This study was aimed to evaluate and to compare the acute and subchronic toxicities of the hydroethanolic extracts of theroot (HECRB) and stem bark (HECSB) of Cassia sieberiana in rats. In acute toxicity study, animals were divided into 2groups (n = 3). Rats received the single dose of 5000 mg/kg of HECRB and HECSB extracts of C. sieberiana orally. Forthe subchronic toxicity, 4 groups (n = 6) were given daily 500 and 1000 mg/kg of extracts for 28 days. Animal behaviorswere observed after each treatment. Results showed that HECRB and HECSB were not toxic at 5000 mg/kg. The LD50was >5000mg/kg for both extracts. After repeated doses of 500 and 1000 mg/kg, both extracts did not significantly affectthe relative organs weight of treated rats. HECSB did not affect biochemical and hematological parameters. However,HECRB significantly increased parameters such as red and white blood cells, hemoglobin, volume globulaire moyen, teneur corpusculaire moyenne en hemoglobine, and triglycerides. This study showed that HECRB and HECSB are nottoxic. It contributes to a better knowledge of the toxicity of C. sieberiana used for the treatment of several diseases in Togo.

RESUMO

OBJECTIVE: To prepare paclitaxel / oleyl chitosan nanoparticles (PTX / OCS-NPs) and to examine its physicochemical properties and acute lung toxicity. METHODS: The morphology and size were determined by transmission electron microscope (TEM). The average diameters and polydispersity index were evaluated by dynamic light scattering (DLS). Encapsulation efficiency (EE) and in vitro release were determined by high performance liquid chromatography (HPLC). RESULTS: The RESULTS of PTX/OCS-NPs were spherical in shape, uniform particle size and dispersed uniformly. The diameters of NPs were(292.4±20)nm, PDI less than 0.246 and Zeta potential was about 25 mV. The drug loading of PTX was 15.06%. The encapsulation efficiency of PTX was 48.96%. In vitro release study showed that the cumulative release of PTX from NPs were 41.41% and 56.23% respectively, within 48 h in phosphate buffered saline (PBS) pH 7.4 and pH 4.5. Acute toxicity showed that there was no change in the number of white blood cells in the blood, and the content of LDH in the bronchoalveolar lavage fluid was not changed. The RESULTS indicated that the drug delivery system has a good biosafety and biocompatibility. CONCLUSION: PTX / OCS-NPs have excellent physicochemical properties, high drug loading, with sustained release, no acute lung toxicity, that is expected to be a good delivery of paclitaxel and other difficult anti-cancer drug delivery system.

RESUMO

OBJECTIVE@#To test fumigant and repellent properties of sesquiterpene-rich essential oil from Teucrium polium subsp. capitatum (L.).@*METHODS@#The fumigant toxicity test was performed at (27±1)°C, (65±5)% relative humidity, and under darkness condition and 24 h exposure time. The chemical composition of the isolated oils was examined by gas chromatography-mass spectrometry.@*RESULTS@#The major compounds were α-cadinol (46.2%), caryophyllene oxide (25.9%), α muurolol epi (8.1%), cadalene (3.7%) and longiverbenone (2.9%). In all cases, considerable differences in mortality of insect to essential oil vapor were observed in different concentrations and exposure times. Callosobruchus maculatus (C. maculates) (LC50=148.9 μL/L air) was more susceptible to the tested plant product than Teucrium castaneum (T. castaneum) (LC50=360.2 μL/L air) based on LC50 values. In the present investigation, the concentration of 3 μL /mL acetone showed 60% and 52% repellency against T. casteneum and C. maculatus adults, respectively.@*CONCLUSIONS@#The results suggests that sesquiterpene-rich essential oils from the tested plant could be used as a potential control agent for stored-product insects.
