RESUMO

OBJECTIVE@#To explore the feasibility of detecting maternal hereditary mitochondrial tRNA@*METHODS@#We performed sequence analysis of mitochondrial DNA in blood samples from 2070 cases of maternal hereditary mitochondrial disease in the First Affiliated Hospital of Wenzhou Medical University, and identified 3 patients with m.15927G>A mutation.Buccal swabs and blood samples were obtained from the 3 patients (mutation group) and 3 normal volunteers (control group).After extracting whole genomic DNA from all the samples, the DNA concentration and purity were analyzed.The PCR products were subjected to dot blot hybridization, Southern blot hybridization, and DNA sequencing analysis to verify the feasibility of detecting m.15927G>A mutation using buccal swabs.@*RESULTS@#There was no significant difference in DNA concentration extracted from buccal swabs and blood samples in either the mutation group or the control group (@*CONCLUSIONS@#Buccal swabs collection accurate is an accurate and sensitive method for the detection of m.15927G>A mutation.

A large quantity of studies related to renal transplantation were reviewed to extract and summarize the international frontier hot spots and difficulties, new transplantation technologies, new methods, new visions and new progress on renal transplantation in 2020.The main contents included rejection, optimization application and regulation of immunosuppression, transplant infection, malignancy after transplantation, non-invasive detection and biomarkers, donor organ preservation, repair and utilization, recurrence of renal disease after renal transplantation, multi-factors affecting long-term survival of transplant kidney, computer and artificial intelligence, etc.Strengthening the reading and thinking of literatures in the field of renal transplantation and broadening horizons in higher starting piont, combined with clinical practice of renal transplantation in China, help to promote the long-term efficacy of renal transplantation.

RESUMO

The use of non-invasive blood glucose detection techniques can help diabetic patients to alleviate the pain of intrusive detection, reduce the cost of detection, and achieve real-time monitoring and effective control of blood glucose. Given the existing limitations of the minimally invasive or invasive blood glucose detection methods, such as low detection accuracy, high cost and complex operation, and the laser source's wavelength and cost, this paper, based on the non-invasive blood glucose detector developed by the research group, designs a non-invasive blood glucose detection method. It is founded on dual-wavelength near-infrared light diffuse reflection by using the 1 550 nm near-infrared light as measuring light to collect blood glucose information and the 1 310 nm near-infrared light as reference light to remove the effects of water molecules in the blood. To satisfy the daily demand of skin condition maintenance, make non-invasive real-time detection, and get proper quantitative evaluation of skin viscoelasticity parameters at the same time, a portable non-invasive detection system to acquire real-time skin tissue viscoelasticity is developed. The system relies mainly on a single-degree-of-freedom forced vibration model, with spring-damp-mass, and on dynamic micro indentation method. The experiment is conducted on two kinds of springs, and on pigskin tissues as well, the system's suitability, accuracy and stability are confirmed. The skin viscoelasticity detection in vivo is also carried out on 20 subjects with different ages, the differences of skin viscoelasticity in various parts of the body are investigated, and the correlations between age and skin viscoelasticity are clarified.

To facilitate noninvasive diagnosis of anemia, high-performance and portable near infrared (NIR) spectrometer for human blood constituents was designed and fabricated based on linear variable filter (LVF).Meanwhile, the performance of support vector regression (SVR) model for quantitative analysis of human hemoglobin (Hb) was investigated.Spectral data were collected noninvasively from 100 volunteers by self-designed LVF-NIR spectrometer, then divided into calibration set, validation set 1 and 2.To establish a robust SVR model, grid search method was applied to optimize the penalty parameter and kernel function parameter c=5.28, g=0.33.Then, Hb levels in the validation 1 and 2 sets were quantitatively analyzed.The results showed that the root mean square error of prediction (RMSEP) were 10.20 g/L and 10.85 g/L, respectively, and the relative RMSEP (R-RMSEP) were 6.85% and 7.48%, respectively.The results indicated that the SVR model had high prediction accuracy to Hb level and adaptability to different samples, and could satisfy the requirements of clinical measurement.Based on the SVR algorithm, the self-designed LVF-NIR spectrometer has a wide application prospect in the field of non-invasive anemia diagnosis.

There contains so much physiological/pathological information in pulse wave, which can reflect the early changes in cardiovascular function parameters. Therefore, it would be valuable to quantify the information revealed by pulse wave as important reference for cardiovascular diseases. In this review, the relationship between physiological/pathological information in cardiovascular system and pulse wave was described from the viewpoint of pulse wave formation mechanism, and to explain how the non-invasive detection indexes supplied by pulse wave could be applied in clinical use by analyzing the waveforms. Cardiovascular health evaluation index established by physiological/pathological information from cardiovascular system with pulse wave theory was also discussed. The development of non-invasive detection methods for cardiovascular system based on pulse wave theory can contribute to cardiovascular health care and provide important reference for the early detection, early prevention and early treatment of cardiovascular diseases. Besides, the non-invasive detection method can simplify the process of clinical testing and reduce the costs so as to achieve health assessment on cardiovascular diseases timely and reduce the mortality and disability rates.

Objective To establish the FCM method of non invasive detection of fetal ABO and Rh(D) blood groups in maternal blood.Methods Using absorption and elution method, we obtained the IgG anti A and anti B from human sera. The IgG anti A, B, D were used as the first antibody to react with RBCs in maternal peripheral blood.The goat anti human IgG F(ab')2 FITC was used as the second antibody to conjugate anti A, B, D antibodies, Meanwhile anti i PE was used to mark fetal RBCs in maternal peripheral blood.The fluorescence dot plot diagrams of maternal and fetal cells acquired by FCM were used to detect fetal ABO and Rh(D) blood groups.Results Peripheral blood from 69 pregnant women between 8 and 39 weeks of gestation were studied.Fetal cells could not be found in 13 samples.Of the remaining 56 samples,fetal red cells were identified successfully with ABO/Rh(D) blood types identical to those tested after the birth of the baby.Conclusion In women with fetomaternal hemorrhage(FMH) during pregnancy,the FCM method established by the author can accurately and non invasively detect the blood groups of fetuses.This method can possibly be used for diagnosis of hemolytic disease of the newborn.

RESUMO

The definition, principle, characteristics of micropower Ultra-Wide-Band (UWB) radar are described and discussed in this paper. The human subject is experimented using the UWB radar developed by our team. The experimental results show that UWB radar is able to detect the life signals of the human subject such as respiration and heartbeat without any sensor and electrode touching the subject.The radar can be applied to non-invasive detection of life signals.
