RESUMO

Therapeutic angiogenesis is an important strategy to rescue ischemic tissues in patients with critical limb ischemia having no other treatment option such as endovascular angioplasty or bypass surgery. Studies indicated so far possibilities of therapeutic angiogenesis using autologous bone marrow mononuclear cells, CD34⁺ cells, peripheral blood mononuclear cells, adipose-derived stem/progenitor cells, and etc. Recent studies indicated that subcutaneous adipose tissue contains stem/progenitor cells that can give rise to several mesenchymal lineage cells. Moreover, these mesenchymal progenitor cells release a variety of angiogenic growth factors including vascular endothelial growth factor, fibroblast growth factor, hepatocyte growth factor and chemokine stromal cell-derived factor-1. Subcutaneous adipose tissues can be harvested by less invasive technique. These biological properties of adipose-derived regenerative cells (ADRCs) implicate that autologous subcutaneous adipose tissue would be a useful cell source for therapeutic angiogenesis in humans. In this review, I would like to discuss biological properties and future perspective of ADRCs-mediated therapeutic angiogenesis.

RESUMO

Ischemic stroke is one of the main diseases which harm the health of the elderly. To promote angiogenesis is of great significance for treating ischemic stroke. In recent years, treatment by TCM based on therapeutic angiogenesis intervention for ischemic stroke has achieved great development. Pharmacological and clinical studies have shown that TCM can promote angiogenesis, thereby improving cerebral ischemia and neurological function. This article reviewed the current situation in this field and provided references for clinical treatment of ischemic stroke.

Diabetic foot (DF) occurs as a concomitant illness of diabetes mellitus (DM). DM is one of the main causes of nontraumatic amputation in Germany with severe peripheral arterial disease (PAD) with critical limb ischemia (CLI) being of major concern. Although modern techniques are available surgical vascularisation and percutaneous intervention are limited. This problem leads increasing numbers of limb amputations in patients with diabetes mellitus. The physiological process of angiogenesis, vasculogenes is and arteriogenesis contribute to the growth of collateral vessels in response to obstructive arterial disease causing limb ischemi. In clinical practice the endogenous angiogenic response is often impaired. Therapeutic angiogenesis is an application of biotechnology to stimulate new vessel formation via local administration of pro-angiogenic growth factors in the form of recombinant protein, or gene therapy, or by implantation of progenitor cells or stem cells that will synthe size multiple angiogenic cytokines. This review summarises the endothelial function and dysfunctionin DM, the mechanism of homing, the transplantation method and the status of clinical trials in stem cell field to treat limb ischemia.
