RESUMO

Aim To investigate changes in expression of steroid receptor coactivator-1 ( SRC-1) gene during the differentiation of mouse Neural Stem Cells (NSCs) in vitro. Methods Primary NSCs were obtained from cerebral cortex of neonatal mouse aged 1 ~ 3 days mechanically and enzymatically. The neurospheres were cultured in vkro, The neurosphere cells were induced to differentiate into neurons and astrocytes by serum after primary and passage culture. The cultured NSCs were identified by neurospheric morphology and Nestin immunofluorescence assay; Neurons and astrocytes were recognized respectively by specific antibodies to detect the cell types on days 3 and 9 of differentiation; The expression of SRC-1 gene mRNA and protein were detected by RT-qPCR and Western blotting on days 0, 3 and 9 of differentiation. Results Nestin-positive NSCs were observed in cultured neurospheres derived from cerebral cortex of neonatal mouse in vitro. During the process of differentiation, neurons were the main cells on day 3 and astrocytes were the, main ones on day 9. Compared with day 0, SRC-1 expression increased on day 3 and decreased on day 9 significantly. Conclusion NSCs could be isolated from cerebral cortex of neonatal mouse and cultured well in vilro. During the differentiation of NSCs, thdre were significant changes in SRC-1 expression, which was the highest in neurons, followed by NSCs and astrocytes.

Objective To investigate the expression change of RACK1 ,Src and Bcl‐2 in gastric carcinoma tissue and adjacent carcinomatous tissue .Methods Eighty specimens of gastric carcinoma and adjacent carcinomatous tissues in our hospital from Au‐gust 1 ,2011 to February 1 ,2014 were collected .The immunohistochemistry staining and Western blotting methods were adopted to detect the expression of RACK1 ,Src and Bcl‐2 protein in gastric carcinoma and adjacent carcinomatous tissues ,and their correlation was analyzed and performed the statistical analysis by combining with the clinicopathological data .Results The immunohistochem‐istry staining and Western blotting detection displayed that the expression positive rate and expression level of RACK 1 in gastric carcinoma tissue were obviously lower than those in the adjacent carcinomatous tissue ,while the expression positive rate and ex‐pression level of Src and Bcl‐2 in gastric carcinoma tissue were obviously higher than those in the adjacent carcinomatous tissue ,the differences were statistically significant (P0 .05) .Conclusion The expression of RACK1 in gastric carcinoma tissue is significantly decreased ,while the expres‐sions of Src and Bcl‐2 are increased .

Adenosine monophosphate-activated protein kinase (AMPK) activation also can inhibit the synthesis of cholesterol and fat.Recent studies have shown that activation of AMPK can also inhibit the synthesis of bile acid and promote bile salt export pump' s generation.All of those illustrated that AMPK played an important role in regulating bile acid metabolism.This article will summarize the AMPK activation pathway,bile acid metabolism,AMPK activity in bile acid synthesis and transfer,and so on.

Objective: To study the spatiotemporal expression pattern of histone acetyltransferase steroid receptor coactivator-1 (SRC1) in developing mouse heart, so as to explore the relationship of SRC1 with heart development. Methods: The normal mouse hearts were collected at E7.5-E18 and 1 day and 3 months after birth; 9 specimens were chosen for each time point to observe the spatial expression of SRC1 by immunohistochemistry technique, and 6 specimens of each time point were used to examine the temporal expression of SRC1 protein by Western blotting technique and to plot the time-dose curve. Results Immunohistochemistry showed no expression of SRC1 in the heart primordium at E7.5; only very weak SRC1 expression was found in the cardiac tube at E8. 5-E9.5. Weak SRC1 expression was found in the trabeculae after E10.5 and relatively strong and widespread expression was found in other heart regions. Western blotting results demonstrated that SRC1 protein expression at E10.5 was significantly lower than those at E11.5 and E12.5 (P0.05). SRC1 expression reached the peak at E11. 5-E12.5, and there was no significant difference between the two time points (P>0.05). SRC1 expression gradually decreased after Ell. 5-E12.5 till the adulthood, and there were no significant differences in the expression after E13.5 (P>0.05). Conclusion: Widespread distribution of SRCl is present in the developing mouse heart after ES. 5, and the expression is in a dynamical spatiotemporal pattern, suggesting that SRC1 may take part in the overall regulation of the heart development, and it might has a closer relationship with the early induction of the heart septa.

Diffuse leiomyomatosis of the uterus is a rare condition that is distinguished from the uterine leiomyoma due to the diffuse involvement of the myometrium by numerous, ill-defined, smooth muscle nodules. We present here a case of diffuse uterine leiomyomatosis in a 34-year-old woman. The hysterectomy revealed a symmetrically enlarged uterus containing numerous, small, ill-defined leiomyomatous nodules. Microscopically, the nodules were composed of compact fascicles and interweaving bundles of uniform benign smooth muscle cells. On the immunohistochemical staining, the progesterone receptor level was higher in the leiomyomatosis than in the adjacent normal myometrial tissue, but the estrogen receptor level and Ki-67 labeling index were equal in both areas. At the twelve months follow-up, this patient has been doing very well with no evidence of pelvic or intraabdominal recurrence of disease.

RESUMO

Angioleiomyoma of the sinonasal area is an extremely rare benign neoplasm. To the best of our knowledge, only 26 cases have been described. Here, we report a case of angioleiomyoma arising in the nasal cavity of a 60-year-old woman. Microscopically, the tumor consisted of proliferating smooth muscle cells punctuated with thick-walled vessels with slit-like lumina. The tumor was negative for estrogen and progesterone receptor by immunohistochemical study. Further studies are needed to clarify whether the growth of this tumor is sex steroid-dependent.

RESUMO

Objective To investigate the effect of steroid receptor coactivator family in initiation, development, treatment and prognosis of breast cancer. Methods The literatures in recent years which have related to the effect of steroid receptor coactivators in breast cancer are reviewed. Results Steroid receptor coactivators are essential for several kinds of steroid hormones binding to steroid receptors, so they are important accessory factors that induce the initiation, development and recurrence of breast cancer, and predictive factors that estimate the prognosis of breast cancer. Conclusion Inhibition of the expression and signaling pathway of steroid receptor coactivators may be effective for breast cancer prevention and treatment.

The role of steroid receptor has been widely studied and well established in the field of breast cancer. In contrast of breast cancer, the clinicopathological roles of steroid receptors in benign breast diseases were not established clearly until now. Authors evaluated steroid receptor expressions of benign breast diseases using the immuno-histochemical staining and analyzed their relationship with pathologic types and the correlation between the positivity of steroid receptor expression and the proliferative activity of ductal epithelial cells. The results were follows; 1) The assayed tissue specimens were 93 cases which consisted with fibrocystic disease, 39 cases and fibroadenoma, 54 cases. 2) The positivity rate of steroid receptor expression in fibroadenoma (ER: 59.2%, PR: 75.9%) seemed to be more higher than in fibrecystic diseases (ER: 46.1%, PR: 55.5%) without a statistical significance. 3) The positivity rate of ER expression of fibrocystic diseases developed in premenopausal women (54.8%) was significantly higher than in postmenopausal women (2.5%). 4) Any significant correlation was not noted between the positivity of steroid receptor expression in the tissue and ductal epithelial proliferative activity. In conclusion, fibrocystic disease and fibroadenomas possessed a significant amount of steroid receptor positive cells in their tissues. However, the positivity of steroid receptor expression in the tissues seemed to have few correlation with the ductal epithelial cellular proliferation.

Objective To investigate the effects of deguelin on the proliferation inhibition and apoptosis induction in RPMI-8226 cells in vitro,as well as the regulation of steroid receptor coactivator-3(SRC-3)to explore the relationship between them.Methods The effect of deguelin on the growth of RPMI-8226 cells was studied by MTT assay.Apoptosis was detected through Hoechst 33258 staining and Annexin-V FITC/PI double-labeled cytometry.RT-PCR Technology was applied to assessment of the mRNA expression of SRC-3,whereas,SRC-3 protein expression and localization were determined by using immunohistochemistry method.Results Deguelin presented striking proliferation inhibition potency on RPMI-8226 cells in vitro and apoptosis induction activity in a time-and dose-dependent manner.The IC50 value for 24 h was(54.55?0.40)nmol/L,(17.04?0.73)% RPMI-8226 cells went apoptotic when treated with 25 nmol/L deguelin for 24 h,with the dose of deguelin increasing to 100 nmol/L,(63.57?1.36)% cells were apoptotic.Over-expression of SRC-3 was found in RPMI-8226 cells,whereas the mRNA and protein expression levels of SRC-3 were significantly downregulated in RPMI-8226 cells induced by deguelin in a dose-dependent manner.The disposition of SRC-3 was situated mainly at the nuclear,occasionally in the cytoplasm.Conclusion Deguelin exhibited potent proliferation inhibition to the human myeloma cell line RPMI-8226 cells,furthermore,deguelin might induce RPMI-8226 cells apoptosis in a dose-dependent manner,which might correspond to the regulation of the expression of SRC-3.

RESUMO

Objective To breed and identify steroid receptor coactivator-3 (SRC-3) knock-out mice. Methods Heterozygote mice were bred and reproduced. Wild genotype, heterozygote genotype, and homozygote genotype would appear in offsprings of parents. Genome DNA extracted from the murine tails was subjected to PCR for genotype identification. Male homozygote mice were selected to mate with the female heterozygote mice for acquiring homozygote baby mice according to Mendel law. Results Breeding and reproducing were successful and more heterozygote genotype mice were reproduced. Conclusion Appropriate methods for breeding, reproducing, and identifying are the effective way for acquiring SRC-3 knock-out mice from heterozygote mice.
