Observation on the effect of Jinqiao-Reduqing granule for hte children with herpetic angina fever / 国际中医中药杂志

Objective To evaluate the effect of Jinqiao-Reduqing granule in the treatment of children with herpes angina fever.Methods A total of 90 children with herpes angina were randomly divided into two groups,with 45 cases in each group.Both groups received the symptom-supported treatment.On this basis,the control group took ribavirin granules,while the observation group took Jinqiao-Reduqing granule.Both groups were treated for 4 days.The symptoms and signs were scored before and after treatment.The fever abatement time,herpes subsidence time,salivation disappearance time and appetite recovery time were observed and recorded.The serum levels of IL-2,IL-6,IL-10 and TNF-α were detected by ELISA,and peripheral blood levels of CD4+,CD8+,CD4+/CD8+ were detected by flow cytometry.Results After treatment,the scores of symptoms and signs in the observation group were significantly lower than those in the control group (3.02 ± 2.89 vs.6.88 ± 2.31;t=6.999,P＜0.01).The time of fever abatement (16.8 ± 3.2 h vs.29.9 ± 4.4 h,t=16.202),herpes subsidence (72.3 ± 5.4 h vs.96.7 ± 6.2 h,t=19.765),salivation disappearance (26.4 ± 2.0 h vs.46.3 ± 3.6 h,t=32.479) and appetite recovery (19.6 ± 4.2 h vs.39.7 ± 4.9 h,t=20.907) were significantly earlier than those in the control group (P＜0.01).After treatment,the serum levels of IL-2,IL-6,IL-10 and TNF-α in the observation group were lower than those in the control group (t were 4.974,4.410,6.447 and 4.767,respectively,all Ps＜0.01);CD4+ levels and CD4+/CD8+ were significantly higher than those in the control group (t were 4.033,6.860,all Ps＜ 0.01),and CD8+ levels was significantly lower than those in the control group (t=4.928,P＜0.01).Conclusions The Jinqiao-Reduqing granule can shorten the time of symptoms subsidence,reduce the level of inflammatory cytokines and improve the immunity of children with herpes angina fever.

Improvement of Quality Standard of Reduqing Oral Liquid / 中国中医药信息杂志

Objective To improve the quality standard of Reduqing Oral Liquid. Methods TLC was used to identify the featured spots of Glycyrrhizae Radix et Rhizoma, Belamcandae Rhizoma and Scutellariae Radix;HPLC method was usded to detect the contents of Baicalin in Reduqing Oral Liquid. Results TLC could identify the featured spots of Glycyrrhizae Radix et Rhizoma, Belamcandae Rhizoma and Scutellariae Radix; Baicalin was detected in range of 53.60–536.00 μg/mL with good linear relationship (r=0.999 99), the average recovery rate was 99.85%, and RSD was 0.63%(n=6). Conclusion The method is simple, accurate and reliable, which can be applied to the quality control of Reduqing Oral Liquid.

Optimization of ultrahigh pressure extraction process of chlorogenic acid and forsythiaside in Jinqiao Reduqing granules by Box-Behnken design-response surface methodology / 中国药学杂志

OBJECTIVE: To optimize the ultrahigh pressure extraction (UPE) process of effective constituents from Jinqiao Reduqing granules by Box-Behnken design-response surface methodology. METHODS: On the basis of single factor screening, a three-factor and three-level Box-Behnken experimental design was employed, with solvent ratio (X1), extraction pressure (X2) and extraction time(X3) as independent variables. Dependent variables were transformed into desirabilities mathematically by Hassan's method. Data of overall desirabilities were fitted to a second order polynomial equation, through which three dimensional response surface graphs were produced. Optimum experimental conditions were selected from the stationary point of the response surfaces. RESULTS: Regression coefficient of binomial fitting complex model was as high as 0.9878. The optimum conditions of UPE were as follows: the materials were extracted for 38 s with 24 times amount of water, the extraction pressure was 112.1 MPa, and the extraction was performed for one tine. The bias between observed and predicted values was 5.27%. CONCLUSION: As a novel extraction technology for Chinese herbal medicine, the UPE procedure has higher extraction yield, lower extracting temperature, shorter extracting time and less power consumption. The UPE has provided a brand new method for extraction of chlorogenic acid and forsythiaside from Jinqiao Reduqing granules.

Experimental and clinical study on inhibitory effects of Reduqing on human cytomegalovirus / 中国新药与临床杂志

AIM:To study the therapeutic effect of traditional Chinese medicine Reduqing on human cytomegalovirus (HCMV). METHODS:The expression of the HCMV late-mRNA in infected cells was measured by quantitative reverse transcription-polymerase chain reaction(RT-PCR) and the development of cytopathic effect (CPE) caused by HCMV was observed before and after treatment with Reduqing. On the basis of its anti-HCMV activity study in vitro, Reduqing was applied to the child-bearing aged women and pregnant women who were HCMV-IgM positive which indicated the active HCMV infection. The serum levels of tumor necrosis factor-alpha (TNF-α) and interleukin-2 (IL-2) before and after treatment were measured by the ELISA method. RESULTS:The results showed that the Reduqing had a strong inhibitory effect on both HCMV-mRNA and CPE. The level of the TNF-α increased while IL-2 decreased significantly in HCMV-IgM positive serum when compared with HCMV-IgM negative cases. In HCMV-IgM positive patients, after treatment with Reduqing, the serum levels of TNF-α and IL-2 returned to normal, and 73 % of them turned to be HCMV-IgM negative, which was significantly higher than that (14 %) in control group. CONCLUSION:It is suggested that serum TNF-α and IL-2 level are closely related to the activity of HCMV. Reduqing can enhance the immunity of the body and has an obvious inhibition on HCMV in vitro and in vivo, and can thus serve as an effective medicine for active HCMV infection therapy, especially for the pregnant women.
