RESUMO

OBJECTIVE@#To observe the effects of auricular thumbtack needle on breast feeding and lactation function in primiparous women with cesarean section, and to explore its mechanism of action from the perspective of lactation-related gene expression.@*METHODS@#One hundred cases of primiparous women with cesarean section were randomly divided into an observation group (50 cases, 3 cases dropped off) and a control group (50 cases, 2 cases were eliminated). The patients in the control group were treated with routine obstetric care. Based on the treatment of the control group, the patients in the observation group were treated with auricular thumbtack needle at Neifenmi (CO18), Xiong (AH10), Xiongzhui (AH11), Shenmen (TF4), and Jiaogan (AH6a), etc., with one side of auricular point selected, only once for a total of 3 d. The lactation initiation time, lactation adequacy rate at postpartum 72 h, exclusive breastfeeding rate at postpartum 42 d, and breastfeeding score after treatment were compared between the two groups. Real-time quantitative PCR and Western blot method were used to detect the mRNA and protein expression levels of TDP-43, Btn1A1 and XDH.@*RESULTS@#After treatment, the lactation initiation time in the observation group was earlier than that in the control group (P<0.01), and breastfeeding score in the observation group was higher than that in the control group (P<0.01). The lactation adequacy rate at postpartum 72 h was 63.8% (30/47) in the observation group, which was higher than 41.7% (20/48) in the control group (P<0.05). The exclusive breastfeeding rate at postpartum 42 d was 72.3% (34/47) in the observation group, which was higher than 47.9% (23/48) in the control group (P<0.05). The mRNA and protein expression levels of TDP-43 and Btn1A1 in breast milk in the observation group were higher than those in the control group (P<0.01), while there was no statistically significant difference in mRNA and protein expression of XDH in breast milk between the two groups (P>0.05).@*CONCLUSION@#The auricular thumbtack needle in addition to routine care could promote lactation initiation, improve lactation adequacy rate and exclusive breastfeeding rate in primiparous women with cesarean section, and the action mechanism may be related to up-regulation of TDP-43 and Btn1A1 expression.

OBJECTIVE: To investigate the time- and dose-dependent effects of 3, 5, 2', 4'-tetrahydroxy chalcone (P40) on the levels of uric acid and the contents of hepatic xanthine dehydrogenase (XDH) and xanthine oxidase (XOD) in the hyperuricemic mice induced by potassium oxonate. METHODS: The hyperuricemic mice were induced by an intraperitoneal injection of uricase inhibitor potassium oxonate. Serum uric acid levels were determined by using tie phosphotungstic acid method. The contents of hepatic XOD and XDH were determined using commercially available Elisa kits. Allopurinol was as a positive control. RESULTS: When orally administrated to the oxonate-induced hyperuricemic mice, compound P40 at doses of 0.5, 1.0, 2.0, 4.0 mg · kg-1 and the allopurinol at a doses of 1.0 mg · kg-1 significantly decreased the uric acid levels and reduced the concentration of XOD compared with model group. As for the study of time-dependent effects: after administration of allopurinol 30 min or P40 60 min effectively reduced serum uric acid levels compared to the model group, respectively. Administration with P40 and allopurinol for 15, 30, 60, 90 min can reduce the concentration of XDH and XOD, compared with model group. CONCLUSION: P40 could reduce the serum uric acid levels in oxonate-induced hyperuricemic mice by reducing the contents of hepatic XDH/XOD.

Objective To analyze the differences between the semi-quantitative RT-PCR and real time quantitative fluorescence RT-PCR assays for detecting XDH/XO mRNA expression in various organ tissues of rhesus monkey, and provide useful reference in methodology of experimental studies.Total RNA was extracted from the myocardium, kidney, testis, skin, and liver tissues, respectively, for detecting XDH/XO mRNA expression in rhesus monkey by semi-quantitative RT-PCR and real time quantitative fluorescence RT-PCR assays.The sensitivity and specificity of the two assays were compared with each other using the same primer sequences and reference genes.Results The expression of XDH/XO mRNA in different organ tissues were detected by both the two PCR assays.The sensitivity of quantitative fluorescence real-time RT-PCR for the XDH/XO mRNA expression in the liver tissue was 39 times higher than that by semi-quantitative RT-PCR.Conclusions Both the quantitative and semi-quantitative fluorescence RT-PCR assays can be used to detect the expression of XDH/XO mRNA in different organ tissues of rhesus monkey.The sensitivity of quantitative fluorescence real-time PCR assay is more sensitive than that of the semi-quantitative RT-PCR assay.

The present study was aimed at investigating the in vivo hypouricaemic activity of the various fractions of the hydromethanolic extract of the leaves of Erythrina stricta roxb (papilionacea) using oxonate-induced hyperuricaemic mice. The leaves of this species was used in traditional medicinal system for the treatment of gout, rheumatism, jaundice, bronchitis, fever, skin eruptions, wounds, etc. The pet-ether, chloroform, ethyl acetate and residual fractions at a dose of 200 mg/kg b.w. orally, were tested for their ability to reduce the serum urate level and inhibitory actions on the XO/XDH enzyme activities in the mouse liver and intestine. Potassium oxonate (280 mg/kg, i.p.), an uricase inhibitor was used to induce hyperuricaemia. Allopurinol (10 mg/kg, p.o.) was used as the positive control. The pet-ether, chloroform and ethyl acetate fractions when administered to hyperuricaemic mice produced a significant reduction in serum urate levels. In addition, these fractions elicited significant inhibitory actions on the XO/XDH enzyme activities in the mouse liver. The hypouricaemic activity may be due to the inhibition of XO/XDH enzymes. The effect of the fractions was less potent than allopurinol. The activity produced by the residual fraction was insignificant (P>0.05). Phytochemical screening of the leaves of Erythrina stricta revealed the presence of tannins, flavonoids, alkaloids and terpenoids. The presence of phytochemical constituents may be partly responsible for the beneficial effect of the fractions on hyperuricaemia and gout. These results suggest that the leaves of Erythrina stricta could be used as a potential source to treat gout and other inflammatory disorders.
