RESUMO

Objectives: The aim of this work is to carry out a phytochemical study and to evaluate in vitro antibacterial potential of the aqueous extract of Phyllanthus niruri, a plant traditionally used in Kasai Oriental (DR Congo) against various bacteriosis and non-bacterial diseases. Study Design: P. niruri, was selected from a list resulting from an ethnobotanical survey carried out in Kasai Oriental because of the number of citations and recipes involving it, the level of preference of the species as well as the diversity of diseases treated and the plebiscite of its effectiveness by local traditional healers. To contribute to the enhancement of this plant traditionally used against various bacteriosis and to confirm its potential antibacterial power, it was subjected to phytochemical screening and its aqueous extract was tested in vitro on bacterial strains. Place and Duration of the Study: The period of this research went from December 2017 to February 2018. The analyzes were carried out at the physico-chemical and microbiological analysis laboratories of the Congolese Control Office of Mbujimayi (DR Congo) and at the Biology and Chemistry laboratories of the ISP Mbujimayi. Methodology: The chemical groups of the bioactive substances were sought using the classic methods of characterization in solution by precipitation, coloring and foam reactions. The diameters of the zones of inhibition, MICs and CMBs of the aqueous extract of this plant were determined in vitro against 20 bacterial strains subjected to the test. Results: P. niruri contains various bioactive chemical groups. Its aqueous extract showed antibacterial activity in vitro against several of 20 tested bacterial strains. According to the MICs and CMBs, the inhibitory action spectrum covers 12 bacterial strains out of the 20 tested. Conclusion: The results found confirm that P. niruri has antibacterial principles and a therapeutic potential by the presence of several bioactive substances and the inhibitory power of its aqueous extract on some bacterial strains tested.

RESUMO

Aims: This study aimed to find out any synergism of gentamicin with the solvent extracts of small tropical herb Phyllanthus niruri to combat methicillin resistant Staphylococcus aureus. Methodology: Bioactive constituents of Phyllanthus niruri were extracted by macerating ground dry powder of the leaves in water, n-hexane, chloroform, methanol and ethanol for 48-72 h followed by filtration and evaporation of solvents. Microdilution method was used to determine the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of these extracts. The synergistic effects between gentamicin and the extracts were evaluated by the checkerboard assay to calculate the fractional inhibitory concentration index (FICI). In all cases, ten hospital associated MRSA strains were used. Results: The MIC of aqueous and methanolic extracts of P. niruri against different MRSA strains varies from 3.125 mg/ml to 12.5 mg/ml. For the MRSA strain the combination of methanolic extract with gentamicin decreased the MIC of extract from 6.25 mg/ml to 0.2 mg/ml and the MIC of gentamicin from 2048 µg/ml to 256 µg/ml showing a strong synergistic effect with a fractional inhibitory concentration index of 0.157. Steroids, flavonoids, phenolic compounds and quinones identified in the extracts may play role in synergistic relation. Conclusion: The present investigation shows that bioactive constituents from Phyllanthus niruri have an excellent synergy with gentamicin against MRSA and can be further explored as an alternative anti-staphylococcal agent.

RESUMO

Objective: To investigate the antibacterial activities of crude ethanol extracts of 12 Philippine medicinal plants. Methods: Crude ethanol extracts from 12 Philippine medicinal plants were evaluated for their antibacterial activity against methicillin-resistant Staphylococcus aureus, vancomycin-resistant Enterococcus, extended spectrum β-lactamase-producing, carbapenem-resistant Enterobacteriaceae and metallo-β-lactamase-producing Pseudomonas aeruginosa and Acinetobacter baumannii. Results: The leaf extracts of Psidium guajava, Phyllanthus niruri, Ehretia microphylla and Piper betle (P. betle) showed antibacterial activity against the Gram-positive methicillin- resistant Staphylococcus aureus and vancomycin-resistant Enterococcus. P. betle showed the highest antibacterial activity for these bacteria in the disk diffusion (16-33 mm inhibition diameter), minimum inhibitory concentration (19-156 μg/mL) and minimum bactericidal concentration (312 μg/mL) assays. P. betle leaf extracts only showed remarkable antibacterial activity for all the Gram-negative multidrug-resistant bacteria (extended spectrum β-lactamase-producing, carbapenem-resistant Enterobacteriaceae and metallo-c-lactamase-producing) in the disk diffusion (17-21 mm inhibition diameter), minimum inhibitory concentration (312-625 μg/mL) and minimum bactericidal concentration (312-625 μg/mL) assays. Conclusions: P. betle had the greatest potential value against both Gram-negative and Gram-positive multidrug-resistant bacteria. Favorable antagonistic activities were also exhibited by the ethanol extracts of Psidium guajava, Phyllanthus niruri and Ehretia microphylla.

RESUMO

The anti-inflammatory and antiallodynic effects of spray dried powders starting from leaves, stems, roots, the mixture of leaves and stems, as well as the whole plant aqueous solutions of Phyllanthus niruri L., Phyllanthaceae, were assessed. Gallic acid, used as chemical marker, was quantified by HPLC in the spray dried powders. Carrageenan-induced inflammatory and allodynic responses in the mouse paw were used as pharmacological models. Quantitative and qualitative differences among chemical composition of different herb parts were observed. The oral administration of leaves or leaves plus stems spray dried powders (100 mg/kg) significantly inhibited the carrageenan-induced allodynic effect (42 ± 5 and 54 ± 3%, respectively). Additionally, the spray dried powders of leaves significantly reduced carrageenan-induced paw oedema (35 ± 6%). The spray dried powders of roots, stems, or the mixture of leaves, stems and roots (100 mg/kg, p.o.) did not exhibit antiallodynic or antioedematogenic effect in the same model. In conclusion, differences in the chemical composition of spray dried powders from P. niruri are reflected in their in vivo pharmacological actions. Despite of a direct relationship of anti-inflammatory and antiallodynic effects with the gallic acid content had been observed, especially in the spray dried powders of leaves, the use of spray dried powders of leaves plus stems showed to be more effective, suggesting a synergic effect between their constituents.

RESUMO

Phyllanthus niruri L., Euphorbiaceae, spray-dried standardized extract was studied for its anti-inflammatory and antinociceptive activities in adult albino rats and mice. The anti-inflammatory effect of spray-dried standardized extract was observed in carrageenan-induced paw edema and thioglycolate-induced leukocyte migration, while antinociceptive effects were observed using Randall & Selitto, tail flick, and hot plate tests. This study showed that intraperitoneal spray-dried standardized extract at 100, 200, 800, or 1600 mg/kg reduced the vascular response in the inflammatory process of paw edema induced by 1% carrageenan. Oral spray-dried standardized extract at 100 or 200 mg/kg inhibited leukocyte migration to the site of inflammation induced by 3% thioglycolate. In rats, at 100 and 200 mg/kg intraperitoneally, the extract exhibited a marked peripheral analgesic effect in a Randall & Selitto assay and showed significant central analgesic activity in a hot plate and tail flick assay. In conclusion, this study suggested that Phyllanthus niruri spray-dried standardized extract has potent inflammatory and antinociceptive activities and that these activities are not modified by standard drying process, making it feasible to use the dry extract standardized to obtain a phytotherapic preparation and thus validating its use for the treatment of pain and inflammation disorders.

RESUMO

Tea from Phyllanthus niruri L., Phyllanthaceae, aerial parts is commonly used by Brazilian folk medicine for its benefits on the treatment of genitourinary disorders, for what the polyphenolic compounds are mainly responsible. The yield of such compounds may be influenced by several variables related with the plant growth. The effects of planting periods and harvesting conditions are investigated in this work, including four different seasons. The cultivation was characterized by dry mass yield of aerial parts, and the effect of pruning was analyzed. Leaves, stems and their mixtures were analyzed after drying and milling. Loss on drying and water soluble extractives were used as physical parameters for quality control. Flavonoid content and gallic acid were chosen as chemical markers for this work. The spectrophotometric trial based on the aluminum chloride complexes was applied to evaluate the total flavonoids content. Gallic acid contents were measured from the water extractive solutions by high-performance liquid chromatography. The pruning caused a positive influence on the amount of leaves and stems. The highest flavonoids and gallic acid contents were found in the leaves, which were developed over the summer and the winter, respectively, both from the second harvesting (after pruning). Chomatographic profile by HPLC was characterizes by the presence of gallic acid and two other major peaks (not identified substances), which relation was peculiar to each aerial part. In conclusion, these results suggest that even under less favorable climatic conditions, in winter, the pruning seems to cause a strong influence over the P. niruri polyphenolics production. Indeed, the total flavonoids content, as well as the HPLC profile, can be used as indicative parameters of the ratio of leaves and stem in the vegetal raw material.

RESUMO

Phyllanthus niruri L. (Euphorbiaceae), known as "quebra-pedra" (Portuguese for "stonebreaker"), is an herb used for kidney disorders. In light of its frequent use by the population, the present study aimed to investigate the genotoxic, antigenotoxic and cytotoxic activities of a standardized P. niruri extract in bone marrow rats. Three groups of 12 animals were treated daily by gavage over a period of 30 days, with 50, 150 or 250 mg/kg of P. niruri extract aqueous solution. The control group (n = 12) received tap water. At the end of treatment (day 31), groups were divided into two minor subgroups (n=6/group) and received cyclophosphamide (50 mg/kg, i.p.) or saline 0.9% (i.p.). After 24 hours, we evaluated the frequency of micronucleated polychromatic erythrocytes for each animal (MNPCE) at 1000 PCE. Cytotoxicity was evaluated with the PCE/NCE ratio (NEC = normochromatic erythrocytes). General toxicity was assessed during treatment using the parameters of body weight gain, ration and water consumption. The dry extract did not provoke changes in body weight, weight gain, ration and water intake or changes in the frequency of MNPCE or cytotoxicity in bone marrow. We propose that the P. niruri extract used here showed no genotoxic, antigenotoxic and cytotoxic activities under the experimental conditions.

Medicinal plants have been used worldwide in the prevention of urolithiasis. The occurrence of this disease has increased in men, ruminants, dogs and cats. With regard to animals, acquired (congenital or hereditary) pathophysiological factors may be involved, a fact that bears similarity to what has been observed in humans. Yet, despite advances in research, the use of medicinal plants in veterinary medicine remains still underutilized. Thus, the purpose of this study is to conduct a literature review on the use of medicinal plants in the control of urolithiasis. The plants used are: Ammi visnaga, Bergenia ligulata, Cynodon dactylon, Herniaria hirsute and Phyllanthus niruri. Antilithogenic activity was related to changes in the ionic composition of urine, diuretic activity and inhibition of crystallization and aggregation of crystals. It is worth highlighting the use of Phyllanthus niruri due to its efficiency in the prevention of uroliths. Phyllanthus niruri extracts normalized high levels of urinary calcium, inhibiting the formation of uroliths in the initial stages. Additionally, they were associated with a higher glomerular filtration and higher urinary excretion of uric acid. These results suggest a potential use due to its lytic and preventive effect, as well as due to its application to hyperuricemic patients having renal insufficiency. We can conclude that the studies that pertain to the current literature can demonstrate the beneficial effects of using medicinal herbs to control urolithiasis. However, studies conducted in domestic animals remain scant, while tests developed in rodents still predominate.

RESUMO

The effects of aqueous (PnAq) and alcoholic (PnAl) extract (50-250 mg/kg) of P. niruri on in vivo gamma radiation induced chromosome aberration and in vitro antioxidant activity (50-500 µg/ml) were studied. The antioxidant activity was studied by measuring inhibition of hydroxyl radicals generated by the fenton reaction along with pro-oxidant and iron chelating ability. PnAl showed highly significant in vitro free radical scavenging ability when compared to DMSO above 250 µg/ml concentration. PnAq showed significant pro-oxidant activity while PnAl was devoid of it at the tested concentrations. Exposure to gamma radiation (4 Gy) caused 29.10 % increase in the frequency of chromosomal aberrations. Administration of PnAl (250 mg/kg) showed highly significant decrease in chromosomal aberrations compared to radiation treated group. Radioprotective potential of alcoholic extract was found to be more effective than the aqueous extract. Qualitative phytochemical investigation of PnAq and PnAl revealed the presence of sugars, flavonoids, alkaloid, lignans, polyphenols, tannins, coumarins and saponins. Higher radioprotective effect of the alcoholic extract may be attributed to rich presence of antioxidant polyphenolic compounds.

Introduction: both Phyllanthus niruri L. and Azadirachta indica A. Juss. are used in Venezuela on an empirical basis for the treatment of diabetes mellitus. It was considered important to verify the potential usefulness to face acute hyperglicemic condition; and taken into account some adverse effects of A. indica, to value possible synergic effects with P. niruri that allow reducing the dose and taking advantage of its therapeutic pharmacological action. Objectives: to assess the acute hypoglycemic or anti-hyperglycemic effect. Methods: aqueous extracts from A. indica leaves and the P. niruri plant were prepared first, then 70 g of vegetal material were weighted and placed into 200 mL of water at 55-60 ºC. Three courses of treatment were organized; two included each extract and the third based on the combination of equal portions of them. Each extract and the combination were measured their content of glycose in order to prepare glycoside solutions of 13.77, 21.54 and 18.82 mmol/L, which acted as controls of the therapies with A. indica, P. niruri and their combination. They were orally administered at a dose of 5 mL/300 g and compared before and after the administration. Likewise, glycemia was measured before and 2 hours after treatment. Results: in the comparison with its glycoside solution control, only the combination of extracts significantly reduced glycemia. As to the comparison before and after the administration, all the treatments managed to decrease in a significant way glycemia; however, in glycoside solution controls, there was not remarkable variation of glycemia. Conclusions: there was proved the hypoglyceemic effect of the whole P. niruri plant, leading to its validation as a Venezuelan traditional medicinal plant and the synergic effect that may be achieved with A, indica leaves was put forward.

RESUMO

In spite of considerable efforts to identify effective treatments for urolithiasis, this is a goal yet to be achieved. This review summarizes experimental and clinical data evaluating the effect of the plant Phyllanthus niruri, a plant with worldwide distribution, as a potential agent to prevent and/or to treat urolithiasis The review is based on data from the literature and on the results obtained by our group from either in vivo/in vitro experiments or clinical studies. Phyllanthus niruri has been shown to interfere with many stages of stone formation, reducing crystals aggregation, modifying their structure and composition as well as altering the interaction of the crystals with tubular cells leading to reduced subsequent endocytosis. The clinical beneficial effects of Phyllanthus niruri may be related to ureteral relaxation, helping to eliminate calculi or to clear fragments following lithotripsy, or also to a putative reduction of the excretion of urinary crystallization promoters such as calcium. No adverse renal, cardiovascular, neurological or toxic effects have been detected in either of these studies. Altogether, these studies suggest a preventive effect of Phyllanthus niruri in stone formation or elimination, but still longer-term randomized clinical trials are necessary to confirm its therapeutic properties.

RESUMO

AIM: To evaluate anti cataract effect of phyllanthus niruri (PN) both in vitro and in vivo galactose induced cataract.METHODS: Aqueous extract of PN was evaluated against galactose-induced cataract both in vitro and in vivo. Galactosaemic cataract was induced in rats by feeding 300g/L galactose diet. PN was administered orally at three-dose levels 75, 150 and 300 mg/kg of body weight. Rat lenses were subjected to osmotic stress in vitro by incorporating galactose (30mmol/L) in the culture medium. The effect of PN (720 and 880μg/mL) on the glutathione (GSH) and polyols levels was studied.RESULTS: PN significantly delayed the onset and progression of cataract in vivo. In addition to the delay in reaching various stages of development of cataract, stage IV did not develop with lower doses till the completion of experimental period. Lenses treated with PN 880μg/mL concentration showed higher levels of GSH and decreased levels of polyols in vitro. In vivo, 75mg/kg significantly delayed the onset and progression of cataract as compared to control.CONCLUSION: PN delayed the process of cataracto-genesis in the experimental models. However, further study is required to extrapolate the use in human beings for the prevention of cataract.

Medicinal plants can be important therapeutic alternative, however microbiological contamination of plants represent a problem to be solved before the production. The technologic process of raw material has many stages, generally, adverse to microbial growth, but its complete elimination depends on the initial and work condition utilized. The aim of this work was to verify the microbial contamination of Phyllanthus niruri aerial parts and derivatives products, such as extractive solution (SE) and spray dried extract (PSA) with the purpose of evaluating the decrease of the contamination after the decoction and the spray dry. The microbiological analysis of the products was performed by total plate count and MPN coliform. The results showed that the contamination of the products was below the limit maxim. The contamination in the SE was significantly lower than in the plant and this fact suggest that the decoction was responsible for a reduction of 98,3 percent of the initial contamination. On the other hand, the PSA contamination was similar the SE thus suggesting that the spray dry did not affect the microorganisms present in the initial product.

RESUMO

Phyllanthus niruri L. has been used in the folk-medicine of Vietnam and many countries in the world. Authors published the results about separation and recognition phyllathin hypothyllathin and beta-sitoscriptase, these are antioxidant, antibacterial activities. It works at 3 bacterial strains, these are: E. coli, B. subtilis and S. cervisiace; both these agents expressed antioxidant activity but much less than quercetin
