RESUMO

BACKGROUND Plasmodium vivax, the major cause of malaria in Latin America, has a large subtelomeric multigene family called vir. In the P. vivax genome, about 20% of its sequences are vir genes. Vir antigens are grouped in subfamilies according to their sequence similarities and have been shown to have distinct roles and subcellular locations. However, little is known about vir subfamilies, especially when comes to their functions. OBJECTIVE To evaluate the diversity, antigenicity, and adhesiveness of Plasmodium vivax VIR-E. METHODS Vir-E genes were amplified from six P. vivax isolates from Manaus, North of Brazil. The presence of naturally acquired antibodies to recombinant PvBrVIR-E and PvAMA-1 was evaluated by ELISA. Binding capacity of recombinant PvBrVIR-E was assessed by adhesion assay to CHO-ICAM1 cells. FINDINGS Despite vir-E sequence diversity, among those identified sequences, a representative one was chosen to be expressed as recombinant protein. The presence of IgM or IgG antibodies to PvBrVIR-E was detected in 23.75% of the study population while the presence of IgG antibodies to PvAMA-1 antigen was 66.25% in the same population. PvBrVIR-E was adhesive to CHO-ICAM1. MAIN CONCLUSIONS PvBrVIR-E was antigenic and adhesive to CHO-ICAM1.

RESUMO

Hepatocellular carcinoma ( HCC) , as one of the most common malignant neoplasms , has a relatively high morbidity and mortality rate worldwide.MicroRNAs (miRNAs),a type of comparatively conserved endogenous small non-coding RNAs, function as pivotal regulators involved in various biological functions through the post -transcriptional regulation of gene expression .Some miRNA genes are arranged into a intandem model and reside close together on the same chromosome , forming miRNA clusters . These clustered miRNAs are mostly located on polycistronic transcripts and expressed at similar levels. In the human imprinted 14q32 region, 52 miRNA genes are organized as a large cluster which spans about 220 kb of the long arm ( q) .Lines of evidence show that dysregulation of miRNAs in this cluster are involved in the development of HCC .This review summarizes the structural characteristics of 14 q32 miRNA cluster as well as its impact on HCC in initiation and progression .

RESUMO

Objective To study whether miR-106b is involved in endothelial cells-mediated angio-genesis .Methods miR-106b cultured and transfected with endothelial cells was divided into miR-106b group ,blank control group and positive control group .RNA was extracted from miR-106b .The transfection efficiency was confirmed by inverse transcription .Endothelial cell tubes formed in matrigel were observed .Apoptosis of transfected miR-106b was assayed by TUNEL assay .Target genes of miR-106b were detected .Expressions of miR-106b and candidate genes were detected by RT-PCR and Western blot ,respectively .Results The number of endothe-lial cell tubes formed in matrigel was significantly less in miR-106b group than in blank control group and positive control group .The ratio of formed tubes ,signal transduction and mRNA ex-pression level were significantly lower in miR-106b group than in positive control group ( P0 .05) .Conclusion miR-106b inhibits endothelial cells-medi-ated angiogenesis by down-regulating the signal transduction and STAT 3 ,which is not directly re-lated with VEGFA .
