Abstract Diclofenac sodium (DF) is a non-steroidal anti-inflammatory drug (NSAID) that possesses antipyretic, analgesic, antinociceptive and anti-inflammatory activities. Like other NSAIDs, DF is known to be associated with renal, cardiovascular, and gastrointestinal complications. The present study was carried out to evaluate the adverse effects of DF in vivo in wistar albino rats and to assess if oral administration of the organic osmolyte betaine mitigates the adverse effect of DF. Eighteen male Wistar rats were divided into three groups, one group of animals was fed orally with 20 mg/kg of DF once/day, and the other group received a combination of 20 mg/kg of DF and 30 mg/kg of betaine, once/day. Apart from the hematological and biochemical parameters, histopathological changes in the liver, lungs, brain, heart and kidney were also investigated. Histopathological alterations that were found in the liver, kidney, and lungs of DF-treated animals were found to be minimal or absent in DF + betaine-treated animals, as compared to untreated control. The results showed that betaine mitigates the adverse effects associated with DF treatment.

Objective: To evaluate the antidiabetic and the antidiarrhoeal effects of ethanolic extracts of Psidium guajava leave (EEPGL) in Wister rats to support its traditional uses. Methods: Oral glucose tolerance test model and alloxan induced diabetic test model were performed to evaluate antidiabetic activity of EEPGL at doses of 1.00, 0.50 and 0.75 g/kg respectively. For antidiarrhoeal effects of EEPGL, castor oil-induced diarrhoea model and gastrointestinal motility test with barium sulphate milk model were also assessed at doses of 750, 500 and 250 mg/kg, respectively. Results: Administration of EEPGL at doses 1.00 and 0.50 g/kg significantly (P<0.05) decreased blood glucose levels in oral glucose tolerance test model as well as 0.75 g/kg dose in alloxan induced diabetic test model in Wister rats (P<0.001). Application of EEPGL at doses of 750 and 500 mg/kg showed antidiarrhoeal effect in castor oil-induced diarrhoeal model (P<0.001 and P<0.01, respectively), and 750 mg/kg (P<0.01), 500 and 250 mg/kg (P<0.05) doses in barium sulphate milk model in aforesaid animals. Conclusions: These results exhibited the significant antidiabetic and antidiarrhoeal activities of ethanolic extracts of Psidium guajava leave in Wister rats

Testicular torsion is a disorder involving the scrotum that results in a compromise of its blood supply. The aim was to investigate the effect of Pausinystallia macroceras (PM) on testicular histology following torsion-detortion at different time intervals ranging from 1 to 4 hours 65 mature male Wister rats allotted randomly into seven groups (A to G; E& F further divided into 4-subgroups). Each group/subgroup comprised 5 rats. Testis maintained in the torted position (T) for 1, 2, 3 and 4 hours in Groups A (AT1+PM), B (BT2+PM), C (CT3+PM) and D (DT4+PM). Group E subgroups (E1+PM, E2+PM, E3+PM, E4+PM -) were sham operated, without torsion served as the sham control. Group F subgroups (F1T1, F2T2, F3T3 and F4T4) were torted as in A. All animals (except groups F & G) treated with PM extract (0.1 g/kg.b.w/day) for 56 days. Group G rats (normal control). Testes processed for histological studies. In AT1+PM showed preserved seminiferous tubules. BT2+PM, revealed varying number of necrosed and apoptotic seminiferous tubules. Group CT3+PM rats were similar to BT2+PM although with a slightly higher proportion of seminiferous tubules had undergone necrosis. In DT4+PM, sections showed few viable spermatozoa within the seminiferous tubules. When compared to the torted group F; showed extensive areas of seminiferous tubular necrosis (F3T3) as well as damage to the interstitium; while in F4T4 there were no viable testicular tissues seen. In conclusion, PM significantly prevented the cellular changes and cell death observed especially in group AT1+PM and BT2+PM.


La torsión testicular es un trastorno que involucra el escroto resultando en un compromiso del suministro sanguíneo. El objetivo fue investigar el efecto de Pausinystallia macroceras (PM) en la histología testicular tras torsión-detorsión a intervalos de tiempo diferentes que van desde 1 a 4 horas en 65 ratas macho Wistar maduras, asignando aleatoriamente en siete grupos (desde A a G, mientras que E y F se dividieron en 4 subgrupos). Cada grupo/subgrupo estuvo compuesto por 5 ratas. Los testículos se mantuvieron en posición torsionada (T) durante 1, 2, 3 y 4 horas en los grupos A (AT1 + PM), B (BT2 + PM), C (CT3 + PM) y D (DT4 + PM). El grupo E, subgrupos (E1 + PM, E2 + PM + PM E3, E4 + PM) fueron operados por modelo sham sin torsión, que sirvió de control. El grupo F, subgrupos (F1T1, F2T2, F3T3 y F4T4) fueron torsionados como en A. Todos los animales (excepto los grupos F y G) fueron tratados con extracto de AM (0,1 g/kg peso corporal/día) durante 56 días. El grupo G fueron ratas control (control normal). Los testículos fueron procesados para el estudio histológico. En AT1 + PM se observó preservación de los túbulos seminíferos. BT2 + PM, reveló un número variable de túbulos seminíferos con necrosis y apoptosis. El grupo de ratas CT3 + PM fue similar a BT2 + PM, aunque un porcentaje ligeramente superior de los túbulos seminíferos mostraron necrosis. En DT4 + PM, los cortes mostraron pocos espermatozoides viables dentro de los túbulos seminíferos. En comparación con el grupo F torsionado mostró extensas áreas de necrosis tubular (F3T3), así como daños en el intersticio; mientras que en F4T4 no hubo tejido testicular viable. En conclusión, PM previno significativamente cambios celulares y la muerte celular observada, especialmente en el grupo AT1 + PM y BT2 + PM.

En el presente trabajo se investiga la inducción de placas ateromatosas en ratas albinas machos, cepa Wistar (Rattus norvegicus) crónicamente infectadas con Trypanosoma cruzi y alimentadas ad libitum con una dieta rica en grasas de origen vegetal, durante tres meses. La infección crónica evidenciada por pruebas sero-parasitológicas, reveló presenciade anticuerpos IgG anti- T. cruzi y ausencia de parasitemiaspatentes. La dieta rica en grasas produjo en las ratas infectadas (A) y sanas (C) un aumento significativo en el peso (P<0,05), en comparación con las ratas controles (B) y sanas (D) sometidas a la dieta normal. El estudio histopatológicode secciones de la arteria aorta de las ratas del grupo A (infectadas/dieta grasa), mostró abundantes depósitoslipídicos, procesos inflamatorios (vasculitis) y placas ateromatosas en formación. En las secciones de corazón y músculo esquelético se evidenció miocarditis y miositis con características de cronicidad sin parasitismo tisular. Las pruebas inmunohistoquímicas aplicadas a los cortes de arteria,corazón y músculo esquelético de las ratas infectadas A (infectadas/dieta-grasa) y B (infectadas/dieta-normal), mostraron abundantes depósitos antigénicos, lo que indica persistencia de antígenos parasitarios. En conclusión, las ratasinfectadas con T. cruzi alimentadas con la dieta rica en grasas, tienen una mayor propección a desarrollar placas ateromatosas.Los resultados demostraron que una dieta hiperlipídicaes un factor de riesgo para el desarrollo de enfermedadateromatosa en individuos con enfermedad de Chagas


This work is focused on the induction of atheromatous plaques in male albino rats (Rattus norvegicus), Wistar, chronically infected with Trypanosoma cruzi and fed ad libitum with diet rich in fats of vegetal origin, during three months. The chronic infection detected by serological and parasitological assays, revealed the presence of antibodies IgG anti- T. cruzi and the absence of patents parasitemias. The diet rich in fats produced in the group of infected rats (A) and the group of healthy rats (C) a significant increase in the weight (P<0,05), in comparison with the control group of infected rats (B) and the group of healthy rats (D), fed with a normal diet. The histopathological study of sections of the aorta arteryof the rats of the group (A) (infected/diet fat), showed abundants lipid deposits, inflammatory processes (vasculitis) and atheromatous plaques in development. The sections of the heart and skeletal muscle showed pictures of a myocarditis and myositis with features of chronic tissue without parasitism. The immunohistochemestry assays applied to the cuts of artery, heart and skeletal muscle of the infected rats A (diet/fat) and B (normal/diet), showed abundants antigen deposits. In conclusion, the rats chronically infected with T. cruzi and fed with a diet rich in fats, have a main propensity to develop atheromatous plaques. The results showed that a hyperlipidic diet is a risk factor for the development of atheromatous disease in individuals with Chagas`disease
