RESUMO

Abstract In drug therapy, it is important to provide therapeutic levels of drug to the site of action and maintain them during the treatment. This work describes the in vitro release of alendronate from sodium alginate cross-linked Montmorillonite (MMT) composite beads. Effect of crosslinking cation, concentration of montmorillonite and media on encapsulation efficiencies, and release profiles of alendronate were studied. Beads were characterized using equilibrium swelling ability study, Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), Energy-dispersive x-ray spectroscopy (EDX) and scanning electron microscopy (SEM). Results indicate that addition of montmorillonite increases the encapsulation efficiencies and slows down the release rates significantly.

RESUMO

Abstract Evaluation of montmorillonite for paraquat by in vitro and in vivo test. In vitro test were evaluated by a batch test, taking the paraquat concentration, adsorbents, reaction environment and time as indices, the absorption rate was screened by orthogonal design. In vivo test was executed with rabbits. Group 1: 4 rabbits dosed with montmorillonite. Group 2: 8 rabbits dosed with 200 mg/kg paraquat. Group 3: 6 rabbits dosed with 200 mg/kg paraquat then gavage with montmorillonite 5 min later. Group 4: 6 rabbits dosed with 200 mg/kg paraquat then gavage with montmorillonite 30 min later. Blood paraquat concentration, serum cytokines, blood gas analysis and histopathology of lung were implemented. In vitro test found that all the four factors influence the absorption rate of paraquat (P < 0.05). In vitro test found that oral montmorillonite could change toxicokinetics parameters of paraquat (P < 0.05); decrease raised serum TGF-ß1 and HMGB1 (P < 0.05) and alleviate the histopathology damage of lung. Montmorillonite might exert its protective effects on paraquat induced damage

RESUMO

Abstract Despite decades of research, wound healing remains a significant public health problem. This study aimed to develop and evaluate a topical sodium alginate gel containing vancomycin (Van) loaded MMT NPs for wound healing applications. Van was loaded in MMT at different conditions (pHs of 6, 7 and temperatures of 40, 50 °C) (Van/MMT NPs). The optimum formulation (with the smallest particle size and a high value of zeta potential; 270.8 ± 77.35 nm and -35.96 ± 2.73, respectively) showed a high drug-loading capacity (entrapment efficacy of 96%) and a sustained release pattern of Van (95%) over 480 min. The optimum Van/MMT NPs were embedded into the sodium alginate (SA) gel (Van/MMT NPs/SA gel). The Van/ MMT NPs/SA gel showed a sustained and slow release pattern of Van (95%) over 50 h. FTIR tests revealed the electrostatic interaction between MMT and Van. The broth macrodilution tube method was used to determine the minimum inhibitory concentration (MIC) of Van, Van/ MMT NPs, and Van/MMT NPs/SA gel against Staphylococcus aureus. The results showed the promising antibacterial activity of Van/MMT NPs/SA gel, thus, this gel can be a promising formulation for the management of infected wounds

RESUMO

The aim of the present work is fabrication of dual cross linked sodium alginate(SA)/montmorillonite(MMT)microbeads as a potential drug vehicle for extended release of curcumin(CUR).The microbeads were prepared using in situ ion-exchange followed by simple ionotropic gelation technique. The developed beads were characterized by Fourier transform infrared spectroscopy(FTIR),differential scanning calorimetry(DSC),thermogravimetric analysis(TGA),X-ray diffraction(X-RD)and scanning electron microscopy(SEM).The effect of MMT on encapsulation efficiency of CUR and intercalation ki-netics was investigated.Dynamic swelling study and in vitro release study were investigated in simu-lated intestinal fluid(pH 7.4)and simulated gastric fluid(pH 1.2)at 37℃.Results suggested that both the swelling and in vitro release studies were influenced by the pH of test media, which might be suitable for intestinal drug delivery. The release mechanism was analyzed by fitting the release data into Korsmeyer-Peppas equation.

RESUMO

Aims@#This research was conducted to develop and characterize polyvinyl alcohol (PVA)/montmorillonite (MONT) clay incorporated with carvacrol (Carva) nanocomposite film as a potential material in wound dressing.@*Methodology and results@#Organophilic MONT clay, which was initially modified from commercial MONT clay by cetyltrimethylammonium bromide (CTAB), was used in the polymerization process using PVA. The synthesized nanocomposites were visualized via transmission electron microscopy (TEM). The developed film (PVA/MONT/Carva nanocomposite film) was characterized via Fourier transform infrared (FTIR). The investigation on mechanical property and antimicrobial activity of the film was also performed. All nanocomposites are spherical, with a size of 92.8 ± 22.1 nm. The -OH stretch, C-H stretch, aromatic group, SiO stretch, and C-O from acetyl group were identified in the PVA/MONT/Carva nanocomposite films. During the chemical release test, carvacrol attained a plateau at 24 h, with a total release of 62.3%. This nanocomposite exhibited a severe detrimental influence on the growth of Gram-bacteria and yeasts, which represented a broad spectrum of antimicrobial agents. All test microorganisms showed approximately up to 82% reduction of microbial growth during the Hohenstein challenge test. Physically, the nanocomposite films were yellowish and apparent. The film was sturdy, flexible, elastic and consisted of excellent water holding capacity.@*Conclusion, significance and impact of study@#PVA/MONT/Carva nanocomposite film may have a useful potential to be merged in the pharmaceutical application, especially in wound dressing production.

Objective:To establish the model of rats with UC by 2,4,6 trinitrobenzene sulfonic acid ( TNBS)/ethanol to understand the changes of IL-17,IL-23,TGF-β1,IFN-γin pathogenesis of ulcerative colitis (UC) and the impacts of mesalazine,mont-morillonite powder and clostridium on that.Methods:85 rats were divided randomly into 6 groups including the model ,mesalazine, montmorillonite, clostridium, mesalazine combined with montmorillonite powder group (15 rats per group ) and an additional control group of 10 normal rats.TNBS/ethanol was used to establish the model of rats with UC.The dosage of model, mesalazine, montmorillonite powder ,clostridium and mesalazine combined with montmorillonite powder were converted by surface area of rat and given to the rats by gastric-tube daily. All rats were executed at the 12th day of the molding and treatment ,and the blood and colon samples were collected. The content of IL-17,IL-23,TGF-β1,IFN-γwas measured in the blood by the ELISA method. The different changes of the four cytokines were compared separately. Results: ( 1 ) TNBS/ethanol was used to establish the model of rats with UC.The obvious inflammation can be observed on distal colon of rats by pathologic sections of HE stained .(2) The levels of plasma IL-17,IL-23 and IFN-γin the model, mesalazine, montmorillonite, clostridium and mesalazine combined with montmorillonite powder group were higher than ones in the control gruop (P<0.05),meanwhile,the levels of plasma TGF-β1 are lower (P<0.05).3.Comparing with the model group, the levels of plasma IL-17,IL-23 and IFN-γin the mesalazine, montmorillonite, clostridium and mesalazine combined with montmorillonite powder group were lower , and the levels of TGF-β1 were higer. The differences have statistic significative ( P<0.05).(4)Comparing the efficacy of treatment among mesalazine ,montmorillonite powder, clostridium and mesalazine combined with montmorillonite powder ,mesalazine and mesalazine combined with montmorillonite powder gets the best results ( P<0.05 ).The efficacy between montmorillonite powder and clostridium is similar.Conclusion:(1) TNBS/ethanol can be used to establish the model of rats with UC and can be successfully approved by the measurement of symptoms and pathologic investigation .(2)The IL-17,IL-23 and IFN-γare very active higher in the blood of rats with experimental ulcerative colitis induced by TNBS /ethanol, and have a positive correlation with inflammation. The TGF-β1 is in decrease in the blood of rats with colitis , and has a Negative Correlation with inflammation.(3)Mesalazine,montmorillonite powder, clostridium and mesalazine combined with montmorillonite powder may renovate the damage of inflammatory tissues of rats with experimental ulcerative colitis , and relieve the symptom of inflammation by reducing exudation of proinflammatory cytokines IL-17, IL-23 and immune regulatory cytokines IFN-γby producing the exudation of proinflammatory cytokines TGF-β1 to be good for recovery of UC.

RESUMO

Objective To observe the therapeutic effects of Reduning combined with montmorillonite powder for infantile viral enteritis. Methods A total of 136 patients with infantile viral enteritis in Shanxi Tongchuan Mining Bureau Central Hospital pediatric clinic from August 2010 to October 2013 were selected and divided into two group by random number table method, a Reduning group (65 cases) and a combined treatment group (71 cases). The Reduning group was treated with Reduning injection and the combined treatment group was treated with Reduning injection plus oral montmorillonite powder. The total efficiency, improvement of symptoms and staying hospital time were compared between the two groups. Results The total effective rate in the combined treatment group was significantly higher than that in the Reduning group (97.1% vs. 59.9%, χ2=28.696, P<0.05). The fever clearance time (2.96 ± 1.32 d vs. 3.84 ± 1.94 d; t=2.587, P=0.011), the time to stop diarrhea (3.12 ± 1.44 d vs. 3.95 ± 1.97 d;t=2.340, P=0.022) and staying hospital time (4.60 ± 1.62 d vs. 5.86 ± 2.35 d;t=3.044, P=0.003) in the combined treatment group were all shorter than those in the Reduning group. Conclusions Reduning combined with montmorillonite powder is more effective in the treatment of infantile viral enteritis than Reduning alone, which shortens the course and staying hospital time.
