RESUMO

Objective@#To evaluate the significance of different clinicopathologic features on prognosis of patients with squamous cell carcinoma of vulva.@*Methods@#We retrospectively analyzed the prognostic relevance of different clinicopathological variables of 201 patients with squamous cell carcinoma of vulva treated in Cancer Hospital, Chinese Academy of Medical Sciences. The data including age, initial symptoms, stage, location, tumor size, histological grade, number and size of metastatic lymph nodes, treatment mode, and presence of leukoplakia vulva was used to evaluate the prognosis of vulvar squamous cell carcinoma.@*Results@#The median age of onset was 62.0 years old, with 74 patients in stage Ⅰ, 27 in stage Ⅱ, 55 in stage Ⅲ and 9 in stage Ⅳ. The median progression-free survival was 90.0 months. The 5-year progression-free survival rate of the total patients was 55.5%, while the 10-year progression-free survival rate was 48.5%. Univariate analysis showed statistically significant prognostic parameters included clinical stage, number of metastatic lymph nodes, tumor size and treatment mode (all P<0.001). Multivariate analysis showed that number of metastatic lymph nodes (P<0.05) was an independent prognostic factor for progression-free survival.@*Conclusion@#The study illustrates that number of metastatic lymph nodes represents important independent factor for progression-free survival of patients with vulvar squamous cell carcinoma.

Aim: To investigate the effect of osthole on inducing apoptosis and inhibiting the proliferation of vulvar squamous cell carcinoma SW962 cells through PDK/Akt signaling pathway and changes of PDK/Akt pathway protein in the coexistence of osthole and IGF-1. Methods: Vulvar squamous cell carcinoma SW962 cells were cultured in vitro and different concentrations of osthole intervention were given. The effect of osthole on the proliferation of SW962 cells was determined by MTT. Cell apoptosis was detected by flow cytometry. DAPI staining revealed the changes in cellular morphology. The expressions of cleaved-caspase-3, PI3K, p-Akt, Bcl-2, Bax protein in SW962 cells were assessed by Western blot. Results: MTT results showed that osthole could inhibit the proliferation of SW962 cells in a concentration-dependent manner. The results of DAPI staining and FCM by Annexin V-FITC and PI staining indicated osthole induced apoptosis of SW962 cells in a concentration-dependent manner. Western blot showed that osthole increased the expression of Bax and cleaved caspase-3 and decreased the expression of Bcl-2, PI3K, p-Akt. IGF-1 induced increased expression of PI3K, p-Akt and Bcl-2, while osthole reversed IGF-1 and down-regulated the expression of three proteins. Conclusions: Osthole induces the apoptosis and inhibits the proliferation of SW962 in vulvar squamous cell carcinoma by inhibiting PI3K/Akt signaling pathway.

RESUMO

Objective To investigate the effects of long non-coding RNA (lncRNA) HOTAIR on the biological behavior of the vulvar squamous cell carcinoma A431 cells.Methods The expression level of HOTAIR in vulvar squamous cell carcinoma A431 cells and normal epidermis HaCaT cells were detected by real-time PCR.A431 cells were transfected with HOTAIR siRNA,then the proliferation,apoptosis,migration activities,and invasion activities were measured by CCK-8 assay,flow cytometry assay,transwell chamber migration test,and transwell chamber invasion test,respectively.Results After down-regulating the level of HOTAIR in A431 cells,the cell proliferation,migration,and invasion abilities were decreased (P < 0.05),while the cell apoptosis rate was increased (P < 0.05).Conclusion lncRNA HOTAIR may play an important role in the proliferation,apoptosis,migration,and invasion in vulvar squamous cell carcinoma.
