RESUMO

To study the effect and mechanism of extract of Quzhou Aurantii Fructus(QAF) on liver inflammation in CCl_4-induced liver fibrosis mice. Totally 60 C57 BL/6 male mice were randomly divided into control group(distilled water, oral), model group(distilled water, oral), colchicines group(Col, colchicines 2 mg·kg~(-1)·d~(-1), oral), low-dose QAF group(QAF-L, QAF 100 mg·kg~(-1)·d~(-1), oral) and high-dose QAF group(QAF-H, QAF 300 mg·kg~(-1)·d~(-1), oral) by random number table method. The model group and each administration group were injected with carbon tetrachloride(CCl_4) 1 mL·kg~(-1)(CCl_4-olive oil 1∶4), twice a week, totally 6 weeks. After the last administration, the mice were sacrificed, and serum and liver tissue were collected. Serum ALT and AST levels were measured in each group to observe the liver function of mice. The pathological changes and inflammatory cell infiltration in liver were observed by HE staining and F4/80 immunohistochemical staining. The mRNA expressions of TNF-α, IL-18 and IL-1β were detected by RT-PCR. The protein expressions of IκBα, p-IKKα/β, p-p65, NLRP3, caspase-1 and cleaved caspase-1 were analyzed by Western blot. The results showed that QAF significantly reduced serum ALT and AST levels, and alleviated the degree of liver damage.The results of immunohistochemistry showed that QAF significantly reduced liver inflammatory cell infiltration in liver fibrosis mice. The results of RT-PCR and Western blot showed that QAF significantly inhibited mRNA expressions of TNF-α, IL-18 and IL-1β in liver of fibrosis mice. QAF also suppressed the degradation of IκBα protein and reduced p-IKKα/β, p-p65, NLRP3 and cleaved caspase-1 protein expressions. In conclusion, QAF improves CCl_4-induced liver fibrosis in mice. The mechanism may be related to the inhibition of NF-κB/NLRP3 inflammasome-mediated inflammation signaling pathway.

RESUMO

[Objective]To sum up LIN Qinfu(the fifth generation heritage of Quzhou LEI's medical )'s academic thoughts on clinical diagnosis and treatment of pediatric diseases. [Method]To study and research LIN Qinfu′s papers and books, to collect and arrange typical cases record, and his method of TCM differentiation of symptoms and signs, by these methods to summarize LIN Qinfu′s clinical experience in the treatment of pediatric diseases. [Result] LIN Qinfu pays attention to learn traceability, inherits two famous schools of medical of Xin'an WANG 's medical and Quzhou LEI's medical;Emphasize the comprehensive use of four diagnostic methods, observation is the most important in the diagnosis of pediatric. Advocate that children's physique is tender Yin-Yang,treatment for children should calm the mind, balance Yin-Yang,therapy and medication should be gentle and take account in the spleen and stomach. [Conclusion]LIN Qinfu has profound knowledge and rich experience of the treatment of pediatric diseases. and his clinical experience academic is medication unique, which is worthy of inheritance and promotion.

RESUMO

[Objective]To study doctor LEI Feng's life story from Zhejiang Province in late Qing Dynasty from the point of view of the social history. [Methods]With the research methods of history and philology, collect relevant historical materials, to study LEI Feng 's life story from the view of social history. [Results]LEI Feng was born in the nineteenth year of the Daoguang Emperor of the Qing Dynasty(1839), died in the fourteenth year of the Guangxu Emperor of the Qing Dynasty(1888), at the age of 49. LEI Feng had a daughter named GONG Yuyun(about 1870-1899), a son named LEI Dazheng(?-1899), two students named ChENG Xi and JIANG Cheng, LEI Feng's father named LEI Huanran(?-about 1862), LEI Huanran's medical teacher was CHENG Zhitian. [Conclusion]LEI's medical family in Quzhou city of Zhejiang Province inherited from father to son and teacher to student, they both had their own medical literatures, the academic origin of LEI's medical family was Xin'an medicine;though LEI Feng called himself an ordinary person, he was good at writing poem, calligraphy and painting even music, he played a special role in traditional Chinese medicine 's social history.
