RESUMO

Objective To evaluate the incidence of four aberrant behaviors in patients with pervasive developmental disorders (PDDs).Methods The abnormal behaviors during the last month in 138 PDDs patients were assessed with Aberrant Behavior Checklist and Conners Index of Hyperactivity.Results The incidence of aberrant behaviors was 61.6% for hyperactivity,81.9% for stereotyped behaviors,13.77% for self-injury,and 58.70% for irritability in PDDs patients.The occurance of irritability in autism patients was higher than that of asperger syndrome (AS) patients (x2 =5.623,P=0.018).Conclusion Stereotyped behaviors,hyperactivity and irritability are common in children with PDDs.Autism patients are more likely to exhibit irritability behaviors than AS patients.

RESUMO

Objective To investigate the effect of vanillin via olfactory pathway on repetitive stereotyped behavior in autistic rats and its possible mechanism.Methods The model of autism was established by the single intraperitoneal injection of 600 mg/kg sodium valproate on day 12.5 of pregnant rats.Twenty-four autism animals were adopted according to the eye opening time,body mass and behavioral test.They were randomly divided into control group and vanillin group with 12 in each group,while the saline group rats as normal control group and the blank group rats as normal blank group.The repetitive stereotyped behavioral were examined in four groups of rats and the concentration of 5-HT in the amygdala was determined by immunohistochemical stainning.Results Compared with the control group,the repetitive stereotyped behaviors of vanillin group decreased (stereotype time (113.31 ± 18.45) s vs (250.52± 13.25) s,P<0.01) and the expression of 5-HT in amygdala increased (5-HT positive neuron number(3.42± 1.32) vs (1.32± 1.04),P<0.05).Conclusions Vanillin via olfactory pathway can improve repetitive stereotyped behaviors of autism rats,which is associated with increased the expression of 5-HT in amygdala.

@#ObjectiveTo observe the effects of Jintong capsule on model rats of Tourette syndrome (TS) and explore its probable pharmacological mechanisms.MethodsSD rats were randomly divided into six groups: blank control, TS model, haloperidol and three Jintong capsule treated groups. Model rats were copied by intraperitoneal injection of iminodipropionitrile (IDPN). The stereotyped behaviors of model rats were recorded. Open field test was used to detect ability of space recognition of rats, high performance liquid chromatography was used to detect content of monoamines, and flow cytometry was used to detect the ratio of T lymphocyte.ResultsJintong capsule can ameliorate the stereotyped behaviors of model rats, decrease content of dopamine in striatum and increase the ratio of CD4/CD8.ConclusionJintong capsule can improve behaviors of model rats. The potential mechanism of Jintong capsule maybe: it can affect the dopaminergic system of model rats, and Jintong can enhance the immune system of model rats.

OBJECTIVE: This study was to investigate in vivo antidopaminergic activities of polygalasaponins which included in the roots of Polygalae tenuifolia, one of the natural plants prescribed to treat psychotic disorders in traditional medicine. METHODS: The chemical separations were conducted by chromatographies with various columns and the chemical structures were identified using spectroscopy, NMR and so on. The apomorphine-induced stereotyped behaviors and climbing behaviors in mice as animal models of psychotic disorders were applied to explore the antidopaminergic activity. RESULTS: The active compounds with antidopaminergic effects in the roots of Polygalae tenuifolia were identified as polygalasaponin A-D. The polygalasaponins showed to suppress the apomorphine-induced stereotyped behaviors and climbing behaviors in dose-related fashions. CONCLUSION: This study suggests that polygalasaponins might have antipsychotic activities. The natural products need to be explored extensively as sources of new medications in psychiatric fields. Further explorations for chemical and pharmacological properties of polygalasaponins are required.

OBJECTIVES: This study was to investigate in vivo activities of Polygalae Radix, a natural plant including active components having affinities to central nervous system receptors, using an animal model for psychotic disorders in terms of behavioral psychopharmacology. METHODS: The extracts of Polygalae Radix in two concentrations of 5.0mg/g and 0..5mg/g were administered to mice in order to explore the suppression of apomorphine-induced stereotyped behaviors visually and to compare with the effects of haloperidol and control. 10 mice were tested in each groups. RESULTS: The capacities of suppressing the apomorphine-induced stereotyped behaviors were observed in decreasing order of haloperidol, Polygalae Radix 5.0mg/g, Polygalae Radix 0..5mg/g and control. The extracts of Polygalae Radix in both concentrations suppressed apomorphine- induced stereotyped behaviors significantly, and Polygalae Radix 5.0mg/g was as potent as haloperidol. CONCLUSION: This suggests that Polygalae Radix contain some effective ingredients to influence the central dopamine pathway in vivo. The identification of the chemical structures and the exploration of the biochemical and pharmacological characteristics of active components are to be required through further studies.
