RESUMO

Osteoporosis is a health problem to cause global concern. A lot of methods have been used to prevent and treat osteoporosis, but there is still a lack of effective treatment for osteoporosis owing to limited understanding of its mechanism. Therefore, the aim of this present study is to explore the underlying mechanism of Wuling Powder, a traditional Chinese medicine on treating osteoporosis. In this study, we firstly screened and identified the common targets between Wuling Powder and osteoporosis through the related databases, and then explored the relationships among these targets, Wuling Powder and osteoporosis by using Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG) and network analyses. Subsequently, the molecular docking was performed by using systemsDock to evaluate the potential binding relationships between the active components of Wuling Powder and their related targets. The results showed that in total of 14 common targets including CREBBP, ADAM17, GOT1, GAPDH, USP8, ERBB2, EEF1A1, MTOR, RAC1, ETS1, DDX58, GCK, EGF and S100A8 were screened. EGF, ERBB2, MTOR and HIF-1 were the potential therapeutic targets for osteoporosis, and they were also the related targets for predicting active components in Wuling Powder. Taken together, we concluded that Wuling Powder might be used to treat osteoporosis through above these targets.

Objective: To explore the mechanism of Wuling Powder in the treatment of chronic heart failure (CHF) based on network pharmacology. Methods: Traditional Chinese Medicine Systems Pharmacology Database (TCMSP) and literature mining were used to search the chemical components and targets of Wuling Powder, and a single drug-active ingredients-target network was established. The related targets of chronic heart failure were collected through Genecards and OMIM databases, the network model of active components-CHF-targets was constructed and analyzed by Cytoscape 3.7.1 software. The protein-protein interaction (PPI) network was constructed by STRING database platform, and the gene oesthetics (GO) function annotation and Kyoto Encyclopedin of Genes and Genomes (KEGG) signal pathway enrichment analysis were performed by DAVID online tools. The molecular docking was performed using Surflex software. Results: Fifty components, 29 potential targets, 8 243 targets related to chronic heart failure, and 27 targets of Wuling Powder-CHF were obtained. The network analysis results showed that the key targets of Wuling Powder in the treatment of chronic heart failure included CASP3, RELA, AR, ESR1, CHRM1 and CASP8, etc. Biological processes mainly involved signal transduction, nervous system development, transcription initiation from RNA polymerase II promoter in response to estradiol, synaptic transmission, cholinergic synaptic transmission, etc. KEGG enrichment involved neuroactive ligand-receptor interaction, PI3K-Akt signaling pathway, cholinergic synapse, etc. The molecular docking results showed that (+)-catechin, taxifolin and other key compounds in Wuling Powder had better binding ability with key targets such as CASP8, CHRM1, and NR3C1. Conclusion: The material basis and mechanism of Wuling Powder in the treatment of chronic heart failure were revealed based on network pharmacology, which provided a certain theoretical basis for the clinical application of Wuling Powder.

Wuling powder, which is from Treatise on Cold Damage and Miscellaneous Diseases of Zhang Zhongjing, is a famous formula from thousand years now on. As people have a deeper understanding of Wuling powder, its application range is more extensive. In Treatise on Cold Damage and Miscellaneous Diseases and Essentials from the Golden Cabinet, there are a total of 11 articles concerning Wuling powder. Wuling powder syndrome can be classified as pulse floatation, fever, thirst, irritability, vomiting and urination, which can also occur in dehydration by sorting out and analyzing these articles. This paper talks about the relationship between Wuling powder syndrome and dehydration from the etiology, the symptom and the clinical application as well as to the thought of Wuling powder syndrome several aspects.

Objective]Analysis of related theory that“the theory of spleen and stomach”treats“diet injury”, to explore principle-method-recipe-medicines’guiding significance for treatment of internal injuries caused by“diet injury”to explore the related theory’s guiding significance for treatment of internal injuries caused by“diet injury”(This is the related theory to“the theory of spleen and stomach”treating“diet injury”).[Method]Through the process of diagnosis and treatment of three cases analysis(They are“drinking wound”case, dizziness case and insomnia case), expound the argumentation thinking to“the theory of spleen and stomach”treating“diet injury”, principle of prescription medication, and matters needing attention, etc. Through the analysis of diagnosis and treatment process, expound the argumentation thinking to“the theory of spleen and stomach”treating“diet injury”, principle of prescription medication, and matters needing attention. Including the prescription of treatise on febrile disease———Wuling powder’s application in the“diet injury”,Gehuajiejiu soup, Zhishu pill, Hezhong pill’s application in the“drinking wound”.[Result] Understand related theory that the“the theory of spleen and stomach”treats “diet injury”, specific therapeutic principle for sweating, the urine and digestion. Including the prescription of treatise on febrile disease—Wuling powder’s application in the“diet injury”,Gehuajiejiu soup, Zhishu pill, Hezhongpill’s application in the“drinking wound”, Wuling powder, Gehuajiejiu soup are dehumidification by sweating and the urine, Zhishu pill, Hezhong pill ,etc are helping digestion.“The theory of spleen and stomach”treating“diet injury”,prescription and symptoms are consistent. Clinical efficacy of three consilias is satisfaction. Among them, drinking wound, shares proprietary Chinese medicine medicinal properties to achieve and anti-nausea, points up and down digestive purpose of alcohol. Gehuajiejiu soup points up and down digestive purpose of alcohol warning patients at the same time; The dizziness, Wuling powder and Banxiazhishu pill wet Xiaoshi with use and effect;Insomnia, emphasis will be placed on the medication method of small amount many times, and“Nursing mind modulation spleen and stomach”, cooperating with use, treatment effect is obvious. [Conclusion]Using the related theory of“the theory of spleen and stomach”is worth clinical exploration to treatment of clinical disease caused by the“diet injury”, because it is dialectical correctly, curative effect is better.

RESUMO

Objective To investigate the effects of Wuling Powder on insulin resistance of C57BL/6J mice induced by high lipid diet, and discuss the mechanism. Methods C57BL/6J mice were randomly divided into six groups:normal group, model group, rosiglitazone group, Wuling Powder in low, middle, and high dose groups, 10 mice per group. Besides the normal group, other five groups were fed with high fat and sugar diet, with a purpose to establish insulin resistance model. Normal group and model group were given pure water. Rosiglitazone group received a gavage with rosiglitazone of 0.75 mg/kg. Wuling Power low, middle, and high groups received gavage with Wuling Power of 1.23, 3.69, 11.07 g/kg, respectively, the does volume was 0.2 mL/10 g, once a day. The weight and abdominal girth were detected every week. At the end of the sixth week, mice were given 12-hour fasting, and their eyeball were taken for blood. The body weight, length, and fat in abdomen and both kidneys were detected. Paraffin section was made with HE staining. FPG and FINS of each group were detected. ISI and IRI were calculated, and TC and TG were detected. Results Compared with the model group, Wuling Powder can significantly reduce the body weight and abdominal girth of mice (P<0.01, P<0.001), improve liver fatty degeneration, lower the FPG, FINS, TCH, TG, IRI, and increase the ISI in mice (P<0.05, P<0.01). Conclusions Wuling Powder has the effect of preventing insulin resistance of C57BL/6J mice induced by high lipid diet.

[Objective] To observe the clinical effect of classical formula Wuling Powder on residual ovary syndrome(ROS). [Method] Choose medical cases for example, explore the clinical effect of revised Wuling Powder on ROS from diagnosis standard, cases data, causa morbi, mechanism, treatment rule and methods, effect standard and result respectively. [Result] The ROS treated with revised Wuling Powder is reasonable, with definite cure effect. [Conclu-sion] The classical formula Wuling Powder treating ROS has correct cure effect, worthy big disease cases study and further proving its efficacy.

RESUMO

Objective To evaluate the therapeutic effect of Wuling Powder (WLP) on adriamycin-induced nephrotic syndrome in rats. Methods Rat models of nephrotic syndrome were established by injection of adriamycin into tail veins of rats. The rats were divided into 7 groups: group A(normal control),group B (high-dosage WLP),group C (moderate-dosage WLP),group D (low-dosage WLP),group E (hydrocortisone),group F (hydrocortisone and moderate-dosage WLP extractive) and group G (model control). The influence of 4-week treatment of WLP on 24-hour urinary protein volume,serum levels of albumin,cholesterol and triglycerides as well as pathological changes of kidney tissue were assayed. Results 24-hour urinary protein volume of group B,C,E and F were significanuly lower than that of group G;Serum albumin level of group B,C,E and F differed from that of group G; Serum albumin level of group E and F were higher than that of group B and C;Serum cholesterol level of WLP groups (group B,C,E,F) was much lower than that of group G and the difference was insignificant between group B,F and group A;As for serum triglycerides level,three WLP groups and group F differed from group E while the group G not. Conclusion WLP not only can eliminate tissue edema and decrease urinary protein volume and blood lipid content,but also can increase serum albumin levels,thus it can protect the kidney from injury. There also have been showen that a synergistic action of WLP and hydrocortisone has been shown in the treatment of nephrotic syndrome.

RESUMO

24 cases of renal failure induced by chemotherapy were treated with Wuling powder plus additives.Theresult of clinical amelioration was 87.5%.with a to-tal effective yielded marked actions of promoting ap-petite in animals,lowering serum urea nitrogen,cre-atinine,2 microglobulin.It also decreased the inhibi-tion of PDD on the enzyme activity of renal Na~+—K~+ATP.Microscopic exam revealed the pathological le-sion of kidney to be milder in the tested group.
