RESUMO

Objective:To optimize proportion of couplet medicine of Bupleuri Radix and Scutellariae Radix in the treatment of anti-depression, and to explore the possible antidepressant mechanism of this couplet medicine. Method:The dosages of Bupleuri Radix and Scutellariae Radix in the 2015 edition of Chinese Pharmacopoeia were taken.According to U7(72) uniform design table, Bupleuri Radix and Scutellariae Radix were carried out into 7 groups.The chronic unpredictable mild depression model mice were induced by intragastric administration of decoction of this couplet medicine and the antidepressant effect was observed by the behavior tests, which included sucrose preference test, tail suspension test, forced swimming test and open field test(crossing scores).The regression equations were established by selecting the effective indexes.The experiments of retest were taken to check the results and the possible antidepressant mechanism was primarily investigated by measuring the phosphorylation level of cyclic adenosine monophosphate(cAMP)-response element binding protein(CREB) and expression of brain-derived neurotrophic factor(BDNF). Result:Compared with the blank group, sucrose preference rate of the model group was significantly decreased(PPPPPPPPPPPPPPConclusion:Compatibility of Bupleuri Radix and Scutellariae Radix can ameliorate depressive-like behavior of model mice, and the best antidepressant compatibility proportion of Bupleuri Radix and Scutellariae Radix is 1:1, the optimal amounts of them are about 5 g.The antidepressant effect may relate to promoting phosphorylation level of CREB and the expression of BDNF in the hippocampus.

Objective: To investigate the effects of compatibility of Radix Ginseng and Radix Scutellariae (CRH) on learning and memory ability in multi-infarct dementia (MID) model rats and to explore the mechanism from the perspective of brain energy. Methods: We established MID rat model by thrombus injection, and learning and memory ability of MID rats was evaluated by Morris water maze; HE staining was used to observe the pathological changes in hippocampal CA1 region of rats; the contents of ATP, ADP and AMP in brain tissues were determined by highperformance liquid chromatography; flow cytometry was used to detect the changes of mitochondrial swelling and mitochondrial membrane potential in hippocampus of rats. Results: Compared with the rats in model group, the escape latency was significantly shortened in both high and low dose groups (P < 0.05) . The times of crossing the platform were increased in both high and low dose groups (P < 0.05) . The disorder of cell arrangement and loss of number of cell layers were significantly improved in high and low dose groups (P < 0.05) . The degree of mitochondria swelling was significantly reduced in low dose group (P < 0.05) . The decreasing trend of mitochondrial membrane potential was improved (P <0.05), and the energy charge of brain tissue was increased in high dose group rats (P < 0.05) . Conclusion: CRH improved the cognitive decline and increased the concentration of energy charge in MID rats which was in relation to protect mitochondrial damage and improve brain energy, and further protect neurons.

Objective:To establish an HPLC method fro the simultaneous determination of baicalin,wogonin,baicalein and wog-nin. Methods:A DiamonsilTMC18(150 mm×4.6 mm,5 μm) column was used. The mobile phase was acetonitrile-0.1% H3PO4 with gradient elution and the detection wavelength was 278 nm.The flow rate was 1.0 ml·min-1,the injection volume was 5μl and the column temperature was 30℃. Results:The 4 flavonoids in wine-processed Radix Scutellariae showed good separation under the chro-matographic conditions with a good linear relationship within the corresponding linear range. The average recovery of each chemical component was 97.71%-98.34%,and the content of baicalin was with RSD<2.0%. The samples from 13 habitats accorded with the requirement of processed Radix Scutellariae in Chinese Pharmacopoeia. Conclusion: Wine can maintain flavonoid glycosides and de-stroy the self-degradation of Scutellaria baicalensis enzyme to the greatest degree,which not only ensures the clinical effects of wine-pro-cessed Scutellaria baicalensis,but also is convenient to the storage of Scutellaria baicalensis Georgi,however,wine can reduce the con-tent of flavonoid glycosides in Radix Scutellariae to varying degrees,so the processing technology needs to be improved further.

Xiaochaihu decoction is a classic prescription of traditional Chinese medicine. Modern research has proved its anti-depression effect. However, its pharmacological mechanism for anti-depression effect is difficult to be unveiled because of the complexity of compound Chinese medicines. Bupleuri Radix and Scutellariae Radix is the core drug pair of Xiaochaihu decoction. In this research, Bupleuri Radix and Scutellariae Radix were analyzed by the integrative pharmacology platform to study its molecular mechanism for anti-depression. One hundred and sixteen active ingredients were predicted, 62 for Bupleuri Radix, mainly including saikosaponins, acids, alcohols, and 54 for Scutellariae Radix, mainly including flavonoids and glycosides. Its anti-depression effect was relevant to 118 core targets, including 22 known disease targets, such as serotonin receptor(HTR2C), activating transcription factor(ATF1, ATF2), δ opioid receptor(OPRD1), μ opioid receptor (OPRM1), κ opioid receptor(OPRK1), inositol monophosphatase(IMPA1), Toll-like receptor 4 (TLR4), histamine H1 receptor(HRH1), neurotrophic factor tyrosine kinase receptor1 (NTRK1), Glycogen synthetase kinase 3β(GSK3β), etc. The antidepressant effect involved positive regulation of transcription from RNA polymerase Ⅱ promoter, transcription factor binding, cytosol, transcriptional regulation of DNA template, enzyme binding, endocrine system, nervous system, neurotrophin signaling pathway, cell growth and death, signal transduction, thyroid hormone signaling pathway and other related biological processes and metabolic pathways. This study provides a scientific evidence for further study of the anti-depression mechanism of this drug pair.

Objective:To compare the clinical effects of gargle fluid containing scutellaria baicalensis with chlorhexidine in the treatment of chronic gingivitis and to observe TNF-α expression in gingival crevicular fluid(GCF) before and after treatment.Methods:90 cases of chronic gingivitis were divided into 3 groups (n =10) based on random number table and treated by scaling and mouth wash with normal saline(group A),chlorhexidine(group B) and scutellaria baicalensis(group C) respectively.PI and GI were measured,TNF-α level in GCF was examined after initial therapy,1 week,2 week and 1 month after treatment.Results:Comprehensive analysis of the whole treatment process showed that there was difference of PI among 3 groups(F =3.506,P =0.034).PI in group B was lower than that in group A and C (P < 0.05;A vs C,P > 0.05);there was difference of GI among the 3 groups (F =6.359,P =0.003).GI in group A was higher than that in group B and C(P <0.05;B vs C,P >0.05),there was difference of TNF-α concentration in GCF among the 3 groups(F =3.780,P =0.027);TNF-α in group A was higher than that in group B and C(P <0.05,B vs C,P >0.05).Conclusion:Radix scutellariae and chlorhexidine gargle fluid are equally effective in the treatment of chronic gigivitis.

OBJECTIVE: To explore the bacteriostasis and plasmid elimination activities of different extracted parts of traditional Chinese medicine Radix Scutellariae Baicalensis on NDM-1 Acinetobacter calcoaceticus. METHODS: Thein vitro antibacterial effect of the extracts from Radix Scutellariae Baicalensis was studied. Inhibition zone and minimum inhibitory concentration(MIC) of the alcohol extract and water decoction were examined by using MH agar plates and microdilution susceptibility testing. The growth curve of the NDM-1 Acinetobacter calcoaceticus was tested after being incubated with alcohol extract and water decoction at sub-MIC. At three time points after incubation with different extracts at sub-MIC, photocopy dish method was used to screen plasmid-cured strains of NDM-1 Acinetobacter calcoaceticus. The plasmid-elimination rates and phenotypic changes were compared. RESULTS: Both the alcohol extract and water decoction of Radix Scutellariae Baicalensis inhibited the growth of NDM-1 Acinetobacter calcoaceticus. The MICs were 1.56 mg·mL-1 for the alcohol extract and 6.25 mg·mL-1 for the water decoction. The growth curve showed that the antibacterial effect of the alcohol extract was more obvious. Both the alcohol extract and water decoction of Radix Scutellariae Baicalensis had some degrees of plasmid elimination effect. The plasmid-elimination rates in the alcohol extract group were higher than those in the water decoction group. The plasmid-elimination rates were 61.27% for the alcohol extract and 49.78% for the water decoction, respectively. CONCLUSION: Radix Scutellariae Baicalensis can inhibit the growth of NDM-1Acinetobacter calcoaceticus and eliminate the drug-resistant plasmid effectively and has the potential to be used to control the spread of pan-drug resistant Acinetobacter strains or be an adjuvant treatment method for clinical infections. Its alcohol extract has better effect.

Results In the model correlation coefficient(R2)was 0.9702,root-mean-square error of calibration(RMSEC)was 0.555,and root-mean-square error of predict(RMSEP)was 1.05. Conclusion In this paper,rapid quality analysis method of Radix Scutellariae granules from different manufacturers is studied using NIRS technology. The model can predict the content of baicalin nondestructively and rapidly. It can give some reference for researching the quality control of Radix Scutellariae granules .

Objective]Observe clinical efficacy of herpes zoster treated by the compound radix scutellariae solution wet application. [Method]50 patients were randomly divided into 25 cases of the compound radix scutellariae group and 25 cases of the nitrofurazone group, respectively given wet application of the compound radix scute-llariae solution and nitrofurazone solution, at the same time given intravenousganciclovir 0.3g/time,1 time/d, oral vitamin B1 20mg/time, 3 times/d, oral cobalt amine 0.5mg/time, 3 times/d, one week as a period. Record all patients' pain, skin lesion scores before the treatment and in the course of the third, fifth, seventh day and record the time of herpes stopping hair, blister drying up, blister beginning to scab and the scabby area not less than 50%.[Result] Pain, skin lesion scores of the compound radix scutellariae group were signifycantly lower than those of nitrofurazone group, the time of herpes stopping hair, blister drying up, blister beginning to scab and the scabby area not less than 50% of the compound radix scutellariae group compared with nitrofurazone group in advance, the difference was statistically significant(P<0.05). [Conclusion]Treatment of the compound radix scutellariae solution to herpes zoster had distinct clinical efficacy, having significant advantages in terms of alleviating pain and promoting skin lesions subsided.

[Abstratct] Objective To investigate the preventive effect of atomization inhalation with mixture of baikal skullcap root and light yellow sophora root for proventing invasive fungal infection on lower respiratory tract infection after chemotherapy. Methods A total of 60 cases of patients with lower respiratory tract infections after chemotherapy were selected and randomized into control groups and treatment groups, there were 30 cases in every group, anti-infection was taken in two groups according to drug sensitivity test,atomamdation inhalation with mixture of baikal skullcap root and light yellow sophora root was taken in treatment groups in addition, clinical effect was contrasted, incidence rate and time of fungal infection were contrasted between control group and treatment group,and index of blood gas analysis was contrasted between control group and treatment group after two weeks. Results Clinical efficiency was 86.67% in treatment group and was 70.00% in control group,clinical efficiency was higher in treatment groups than in control group,incidence rate of fungal infection was 6.67% in treatment group and was 23.33% in control group,incidence rate of fungal infection was lower in treatment group than in control goup,time of fungal infection was (11.58±1.31)days in treatment group and was (9.41±1.10)days in control group,time of fungal infect was later in treatment group than that in control group. There were no significant differences of the levels of pH,SaO2,PaCO2 and PaO2 between the two groups before treatment, the levels of pH,SaO2,and PaO2 were higher and PaCO2 was lower in treatment group than that in control goup after treatment. Conclusion Atomization inhalation with mixture of baikal skullcap root and light yellow sophora root could improve clinical curative effect of lower respiratory tract infection after chemotherapy,and im-prove respiratory function,provent invasive fungal infections and has good clinical effect.

Objective To explore the rules of the compatible application of antagonistic combination Ginger and Radix Scutellariae in clinical.Methods The prescriptions included the compatibility application of Ginger and Radix Scutellariae were systematically retrieved in literature. Using Excellas a storage and analysis tool, the information about treatments, dosage, compatibility proportion, and dosage form were analyzed with the method of bibliometrics.Results The prescriptions included the compatibility application of “Ginger and Radix Scutellariae” were mainly used for the treatment of internal diseases (746 cases, 76%), followed by gynecological diseases (62 cases, 6%), ear-nose-throat diseases (43 cases, 4%), etc. The largest number of internal diseases was spleen-stomach diseases, which accounted for 26% (193 cases) of the total internal diseases. And the others were diseases of liver-gallbladder 189 cases (25%), and diseases of lung (185 cases, 25%). In the prescriptions, the dosages of Radix Scutellariae were primarily 10, 12, 9, 15 g, 69% (532/772) of which were in the dose range provided by Chinese Pharmacopoeia. The dosages of Ginger were mainly 10 g and 6 g, 84% (356/424) of which were in the dose range provided by Chinese Pharmacopoeia. The compatibility proportion of Ginger and Radix Scutellariae was mostly 1∶1, which accounted for 32%.Conclusion The antagonistic combination Ginger and Radix Scutellariae was widely used in a variety of interior, exterior, gynaecology, paediatrics and other syndromes. The two drugs were contrary in the property (one cold and the other hot) and the function (one warming lung and the other clearing away the lung-heat), but their compatibility application can achieve a satisfactory efficacy.

Objective:To optimize the extraction technology of radix scutellariae. Methods: The extraction of radix scutellariae was scanned by ultraviolet spectrophotometry from 200 to 400nm. The content of baicalin was determined by HPLC. The ultraviolet ab-sorption, baicalin content and extraction rate were used as the indices, and the optimal extraction conditions were investigated by single factor experiments and orthogonal design tests. Results: The optimal extraction conditions were as follows: the ethanol concentration was 60%, the solid-liquid ratio was 1∶40, ultrasound extraction time and temperature was 40 min and 60℃, respectively. Conclusion:The extraction of radix scutellariae has good sunscreen with promising ultraviolet absorption in UVB. Ultrasound extraction has high ex-traction yield with short time, which can be used to extract sun-screening constituents from radix scutellariae.

Objective To obtain the optimal extraction process of Prunella vulgaris L.Methods With the dependent variables of ethanol concentration,reflux time and solvent fold,and dependent variable of the amount of baicalin,Central Composite Design/Response Surface Method was applied to optimize the reflux extraction process.Predictions were carried out by comparing the observed and predicted values.Results The optimum conditions of extraction process were 45% ethanol,2 hours for reflux,4 fold sovent.Regression coefficients of binomial fitting complex model was 0.9759.Bias between observed values and predicted values was-3.86%.Conclusion The optimum model is simple,precise and highly predictive.

Objective To establish the quality standard for Kangganmao Granule.Methods Radix scutellariae,radix et rhizome glycyrrhizae and radix platycodonis in the prescription were identified by TLC.The content of the baicalin was determined by HPLC.The column of AichromBond-AQ-C18(4.6 mm? 150 mm,5 ?m)was used.The mobile phase was CH3OH-H2O-H3PO4(43:57:0.2),with the flow of 1.0 mL/min,the column temperature of 30 ℃,and peaks were detected at 280 nm.Results The characteristic of identification by TLC was distinct and highly specific.The linear range of baicalin was 0.049 6~0.297 6 ?g,r =0.999 7,the average recovery was 101.80%,RSD=1.86%(n=6).Conclusion The method was easy to operate with accurate result and good reproducibility.It can be used effectively for the quality control of the preparation.

OBJECTIVE:To evaluate the effect of single decoction vs.mingled decoction on the content of Baicalin in Radix Scutellariae.METHOD:The separation of samples was performed on Agilent C18 column(250 mm?4.6 mm,5 ?m)with a column at room temperature.The mobile phase consisted of 0.5% formic acid-methano and 0.5% formic acid solution(55∶45)with flow rate of 1.0 mL?min-1 and detection wavelength of 274 nm.RESULTS:The linear range of Baicalin was 0.258 5~2.326 5 ?g(r=0.999 9)with an average recovery of 97.8%(RSD=1.1%,n=9).CONCLUSION:The content of Baicalin in Radix Scutellariae decocted alone is higher than in the mixture of all the single decoctions,and significantly higher than in Radix Scutellariae decocted by mingled decoction,all showing significant differences.

Objective To establish a TLC method for the identification of herbal medicines in Fuke Zhixue Granules.Methods Fructus Gardeniae,Radix Scutellariae,Flos Sophorae,Radix Angelicae Sinensis and Cortex Moutan were identified by TLC.Results Under the selected condition,clear and distinguishable spots were detectable and the separating result was satisfactory.Conclusion The methods are proved to be simple and specialized with a good reproductivity and can be used as one of the quality control for the preparation.

Objective To establish the standard for the quality control of Compound Huangqin Tablets. Methods The identification of Radix Scutellariae,Rhizoma Polygoni Cuspidati and Caulis Mahoniae in compound Huangqin tablets were carried out by TLC.The content of baicalin in the preparation was determined by HPLC.Chromatocolumn was adopted,methanol-0.4 %H3PO4(50 ∶50) used as the mobile phase,flowrate 1.4mL/min,column temperature at 40 ℃and detection wavelength 278 nm . Results The TLC spots were highly clear without the interference of negative control and were reproductive. The calibration curve for baicalin was linear (r=0.9996) in the range of 0.12~0.72 ?g and the mean recovery was 97.17 %and RSD=1.78 %(n=5). Conclusion This method can be used for the quality control of Compound Huangqin Tablets.

The content of scutellarin in Radix Scutellariae and Shuanghuanglian Injection was determined with dual wavelength TLC scanner. This method can be used as a rapid quantitation one in quality verification because of its simplicity involving hign sensitivity, good reproducibility and stable result.
