RESUMO

Initiation of rDNA transcription requires the assembly of a specific multi-protein complex at the rDNA promoter containing the RNA Pol I with auxiliary factors. One of these factors is known as Rrn3P in yeast and Transcription Initiation Factor IA (TIF-IA) in mammals. Rrn3p/TIF-IA serves as a bridge between RNA Pol I and the pre-initiation complex at the promoter. It is phosphorylated at multiple sites and is involved in regulation of rDNA transcription in a growth-dependent manner. In the early branching parasitic protist Entamoeba histolytica, the rRNA genes are present exclusively on circular extra chromosomal plasmids. The protein factors involved in regulation of rDNA transcription in E. histolytica are not known. We have identified the E. histolytica equivalent of TIF-1A (EhTIF-IA) by homology search within the database and was further cloned and expressed. Immuno-localization studies showed that EhTIF-IA co-localized partially with fibrillarin in the peripherally localized nucleolus. EhTIF-IA was shown to interact with the RNA Pol I-specific subunit RPA12 both in vivo and in vitro. Mass spectroscopy data identified RNA Pol I-specific subunits and other nucleolar proteins to be the interacting partners of EhTIF-IA. Our study demonstrates for the first time a conserved putative RNA Pol I transcription factor TIF-IA in E. histolytica.

INTRODUCTION: Occurrence of apoptosis and expression of proliferative markers are powerful tools to establish a prognosis in the follow-up of cancer. OBJECTIVE: To evaluate the growth fraction in papillomas and laryngeal squamous cell carcinomas with three degrees of differentiation through apoptosis and the expression of nucleolus organizer regions. METHODS: Retrospective study from which paraffin material was submitted to microtomy and hematoxylin-eosin and silver staining. Stained slides were used to quantify the apoptotic index and the number of nucleolus organizer regions by morphometry. RESULTS: Apoptosis was significantly more frequent in well differentiated carcinomas and in papillomas, and a higher growth fraction of expressed nucleolus organizer regions and cells that expressed a greater than average number of nucleolus organizer regions were more frequently noted in undifferentiated carcinomas. CONCLUSIONS: Thus, it was possible to verify that a high apoptotic index was associated with a lower chance of tumor differentiation in carcinomas, while a greater number of total nucleolus organizer regions, cells expressing nucleolus organizer regions above average and a higher growth fraction were associated with greater likelihood of abnormal cell proliferation and increased tumor differentiation. .


INTRODUÇÃO: A ocorrência de apoptose e a expressão de marcadores proliferativos são ferramentas poderosas no estabelecimento do prognóstico do câncer. OBJETIVO: Avaliar a fração de crescimento de papilomas e carcinomas laríngeos de células escamosas, com três graus de diferenciação, através da apoptose e expressão de regiões organizadoras de nucléolo. MÉTODO: Estudo retrospectivo, cujo material em blocado em parafina foi submetido à microtomia e coloração em hematoxilina-eosina, e pela prata. As lâminas coradas foram utilizadas para quantificar o índice apoptótico e o número de regiões organizadoras de nucléolo (NORs) através da morfometria. RESULTADOS: A apoptose foi significativamente mais frequente em carcinomas bem diferenciados e em papilomas; enquanto que uma maior fração de crescimento, de NORs expressos e de células que expressaram maior número de NORs, foram mais frequentes nos carcinomas indiferenciados. CONCLUSÕES: Foi possível verificar que o índice apoptótico elevado indica menores chances de diferenciação tumoral nos carcinomas, enquanto que um maior número de NORs totais e células expressando NORs acima da média, e uma maior fração de crescimento, determinam maiores chances de proliferação celular anormal e maior diferenciação tumoral. .

The objective of this study was to evaluate the cellular proliferative potential of oral lichen planus (OLP) lesions from patients without hepatitis C virus (HCV) by means of AgNOR method, as well as the cellular proliferative potential of the normal oral mucosa from patients with HCV, treated or untreated by interferon and ribavirin. A cross-sectional study was developed to investigate four groups: 10 HCV+ patients without clinical signs of OLP who had never been treated for HCV infection - Group 1; 10 HCV+ patients that were under interferon and ribavirin treatment - Group 2; 15 patients with reticular OLP lesions histopathologically confirmed, without HCV - Group 3; and 15 blood donors without HCV infection and no clinical signs of OLP GROUP 4 Control Group. The cytological material of all groups was collected by the liquid-based cytology technique. Then, the sedimented material from each patient was filled with the Nucleolar Organizer Regions impregnation by silver method (AgNOR). The count of NORs was performed on 100 epithelial cell nuclei per patient using the Image Tool(tm) software. The Tukey HSD test was used to compare the median value of NORs among the groups and showed that the oral mucosa of HCV+ patients previously treated with anti-HCV drugs (GROUP 2), presented a higher average number of NORs in relation to others (p<0.05). The anti-HCV treatment may be related to increased cell proliferation of oral mucosa, indicating a possible relationship between OLP and HCV+ patients treated with interferon and ribavirin.


O propósito deste estudo foi avaliar o potencial proliferativo celular das lesões de líquen plano bucal (LPB) de pacientes sem vírus da hepatite C (VHC) por meio do método AgNOR, comparando-o ao potencial proliferativo celular da mucosa bucal normal de portadores de VHC, tratados ou não com interferon e ribavirina. Um estudo transversal foi realizado para investigar 4 grupos: 10 pacientes VHC+ sem sinais clínicos de LPB que nunca haviam sido tratados para a infecção por VHC - Grupo 1; 10 pacientes VHC+ que estavam sob tratamento com interferon e ribavirina - Grupo 2; 15 pacientes com LPB reticular histopatologicamente confirmado, sem VHC - Grupo 3; e 15 doadores de sangue sem infecção por VHC e sem sinais clínicos de LPB (Grupo 4 - Grupo de Controle). O material celular de todos os grupos foi coletado pela técnica da citologia em base líquida. Então, o material sedimentado de cada paciente foi submetido ao método da impregnação das regiões organizadoras nucleolares pela prata (AgNOR). A contagem das NORs foi realizada em 100 núcleos celulares epiteliais por paciente por meio do programa Image Tool(r). O teste Tukey HSD foi utilizado para comparar o valor médio de NORs entre os grupos e mostrou que a mucosa bucal dos pacientes VHC+ previamente tratados com fármacos anti-VHC (Grupo 2) apresentou maior número médio de NORs por núcleo em relação aos outros (p<0,05). O tratamento anti-VHC pode estar relacionado ao aumento da atividade proliferativa celular da mucosa bucal, aventando uma possível relação entre LPB e pacientes VHC+ tratados com interferon e ribavirina.

As Regiões Organizadoras de Nucléolo (NORs - nucleolar organizer regions) são utilizadas para descrever regiões de cromatina coradas por Nitrato de Prata e estão relacionados com a atividade de síntese de RNAr e com a agilidade e rapidez na proliferação celular nos tecidos estudados. O objetivo deste trabalho foi relacionar a quantidade de AgNORs, a atividade proliferativa e o estágio da gestação em equinos, utilizando a coloração de Nitrato de Prata. Os anexos embrionários foram coletados, fixados em solução de formaldeído tamponado 10%, emblocadas em paraplast e submetidos à coloração de Nitrato de Prata. Os grupos foram determinados de acordo com a idade gestacional. A quantidade de NORs encontrada no cório no começo da gestação indica início da atividade celular e na medida em que a gestação avança, a quantidade de NORs aumenta, sugerindo maior atividade de síntese e aumento da sua importância na manutenção do feto. Ao contrário do que ocorre no cório, a quantificação das NORs foram maiores no final da gestação do que no inicio, sugerindo a estabilização destas membranas no final da gestação. A cinta coriônica e o saco vitelino foram encontrados no início da gestação e apresentaram grande quantidade de NORs, sugerindo função de síntese e proliferação no inicio da gestação, visto que suas funções é manutenção do embrião até a formação completa da placenta verdadeira (cório-alantoide). Concluímos que as membranas que se desenvolvem de maneira progressiva de acordo com o crescimento embrionário/fetal (cório, alantoide e âmnio) têm aumento no número de NORs e as membranas que involuem após a formação do embrião/feto (saco vitelino e cinta coriônica) têm um decréscimo neste número, sugerindo a diminuição da atividade proliferativa nestas membranas.


The Nucleolar Organizer Regions (NORs - nucleolar organizer regions) are used to describe regions of chromatin stained with silver nitrate and are related to the activity of rRNA synthesis and to the agility and speed of cell proliferation in the tissues studied. The objective of this study was to relate the amount of AgNORs, proliferative activity and stage of pregnancy in horses, using the coloring of Silver Nitrate. The embryonic attachments were collected, fixed in 10% buffered formaldehyde, embedded in paraplast and stained by silver nitrate. The groups were determined according to the gestational age. The amount of the corium NOR found in early pregnancy indicates the onset of cell activity, and in that the pregnancy progresses, the amount of NOR increases, suggesting higher activity and increased synthesis of their importance in maintaining the fetus. Contrary to what occurs in the corium, the quantification of NORs was higher in late pregnancy than in the beginning, suggesting the stabilization of these membranes in late pregnancy. The chorionic girdle and the yolk sac were found in early pregnancy and had lots of NORs, suggesting synthesis function and proliferation in early pregnancy, since their functions is maintenance of the embryo until the complete formation of the true placenta (chorio-allantoic membranes). We conclude that the membranes that develop in a progressive manner in accordance with the growing embryo/fetal (chorion, amnion and allantoic membranes) have an increased number of NORs and the membranes that involute after the formation of the embryo/fetus (yolk sac and chorionic girdle) have a decrease in number, suggesting a reduction in proliferative activity in these membranes.

Antecedentes: El proceso biológico de diferenciación celular es la traducción de múltiples procesos nucleares y citoplasmáticos que determinan cambios complejos y fundamentales en la ultraestructura, bioquímica y fsiología celular, los cuales pueden ser cuantifcados mediante técnicas morfométricas. Objetivo: Evidenciar en términos cuantitativos y morfológicos las variaciones experimentadas por los nucleolos pertenecientes a células mamarias de la línea HC11 tanto normales como en mecanismo de diferenciación. Método: Se estudió a nivel de la microscopía electrónica de transmisión los tipos celulares en etapa de proliferación (HC11 GM) en comparación con células en estadio de diferenciación (HC11 IM), cuantifcando las variaciones de los nucleolos y su relación con estructuras involucradas en síntesis proteica. Resultados: Se evidencian diferencias estadísticamente signifcativas referentes al área, volumen y longitud entre los nucleolos pertenecientes al tipo celular normal-proliferante y el que se encuentra en proceso de diferenciación. Conclusión: Las células mamarias en proceso de diferenciación presentan una notable disminución de sus nucleolos, y sus ribonucleoproteínas constitutivas generarán básicamente ribosomas adheridos al retículo endoplasmático rugoso, sintetizando proteínas de exportación.


Background: The biological process of cell differentiation is the traslation of multiple nuclear and cytoplas-mic processes that determine complex and fundamental changes in ultrastructure, biochemistry and cell physiology, which can be quantifed using morphometric techniques. Objective: To show in quantitative and morphological terms changes experienced by the nucleolus belonging to HC11 line mammary cells both, in proliferating and differentiation process. Methods: A study at the level of transmission electron microscopy of cell types in stage of cell proliferation in comparison with stage of differentiation was designed to quantify variations of nucleolus and their relation to structures involved in protein synthesis. Results: Marked differences in the area, volume and length of the nucleolus were found between normal-proliferating cell types and those in mechanism of differentiation. Conclusion: The mammary cells in differentiation process show a dramatic decline in its nucleoli and their ribonucleoproteins generate basically ribosomes attached at endo-plasmic reticulum, synthesizing export proteins.

The nucleolus is a subnuclear structure of eukaryocytes. It was thought that nucleolus only participates in the biogenesis and processing of rRNA. However, more and more evidence shows that it has many other functions, such as tRNA precursor processing, stress sensing and it is also involved in gene silencing, senescence and cell cycle regulation. Here, we summarize the recent understandings about the nucleolar functions, the regulation of nucleolar localization of proteins and the role that the nucleolus plays in virus infection, in which some related studies of Herpes simplex virus type 1 (HSV-1) US11, UL24 and bovine herpesvirus-1 infected cell protein 27 (BICP27) carried out in our lab will also be included.

Hypostomus is the most speciose genus in the family Loricariidae, with approximately 120 species. These fish show a wide morphological and color variation, which hinders the identification of species, mainly of widely distributed representatives. The aim of this study was to contribute to the current knowledge on cytogenetic features of Hypostomus nigromaculatus. Three specimens of H. nigromaculatus, collected in two tributaries of rio Tibagi, Paraná, and in Cachoeira de Emas, rio Mogi-Guaçu, São Paulo, the latter being the type locality of H. nigromaculatus, were studied. Chromosomal preparations were submitted to Giemsa staining, silver nitrate impregnation, C-banding and CMA3 and DAPI fluorochromes staining. All samples presented 2n = 76, but the rio Mogi-Guaçu sample differed from those from tributaries of rio Tibagi in relation to karyotype formulae, distribution and composition of heterochromatin, and NOR location. The silver nitrate staining revealed the presence of multiple Ag-NORs for all samples, but with differences on the location on chromosomes. CMA3 staining reveled bright signals equivalent to NOR-bearing chromosomal segments; such sites were characterized by negative, i.e. unstained, marks after DAPI staining. The pattern of heterochromatin distribution was distinctive among samples from rio Mogi-Guaçu and tributaries of rio Tibagi. The differences observed between the sample from rio Mogi-Guaçu and the ones from tributaries of rio Tibagi allow us to suggest that these samples are presently isolated. Further analyses are necessary to ascertain whether such isolation refers to distinct populations or characterizes true different species.


O gênero Hypostomus é um dos mais especiosos na família Loricariidae, tendo aproximadamente 120 espécies. Apresenta uma ampla diversidade quanto ao padrão de coloração e morfologia, o que dificulta a identificação de determinadas espécies, principalmente aquelas com ampla distribuição geográfica. Para isso os dados obtidos neste trabalho contribuem para os estudos citogenéticos de Hypostomus nigromaculatus. Foram analisados três exemplares de H. nigromaculatus de afluentes do rio Tibagi, Paraná e Cachoeira de Emas, rio Mogi-Guaçu, São Paulo, sendo esta última, a localidade tipo de H. nigromaculatus. Os cromossomos foram submetidos à coloração convencional (Giemsa), impregnação por nitrato de prata, bandamento-C e coloração com os fluorocromos CMA3 e DAPI. Todos os exemplares apresentaram 2n = 76, no entanto com diferença quanto às fórmulas cariotípicas, distribuição e composição da heterocromatina. O nitrato de prata detectou RONs múltiplas para as amostras, porém com diferenças quanto à localização nos cromossomos. A coloração com fluorocromo CMA3 foi correspondente aos cromossomos Ag-RONs, na coloração com DAPI foram observadas bandas negativas, ou seja, não coradas. O padrão de distribuição da heterocromatina foi diferente para as amostras do rio Mogi-Guaçu e dos tributários do rio Tibagi. As diferenças observadas entre as amostras de rio Mogi-Guaçu e afluentes do rio Tibagi permitem-nos sugerir que essas amostras estejam atualmente isoladas. Outras análises são necessárias para determinar se as amostras analisadas são populações distintas ou trata-se de espécies diferentes.

The pattern of silver nitrate (Ag)-staining differed among testicular lobes of Antiteuchus tripterus. In general, these differences are in regard to the number, size, shape, coloring intensity, and location of the stained bodies or masses, observed during meiosis and spermiogenesis. These characteristics were similar in lobes 1-3. Lobes 4-6, however, differed from each other and from lobes 1-3 as well. Because the Ag-staining method is specific for nucleolar organizing regions and nucleolar material, the observations in meiosis of lobes 1-3 suggested the presence of a single pair of nucleolar organizing region-bearing chromosomes in A. tripterus, as previously found in other Pentatomidae species. In general, the amount of Ag-stained material seen in meiosis of the testicular lobes 1-3 of A. tripterus is smaller than in the other lobes. The differences among Ag-staining in testicular lobes of Antiteuchus tripterus lobes observed during spermiogenesis included a striking variation in morphology of the Ag-stained material found in the head and tail of the spermatids. Given that the key role of the nucleolar material is to participate in protein synthesis, interlobular variations seem to be related to the different functions attributed to each lobe (reproduction to lobes 1-3 and basically nutrition to lobes 4-6). To our knowledge, this is the first time that the nucleolar material was studied in each testicular lobe during spermatogenesis. The present observations encourage further studies since, in addition to being of basic biological interest, several Pentatomidae species are agricultural pests and added knowledge of their biology, mainly in reproduction, may be important for the development of control strategies.

Euphorbia heterophylla L. (Euphorbiaceae) is a herbaceous species of great economic importance due to its invasive potential and consequent damage to agriculture and pasture land. For the first time, we provide information on its chromosome number, morphology, and behavior of mitotic chromosomes. Seeds were germinated and submitted to four treatments to obtain metaphases: 0.5% colchicine for 2 to 5 h, at ambient temperature; 0.5% colchicine for 16 to 24 h; 0.0029 M 8-hydroxyquinoline (8-HQ) for 2 to 5 h at ambient temperature, and 0.0029 M 8-HQ for 16 to 24 h at 4°C. The material was then fixed in methanol:acetic acid (3:1) and kept at -20°C for 24 h. Roots were macerated in the enzyme solution of Flaxzyme™ (NOVO FERMENT™)-distilled water (1:40) at 34°C for 2 h and later fixed again. Chromosome preparations were obtained by the dissociation of the apical meristems. The best chromosome preparations were obtained with the use of 8-HQ for 21 h 30 min at 4°C. E. heterophylla showed 2n = 28 chromosomes. The short arm of the largest pair of chromosomes of the complement (pair number 1) displayed a secondary constriction while the nucleolus was observed in the interphasic cell. Structural rearrangements were also observed in the E. heterophylla L. genome. The genomic instability associated with polyploidy may be the result of selection shaped by environmental adaptations and/or human-induced manipulation through agricultural practices.

Triatomines are of great concern in public health because they are vectors of Chagas’ disease. This study presents an analysis of the species Triatoma melanosoma. The cytogenetic characteristics of triatomines include holocentric chromosomes, post-reductional meiosis in the sex chromosomes and nucleolar fragmentation in the meiotic cycle. The methodology utilized consisted of the techniques of lacto-acetic orcein staining and silver ion impregnation. The organs analyzed were adult testicles. The results enabled to classify the chromosomes by number and size, being three large, eight medium and one small heterochromosome. The three largest chromosomes and the heterochromosomes showed heteropyknotic chromatin in meiosis. The heterochromosomes in 8.05% of the cells in metaphase I behaved as pseudobivalents, contrasting with 91.95% of the cells with individualized sex chromosomes, confirming the achiasmatic nature of these chromosomes. However, the pseudobivalents occurred prominently in metaphase II (78.38%), this fact probably is related to the post-reductional nature of the sex chromosomes. The nucleolus in T. melanosoma persisted until the diplotene phase after which it began to fragment. Nucleolar corpuscles were observed in metaphases I and II and during anaphases I and II, these characteristics being related to the phenomenon of nucleolar persistence. In the initial spermatids, peripheral silver ion impregnation occurred, which could be analogous to the pre-nucleolar corpuscles observed after fragmentation. Thus, this study extends our knowledge of the characteristics of triatomines, in particular, heteropyknotic degree, kinetic activity, formation of sex chromosome achiasmatic pseudobivalency, confirmation of the fragmentation phenomenon, and post-meiotic nucleolar reactivation.

A cytogenetic study was conducted on an Astyanax eigenmanniorum population from the Caetano Stream (18º 44' 56" S/ 048º 18' 39" W) - in Uberlândia, MG, Brazil - showing a modal diploid number of 48 chromosomes in the standard male and female karyotypes. However, in several specimens it was also possible to observe metaphases with one or two B chromosomes, increasing the diploid number to 49 or 50 chromosomes, respectively. The supernumerary chromosomes were totally heterochromatic and highlighted after C-banding. The silver-stained nucleolus organizing regions (Ag-NORs) were located in at least five chromosomes of the standard karyotype, thus characterizing a multiple NOR system in the species. This is the first occurrence of an A. eigenmanniorum population with 2n = 48 chromosomes, bearing supernumerary chromosomes.

Deltamethrin is a widely used insecticide belonging to the class of pyrethroid. Although the neurotoxicity of pyrethroids including deltamethrin is well established, it is still unclear whether exposure to deltamethrin during neonatal period has any deleterious effect on the survival of the Purkinje cells in the cerebellum. In the study, we investigated the total number of Purkinje cells in experimental rats exposed to deltamethrin using a stereological method, the fractionator. Deltamethrin in a dose of 1 mg/kg/day (corresponds to 20% of LD50 ) was administered through oral gavage to male pups from 2nd to 5th postnatal day (PND). At PND 21 the animals were sacrificed and their cerebelli were removed. The cerebelli were systematically sampled using the fractionator method and stained with cresyl fast violet. The number of the Purkinje cells was counted for each cerebellum. The results showed that there was no significant difference in the total number of Purkinje cells in the deltamethrin-treated group as compared to the control animals. This suggests that deltamethrin exposure at the current dosage during the neonatal period do not have any significant effect on the survival of the Purkinje cells in the cerebellum.

Pentatomidae is a family of Heteroptera which includes several agriculture pests that have had different aspects of their meiosis and spermiogenesis analyzed. In the present study we analyzed the morphological patterns of the heteropycnotic chromatin and the nucleolar material of Mormidea v-luteum, Oebalus poecilus and Oebalus ypsilongriseus. The three species presented multilobate testes, with three lobes in M. v-luteum and four in the Oebalus species. A karyotype with 2n = 14 chromosomes (12A + XY) was observed in the three species. Several characteristics were common to the three species, such as the absence of a testicular harlequin lobe (a lobe which produces different types of spermatozoa, previously considered a general characteristic of this family), late migration of the sex chromosomes and semi-persistence of the nucleolus. The three species also shared some characteristics regarding the patterns of the heteropycnotic chromatin and nucleolar material, but differed in others mainly related to the location of the heteropycnotic chromatin in the spermatids and the morphology and distribution of the nucleolar material at zygotene. The differences were always between species from different genera, suggesting a relationship with their genetic divergence.

Triatoma klugi is a Chagas disease vector in the Rio Grande do Sul State. Triatominae chromosomes are holocentric and sex chromosomes segregation is post-reductional. In this paper we describe the karyotype of male T. klugi and a meiotic analysis including the nucleolar behavior during spermatogenesis. Testis cells were analyzed after lacto-acetic orcein and silver nitrate staining. Two autosomes and the heterochromosomes presented nucleolar activity (Ag-NORs) during diplotene-diakinesis. The analysis of metaphase I and II revealed a karyotype with 2n = 20+XY. In metaphase I a prominent nucleolar mass was observed in the cell periphery and small silver grains were detected in metaphase II. During anaphase, the chromosomes segregated in parallel and a typical holocentric late migration behavior was observed. The restoration of the nucleolus was an important feature in this phase. During telophase nucleolar masses persisted and in early spermiogenesis the spermatids presented a small peripheral mass until elongation. The present study is a contribution to the study of chromatin behavior and nucleolar persistence in meiosis.

El nucléolo, considerado únicamente como el sitio de síntesis de los ribosomas, actualmente representa una estructura nuclear dinámica que participa en la regulación de importantes procesos celulares. Numerosas evidencias han demostrado que el envejecimiento celular es una de las diversas funciones que son controladas por el nucléolo. Las mutaciones en las proteínas de localización nucleolar promueven el envejecimiento prematuro en levaduras y humanos. La carencia de represión en la transcripción de genes que codifican para el ARNr que se encuentran dañados, y las mutaciones en las helicasas del ADN encargadas de minimizar la formación de círculos extra-cromosómicos del ADN que codifica para el ARNr, provocan modificaciones en la estructura del nucléolo e inducen envejecimiento prematuro en levaduras. De igual manera, en los humanos la carencia de las helicasas del ADN localizadas en el nucléolo y que participan en el mantenimiento de la integridad genómica, favorecen el desarrollo de aquellas enfermedades asociadas con el envejecimiento acelerado. Además, la presencia de algunos componentes de la telomerasa en el nucléolo, indica que parte de la biosíntesis de esta enzima se realiza en esta estructura nuclear, sugiriendo una conexión entre el nucléolo y la síntesis de los telómeros en la regulación del envejecimiento celular. Por otra parte, el nucléolo secuestra proteínas para regular su actividad biológica durante el inicio o término de la vida replicativa celular.


The nucleolus has been considered originally only as the site for the ribosome synthesis, but now it is well known that it represents a dynamic nuclear structure involved in important cellular processes. Several evidences have demonstrated that the nucleolus regulates the cellular senescence. Specific mutations on the DNAs codifying for nucleolar proteins induced premature senescence from yeast to human. The failure to repress the genes transcription codifying for damaged rRNA, and the mutations in DNA helicases, which minimizes the formation of DNA extra-chromosomal circles codifying for rRNA, modify the nucleolar structure and induce premature senescence in yeast. Similarly, in humans, the reduction of these DNA helicases levels, which are localized in the nucleoli and participate in maintenance of genomic integrity, helps to the development of those diseases associated with premature senescence. Furthermore, the presence in the nucleolus of some telomerase components, indicates that part of the biosynthesis of this enzyme occurred in this nuclear structure; suggesting a communication between the nucleolus and the synthesis of the telomeres in the regulation of cell senescence. On the other hand, the nucleolus sequesters proteins to regulate its own biological activity, from the start to the end of cellular replication. In addition this nuclear structure is involved in the biosynthesis of most cellular ribonucleoprotein particles, as well as in cell cycle regulation, making it central to gene expression. In conclusion, the nucleolus became a multifunctional subnuclear structure involved from cell proliferation to cell senescence.

Objective To investigate the value of quantitative on analysis of argyrophil nucleolar organizer regions(AgNOR),carcinoembryonic antigen(CEA)and ferritin(Ft) for differentiation diagnosis of benign and malignant serous cavities effusion.Methods HPIAS-2000,a highly-clear image analysis system,was used to quantify 12 parameters of AgNOR particles of falling-off cells from 57 cases of benign and malignant serous cavities effusion.The shapes of AgNOR were observed and sorted.Meanwhile,CEA and Ft were examined with chemiluminescent assay.Results Among 12 parameters of AgNOR quantification,9 parameters were significantly distinct between benign and malignant serous cavity effusion(P

Drosophila mulleri (MU) and D. arizonae (AR) are cryptic species of the mulleri complex, mulleri subgroup, repleta group. Earlier cytogenetic studies revealed that these species have different regulatory mechanisms of nucleolar organizing activity. In these species, nucleolar organizing regions are found in both the X chromosome and the microchromosome. In the salivary glands of hybrids between MU females and AR males, there is an interspecific dominance of the regulatory system of the D. arizonae nucleolar organizer involving, in males, amplification and activation of the nucleolar organizer from the microchromosome. The authors who reported these findings obtained hybrids only in that cross-direction. More recently, hybrids in the opposite direction, i.e., between MU males and AR females, have been obtained. The purpose of the present study was to evaluate, in these hybrids, the association of the nucleoli with the chromosomes inherited from parental species in order to cytogenetically confirm the dominance patterns previously described. Our results support the proposed dominance of the AR nucleolar organizer activity over that of MU, regardless of cross-direction.

Objectives To study the relationship between gastrin expression and proliferating cell nuclear antigen (PCNA),argyrophilia nucleolar organizer regions(AgNORs) in colorectal cancer (CC) tissue. Thioacetamide (TA) is converted into a hyperacetylating agent which causes hepatic necrosis, regeneration, cirrhosis and cancerous transformation. One of the most characteristic toxicities of TA in rat is observed with a 50 mg/kg per day which induces nucleolar enlargement different from that in regenerating liver. From TA-treated liver, the nucleoli were isolated and characterized for an altered nucleolar signal transduction system. Immunochemistry revealed that the poisoned nucleoli had increased levels of both nucleolus specific proteins (nucleophosmin and nucleolin) and various signal molecules (CK2, Erk1/2, p38, protein kinases A and C, and cyclin A). Using flow cytometry, the nucleoli were found to be in G2-arrested nuclei. Manifestation of the nucleolar enlargement could be readily observed using an ex vivo hepatocyte culture. There were two types of nucleolar enlargement. One was observed in normal hepatocytes with light density of enlarged nucleoli. The other was in TA-treated hepatocytes with dense and compact density of enlarged nucleoli, which contained a 3 to 5-fold higher nudeophosmin content than the control. In vitro induction of nucleolar enlargement with TA was possible. As soon as the hepatocytes anchored on a collagen coat, exogeneous TA (higher than 1 microg/mL) could induce dense and compact nucleoli. However, when an exogeneous drug was added after monolayer formation (1 day), no drug-induced nucleolar enlargement was observed.

Since there have been very few studies on nucleolar signaling, an attempt was made to establish nucleolar signal pathways which link the cell membrane to the nucleolus for the transfer of extracellular signals. Two pathways were studied. One was the G alpha s mediated cAMP pathway where two signal molecules were yielded, including RII and protein kinase A. The other was the G alpha q mediated DAG/IP3 pathway which yields two signals including protein kinase C and IP3/Ca2+. By the studying isolated nucleoli from resting liver, regenerating liver or weak carcinogen thioacetamide treated liver, it was possible to detect protein kinase A (PKA), protein kinase C (PKC) and RII subunits. In addition, CK2 was detected. It was found that external signals transmitted through G protein coupled receptors could reach into the nucleolus and that physical translocation of signal molecules was an integral step involved in membrane-nucleolus linked pathways. When an in vitro assay of the above signal molecules was carried out using [gamma-32P]-ATP, most kinase dependent phosphorylation was via the major CK2 (more than 95%). Therefore, it is suggested that the major CK2 dependent pathway is involved in 'house keeping' for nucleolar integrity and the minor pathways, dependent on PKA, PKC and others, are involved in subtle regulatory mechanisms such as 'extra-house-keeping' activities by nucleolar chromosomal remodeling.
