Equine piroplasmosis is the most important tick-borne disease to affect horses in Brazil. Theileria equi is one of the causative agents of equine piroplasmosis. Chronic cases are expected, in which the animals show no apparent signs of infection and remain asymptomatic but constitute a source of the infectious agent that ticks can spread. This study was conducted across 81 ranches located in the municipality of Sinop, State of Mato Grosso, Brazil. A sample calculation was performed to estimate the apparent prevalence of T. equi among horses. A total of 1,853 animals were included in the sampling analysis based on the information available from the Institute of Agricultural and Livestock Defense of Mato Grosso State. The serological analysis of 367 serum samples using an indirect enzyme-linked immunosorbent assay (ELISA) to detect anti-T. equi antibodies revealed that 337 animals were positive, representing a frequency of 90.70%. The molecular analysis to amplify the EMA-1 gene showed positivity in 20 of 89 tested samples. The fragments of four samples were sequenced and analyzed to determine their similarities to sequences from other species, based on sequences deposited at GenBank. All showed 100% similarity with T. equi. Our study represents the first report of T. equi antibodies among the equids in north-central region of Mato Grosso, revealing the widespread distribution of seropositive animals.


A piroplasmose equina é a doença transmitida por carrapatos mais importante em cavalos no Brasil. Theileria equi é um dos agentes causadores da piroplasmose equina. São esperados casos crônicos, nos quais os animais não apresentam sinais aparentes de infecção e permanecem assintomáticos, mas constituem uma fonte de infecção e disseminação por carrapatos. Este estudo foi realizado em 81 fazendas localizadas no município de Sinop, Estado de Mato Grosso, Brasil. Um cálculo amostral foi realizado para estimar a prevalência aparente de T. equi entre cavalos. No total, 1.853 animais foram incluídos na análise amostral com base nas informações disponíveis no Instituto de Defesa Agropecuária do Estado de Mato Grosso. A análise sorológica de 367 amostras de soro por meio de ensaio imunoenzimático indireto (ELISA) para detecção de anticorpos anti-T. equi revelou que 337 animais eram positivos, representando uma frequência de 90,70%. A análise molecular para o gene EMA-1 mostrou positividade em 20 das 89 amostras testadas. Os fragmentos de quatro amostras foram sequenciados e analisados para determinar suas semelhanças com sequências de outras espécies, a partir das sequências depositadas no GenBank. Todos mostraram 100% de similaridade com T. equi. Nosso estudo representa o primeiro relato de anticorpos contra T. equi entre os equídeos na região centro norte de Mato Grosso, revelando a ampla distribuição de animais soropositivos.

This study was designed to detect equine piroplasmosis using the molecular technique in Al-Najaf province during the season that showed an increment in tick activities. Blood samples were collected from 110 horses with more than two signs of piroplasmosis. After DNA extraction, the product was examined by a polymerase chain reaction to amplify 18SrRNA. The results showed that the overall percentage of equine theileriosis was 38.18%. According to gender, the percentage of infection was 43.48% and 29.27% in females and males, respectively. Significant variations appeared between infected horses according to age, and the percentage of infection was 50% and 35.22% in less than 2 years and more than 2 years age, respectively. Moreover, the percentage of infection was 62.5% and 19.35% in animals with and without acariasis, respectively. Significant variations were also seen in equine theileriosis according to geographical areas, and the higher percentage was reported in Hera district (60.87%), while the lowest percentage was in the center of Al-Najaf (21.43%). This difference may be due to different distribution of vector of disease (tick), which may be the availability of the suitable weather that helped in the multiplication of the intermediate vectors. In conclusion, this study proved the variations in the occurrences of equine piroplasmosis according to gender, age, and geographical areas.(AU)


Este estudo foi desenvolvido para detectar piroplasmose equina usando a técnica molecular na província de Al-Najaf durante o período do ano com maior ocorrência de carrapatos. Foram coletadas amostras de sangue de 110 cavalos que apresentaram mais de dois sinais de piroplasmose. Após a extração do DNA, o produto foi examinado por reação em cadeia da polimerase para amplificar o 18SrRNA. Os resultados mostraram que a porcentagem geral de teileriose equina foi de 38, 18%. De acordo com o sexo, o percentual de infecção foi 43,48% e 29,27% no sexo feminino e masculino, respectivamente. Apareceram variações significativas entre os cavalos infectados de acordo com a idade, e a porcentagem de infecção foi 50% e 35,22% em menos de 2 anos e mais de 2 anos, respectivamente. Além disso, as porcentagens de infecção foram 62, 5% e 19, 35% em animais com e sem acariasis, respectivamente. Também foram observadas variações significativas na teileriose dos equídeos, de acordo com as áreas geográficas, e o maior percentual foi relatado no distrito de Hera (60, 87%), enquanto o menor percentual foi no centro de Al-Najaf (21,43%). Essa diferença pode ser devido à distribuição diferente do vetor da doença (carrapato), que pode ser a disponibilidade do clima adequado que ajuda na multiplicação dos vetores intermediários. Em conclusão, este estudo provou as variações nas ocorrências de piroplasmose eqüina de acordo com sexo, idade e áreas geográficas.(AU)

The present study was done with the objective of comparative efficacy of arteether and buparvaquone against theileriosis in cattle. Total 67 cattle suspected for theileriosis were screened on the basis of clinical and blood smear examination. Group I (n=6) was treated with Inj. buparvaquone @ 2.5 mg/kg body weight once and Group II (n=6) was treated with Inj. arteether @5 mg/kg body weight intramuscularly for three consecutive days. Rectal temperature, heart rate and respiration rate were significantly increased before treatment and in both the groups after treatment showed significant improvement. Haematological parameters showed significant decreased in Hb, PCV, TEC, TLC and neutrophil and significant increase in lymphocytes, monocytes and eosinophil. Non significant difference in basophil count was observed. After treatment, significant improvement was observed in mean hemoglobin, PCV, TEC, TLC, neutrophil, lymphocyte and eosinophil. Non significant improvement was observed in monocyte and basophil count. Present study revealed percent efficacy of arteether was 66.66% and buparvaquone 100%

Bovine theileriosis, a tick borne haemoprotozoan disease, caused by, Theileria annulata and Theileria orientalis and responsible for heavy economical losses in dairy industry. Diagnosis of theileriosis is mainly based on clinical signs and conventional techniques. Polymerase chain reaction using whole blood is a modern method of diagnosis of theileriosis as it can detect infection even in low parasitemia but only concern is that its very time consuming and chances of loss of DNA is more. Hence, to overcome this, the present study reports the diagnosis of T. annulata and T. orientalis by comparing Whole blood PCR and FTA card PCR whereas a comparison between blood smear examination, whole blood PCR and FTA card PCR was done to determine the sensitivity of the techniques. The study was conducted on 50 blood samples of cattle having typical clinical signs and blood smear after staining with Giemsa’s stain were examined for presence of Theileria spp. in R.B.C. The primer sequences were used as per Kolte et al and Kakati et al. The assay employs primers specific for the gene encoding the 30-kDa major merozoite surface antigen of T. annulata and Major merozoite surface protein gene of T. orientalis. Out of total 50 blood samples, 16 were positive for Theileria sp. by blood smear examination, 19 were positive for T. annulata by Whole blood PCR and FTA card PCR whereas for T. orientalis, 26 were positive by whole blood and 27 samples by FTA card PCR. So the diagnosis of theileriosis by using FTA card PCR is found to be sensitive and less time consuming method as compared to Whole blood PCR.

Equine piroplasmosis is a tick-borne disease caused by the protozoan parasites Babesia caballi and Theileria equi. We report a case of abortion in association with transplacental Theileria equi infection in a mare from the State of Espírito Santo, southeast Brazil. An apparently healthy mare aborted at the tenth month of gestation. At necropsy, the subcutaneous tissue, skeletal muscles, and visible mucosae of the aborted fetus were pale, and there was moderate hydrothorax and marked splenomegaly. Microscopic findings included splenic lymphoid hyperplasia and nephrosis. Merozoite-infected erythrocytes were found within blood vessels of all organs examined and were most numerous in the brain. DNA extracted from the spleen, liver, kidney, and thymus was used as a template for PCR. Generic primers were employed for the detection of piroplasm 18S ribosomal gene. All samples were positive for piroplasm DNA by PCR. Amplicons were purified and then sequenced. Sequencing analysis of these amplicons revealed 98% identity to T. equi sequences. Based on our findings, we suggest that abortion in this case resulted from transplacental Theileria infection.(AU)


A piroplasmose equina é transmitida por carrapatos, sendo causada pelos protozoários Babesia caballi e Theileria equi. O objetivo deste trabalho foi relatar um caso de aborto equino associado à piroplasmose fetal. Uma égua gestante, aparentemente saudável, abortou no 10º mês de gestação. Na necropsia do feto, foi observada palidez de subcutâneo, da musculatura esquelética e das mucosas visíveis, moderado hidrotórax e acentuada esplenomegalia. Histologicamente, havia hiperplasia linfoide esplênica e nefrose. Muitos merozoítos foram observados em eritrócitos nos vasos sanguíneos de todos os órgãos examinados, com maior intensidade no encéfalo. Amostras de DNA do baço, fígado, rim e timo foram utilizadas para PCR com primers genéricos para detecção de sequências do gene 18S ribossomal de piroplasmas, com resultado positivo para todas as amostras. Em seguida, os amplicons das amostras de baço foram purificados, sequenciados e, após análise, mostraram 98% de identidade com a sequência de T. equi. Portanto, as alterações microscópicas, os resultados de PCR e sequenciamento confirmam aborto associado à T. equi por transmissão transplacentária.(AU)

Theileriosis is a disease caused by protozoan parasites which belong to genus Theileria. It can be transmitted through ixodid ticks. Mostly the distribution of Theileria species are restricted to subtropical and tropical regions. Theileria species cause economically significant diseases in cattle, sheep, and goats in Pakistan by infecting domestic and wild ruminants. This is a neglected disease in Pakistan because limited research work has been done yet on it. Ovine theileriosis cases were recorded from published data from 2013-2016 in different geographic regions of Pakistan. The intensity and risk factors were also determined in KPK Province of Pakistan. Theleriosis have been reported from all over Pakistan during period 2013-2016. The prevalence of Theleria annulata in buffaloes ticks was 72.91% (875) in urban regions of Hyderabad and it was 80.8% (970) in periurban regions during period 2013-2014. In 2016, prevalence of ovine theileriosis in KPK districts was as 27.5% in Dera Ismail Khan, 13% in Tank and 19.5% in Bannu. It is concluded that paying close attention to animal keeping, feeding and housing may diminish the occurrence of theileriosis in sheep. The higher prevalence might be due to lack of awareness by the community, farming system of the rural livestock of, absence of diagnostic facilities, expensive acaricidal drugs and poor economic condition of farmers.

Serum samples from 714 equids of Itaguaí and Serrana microregions, Rio de Janeiro, southeastern Brazil, were examined by indirect fluorescent antibody test (titer 1:80) for Theileria equi. The prevalence in the microregions and factors associated with seropositivity were evaluated and the prevalence ratio (PR) calculated. The overall prevalence of T. equi infection was 81.09 percent (n = 579), with higher prevalence (p < 0.05) in the Itaguaí (85.43 percent) when compared to Serrana microregion (76.92 percent). The geographic area, altitude, farming condition and area of origin of equids were associated (p < 0.05) with seropositivity for T. equi. Equids reared in the Itaguaí microregion (PR = 1.11, p = 0.003) and at altitudes below 500 m (PR = 1.10; p = 0,014) were more likely to be seropositive for T. equi. Furthermore, when equids were born in the farm (PR = 1.10, p = 0.008) and reared with poor farming conditions (PR = 1.13, p = 0.018) they were more likely to be exposed to T. equi. The main ticks found on equids were Amblyomma cajennense and Dermacentor (Anocentor) nitens. The microregions studied are endemic areas for equine theileriosis and there exists enzootic stability for T. equi. Only factors related to the collection area of serum samples influenced the seropositivity of equids for T. equi in that region.


Amostras de soro de 714 equídeos das microrregiões de Itaguaí e Serrana, Rio de Janeiro, Brasil, foram submetidas ao teste de imunofluorescência indireta (título 1:80) para Theileria equi. A prevalência entre as microrregiões e os fatores associados à soropositividade foram avaliados e a razão de prevalência (RP) calculada. A prevalência geral para T. equi foi de 81,09 por cento (n = 579), com maior prevalência (p < 0,05) para microrregião de Itaguaí (85,43 por cento), quando comparado a Serrana (76,92 por cento). A região, altitude, nível da propriedade e origem dos equídeos foram associados (p < 0,05) com a soropositividade para T. equi. Equídeos criados na microrregião de Itaguaí (RP = 1,11; p = 0,003) e em altitudes abaixo de 500 m (RP = 1.10; p = 0,014) apresentaram maior chance de se tornarem soropositivos para T. equi. Além disso, quando são criados em propriedades de nível ruim (RP = 1,13; p = 0,018) e nascidos na propriedade (RP = 1,10; p = 0,008) apresentaram mais chance de terem contato com T. equi. As principais espécies de carrapatos encontradas parasitando os equídeos foram Amblyomma cajennense e Dermacentor (Anocentor) nitens. As microrregiões estudadas são endêmicas para theileriose equina e de estabilidade enzoótica para T. equi. Apenas os fatores relacionados à área de coleta das amostras de soro influenciaram a soropositividade dos equídeos para T. equi naquela região.

This study was carried out to investigate fifteen cases of acute lethal infection of calves (< or = 4 months of age) by the protozoan parasite Theileria (T.) annulata in the south of Portugal. Calves developed multifocal to coalescent nodular skin lesions, similar to multicentric malignant lymphoma. Infestation with ticks (genus Hyalomma) was intense. Theileria was seen in blood and lymph node smears, and T. annulata infection was confirmed by isolation of schizont-transformed cells and sequencing of hypervariable region 4 of the 18S rRNA gene. At necropsy, hemorrhagic nodules or nodules with a hemorrhagic halo were seen, particularly in the skin, subcutaneous tissue, skeletal and cardiac muscles, pharynx, trachea and intestinal serosa. Histologically, nodules were formed by large, round, lymphoblastoid neoplastic-like cells. Immunohistochemistry (IHC) identified these cells as mostly CD3 positive T lymphocytes and MAC387 positive macrophages. A marker for B lymphocytes (CD79alphacy) labeled very few cells. T. annulata infected cells in these nodules were also identified by IHC through the use of two monoclonal antibodies (1C7 and 1C12) which are diagnostic for the parasite. It was concluded that the pathological changes observed in the different organs and tissues were caused by proliferation of schizont-infected macrophages, which subsequently stimulate a severe uncontrolled proliferation of uninfected T lymphocytes.

Piroplasms are tick-transmitted, intracellular, hemoprotozoan parasites that cause anorexia, fever, anemia, and icterus. Theileriosis is caused by Theileria sergenti and causes major economic losses in grazing cattle in Japan and Korea. In May 2003, we examined the antigenic diversity of the major piroplasm surface protein (MPSP) gene in 35 healthy Jeju black cattle that were born and raised at the National Institute of Subtropical Agriculture. On microscopic examination of Giemsa-stained blood smears, 9 of 35 cattle had intra-erythrocytic piroplasms. Hematological data were within normal range for all 35 cattle. Amplification of DNA from all blood samples using universal MPSP gene primers showed mixed infections with C, I, and B type Theileria spp. Type C was identified in 20 of 35 blood samples, and type B was identified in 17 samples. Allelic variation was seen in type B.

RESUMO

Theileria annulata, a protozoan parasite of cattle and domestic buffaloes, is transmitted by ticks of the genus Hyalomma, and causes a disease named Mediterranean or tropical theileriosis. In this research 50 cattle naturally infected with Theileria annulata were treated with the extract of the plant Peganum harmala. The treatment was continued for 5 days, the dose of the extract being 5 mg/kg per day. After the treatment, 39 cattle responded to the treatment and recovered, but 11 did not respond to the treatment and died. The recovery rate of animals treated with the extract of the plant Peganum harmala was 78%.

This study was performed to investigate the prevalence of theileriosis and to compare the prevalence of this disease in Korean native cattle reared under different environmental conditions, namely, in a grazing area and a non-grazing area by polymerase chain reaction. Three hundred and one Korean native cattle (276 cows and 25 bulls) that had not received prior treatment or been vaccinated to prevent theileriosis were examined by PCR for Theileria sergenti infection from 2001 to 2002. In our study, the parasitemia range in T. sergenti-positive cattle by microscopy were from 0.1 to 3% (mean 0.8%). In terms of mean prevalence, 204 of the 301 Korean native cattle (67.8%) were positive reaction by PCR. Our results also revealed that the infection rate among cows (70.3%) was significantly higher than that among bulls (40.0%) (p < 0.01). T. sergenti infection among the over 3 year-old-group (75%) had a significant higher prevalence than that among the less than 3 year-old-group (61.8%) (p < 0.05). Our data also showed that grazing areas (76.1%) had the significant higher prevalence than non-grazing areas (51%) (p < 0.001). In conclusion, this study demonstrates that the prevalence of T. sergenti infection is high and that its prevalence in grazing cattle is higher than that in non-grazing cattle. Therefore, life-long treatment and the development of an optimal vaccine are needed to reduce the numbers of bovine theileriosis in both grazing and non-grazing areas.
