RESUMO

Objective To explore the relevance between the quality of standard decoction and the marker component amygdalin of decoction slices of blazing Armeniacae Semen Amarum (BASA) by mathematical model. Methods The BASA standard decoction was prepared, and three linear regression models of the amygdalin content and dry extract rate, the content of amygdalin in standard decoction and decoction slices, and the transfer rate in standard decoction and the content of amygdalin in decoction slices were established by using Design-Expert 8.0.6 software, respectively. Results The dry extract rate of BASA was 8.97%-12.12%; The content of amygdalin in standard decoction was 21.74%-30.32%; And the transfer rate of amygdalin was 70.39%-90.54%. The R2 values of three linear regression models were all greater than 0.8. The P values of each partial regression coefficient were less than 0.05, which indicated that the three models were significant. Then the accuracy of three linear regression models was verified. The relative deviation between the predicted and measured values was less than 10%, and the average relative deviation was not greater than 5%. Conclusion The models established in this study could predict the quality of standard decoction prepared from different BASA, and provide a certain reference value for the establishment of standard decoction quality standard.

RESUMO

Objective To study effects of bitter almond on interstitial cells of Cajal (ICC) in old rats with slow-transit constipation (STC) and analyze mechanism of the gut purge of bitter almond.Methods Forty-five old SD rats were randomly divided into blank control group,model control group,bitter almond-treated group randomly (n=15,each).Compound phenanthroline was used to build the model of STC model.The bitter almond-treated group was given with bitter almond apozem,the other two groups were given with the same amount of saline.After 10 days,the carbon powder propulsion rates in the intestine were measured and specimens were taken.The ICC changes in terms of the number and distribution were observed by immunohistochemical method.The protein and mRNA expressions of c-kit in ICC were measured by Western blot and reverse transcription-polymerase chain reaction(RT-PCR).Results Compared with blank control group [(68.6 ± 6.0) %,the carbon powder propulsion rates in the intestine in model control group[(47.7±± 1.7)%]were declined (P< 0.01).The areas of c-kit positive cell in colon were declined[(638.0 ± 23.5) μm2 vs.(723.7 ± 30.8)μm2]in model vs.blank control (P<0.05).The mRNA expression of c-kit gene in ICC was declined [(0.3±0.1) vs.(1.0±0.1)] (P<0.05),the expression of c-kit protein in ICC were declined[(0.2±0.2) vs.(0.5±0.2)] (P<0.05).Compared with model control group[(47.7± 1.7)%],the carbon powder propulsion rates in the intestine in bitter almond group were increased[(63.4± 3.3)% vs.] (P <0.01).The mRNA expression of c-kit gene in ICC were increased in bitter almond group (0.9 ±0.2) vs model control group[[(0.3±0.1)] (P< 0.01).The expression of c-kit protein in ICC were increased in bitter almond group[(0.5±0.3) vs.in model control 0.2±0.2)] (P<0.01).Conclusions The bitter almond can promote the expression of c-kit mRNA and protein in colonic tissue of old STC rat interstitial and repair a fraction of ICC of STC rat interstitial,which promote intestinal peristalsis in slow-transit constipation rats.

RESUMO

Objective To observe the effects of storage condition on the quality of Semen Armeniacae Amarum. Methods Orthogonal design was adopted. The storage condition was optimized with temperature,humidity and storage time as the observation factors,with the content of amygdalin,and the content,acid value and peroxide value of fatty oil as the indexes.The content of fatty oil,acid value and peroxide value were measured according to the methods provided by 2005 edition of Chinese Pharmacopoeia. The content of amygdalin was measured by HPLC. Results Temperature was the main influencing factor of amygdalin content,then came humidity and storage time. Temperature was also the main influencing factor of the concent and the acid value of fatty oil,and the humidity was the second. For peroxide value,humidity was the main factor,then came temperature and storage time. Conclusion The optimal conditions of storing Semen Armeniacae Amarum as follows:storage in dry place with the temperature of 2~ 8 ℃,and in short storage time.

RESUMO

The content determination of amygdalin in seed coat of Semen Armeniacae Amarum and acute toxic test have been performed. Some issues such as, the effect of removing coat processs on amygdalin content and of seed coat on decoction rate, etc. were studied in experiments. It was suggested not to remove coat after it is processed and to have to be cut into piece before it is used.
