tissue were determined by RT-PCR and ELISA respectively. Results (1) Compared with control group, the lung tissue of model group showed a large number of inflammatory cell infiltration, space congestion, and its inflammation scores increased at 7 and 14 days after modeling (P<0.05). At the same time, compared with model group and vitamin C group, inflammation scores were significantly decreased in medium concentration of SCE group and high concentration of SCE group (P<0.05). (2) Compared with control group, collagen fibers and the degree of fibrosis were significantly increased in model group ,while pulmonary fibrosis were decreased in medium concentration of SCE group and high concentration of SCE group at 14 and 21 days after modeling (P<0.05). (3) With the extension of modeling time, both mRNA and protein expressions of TGF-β1 were obviously elevated, IL-6 decreased and IL-17 reduced after the first increase in PQ group. Compared with PQ group, levels of three cytokines mRNA and protein expression in medium concentration of SCE group and high concentration of SCE group changed as follows:IL-6 level was markedly decreased at 7 and 14 days after modeling;TGF-β1 level was markedly increased at 14 and 21 days after modeling. However, IL-17 level was markedly decrease at three time points(P<0.05). Conclusion SCE can relieve PQ-induced lung inflammation and fibrosis by suppressing TGF-β1, IL-6, and IL-17 expressions.

RESUMO

A protocol has been developed for induction of somatic embryogenesis from whole inflorescence explants of Chamomilla recutita L. (chamomile). Chamomile is a well-known medicinal plant from the Asteraceae family often referred to as the “star among medicinal species.” Nowadays, it is a highly favoured medicinal plant in folk and traditional medicine. Its multitherapeutic, cosmetic and nutritional values have been established through the years of traditional and scientific use and research. Chamomile has an established domestic (Indian) and international market, which is increasing day by day. Among the various major constituents, α-bisabolol and chamazulene have been reported to be more useful than others. Chamazulene occurs in the capitula of the flowers in minute quantities and has been demonstrated to exert antiinflammatory activity in-vivo. Moreover, chamomile is a seasonal 4-5 months winter crop in India but is extensively required in various medicinal applications. Therefore, to increase the overall yield of this plant, its in-vitro propagation is needed. In the present study, somatic embryos were developed from capitulum explants after 2-4 weeks of culture on MS medium supplemented with 26.8 µM NAA and 11.5 µM Kin. The somatic embryos were further subcultured in-vitro, where new plantlets regenerated from embryos. It is concluded that in-vitro propagation is possible in case of chamomile and can be used to increase the overall yield of chamazulene present in the capitula of flowers as well as augment the overall yield of this important plant, which is conventionally propagated by seeds.

RESUMO

Tissue culture can be used in developing an efficient method for production of valuable and enduring indigenous cultivars. A short-term protocol for plantlet proliferation by using nodal segments was developed for Cucumis sativus, local cultivar from Isfahan; known as Dastgerdi as a tolerant cultivar to the root-knot nematode. Nodal explants were cultured on MS medium containing various concentrations of KIN (0, 0.5, 1 and 1.5mg.l-1) in combination with IBA (0, 0.025 and 0.5mg.l-1). The best response for proliferation rate (100%), mean number of nodes per plantlet (6.01) and percentage of rooted In vitro propagated shoots were observed on medium supplemented by KIN (1.5mg.l-1) after about 3-4 weeks. The maximum length (5.43cm) of micro-shoots obtained on MS medium with KIN (1mg.l-l) + IBA (0.025mg.l-1).

Objective To observe the effect of laparoscopic operation( LPS) for the treatment of endometrio-sis(EMS) infertile serum matrix metalloproteinase in patients of matrix metalloproteinases-9(MMP-9),tissue inhibi-tors of metalloproteinase-1(TIMP-1) and interleukin-2 (IL-2),interleukin-10(IL-10) .Methods 80 patients with EMS in 2012 August to 2013 August to accept the choice of operation cases, according to the different operation modes for the observation group(received LPS operation therapy) and control group(received open operation treatment), 40 cases in each group. The changes of serum MMP9, TIMP-1, MMP-9/TIMP-1 and IL-2, IL-10, IL-2/IL-10 and 6 months after operation in the two groups were observed before and after treatment, the pregnancy situation were ob-served.Results The MMP-9 levels of the patients in the observation group[(51.21 ±24.01)μg/L] than before treatment[(261.88 ±190.11)μg/L] decreased significantly, and the observation group decreased more obviously than that in the control group;the observation group the level of TIMP-1[ (45.88 ±11.02)μg/L] than in the control group[(25.32 ±6.67)μg/L]increased significantly (t =4.846,P <0.05);observation group MMP-9/TIMP-1 decreased significantly compared with the control group, the difference was statistically significant ( t=3.636,P<0.05);the two group after treatment,IL-2,IL-10,IL-2/IL-10,the difference was statistically significant ( t=4.228, 4.415,3.396,all P<0.05);the observation group the pregnancy rate was 65%,the abortion rate 7.7%,30.0%and the 33.3%was better than that of the control group (χ2 =9.825,4.060,all P<0.05).Conclusion LPS is an effective method in treating EMS,which can increase the serum TIMP-1 and IL-2 levels, reduce MMP9 and IL-10 level;improve the immune index, improve the patient's fertility.

RESUMO

Kin discrimination was tested in the cannibalistic H. tigerinus tadpoles to know whether cannibalism is selectively directed towards non-kin members or it is indiscriminate. The association choice tests were conducted using satiated as well as starved subjects with the assumption that they will associate near non-sibs rather than near sibs with the intention of preferentially cannibalizing them. However, test tadpoles, fed or starved showed a random association choice with sibs and non-sibs, as in the end-bias stimulus blank tests. Therefore it is suggested that cannibalistic H. tigerinus tadpoles do not discriminate sibs from non-sibs and cannibalize on both rather indiscriminately.
