RESUMO

OBJECTIVE:To establish a method for the contents determination of triamcinolone acetonide acetate and tretinoin in Dimensional ointment. METHODS:HPLC was performed on the column of Diamonsil C18 with mobile phase of methanol-0.1%Phosphoric acid solution(87:13,V/V),detection wavelength was 254 nm(0-5 min)and 350 nm(5-20 min),at a flow rate of 1.0 ml/min,column temperature was 25℃,and volume injection was 10 μl. RESULTS:The linear range was 0.5-5 μg/ml for triamcin-olone acetonide acetate(r=0.999 9)and 4.96-49.6 μg/ml for tretinoin(r=0.999 8);RSDs of precision, stability and reproducibility tests were lower than 1%;recoveries were 98.37%-100.03%(RSD=0.58%,n=6)and 98.21%-100.38%(RSD=0.78%,n=6). CON-CLUSIONS:The method is simple, accurate and fast, and suitable for the contents determination of triamcinolone acetonide ace-tate and tretinoin in Dimensional ointment.

RESUMO

Objective To prepare triamcinolone acetonide acetate(TAA)thermosensitive hydrogel for intra-articular injection, and to investigate its release in vitro. Methods TAA suspending thermosensitive hydrogel was prepared by using poly lactic-co-glycolic acid(PLGA)-polyethylene glycol(PEG)-PLGA as gel matrices, xanthan gum as suspending agent and NaCl as flocculant. It was evaluated preliminarily by determining its contents and its in vitro release. Results The best compositions for preparation of TAA suspending thermosensitive hydrogel were 25% PLGA-PEG-PLGA,0.05% xanthan gum, 0.9% NaCl and 4 mg·mL-1 TAA.The gelation temperature was 35.3 ℃ .The average recovery was(98.98±0.31)%. By the method of membraneless dissolution, the accumulative drug release was up to 83. 31% at 16 days. The drug release followed Ritger-Peppas methematical model. Conclusion The TAA thermosensitive hydrogel with obviously sustained release is expected to become a new drug delivery system for intra-articular injection.

RESUMO

Objective To summarize the clinical experience about therapy for ear keloid by local injection of triamcinolone acetonide acetate combined with surgical resection to control the growth of keloid. Methods After 3~4 times injecting the triamcinolone acetonide ac-etate,the keloid was removed by surgery, some edge of keloid skin was kept and sutured without tension. Results The patients were followed up for 6~24 months, all of 31 ears were primary healing, 26 ears were cured, 4 ears were effective, only one ear was invalid, the effective cure rate was about 96. 8%. Conclusion Local injection with triamcinolone acetonide acetate combined with surgical resection can treat ear keloid.

RESUMO

OBJECTIVE: To establish an HPLC method with gradient elution for the determination of resorcinol and triamcinolone acetonide acetate in compound resorcinol cream. METHODS: The HPLC was conducted on a ZORBAX Eclipse XDB-C18 column (4.6 mm × 250 mm, 5 μm) with acetonitrile (A)-0.5% ammonium acetate solution (B) as the mobile phase. The gradient elution program was as follows: 0-3 min (25% A), 3-5 min (25% A-55% A), and 5-15 min (55% A). The column temperature was 40°C. The flow rate was 1.0 mL · min-1 and the detection wavelength was 240 nm. RESULTS: The linear ranges of resorcinol and triamcinolone acetonide acetate were 103.18-515.90 μmg · mL-1 (r=1.0000) and 1.336-6.680 μg · mL-1 (r=0.999 4), respectively. The average recoveries were 99.3% (RSD=0.62%) and 99.1% (RSD=0.73%), respectively. CONCLUSION: The proposed method is simple, rapid and accurate. It has been applied in the assay of samples with satisfactory results. Copyright 2012 by the Chinese Pharmaceutical Association.

RESUMO

Objective To prepare a biodegradable sckeral implant containing triamcinolone-acetonide-acetate(TAA) and polylactic acid(PLA) and investigate its release in vitro. Methods The TAA-PLA implant was prepared by melt-extrude technique. The drug release of TAA in vitro was determined by high performance liquid chromatography(HPLC).Results The implant was white pillar with 8 mm in length,8 mg in weight, and 0.8 mm in diameter. The drug loading was 30%.The drug was released at a steady and slow rate. Its accumulation release rate is 75.84% on the 30th day.Drug release profile in vitro was in accordance with Higuchi equation Q=0.137(t-0.752)1/2.Conclusion A sleral implant containing TAA-PLA is prepared, which has the evident feature in delaying the release of TAA and is biological degradable. It might be a novel vehicle for the topical use of TAA.

RESUMO

Objective:To observe and summarize the therapeutic effects of intratumoral injection with Triamcinolone Acetonide Acetate and Pingyangmycin on infant maxillofacial hemangioma. Methods:Local injection with Triamcinolone Acetonide Acetate(TAA)and Pingyangmycin(PYM)was executed to treat infant maxillofacial hemangioma. If the lesion did not disappear, the treatment would be performed again every four to six weeks at intervals, and the total dose of TAA and PYM could not be more than 100mg and 40mg respectively. The injection treatment was repeated 4 times for one course and the therapeutic effects of maxillofacial hemangioma were detected. Results: 84 cases with maxillofacial hemangioma, treated by local injection with TAA and PYM,had been followed up for one to three years. The cure rate was 90.4%,the notable rate was 7.1%,and the ineffective rate was 2.4%.Conclusions:The intratumoral injection with Triamcinolone Acetonide Acetate and Pingyangmycin is the reliable therapy method with less side effects. It is valuable for treatment of infant maxillofacial hemangioma.

RESUMO

Objective To investigate the effects and complications of injection with triamcinolone acetonide acetate and Pingyangmycin on maxillofacial and perineal hemangioma. Methods Local injection with triamcinolone acetonide acetate (TAA) and Pingyangmycin (PYM) was performed to treat infant maxillofacial and perineal hemangiomas: strawberry hemangioma, cavernous hemangioma and mixed hemangioma, in 107 infants admitted to our hospital from 1996 to 2005. If the lesion did not disappear after the first treatment, the injection would be performed again every four to six weeks interval, and the total dose of TAA and PYM could not be more than 100 mg and 40 mg respectively. The injection treatment was repeated 4 times for one course and the therapeutic effects on maxillofacial and perineal hemangioma was evaluated. Results All cases were followed up for one year after local injection of TAA and PYM. The cure rate was 87.8%, the effective rate was 9.3%, and the ineffective rate was 2.8%. Conclusion The injection of TAA and PYM is a reliable and less side effect therapeutic method, and valuable for infant maxillofacial and perineal hemangioma.
