In the present study a series of 34 synthetic ligustrazine-containing α, ß-Unsaturated carbonyl-based compounds and oximes, recognized as anticancer compounds were assessed against protozoa of the Trypanosoma and Leishmania species. Ligustrazine, chemically known as tetramethylpyrazine (TMP), was selected as the core moiety for the synthesis of α, ß-Unsaturated carbonyl-based compounds and these compounds were selected as precursors for the synthesis of new oximes. Some derivates, including 5f and 6i, showed multiple activities against all tested strains. In particular compounds 5f and 8o are the most potent and they are, therefore, potential candidates for trypanosomiasis and leishmaniasis

Background: Sarcocystis species and Toxoplasma gondii are both zoonotic obligatory intracellular protozoan organisms and cyst-forming coccidian parasites that occur in domestic animals and human throughout the world.Methods: Forty local breed rabbits were divided into four groups, each group ten. Group one were infected with Sarcocystis, group two with Toxoplasma and group three with both parasites and last group was non-infected control group. The LAT serological test was used for detection of anti-toxoplasma antibody in serum of Toxoplasma infected rabbits. The direct impression smears stained with Giemsa was prepared from different body organs including; liver, lung, heart, brain and skeletal muscle for detection of tissue cysts (Bradyzoites) of T. gondii and microcysts of Sarcocystis.Results: In group one, 70% of infected rabbits were positive for toxoplasmosis by serological test; both are and by impression smear method 80% of the rabbits were positive for T. gondii with tissue cysts. Fifty percent of rabbits were positive for microcysts of Sarcocystis by direct impression smear method in group two. In group three, the impression smear and latex agglutination method were positive in 40% and 60% of rabbits, respectively. Statistically, there was no significant difference in detection of toxoplasmosis and sarcocystosis by LAT and impression smear method in group one and three.Conclusions: Rabbits could be source of toxoplasmosis and sarcocystosis and have public health implications and hazard as source of food. They might be source of infection for cats and shed environmentally resistant oocysts.

Extracellular vesicles (EVs) are small membrane-bound vesicles of growing interest in vetetinary parasitology. The aim of the present report was to provide the first isolation, quantification and protein characterization of EVs from buffalo (Bubalus bubalis) sera infected with Theileria spp. Methods: Infected animals were identified through optical microscopy and PCR. EVs were isolated from buffalo sera by size-exclusion chromatography and characterized using western blotting analysis, nanoparticle tracking analysis and transmission electron microscopy. Subsequently, the proteins from isolated vesicles were characterized by mass spectrometry. Results: EVs from buffalo sera have shown sizes in the 124-140 nm range and 306 proteins were characterized. The protein-protein interaction analysis has evidenced biological processes and molecular function associated with signal transduction, binding, regulation of metabolic processes, transport, catalytic activity and response to acute stress. Five proteins have been shown to be differentially expressed between the control group and that infected with Theileria spp., all acting in the oxidative stress pathway. Conclusions: EVs from buffaloes infected with Theileria spp. were successfully isolated and characterized. This is an advance in the knowledge of host-parasite relationship that contributes to the understanding of host immune response and theileriosis evasion mechanisms. These findings may pave the way for searching new EVs candidate-markers for a better production of safe biological products derived from buffaloes.(AU)

Metabolomics is a new discipline which has developed rapidly following post-genomic,transcriptomics and proteomics.It has become an important research method because of its unique advantages.It is mainly used in drug research and development,disease surveillance and other fields.In addition,with the gradual maturation of metabolomics technology and the gradual development of new analytical methods,the application of metabolomics has been more and more widely,and metabolomics technology has been gradually applied to the field of parasite research,especially for protozoan parasites.This paper summarizes metabolomics,metabolomics research methods,and the application of metabolomics in research of protozoan parasites,with the aims of further understanding of metabolomics technology,and providing reference basis for solving problems of protozoan parasites.

A avicultura brasileira atualmente ocupa o terceiro lugar, com uma produção anual de aproximadamente, 10,9 milhões de toneladas de carne de frango. Contudo, severas perdas econômicas são relatadas, devido à coccidiose em granjas de frangos de corte, matrizes e postura. As Eimerias são classificadas como protozoários, sendo que os mesmos se multiplicam nas células intestinais diminuindo a absorção de nutrientes, levando à desidratação, perda de sangue e susceptibilidade para infecção por outros micro-organismos. Com o desenvolvimento da pesquisa objetivou-se determinar os índices de produtividade zootécnica (ganho de peso, consumo de ração, conversão alimentar, taxa de mortalidade e índice de eficiência produtiva) bem como, mensurar o nível residual do Diclazuril nos tecidos de frangos de corte, comparando com os padrões internacionais de Limites Máximos de Resíduos determinados pelo Codex Alimentarius. Para a realização do estudo, utilizou-se 624 frangos de corte, onde metade do grupo foi inoculado experimentalmente com E. acervulina, E. maxima e E. tenella. O estudo foi composto por grupos tratados e não tratados com diclazuril. O uso do diclazuril expressou efeito positivo, no desempenho zootécnico das aves inoculadas artificialmente; a análise residual do medicamento apresentou um período de carência zero, sendo considerada segura para alimentação humana a carne de frangos medicados com Diclazuril.

A ocorrência de Giardia, Cryptosporidium e microsporídios foi investigada por meio da análise de 98 amostras fecais de animais silvestres capturados em uma área de desmatamento para a construção das barragens de Paraitinga e Biritiba, localizadas nos Municípios de Mogi das Cruzes, Salesópolis e Biritiba-Mirim, no Estado de São Paulo. As amostras foram obtidas de 46 roedores, 21 marsupiais, 16 sapos, nove morcegos, três primatas e três lagartos. As técnicas de centrífugo-flutuação com sulfato de zinco, de Kinyoun e a coloração de Gram-Chromotrope foram utilizadas, respectivamente, para a pesquisa de Giardia, de Cryptosporidium e de microsporídios. O total de animais parasitados por um dos protozoários investigados foi de 17,35 por cento (17/98). Cistos de Giardia foram encontrados em amostras fecais de dois pequenos roedores da espécie Coendou villosus (ouriço-cacheiro). Os três animais positivos para Cryptosporidium foram roedores das espécies Akodon montensis, Thaptomys nigrita (ambos conhecidos como ratos do mato) e Sciurus aestuans (serelepe ou caxinguelê). Esporos de microsporídios foram encontrados nas fezes de 12 animais, sendo seis roedores das espécies Oligoryzomys sp.(um), Akodon montensis (três) e Coendou villosus (dois), três marsupiais pertencentes às espécies Didelphis aurita (dois) e Marmosops incanus (um) e três morcegos da espécie Diphylla ecaudata. Este é o primeiro relato de microsporidiose em animais silvestres no Brasil. A presente investigação enfatiza a importância de animais silvestres, particularmente pequenos mamíferos, como potenciais fontes de infecção desses protozoários para outras populações animais, incluindo o homem, em áreas de desmatamento.


The occurrence of Giardia, Cryptosporidium and microsporidia was investigated in 98 faecal specimens from wildlife animals, captured in an area of deforestation for the construction of two water reservoirs (Paraitinga and Biritiba), located in the municipalities of Mogi das Cruzes, Salesópolis and Biritiba-Mirim, in the state of São Paulo (Brazil). Samples were obtained from 46 rodents, 21 marsupials, 16 frogs, 9 bats, 3 tamarins and 3 lizards. For the detection of Giardia, Cryptosporidium and microsporidia it was used, respectively, the floatation technique with lead sulphate, the Kinyoun method and the Gram-Chromotrope staining. The total number of parasitized animals by one of these protozoans was 17.35 percent (17/98). Cysts of Giardia were found in faecal samples from 2 prehensile-tailed porcupines (Coendou villosus). The three positive animals for Cryptoporidium were rodents - 1 montane akodont (Akodon montensis), 1 ebony akodont (Thaptomyces nigrita) and 1 guainan squirrel (Sciurus aestuans). Microporidia spores were seen in the stools of 12 animals - 6 small rodents, including 3 montane akodonts, 1 prehensile-tailed porcupine and 2 pigmy rice rats (Oligoryzomys sp.); 3 marsupials, including 1 gray slender mouse opossum (Marmosops incanus) and 2 big eared opossums (Didelphis aurita); 3 hairy-legged vampire bats (Diphylla ecaudata). This is the first description of microsporidiosis in wildlife animals in Brazil. The present study emphasizes the importance of these animals, particularly small mammals, as potential sources of protozoan infection to other animal populations, including man, in areas of deforestation.
