RESUMO

Up to 15% of male infertility has an immunological origin, either due to repetitive infections or to autoimmune responses mainly affecting the epididymis, prostate, and testis. Clinical observations and epidemiological data clearly contradict the idea that the testis confers immune protection to the whole male genital tract. As a consequence, the epididymis, in which posttesticular spermatozoa mature and are stored, has raised some interest in recent years when it comes to its immune mechanisms. Indeed, sperm cells are produced at puberty, long after the establishment of self-tolerance, and they possess unique surface proteins that cannot be recognized as self. These are potential targets of the immune system, with the risk of inducing autoantibodies and consequently male infertility. Epididymal immunity is based on a finely tuned equilibrium between efficient immune responses to pathogens and strong tolerance to sperm cells. These processes rely on incompletely described molecules and cell types. This review compiles recent studies focusing on the immune cell types populating the epididymis, and proposes hypothetical models of the organization of epididymal immunity with a special emphasis on the immune response, while also discussing important aspects of the epididymal immune regulation such as tolerance and tumour control.

ABSTRACT Introduction: Fabry disease (FD) is a disorder caused by mutations in the gene encoding for lysosomal enzyme α-galactosidase A (α-GAL). Reduced α-GAL activity leads to progressive accumulation of globotriaosylceramide (Gb3), also known as CD77. The recent report of increased expression of CD77 in blood cells of patients with FD indicated that this molecule can be used as a potential marker for monitoring enzyme replacement therapy (ERT). Objective: The purpose of this study was to evaluate the CD77 levels throughout ERT in FD patients (V269M mutation). Methods: We evaluated the fluctuations in PBMC (peripheral blood mononuclear cell) membrane CD77 expression in FD patients undergoing ERT and correlated these levels with those observed in different cell types. Results: A greater CD77 expression was found in phagocytes of patients compared to controls at baseline. Interestingly, the variability in CD77 levels is larger in patients at baseline (340 - 1619 MIF) and after 12 months of ERT (240 - 530 MIF) compared with the control group (131 - 331 MFI). Furthermore, by analyzing the levels of CD77 in phagocytes from patients throughout ERT, we found a constant decrease in CD77 levels. Conclusion: The increased CD77 levels in the phagocytes of Fabry carriers together with the decrease in CD77 levels throughout ERT suggest that measuring CD77 levels in phagocytes is a promising tool for monitoring the response to ERT in FD.


RESUMO Introdução: A doença de Fabry (DF) é um distúrbio causado por mutações no gene que codifica a enzima lisossômica α-galactosidase A (α-GAL). A redução da atividade de α-GAL leva ao acúmulo progressivo de globotriaosilceramida (Gb3), também conhecida como CD77. O recente relato de aumento da expressão de CD77 em células sanguíneas de pacientes com DF indicou que essa molécula pode ser utilizada como um potencial marcador para o monitoramento da terapia de reposição enzimática (TRE). Objetivo: O objetivo deste estudo foi avaliar os níveis de CD77 ao longo da TRE em pacientes com DF (mutação V269M). Métodos: Foram avaliadas as flutuações na expressão de CD77 nas membranas das CMSP (células mononucleares do sangue periférico) em pacientes com DF submetidos à TRE e correlacionados com aqueles observados em diferentes tipos de células. Resultados: Uma maior expressão de CD77 foi encontrada em fagócitos de pacientes em comparação aos controles no início do estudo. Curiosamente, a variabilidade nos níveis de CD77 é maior em pacientes no início do estudo (340 - 1619 MIF) e após 12 meses de TRE (240 - 530 MIF) em comparação com o grupo controle (131 - 331 MFI). Além disso, analisando os níveis de CD77 em fagócitos de pacientes ao longo da TRE, encontramos uma diminuição constante nos níveis de CD77. Conclusão: O aumento nos níveis de CD77 nos fagócitos de portadores de Fabry, juntamente com a diminuição nos níveis de CD77 ao longo da TRE, sugerem que medir os níveis de CD77 nos fagócitos é uma ferramenta promissora para monitorar a resposta à TRE na DF.

Berberis darwinii Hook es una especie que habita el sur de Chile y la Patagonia. Siendo utilizada por la etnia mapuche para el tratamiento de procesos inflamatorios, estados febriles, y dolor estomacal. El propósito del siguiente estudio fue evaluar in vitro las propiedades del extracto de alcaloides de raíz de B. darwinii sobre respuestas celulares en monocitos desde sangre periférica de rata. Los resultados de la cuantificación del extracto muestran una concentración de alcaloides totales de 1,67 mg/g y la caracterización por HPLC- MS determinó la presencia de berberina y palmatina. In vitro se observó que los extractos disminuyeron la capacidad de adhesión y la actividad fagocítica de los monocitos e inhibieron la translocación del factor nuclear NF-κB asociado a la modulación de la inflamación, pero no así la producción de anión superóxido. Estos resultados indicarían que los alcaloides totales de B. darwinii inhiben algunos mecanismos específicos de defensa celular.


Berberis darwinii Hook is a species that inhabits southern Chile and Patagonia. This is being used by the Mapuche ethnic group for the treatment of inflammatory processes, febrile states, and stomach pain. The purpose of the following study was to evaluate in vitro the properties of an alkaloid extract of B. darwinii root on cellular responses in monocytes from the rat peripheral blood. The results of the quantification of the extract showed a total alkaloid concentration of 1.67 mg/g and the characterization by HPLC-MS determined the presence of berberine and palmatine. In vitro, it was observed that the extracts decreased the adhesion capacity and phagocytic activity of the monocytes and inhibited the translocation of the nuclear factor NF-κB associated with the modulation of inflammation, but not the production of superoxide anion. These results indicate that the total alkaloids of B. darwinii inhibit some specific mechanisms of cellular defense.

RESUMO

Brucellosis is one of the common zoonoses caused by Brucella abortus (B. abortus). However, little has been reported on factors affecting invasion of B. abortus into host cells. To investigate cell-type dependent invasion of B. abortus, phagocytic RAW 264.7 and THP-1 cells and non-phagocytic HeLa cells were infected with wild-type and mutant B. abortus, and their invasion efficiencies were compared. The invasion efficiencies of the strains were cell-type dependent. Wild-type B. abortus invasion efficiency was greater in phagocytic cells than in epithelial cells. The results also indicated that there are different factors involved in the invasion of B. abortus into phagocytic cells.

La enfermedad granulomatosa crónica es una inmunodeficiencia primaria, con una incidencia de 1/200 000-250 000recién nacidos vivos. Afecta, principalmente, a varones; la mayoría de las mutaciones son ligadas al cromosoma X y las formas autosómicas recesivas ocurren, con más frecuencia, en comunidades con mayor número de matrimonios consanguíneos. Se caracteriza por sensibilidad a infecciones recurrentes y graves, bacterianas y fúngicas, con formación de granulomas, debido a la incapacidad de los fagocitos para generar compuestos reactivos de oxígeno, necesarios para la muerte intracelular de microorganismos fagocitados. Se presentan tres casos de enfermedad granulomatosa crónica en los que se aisló Serratia marcescens y, tras una anamnesis minuciosa y obtener resultados de pruebas de funcionalidad de neutrófilos, se llegó a un diagnóstico molecular de la enfermedad. La enfermedad granulomatosa crónica puede manifestarse de formas muy variadas, por lo que el alto índice de sospecha y una buena anamnesis son fundamentales para alcanzar un diagnóstico.


Chronic granulomatous disease (CGD) is a primary immunodeficiency with an incidence of 1/200,000-250,000 live births. CGD affects mainly male patients, most of the mutations being X-linked, and autosomal recessive forms occur more frequently in communities with greater numbers of consanguineous marriages. CGD is characterized by sensitivity to recurrent and severe bacterial and fungal infections, with formation of granulomas due to the inability of phagocytes to generate reactive oxygen compounds, necessary for the intracellular death of phagocytic microorganisms. We report three cases of CGD in which Serratia marcescens was isolated, and after detailed anamnesis and performance of neutrophil function tests, a molecular diagnosis of the disease was reached. CGD can be manifested in a wide variety of ways, so that high suspicion and a meticulous anamnesis are essential to reach a diagnosis.

Objective:To investigate the leishmanicidal effects of two antioxidants,caffeic acid and quercetin on Leishmania major (L.major) promasdgotes in vitro,and their immunomodulatory effects on infected phagocytes derived from susceptible BALB/c mice.Methods:Caffeic acid and quercetin-induced cell death was examined by Pi-Hoechst double staining of L.major promastigotes and MTT assay,in the presence or absence of protease inhibitors in vitro.Caffeic acid or quercetin were administered subcutaneously to BALB/c mice infected with L.major promastigotes through a dorsal air pouch.Nitric oxide and superoxide anion production by phagocytes infiltrating the air pouch and the expression of inducible nitric oxide synthase (iNOS),tumor necrosis factor alpha (TNF-α) and nuclear factor kappa B in the air pouch membrane were therefore evaluated using appropriate methods.Results:Caffeic acid and quercetin displayed a dose-dependent cytotoxic effect against L.major promastigotes,and induced cell death via caspases-independent pathways.In vivo,L.major promastigotes inoculation into air pouch cavity of BALB/c mice leads to a sequential influx of neutrophils (hours),followed by macrophages (days).Results showed that L.major delayed apoptosis of infected neutrophils and macrophages by the cleavage of the nuclear factor kappa B p65RelA subunit,and persisted by inhibiting TNF-α and iNOS expression and reactive oxygen species generation.Caffeic acid or quercetin restored reactive oxygen species production and TNF-α-induced iNOS activity,and abrogate apoptosis delay of infected phagocytes.Conclusions:The leishmanicidal effect of caffeic acid and quercetin on promastigotes and amastigotes,as well as reactivation of infected phagocytes apoptosis,suggested a potential therapeutic role against cutaneous leishmaniasis.

OBJETIVO: Analisou-se o impacto do treinamento físico moderado (TFM) e intenso (TFI) sobre o perfil leucocitário e a atividade microbicida de macrófagos alveolares, em 29 ratos machos Wistar. MÉTODOS: Foram formados três grupos: controle-sedentário (CS), treino-moderado (TM) e treino-intenso (TI). Os TFM e TFI foram efetuados através da natação, com aumento progressivo de carga conforme o peso corporal, até um máximo de 3% para o TFM, e 5% para o TFI. As coletas de sangue para contagem total e diferencial dos leucócitos foram automatizadas através do analisador hematológico Sysmex XT- 1800i (Roche®) antes e após o treino. E ao final realizou-se o lavado broncoalveolar para determinar a taxa de fagocitose e a produção de óxido nítrico (ON) de macrófagos. RESULTADOS: O grupo TM apresentou valores maiores para o número de leucócitos (12,77±2,0 x 17,25±2,4 10³/mm³), linfócitos (8,87±1,0 x 12,5±2,1 10³/mm³) e neutrófilos (0,99±0,5 x 3,18±1,0 10³/mm³), p<0,05. Apresentou também uma maior produção de ON (15,77±4,9µmols/mL) e da taxa de fagocitose (38,6%±8,65) em relação ao CS (6,58±1,9µmols/mL e 24,4%±7,40, respectivamente). Enquanto que, o grupo TI apresentou menor taxa de fagocitose (13,1%±1,52 x 24,4%±7,40), maior produção de ON (38,40± 2,1x 6,58±1,9 µmols/mL) e aumento apenas, no valor dos neutrófilos (2,6±1,4x 0,99±0,5 10³/mm³) quando comparado ao CS, p<0,001. CONCLUSÕES: O TFM proporcionou melhora nos mecanismos de defesa dos animais adultos. Enquanto que o TFI reduziu a taxa de fagocitose o que poderá implicar em prejuízo da atividade microbicida dos animais.


OBJECTIVE: To assess the impact of moderate (MPT) and intense physical training (IPT) on the white blood cell profile and the microbicide activity of alveolar macrophages in 29 Wistar male rats. METHODS: The sample was separated into three groups: control group (sedentary), moderate training (MT), and intense training groups (IT). Swimming was the tool used to classify both moderate and intense training groups with a progressive increase of exercise load regarding body weight (up to 3% for the moderate physical training and 5% for the intense training group). The blood samples used to count total and differential leucocytes were automated by a hematological analyzer Sysmex XT-1800i (Roche®) before and after training sessions. At the end of the process, a bronchoalveolar lavage was carried out so to determine the amount of phagocytosis and the production of nitric oxide (NO) of the macrophages. RESULTS: The MT showed an increase in the number of leucocytes (12,77±2,0x17,25±2,4 10³/mm³), lymphocytes (8,87±1,0x 12,5±2,1 10³/mm³) and neutrophils (0,99±0,5x3,18 ±1,0 10³/mm³), p<0,05. There was also a higher production of NO (15,77±4,9µmols/mL) as well as a higher phagocytosis rate of (38,6%±8,65) as to the control group (6,58±1,9 µmols/mL and 24,4%±7,40, respectively). Whereas the IT showed a smaller phagocytosis rate (13,1%±1,52x 24,4%±7,40), a greater production of NO (38,40±2,1 x 6,58±1,9 µmols/mL) and an increase only in the number of neutrophils (2,6±1,4 x 0,99±0,5 10³/mm³) compared to the control group, p<0,001. CONCLUSIONS: The MPT induced a significant improvement in the defense mechanisms of the adult rats. On the other hand, the IPT induced a reduction in phagocytosis rate which could lead to the microbicide activity of the rats being damaged.

A Artrite Encefalite Caprina (AEC) e a Linfadenite Caseosa (LC) possuem alta incidência e transmissibilidade em pequenos ruminantes. Como ambas possuem tropismo por monócitos-macrófagos e afetam mecanismos da resposta inata do hospedeiro, acredita-se que a AEC predispõe o animal a infecções por Corynebacteruim pseudotuberculosis, agente etiológico da LC. Para confirmar esta hipótese, avaliou-se a fagocitose de células da série monócito-macrófago de cabras naturalmente infectadas pelo vírus da AEC (VAEC). Para tanto, foram utilizadas 30 cabras da raça Saanen, alocadas em dois grupos distintos, com 15 animais cada, conforme a sororreatividade de anticorpos séricos antivírus da AEC. Células mononucleares de sangue periférico foram isoladas por gradiente de densidade e plaqueadas para isolamento de células da série monócito-macrófago. Posteriormente, o ensaio de fagocitose de C. pseudotuberculosis foi realizado, após incubação por duas horas a 37ºC a 5% de CO2, e a visualização da fagocitose foi identificada por microscopia óptica. O presente estudo não encontrou diferença na porcentagem de monócito-macrófagos que realizaram fagocitose entre os diferentes grupos (P = 0,41). Todavia, a análise quantitativa de bactérias fagocitadas, demonstrou maior capacidade fagocítica pelos macrófagos-monócitos do grupo sororreagente ao vírus da AEC. Correlação entre monócitos fagocitando e macrófagos que fagocitaram mais de 12 bactérias foi observado neste grupo (r = 0,488; P = 0,006), não sendo o mesmo encontrado no grupo de animais sorroreagentes negativos. Os dados demonstram aumento na intensidade da fagocitose de macrófagos de animais infectados com o vírus da AEC.


Caprine arthritis encephalitis (CAE) and caseous lymphadenitis (CL) have high incidence and transmissibility in small ruminants. Since both virus have tropism for macrophages and monocytes and affect the innate immune response, it is believed that CAE can predispose the animal to infection by Corynebacteruim pseudotuberculosis, the etiological agent of CL. To confirm this hypothesis, we evaluated phagocytosis from the monocyte-macrophage cells from 30 Saanen goats. Goats were uniformly divided in two groups according to results of agar gel immunodiffusion test for CAE virus (CAEV). Peripheral blood mononuclear cells were isolated by density gradient centrifugation and the monocyte-macrophage cells were isolated from the mononuclear cells by their adhesion properties in plaques. Afterwards, phagocytosis of C. psudotuberculosis was performed for two hours at 37ºC, 5% of CO2, and assessed by microscopic visualization. There was no difference in the percentage of monocyte-macrophage cells that phagocytozed C. bovis between groups (P=0.41). However, when phagocytosis rates were classified according to the number of C. pseudotuberculosis phagocyted, the percentage of monocyte-macrophage cells that internalized more than 12 bacteria were higher in serologically CAEV positive animals compared to the serologically negative ones (P<0.001). Furthermore, a positive and significant correlation (r = 0.488; P = 0.006) between the percentage of monocyte-macrophage cells that internalized more than 12 bacteria and the percentage of monocyte that were carrying out phagocytosis was also encountered in serologically CAEV positive goats, however the same were not observed in serologically negative ones. These results demonstrated an alteration in the intensity of C. pseudotuberculosis phagocytosis by monocytes-macrophages from goats infected by CAEV. Thus, these results indicated that goats infected with CAEV may be more susceptible to CL.

O objetivo deste trabalho foi avaliar a atividade funcional de macrófagos de ratos diabéticos, através da liberação do ânion superóxido, na presença do composto "mais vida". Os animais foram divididos em dois grupos, controle (N=20) e diabético (N=20). Avaliou-se a glicemia, massa corpórea e a liberação de superóxido pelos macrófagos de baço de ratos. O composto "mais vida" foi obtido através da mistura de extratos de sete plantas, sendo Orbignia martiana Rodr., Tabebuia avellanedae L.G., Arctium lappa L., Rosa centifolia L., Maytenus ilicifolia Mart., Vernonia condensata Baker e Thuja occidentalis L. Observou-se que glicemia foi maior no grupo diabético. A liberação espontânea do ânion superóxido pelos macrófagos foi menor no grupo diabético. O composto "mais vida", independente dos níveis glicêmicos, aumentou a liberação de superóxido dos macrófagos. Quando as células foram estimuladas pelos extratos vegetais isolados, também houve aumento na liberação do ânion superóxido pelos macrófagos em ambos os grupos. As maiores liberações de superóxido ocorreram quando os macrófagos foram estimulados pela Thuja occidentalis L., Rosa centifolia L., Tabebuia avellanedae L.G. e Maytenus ilicifolia Mart. Estes dados sugerem que a ativação de macrófagos pelo composto "mais vida" pode representar um mecanismo alternativo de defesa para infecções em indivíduos diabéticos.


This study investigated the effects of "mais vida", a commercial natural mix, on macrophages functional activity as evaluated by the superoxide release in diabetic rats. The animals were divided into two groups, control (N = 20) and diabetic (N = 20). This was achieved by determining blood glucose weight and the superoxide released by spleen macrophages. The "mais vida" mix was obtained by the combination of extracts from seven medicinal species, which were: Orbignia martiana Rodr., Tabebuia avellanedae L.G., Arctium lappa L., Rosa centifolia L., Maytenus ilicifolia Mart., Vernonia condensata Baker and Thuja occidentalis L. Blood glucose levels were significantly higher (p<0.05) in the diabetic group, as compared to blood glucose levels in the control group. Superoxide levels in macrophages isolated from normoglycemic rats were higher than those obtained from diabetic animals. The "mais vida" mix, independently of glycemic status, increased significantly the superoxide release in the macrophages. Each extract by itself also increased the superoxide release by phagocytes in the macrophages in both groups. The largest superoxide release occurred when the phagocytes were stimulated by Thuja occidentalis L., Rosa centifolia L., Tabebuia avellanedae L.G. and Maytenus ilicifolia Mart. In addition, the activation of macrophages by the "mais vida" mix may represent an additional protection mechanism for diabetic individuals against infections.

Avaliou-se a celularidade do colostro de 53 vacas da raça Holandesa, utilizando-se 171 amostras, colhidas após a primeira e a segunda ordenha pós-parto. Para a análise citológica quantitativa e qualitativa, foram utilizadas as técnicas de microscopia direta e citocentrifugação. O total de leucócitos obtido por contagem microscópica direta foi de 0,878x10(6) e 1,260x10(6), antes da primeira e da segunda ordenha, respectivamente, e os valores da mediana dos fagócitos obtidos antes da primeira e da segunda ordenha foram de 0,657 e 0,828x10(6) leucócitos mononucleares, e 0,178 e 0,392x10(6) leucócitos polimorfonucleares (P<0,0001). Os valores da mediana dos tipos leucocitários, antes da primeira e da segunda ordenha, foram: 0,640x10(6) e 0,772 monócitos/células epiteliais; 0,136x10(6) e 0,098 linfócitos; 0,045x10(6) e 0,203x10(6) neutrófilos, respectivamente. Concluiu-se que o colostro bovino da segunda ordenha apresentou aumento da quantidade de células somáticas e leucócitos polimorfonucleares/mL de colostro. Essas variações podem estar relacionadas à importância do colostro aos neonatos, ou a modificações fisiológicas e de defesa da glândula mamária, durante o período de adaptação dela.


Colostrum cellularity was studied in 53 Holstein cows using 171 samples, collected before the first and second postpartum milking. Direct microscopic counts and cytocentrifugation were used in the quantitative and qualitative cytological analyses. The total number of leukocytes obtained by means of direct microscopic count was equal to 0.878 x 10(6) and 1,260 x 10(6) , before the first and second milking, respectively. The median of phagocyte counts before the first and second milking were, respectively,0.657 and 0.828 x 10(6) mononuclear leukocytes, and 0.178 and 0.392 x 10(6) polymorphonuclear leukocytes (P<0.0001). The median of leukocyte counts, before the first and second milking were, respectively, 0.640 x 10(6) and 0.772 monocytes/epithelial cells; 0.136 x 10(6) and 0.098 lymphocytes; 0.045 x 10(6) and 0.203 x 10(6) neutrophils. It was concluded that bovine colostrum of the second milking showed an increase in the number of somatic cells and polymorphonuclear leukocytes and polymorphonuclear leukocytes/mL. This variation may be either related to the importance of colostrum to neonates, or to physiological and defense changes that occur in the mammary gland during immediate after postpartum.

In this study, we isolated 12 of Brucella (B.) spp. from cattle, which have been positive in Rose Bangal test and tube agglutination test in Gyeongbuk province in 2009. According to AMOS PCR analysis, isolated 12 strains were identified as B. abortus. Murine derived macrophage, RAW 264.7 cells, were infected with isolated 12 strains or reference strain (B. abortus 544), and bacterial internalization were characterized. According to these results, we divided the isolated strains into the following three groups: class I, lower internalization than that of B. abortus 544; class II, similar internalization to that of that of B. abortus 544; class III, higher internalization than that of B. abortus 544 within RAW 264.7 cells. Furthermore, intracellular growth, bacterial adherent assay, LAMP-1 colocalization, virulence in mice and surface protein pattern were characterized. From these results, representative strains of class III showed lower LAMP-1 colocalization, higher adherent efficiency, higher virulence in mice than those of B. abortus 544, and showed different pattern of surface proteins. These results suggest that B. abortus field strains, isolated from cattle in Korea, possess various virulence properties and higher internalization ability of field strain may have an important role for its virulence expression.

A pele e as mucosas constituem as primeiras barreiras na defesa contra infecções e os macrófagos são componentes essenciais do sistema imune inato, importante neste aspecto. O envolvimento destas células pode ser verificado em grande percentual das imunodeficiências primárias. Desta forma, a avaliação da função fagocitária é de extrema relevância para o reconhecimento dos distúrbios imunológicos que acometem a pele. O objetivo do presente estudo foi avaliar a metodologia laboratorial para a detecção de defeitos funcionais dos fagócitos. Para isto foram estabelecidos os seguintes testes laboratoriais: Nitro Blue Tetrazolium (NBT), Dihidrorodamina (DHR), quimiotaxia, fagocitose e a aderência de S. aureus e C. albicans por citometria de fluxo (CF), além de morte intracelular de S. aureus e C. albicans (CF). Para verificar a integridade do sistema complemento realizou-se ensaios hemolíticos para as vias clássica e alternativa (CH50 e AP50). A metodologia proposta foi aplicada em indivíduos normais para a padronização dos testes. O burst oxidativo avaliado pelo teste da dihidrorodamina (DHR) foi aplicado em 101 indivíduos saudáveis e em paralelo, 50 indivíduos sadios para o teste do NBT. Os mesmos testes foram realizados em pacientes com Candidíase mucocutânea crônica (CMC) (n=9 ), Candidíase persistente (n=5), Suspeita de distúrbios de fagócitos (SDF) (n=14), Doença Granulomatosa Crônica (DGC)(n= 7) e portadores de DGC (n=5)...


Skin and mucosa are part of the first barriers in the defense against infections, and the macrophages are essential components of the innate immune system, important when related to this aspect. The involvement of these cells can be seen in a large percentage of the primary immunodeficiencies. Therefore, the assessment of the phagocitary function is extremely important for the recognition of immunological disorders which affect the skin. The present study focus on the evaluation of the laboratorial methodology for the detection of functional defects of phagocytes. For this the following laboratorial tests were established: Nitro Blue Tetrazolium (NBT), chemotaxis, phagocytosis and adherence of S. aureus and C. albicans through flow cytometry (FC), besides the intracellular death of S. aureus and C. albicans (FC). To assess the integrity of the complement system hemolytic assays were performed for the classic and alternative pathways (CH50 and AP50). The proposed methodology was applied to normal individuals for the standardization of the assays. The oxidative burst evaluated through the dihydrorodamine essay (DHR) was applied to 101 healthy individuals and in parallel, 50 healthy individuals for the NBT assay. The same assays were performed on patients with Chronic mucocutaneous candidiasis (CMC)(n=9), persistent candidiasis (n=5), Phagocytes disorders suspicious (PDS) (n=14), Chronicle granulomatous disease (CGD)(n=7) and CGD carriers (n=5). As imunodeficiências primárias são um grupo de doenças geneticamente heterogêneas que afetam diferentes componentes da imunidade inata e adaptativa, como neutrófilos, macrófagos, células dendríticas, proteínas do sistema complemento, células natural killer e linfócitos B e T. O estudo dessas doenças tem fornecido importantes entendimentos sobre o funcionamento do sistema imune. Mais de 120 diferentes genes já foram identificados, cujas anormalidades são responsáveis aproximadamente 180 diferentes formas de imunodeficiências primárias. Pacientes com imunodeficiências primárias são frequentemente reconhecidos pela sua elevada suscetibilidade a infecções; porém, esses pacientes podem apresentar também várias outras manifestações, como doenças autoimunes, doenças inflamatórias e câncer. O propósito deste artigo é atualizar os principais aspectos das imunodeficiências primárias, especialmente em relação às manifestações clínicas relacionadas ao diagnóstico, enfatizando a necessidade do reconhecimento precoce dos sinais de alerta para essas doenças.


Primary immunodeficiency diseases comprise a genetically heterogeneous group of disorders that affect distinct components of the innate and adaptive immune system, such as neutrophils, macrophages, dendritic cells, complement proteins and natural killer cells, as well as T and B lymphocytes. The study of these diseases has provided essential insights into the functioning of the immune system. Primary immunodeficiency diseases have been linked to over 120 different genes, abnormalities in which account for approximately 180 different forms of these diseases. Patients with primary immunodeficiency diseases are most often recognized because of their increased susceptibility to infections. However, these patients can also present with a variety of other manifestations, such as autoimmune diseases, inflammatory diseases and cancer. The purpose of this article is to update the main aspects of primary immunodeficiency diseases, especially regarding the clinical manifestations related to the diagnosis, emphasizing the need for the early recognition of warning signs for these diseases.

In this study, we observed that lysophosphatidylglycerol (LPG) completely inhibited a formyl peptide receptor like-1 (FPRL1) agonist (MMK-1)-stimulated chemotactic migration in human phagocytes, such as neutrophils and monocytes. LPG also dramatically inhibited IL-1beta production by another FPRL1 agonist serum amyloid A (SAA) in human phagocytes. However, LPG itself induced intracellular calcium increase and superoxide anion production in human phagocytes. Keeping in mind that phagocytes migration and IL-1beta production by FPRL1 are important for the induction of inflammatory response, our data suggest that LPG can be regarded as a useful material for the modulation of inflammatory response induced by FPRL1 activation.

PURPOSE: Diabetes mellitus is a clinically and genetically heterogeneous group of metabolic disorders manifested by abnormally high levels of glucose in the blood. Mounting evidence demonstrates that diabetes is a risk factor for gingivitis and periodontitis. The circulating mononuclear phagocytes in diabetic patients with hyperglycemia are chronically exposed to high level of serum glucose. Thus, this study attempted to determine the effect of pre-exposure of monocytes and macrophages to high concentration of glucose on lipopolysaccharide (LPS)-induced production of pro-inflammatory mediators. MATERIAL AND METHODS: For this purpose, cells were cultured in medium containing normal (5 mM) or high glucose (25 mM) for 4-5 weeks before treatment for 24 h with LPS. LPS was highly purified from Porphyromonas gingivalis or Prevotella intermedia by phenol extraction. RESULT: Results showed that prolonged pre-exposure of cells to high glucose markedly increased LPS-stimulated NO secretion when compared to normal glucose. In addition to NO, high glucose also augmented LPS-stimulated IL-6, IL-8, and TNF-alpha secretion after cells were exposed to high glucose for 4 weeks. CONCLUSION: The present study demonstrates that pre-exposure of mononuclear phagocytes with high glucose augments LPS-stimulated production of pro-inflammatory mediators. These findings may explain why periodontal tissue destruction in diabetic patients is more severe than that in non-diabetic individuals.

As known by other name (natural immunity), the innate immune system comprises all those mechanisms for dealing with infection that are constitutive or built in, changing little with age or with experience of infection. Though in some ways less sophisticated than adaptive immunity, innate immunity should not belittled, since it has evidently protected thousands of species of invertebrates sufficiently to survive for up to 2 billion years. In the innate immune system, molecules of both cellular and humoral types are involved, corresponding to the need to recognize and dispose of different types of pathogen, to promote inflammatory responses and to interact to the adaptive immune system. A major features of innate immunity are the presence of the normal gut flora, complements, macrophages, dendritic cells, natural killer cells and many cytokines that can block the establishment of infection. Both phagocytic cells and complement system have tremendous potential for damaging host cells, but fortunately they are normally only triggered by foreign materials, and usually most of their destructive effects are focussed on the surface of these or in the safe environment of the phagolysosome. This article addreses the comprehensive mechanisms of the major components of the innate immune system to prevent the infection.

The purpose of this study was to determine whether phagocytic activity is measurable by dynamic superparamagnetic iron oxide-enhanced MR imaging. For these experiments on New Zealand White rabbits, which were randomly allocated to normal and silica treated groups, we performed a dynamic MR study and radioisotope study with Tc99m-phytate. In this dynamic MR study, the ratio (Rv) of the distribution volumes of iron oxide (Vm/Ve) could be obtained by applying three- compartment model to the data obtained from the kidney and liver simultaneously. Changes in Rv caused by silica injection and by dosing superparamagnetic iron oxide, AMI-25, were evaluated. In the dynamic MR study using a Beagle dog model the input function could be calculated from data obtained from the hepatic artery and portal vein. Rv's reached maximum values at around 80 minutes after the AMI-25 injection. The Rv of the normal group was 5.06 +/- 1.53 whereas the Rv of the silica treated group was 2.13 +/- 1.20. The results were similar to tissue count data obtained by radioisope study. The Rv value was not dependent on the injected dose of AMI-25. The rate of transport constants (k1, k2, k3) could not be estimated using the 3 compartment model regardless of obtaining the input function. We conclude that Rv may be an quantitative index of decreased phagocytic activity in the liver as determined by dynamic superparamagnetic iron oxide- enhanced MRI.

RESUMO

The phagocytosis of fluorescence microbeads by washed pulmanary marophages of rabbit was determined by the flow cytometry at different postmortem interval. The results showed that the phagocytosis of the pulmonary macrophages of rabbit was markadly dropped with time elapsed after death. It was shown that the rate of phagocytic cell and the number of fluorescence microbeads per cell were decreased along with the prolongation of time after death. This method is helpful to estimate the time of death.
