RESUMO

Objective:To explore the establishment of an animal model of abdominal aortic aneurysm(AAA),and elaborate the role of iNOS inhibitor in the smooth muscle apoptosis of abdominal aortio aneurysm in rats,to find a new theoretical basis for the clinical treatment of small drugs AAA.Methods:SD rats underwent intra-aortic elastase (25U/mL) perfusion to induce AAAs,the positive control group (30) and experiment group (30) use elastase perfusion while the negative control group(30) gives the saline perfusion.After operation the positive and negative control groups were treated with intraperitoneal injections of saline,experimental group injects the iNOS inhibitor Aminoguanidi hydrochloride;Postoperative second,7,and 14 days,The NO content in the serum and specimen of abdominal aortic aneurysm was detected by iNOS Immuno histochemistry and Terminal Transferase-mediated dUTP Nick End-Labeling (TUNEL) to evaluate distribution of smooth muscle apoptosis in abdominal aortic aneurysm.Results:Underwent intra-aortic elastase perfusion to induce AAAs have a high-success-rate.Rate of AAA formation in positive control group 10%,60%,80%,respectively.The treatment group was 0%,10%,20%,and the negative control group was not formed.The treatment group and the negative control group were lower than the positive control group,there were significant differences.In the positive control group,NO content increased gradually from second days,7 days to reach the peak and maintained at a higher level,the treatment group serum NO content was lower than the other two groups,there was significant difference (P<0.05),iNOS was strong expression in the positive control group,in the other two groups of mild expression.TUNEL results showed that a lot of apoptotic cells in the positive control group,after 7 days showed a significant increase trend,to observe the end (2 weeks) gradually increased.,The positive control group was higher than the negative control group and the negative control group,there was significant difference(P<0.05).Conclusion:iNOS inhibitors significantly decreased the content of NO in serum,reduced the apoptosis of smooth muscle cells,and inhibited the formation of abdominal aortic aneurysm.To provide theoretical basis for clinical application of iNOS inhibitors in the treatment and control of AAA.

RESUMO

@#The factors affecting cerebral ischemia model of rat established by middle cerebral artery occlusion include animal selection, drugs of anesthesia, surgical site, thread affect building, and etc. To find and manufacture an appropriate thread is the key to success. It includes the species, length, diameter, hardness, and placing method of the thread. The improvement of these factors greatly increases the successful rate in the establishment of focal cerebra ischemia model of rat caused by middle cerebral artery occlusion.

@#Objective To establish animal model of prolactinoma with Fischer 344 (F344) rats.MethodsFemale F344 rats were injected with estradiol benzoate-treated and 0.9% sodium chloride respectively. After 50 days of treatment, MRI scan was performed and animal's living status was observed, and pituitary weight, serum prolactin level, pathological and PRL immunohistochemistry changes were also detected.ResultsAll estradiol benzoate-treated rats formed the prolactinoma after treatment for 50 days and the growth of body weight was retarded obviously. Pituitary weight and serum PRL increased significantly in estradiol benzoate-treated animals compared with those treated with 0.9% sodium chloride. Pituitary adenomas were found in MRI scan and HE stain was positive. The forming rate of tumor was 100%.ConclusionThe animal model of pituitary adenoma established with F344 rat has more characters such as having a short tumor forming period, easy and stable, and coincided with features of prolactinoma.

@#Objective To establish the model of abdominal heterotopic heart transplantation and grope steady operation methods.Methods The heart transplantation was performed in the control group(group A) and experiment group(group B) with 30 rats in each group.Another 30 animals not performed the operation were as the blank group(group C).During the procurement of the donor heart saline solution containing heparitin was injected into the inferior vena cava of liver.At the same time abdominal aorta was transected.Then heart was perfused until it stopped.The activity of superoxide dismutase(SOD) and content of malondialdehyde(MDA) in blood was measured after transplantation.Results The total perfusion time of the graft was shortened within 2 min and hot ischemia time was shortened within 25 s in the group B.The activity of SOD and content of MDA between two groups was significantly different(P<0.05).Conclusion The perfusing method improved with new technique is quicker and more effective than the traditional one and results in the successful rate improvement.

Objective:Study on establishing HA animal model,which isoperated simply and have short model building period and high successful rate,to provide favorable test materials for HA-related scientific research.Methods:10 SD rats were randomly selected to set up the model by using APH intraperitoneal injection,meanwhile rats intraperitoneally infused with saline served as controls group.After 8 days of experimental treatment,we evaluated the animal model by using blood analysis,reticular count,myelogram and other laboratory experimental results.Results:On eighth day after experimental treat,the results of RBC、HGB、PLT、Ret% in Rats'peripheral blood are respectively(2.98?0.623)?1012/L、(100?14.2)g/L、(320?358.8)?109/L、90.7%?4.23%,the concentration of indirect bilirubin and free-haemoglobin in plasma are respectively(10.3?3.30)?mol/L、(1 248?282.1)mg/L.Compared to the control group,the difference of the results all have the statistical significance;bone marrow exhibited that karyocytes proliferate obviously,mainly the erythroid system.Conclusion:APH intraperitoneal injection can set up HA animal model shortly and conveniently,and the animal model has the clinical features.So the method of APH intraperitoneal injection is an ideal way to establish HA animal model

Objective To study the effects of malondialdehyde (MDA), superoxide dismutase (SOD) and tumor necrosis factor-? (TNF-?) on brain tissue in rats with pancreatic encephalopathy (PE). MethodsThirty-six Wistar rats were randomly divided into control group (n=6) and PE model group (n=30). In control group, rats were injected with normal saline by internal carotid artery (0.1 ml/100 g) and were killed on the first day after the injection. In PE model group, rats were injected with phospholipases A2 (0.1 ml/100 g, 1 000 U/0.1 ml) by internal carotid artery, to establish animal model of PE in rat and 10 rats were killed on day 1, 3, 7 respectively after the injection. The changes of water content in the brain were measured. Leucocytes aggregation and margination in the microvessels, and the changes of cerebral cells and nerve fibers were observed. The levels of MDA, TNF-? and the activity of SOD were tested in the brain homogenate in rats. ResultsIn PE model group, water contents of brain increased; The phenomena of leucocytes accumulation and margination, cellular edema of neurons and demyelination of nerve fibers became more obvious; The levels of MDA and TNF-? increased significantly than those in the control group, while the activity of SOD reduced (P
