RESUMO

Aims: The objective of this study is to monitor the evolution of organochlorine residues levels in kola nuts collected from various actors in the eastern producing region of Côte d'Ivoire. Study Design: Kola nuts samples were collected from farmers, collectors and urban stores in the Eastern of Côte d’Ivoire. Place and Duration of Study: Health Department of Hydrology Health and Toxicology, Training and Research Unit of Pharmaceutical and Biological Sciences, Abidjan, Côte d’Ivoire, running 2016-2019. Methodology: A total of 225 samples were analyzed using Gas chromatography–tandem mass spectrometry (Agilent 7010B Triple Quadrupole GC/MS System). Results: Data showed that all 21 pesticides analyzed were detected in kola nuts samples. Statistical analysis indicated no significant difference in OCPs sub-group used by actors during the three-crop studied. The mean levels of HCHs, DDTs and cyclodienes were ranged from 5±1– 136.67±77.3 µg/kg FW, 5±1 – 116.67±63.2 µg/kg FW and 5±1 - 145±63.2 µg/kg FW, respectively. Otherwise, farmer’s levels of OCPs were lower than those detected in kola nuts from collectors and urban stores samples. Thus, compared to the MRL set by the World Health Organization/Food and Agricultural Organization, the farmer’s samples are lower than limits fixed, unlike the contents registered with collectors and urban stores. In the latter, nearly 80% to 100% of kola nuts collected are contaminated with hexachlorinated residues (HCHs) and heptachlor. Conclusion: There is the need to keep monitoring ecotoxicological chemical substances in kola nuts produced in Côte d’Ivoire and take steps that ensure health safety of end users. Care should be taken since residues could pose chronic health risk for adults and children.

RESUMO

@#Plants contain bioactive compounds and are constantly explored as safer alternatives to conventional insecticides. Despite numerous studies on many plants, information on the insecticidal potential of underutilised plants like tiger nut, Cyperus esculentus L., are scant, although their pharmacological potentials are well known. Hence, this study investigated the larvicidal potential of crude aqueous extracts of two C. esculentus varieties (black and yellow) on the mosquitoes Aedes aegypti (L.) and Culex quinquefasciatus (Say). Mosquito larvae were exposed to C. esculentus crude extracts using the larval bioassay technique of the World Health Organization. Differential larvicidal responses were observed in the test mosquitoes and extracts of Black Dried Tiger nuts (BDT) were more larvicidal than Yellow Dried Tiger nuts (YDT). Acute larval toxicity of the extracts was more pronounced on Cx. quinquefasciatus than Ae. aegypti. The results indicate the potential of C. esculentus (particularly BDT) as a source of mosquito bioinsecticide and merits further studies as a safer alternative to conventional insecticide-based vector control.

RESUMO

Oral Sub Mucous Fibrosis (OSMF) is a common condition found in India due to unique habit of chewing areca nut mixed with pan masala, gutkha, tobacco and spicy food etc. Once started and if neglected it can get transformed into malignancy. The disease once stared can only be controlled with life style changes. We wanted to study the effectiveness of intralesional injection of triamcinolone acetonide, oral llycopene alone and a combination of both in the treatment of oral submucous fibrosis. METHODSThis is a randomised comparative study. All cases with history of chewing of areca nut, pan masala, gutkha, tobacco and associated intolerance to spicy food, restricted mouth opening (grade III) and of age ranging between 15 - 70 years irrespective of gender were included in this study. Patients with previous history of treatment for oral sub mucus fibrosis, disorders of temporomandibular joint and pre-malignant conditions such as leukoplakia, drug allergy were excluded. A total of 150 enrolled cases were randomized into group I, II and III and treated with 1 mL of intralesional injection of triamcinolone acetonide weekly, lycopene capsule twice daily and both in combination respectively for a period of two months. Group II and III cases received oral lycopene throughout the study period, whereas group I and III cases received weekly injection of triamcinolone acetonide till mouth opening of 36 mm or more was achieved. All cases were followed up every two weeks for entire study period of two months, final follow up after two months and oral cavity was assessed for mucosal ulcerations, palpable fibrous bands, mouth opening in millimetres, and tolerance to spicy foods. RESULTSThe number of males and females amongst all groups was 71 and 79 respectively. The most common age group was 51 - 70 years. The mouth opening in 150 cases (grade III) of all groups showed significant improvement (p < 0.001) to grades I and II, of which the group III had maximum mouth opening of 20.22 ± 0.07mm. Amongst all groups in 143 cases, painful mucosal ulcerations disappeared in 136 (95.10 %) and majority (49) were from group II. Similarly, amongst all groups in 142 cases the symptom of burning sensation disappeared in 130 (91.5 %), and majority (49) were from group III. Also, amongst all groups in 150, palpable fibrous bands in oral cavity disappeared significantly in 138 (92 %) (p < 0.15), and the majority (49) were from group III, in remaining 12 it became supple. CONCLUSIONSGroup III agent lycopene in combination with triamcinolone acetonide was more effective than triamcinolone acetonide and lycopene of group I and II respectively when used singly in improving the symptoms and signs of OSMF.

RESUMO

Cardiovascular diseases (CVD) are the main cause of death globally and most CVD can be prevented by addressing their risk factors, such as an unhealthy diet. Many authors have studied the benefits of nut consumption on CVD. Nuts contain high amounts of vegetable protein, unsaturated fatty acids, dietary fibers, vitamins, minerals and many other bioactive compounds, like phytosterols and phenolic compounds, which are able to reduce cholesterol levels and promote antioxidant and anti-inflammatory effects, thereby reducing cardiovascular risks. This review aims to describe studies involving the consumption of nuts, including Brazil nuts and CVD risk factors with positive results in the improvement of lipid profile, glucose metabolism, vascular function, and inflammatory and oxidative stress biomarkers

RESUMO

ABSTRACT High-fat diet-induced obesity is associated with metabolic disorders. The Brazil nut has bioactive substances and has been used to control the damage caused by obesity in several organs. The work intended to show the damage caused by high-fat diet in the bladder wall and if the Brazil nut oil added to the diet could ameliorate or reverse this effect. Sixty-day-old rats were divided into two groups: C (control, n = 30) and HF (high-fat, n = 30) diets. At 90 days, 10 animals of each group were sacrificed. The others were divided into 4 groups: C and HF (animals that maintained their previous diet, n = 10 for each group) and C / Bno and HF / Bno (animals whose control or high-fat diet was supplemented by Brazil nut oil, n = 10 for each group). Sacrifice occurred at 120 days, and the bladders were removed and analyzed. Epithelial height was increased in the HF compared to the C group. In contrast, the C / Bno had a lower epithelial height compared to the others. The percentage of collagen between the detrusor muscle fibers was significantly greater in C / Bno, HF and HF / Bno than in control group. The HF had a larger muscle fiber diameter than the C group, while the C / Bno presented lower values than the HF and HF / Bno groups. HF diets induced bladder wall damage. These changes in the rat's bladder wall were partially reversed by the Bno.

A análise bioinformática foi conduzida usando o Quantitative Insight into Microbial Ecology (QIIME, versão 1.9.0).

Type 2 diabetes mellitus is closely linked to the formation of reactive oxygen species, such as those that increase advanced glycation products (AGEs) and, consequently, inflammation and the development of complications associated with this disease. In this context, the study of the intestinal microbiome is of extreme importance, since it may be able to alter the factors related to the immune system and its modulation may be a good alternative for the improvement of the clinical and inflammatory status. Brazil nut is rich in selenium, in addition to other nutrients, which could act in the anti-inflammatory and antioxidant defense system and influence intestinal microbiota composition. Therefore, this study had the objective of determining the supplementation with type 2 diabetes mellitus. This longitudinal clinical trial was designed with 29 patients with type 2 diabetes mellitus, attended at the Endocrinology Outpatient Clinic of the Hospital das Clínicas, Faculdade de Medicina of the University of São Paulo. The intervention with Brazil nuts lasted 60 days and the participants were evaluated before and after by a collection of feces (intestinal microbiota characterization and selenium concentration) and blood collection (analyzes of selenium status, inflammatory markers, glycemic and lipid levels, and intestinal permeability), and dietary intake by means of a 3-day Food Registry. Initially, the centesimal composition and the selenium concentration of the nut were determined used. Bioinformatic analysis was conducted using the Quantitative Insight into Microbial Ecology (QIIME, version 1.9.0). Statistical analysis was performed with the software Statistical Package for the Social Sciences, version 23.0, and R version 1.1.463. The concentration of selenium in the nut in natura was 79.8 µg / g. The mean age of the participants was 54.9 ± 3.6 years, the mean time of diagnosis of the disease was 11.7 ± 6.9 years. The Brazil nut intake was able to significantly improve the parameters: plasma selenium concentrations (Δ 95,82, p<0,001), nem da circunferência da cintura (Δ-1 p=0,217) and GPX (Δ30.61 p <0.040) and reduction in HbA1c (Δ -0.8, p = 0.001). However, there were no changes in the lipid profile, inflammation, food intake and anthropometric markers. Although we did not verify a change in the global composition of the intestinal microbiota, the interpersonal diversity (ß diversity) varied according to the concentration of SELENOP (p = 0.03) and HbA1c percentage (p = 0.04). We conclude that brazil nut supplementation improved the glycemic profile and the status of selenium. Although no influence was observed on the overall composition of the microbiota, the observed segregation indicates some resistance of the microbiota to the intervention with the chestnut

Brazil nut is a food of high nutritional complexity, due to the high selenium content, micronutrient with antioxidant and anti-inflammatory action, and other important components such as fibers, unsaturated fatty acids, phenolic compounds and various minerals, like zinc, copper and magnesium. This nut is widely consumed by the Brazilian and world population and its health effects must be understood in all aspects. To date, the effects of ingestion of this nut on the constitution of intestinal bacteria were unknown, whose relevance has been exponentially proven in the national and world literature due to its participation in important processes of health and disease. In this sense, the objective of this work was to evaluate the composition of the intestinal microbiota and the mineral profile (metalloma) in the organism after ingestion of Brazil nuts (Bertholletia excels H.B.K.). This prospective study consisted of 97 subjects without chronic noncommunicable diseases of both genders, with a mean age of 28.7 ± 8.5 years. Participants were evaluated before and after supplementation with one unit of Brazil nut (~ 370 µg selenium) per day for 60 days for mineral status (plasma, erythrocytes and feces), glutathione peroxidase (GPX), selenoprotein P (SEPP) concentration, assessment of food intake (24-hour dietary recall) and characterization of the intestinal microbiome. Raw microbiome data were analyzed using Quantitative Insight into Microbial Ecology software (v1.9.0) and statistics were performed using the R package (v1.1.463). Our results indicated that Brazil nuts significantly improved the selenium status of the population, according to the significant increase in concentrations in the body (plasma, erythrocytes and feces) and its functional markers (selenoprotein P and GPX activity) (p <0.0001). The intervention also altered the concentration of other chemical elements (cobalt, arsenic, manganese, barium, lead, copper, zinc, aluminum, iron and magnesium) in plasma, erythrocytes and feces (p <0.001). This study was unprecedented in reporting the effects of Brazil nut intervention on the intestinal bacterial community. Although the overall composition of the intestinal microbiome did not change after ingestion of nuts, the abundance of genera important to human health, such as Faecalibacterium, Bilophila and Bulleidia, was increased. In addition, the metallomic profile in faeces also influenced the constitution and distribution of bacterial groups after nut consumption. We also observed significant correlations between the evaluated minerals and short chain fatty acid producing bacteria, such as Prevotella, Ruminococcus, Oscillospira and Coprococcus. Therefore, it can be concluded that the intervention with Brazil nuts benefited the constitution and distribution of the intestinal microbiota and evidenced the influence of the fecal metallomic profile on bacterial groups important to human health

RESUMO

ABSTRACT Introduction: Ischemia-reperfusion (IR) injury results from inflammation and oxidative stress, among other factors. Because of its anti-inflammatory and antioxidant properties, the Brazil nut (BN) might attenuate IR renal injury. Objective: The aim of the present study was to investigate whether the intake of BN prevents or reduces IR kidney injury and inflammation, improving renal function and decreasing oxidative stress. Methods: Male Wistar rats were distributed into six groups (N=6/group): SHAM (control), SHAM treated with 75 or 150 mg of BN, IR, and IR treated with 75 or 150 mg of BN. The IR procedure consisted of right nephrectomy and occlusion of the left renal artery with a non-traumatic vascular clamp for 30 min. BN was given daily and individually for 7 days before surgery (SHAM or IR) and maintained until animal sacrifice (48h after surgery). We evaluated the following parameters: plasma creatinine, urea, and phosphorus; proteinuria, urinary output, and creatinine clearance; plasmatic TBARS and TEAC; kidney expression of iNOS and nitrotyrosine, and macrophage influx. Results: Pre-treatment with 75 mg of BN attenuated IR-induced renal changes, with elevation of creatinine clearance and urinary output, reducing proteinuria, urea, and plasmatic phosphorus as well as reducing kidney expression of iNOS, nitrotyrosine, and macrophage influx. Conclusion: Low intake of BN prior to IR-induced kidney injury improves renal function by inhibition of macrophage infiltration and oxidative stress.

A multi-residue analysis method was developed for the determination of 38 kinds of pesticides in nuts (almonds, peanuts, cashew nuts and walnuts) by QuEChERS-ultra-high performance liquid chromatography-tandem mass spectrometry. The pesticide residues were extracted with acetonitrile. The extract was cleaned up with PSA, C18 and Oasis PRiME HLB, and then analyzed by UPLC-MS/MS with multiple reaction monitoring (MRM) mode.External standard method was employed to quantify. The limits of detection (LODs, S/N=3) of this method were between 0.01 and 10 μg/kg, and the limits of quantitation (LOQs, S/N=10) were between 0.05-20 μg/kg.All of the tested pesticides showed good linear relationship (r>0.991).The practical samples were determined at three spiked levels and the average recoveries were between 51.0% and 126.0%.The RSDs were less than 20%.This method was simple, sensitive and accurate, and could be used for the routine analysis of pesticide residues in nuts.

RESUMO

ABSTRACT: The present study evaluated the effects of baru nut consumption on body weight, percent adiposity, amount of adipose tissue and blood levels in obese male Swiss mice. After inducing obesity by providing high-glucose diet (60 days), the mice were divided into 4 groups (7 animals per group) and were fed on a control diet (C), high-glucose diet (HG) or high-glucose diet added with baru (HGBA) or soybean oil (HGSO). Groups fed with diet HGBA had a decrease in the weight gain and glucose and triglyceride levels when compared to diet HG. Aimals fed with HG exhibited a higher proportion of epididymal and retroperitoneal adipose tissue. The inclusion of baru nut in the diet improved the control of weight gain and glucose and triglyceride levels in obese mice.

Aflatoxin contamination has been considered as a public health problem, especially in tropical countries, including Brazil. In order to investigate the presence of type B aflatoxins in products marketed in the city of Fortaleza, 23 samples were analyzed by thin layer chromatography. Visible fungal contamination in food was identified according to their macroscopic and microscopic features. The contamination by aflatoxins was detected in 8.7 % of 23 analyzed samples, and 12.5 % of Brazilian nuts samples were positive for AFB1 (<8 µg/kg) and for AFB2 in a contamination rate above the allowed value (16 µg/kg). Among the peanut samples, 33.3 % were positive to AFB1 and AFB2, also in a contamination rate (317.1 µg/kg) which was higher than that recommended by ANVISA. The isolation and morphological characteristics of fungi detected mainly in Brazilian nuts with peel showed the occurrence of the following species: Aspergillus flavus, Aspergillus niger, Mucor sp, Cladosporium sphaerospermum, Cladosporium cladosporoides, Fusarium sp, Aspergillus terreus and bacteria of Actinomycetes phylum. These findings indicate that it is needed to apply the mostly effective quality monitoring of food available to consumers.

RESUMO

The micronutrient selenium is essential to human physiology. As the amino acid selenocysteine, it is inserted into selenoproteins with a wide range of functions including antioxidant capacity, thyroid hormone metabolism, improvement of immune system, brain function, fertility and reproduction. Low selenium status has been associated with increased risk for chronic diseases, such as cancer, type-2 diabetes and cardiovascular disease. In this context, several studies have been conducted in order to investigate if selenium supplementation could reduce the risk of such diseases. However, genetic variations may interfere in the response of individuals to a dietary intervention and must be considered as a important source of inter-individual variation. Therefore, this study was conducted was conducted to investigate the influence of genetic variations in selenoproteins genes on the response to an intervention with Brazil nuts, the richest source of selenium known in nature. The study included 130 healthy volunteers with both genders, aged 20 to 60 years old selected in University of São Paulo. They received nuts for 8 weeks, eating one nut a day, and did a washout period for more 8 weeks. All volunteers had a blood sampling collection every 4 weeks during 4 months, in a total of 5. The following analysis were done: anthropometric measurements, lipid profile, plasma malondialdehyde, plasma and erythrocyte Se, selenoprotein P, plasma and erythrocyte GPx activity, gene expression of GPX1, SEPP1, SELS and SEP15. The volunteers were also genotyped for SNPs rs1050450, rs3811699, rs1800699, rs713041, rs3877899, rs7579, rs34713741 and rs5845. Each unit of Brazil nut provided an average of 300 µg of selenium. All 130 volunteers completed the protocol. The concentrations of total cholesterol and glucose decreased after 8 weeks of supplementation. Moreover, HDL concentrations were higher for carriers of the variant T allele for GPX4_rs713041. The frequencies of the variant genotypes were 5,4% for rs1050450, rs3811699 e rs1088668, 10% for rs3877899, 19,2% for rs713041 e rs7579, 11,5% for rs5845 and 8,5% for rs34713741. The levels of the five biomarkers increased significantly after supplementation. In addition, erythrocyte GPx activity was influenced by rs1050450, rs713041 and rs5845; erythrocyte selenium was influenced by rs5845 and plasma selenium by rs3877899. Gene expression of GPX1, SEPP1 and SEP15 were higher after supplementation. The SNP rs1050450 influenced GPX1 mRNA expression and rs7579 influenced SEPP1 mRNA expression. Therefore, it can be concluded that the supplementation with one of Brazil nut for 8 weeks was efficient to reduce total cholesterol and glucose levels and to increase the concentrations of the main biomarkers of selenium status in healthy adults. Furthermore, our results suggest that GPX4_rs713041 might interfere on HDL concentrations and GPx1 activity, GPX1_rs1050450 might interfere on GPx1 activity, SEP15_rs5845 might interfere on GPx1 activity and erythrocyte selenium and SEPP1_3877899 might interfere on plasma Se levels. Therefore, the effect of genetic variations should be considered in future nutritional interventions evaluating the response to Brazil nut supplementation

An efficient in vitro protocol was standardized for Almond (Prunus dulcis) propagation using dormant axillary buds as explants. Explants were cultured on Murashige and Skoog (MS) and woody plant medium (WPM) supplemented with different concentration/combination(s) of phytohormones. MS basal medium showed lowest shoot induction and took longest duration for shoot initiation. Multiple shoots were induced in MS medium supplemented with the combination of BAP (0.5 mgL-1). Cultures showed poor response for rooting in all combinations of plant growth regulators (PGRs) and took 90 days for initiation. Rooting was higher in half strength of MS than in full-strength. The highest root induction (33.33%) was recorded in half MS medium supplemented with 0.1 mgL-1 IBA (indole-3-butyric acid) followed by full strength of MS medium (20%) supplemented with IBA (0.1 mgL-1). α-Naphthalene acetic acid (NAA) was less effective for rooting than IBA. The highest root induction (25%) was found in half strength of MS medium supplemented with 0.1 mgL-1 NAA followed by full strength of MS medium (20%). The protocol developed would be of use in mass propagation of almond and also support in vitro conservation.
