RESUMO

Nabumetone is used to reduce the pain and inflammation in rheumatoid arthritis. In the current study, immunomodulatory effect of Nabumetone is investigated in mice. The control group was administered normal saline orally as placebo. Nabumetone was administered orally via gavage in two treatment groups at 14mg/kg.b.w. doses and 28mg/kgb.w., respectively. Haemagglutination (HA) assay, Jerne hemolytic plaque and mice lethality assays were applied. In HA assay, the titer was significantly decreased in Nabumetone treatment groups (P< 0.001). In Jerne hemolytic plaque formation assay, there was a significant reduction (P< 0.001) in number of plaques in Nabumetone treated groups when compared with control. In mice lethality assay, there was a significant difference in mortality ratio of mice in control and Nabumetone treated groups (P< 0.001). Therefore, it is concluded that Nabumetone suppresses the humoral immune response in mice.(AU)


A nabumetona é usada na redução da dor e inflamação da artrite reumática. No presente estudo, o efeito imunomodulador é investigado em camundongos. O grupo de controle recebeu solução salina via oral como placebo. Nabumetona foi administrada oralmente via gavagem em dois grupos de tratamentos com doses de 14mg/kg.b.w. e 28mg/kgb.w., respectivamente. Foram realizados ensaios de hemaglutinação (HA), placa hemolítica de Jerne e letalidade dos camundongos. No ensaio HA, o grau foi significativamente menor nos grupos de tratamento com nabumetoma (P< 0.001). No ensaio de formação de placa hemolítica de Jerne houve redução significativa (P< 0.001) no número de placas em grupos tratados com nabumetoma comparado ao controle. No ensaio de letalidade dos camundongos houve diferença significativa no grau de mortalidade de camundongos no grupo de controle e grupos tratados com nabumetoma (P< 0.001). Portanto, conclui-se que a Nabumetoma suprime a resposta imune humoral em camundongos.(AU)

RESUMO

Objective To investigate the preventive effect of nabumetone on the damaged esophageal mucosa in the model of reflux esophagitis, and the effect of expression of cyclooxygenase-2(COX-2), proliferating cell nuclear antigen(PCNA), cyclin D_ 1 (CD_ 1 ) on the progress of reflux esophagitis. Methods A total of 100 SD rats were divided into three groups in random: group Y: operation + saline as positive controls(46); group R: operation + nabumetone(46); and group C: normal control(8). The lesions of esophageal mucosa were observed in the 5th, 17th, 28th , 40th week in groups Y and R and 40th week in group C, respectively. The change of COX-2, PCNA, and CD_ 1 by immunoperoxidase staining in the progress of reflux esophagitis(RE) and the effect of nabumetone were evaluated. RESUMO

OBJECTIVE: To evaluate the clinical curative effect and safety of nabumetone for rheumatoid arthritis. METHODS: The researches about the study on the curative effect and ADRs of nabumetone for rheumatoid arthritis were retrieved and carried out by Meta-analysis. RESULTS: 9 researches were up to the including criteria. As compared with the control group, nabumetone has better effect [RR=1.11,95% CI(1.02, 1.20), P=0.01] but fewer ADRs [RR=0.71,95% CI(0.41, 1.22), P=0.007] in clinical treatment of rheumatoid arthritis. CONCLUSION: The evidence currently available shows that nabumetone has better efficacy and safety. Because the RCTs available for this systematic review are too small, more and further high-quality large-scale RCTs with long-term follow up are required to provide more reliable evidence

OBJECTIVES: Bee venom contains a potent antiinflammatory peptide 401 as well as mellitin. The purpose of this study was to see the efficacy and safety of purified bee venom injection therapy for knee or spinal osteoarthritis patients. METHODS: One hundred and one osteoarthritis patients were randomly assigned to bee venom injection therapy or oral nabumetone medication group. Bee venom injection group was subdivided into 3 groups according to different dosing schedule(group A: gradual increase up to 0.7mg, group B: up to 1.5mg and group C: up to 2.0mg). Control group patients(group D) were given 1000mg nabumetone daily for 6 weeks. There were 25, 26, 25, and 26 patients assigned to A, B, C, or D group. The efficacy of treatment was evaluated by measuring instruments developed by authors, and the safety of bee venom injection was evaluated by hematology and chemistry laboratory examination. RESULTS: Among 101 patients, eighty-one patients completed the study, but twenty patients were dropped out and two of these patients were dropped out due to adverse drug reaction. The efficacy in bee venom group showed better improvement than nabumetone group(p<0.01). Within bee venom group, group B and C showed better improvement than group A(p<0.01). Itching around injection site occurred in most patients, and bodyache occurred in 49 patients (81.7%). Hemoglobin was decreased(0.3g/dl) in group C, but no significant changes were observed in other laboratory values. CONCLUSION: The efficacy of bee venom injection in the control of knee or back pain in osteoarthritis patients was better than nabumetone medication. No severe allergic or adverse reaction was observed in bee venom treatment patients, but problems related with bee venom injection, such as pruritis, bodyache, and the possibility of anaphylaxis, should be considered for the use of bee venom injection.

0.05 ). On the contrary, the COX-2 score in nabumetone group was obviously lower than that in the operation group (P 0.05 ). Conclusion Nabumetone has obvious therapeutic effects on bile-reflux gastritis caused by duodenogastric reflux. Nabumetone can significantly decrease the level of COX-2 in the gastric mucus membrane.
