RESUMO

Objective To investigate the application effect of the Beckman Coulter PK 7300 automatic blood group analyzer and the microplate method in ABO blood group screening .Methods A total of 12000 EDTA anticoagulation whole blood samples from January to May 2015 were collected from voluntary blood donors .The Beckman Coulter Pk7300 automatic blood group analyzer and STAR sampling microplate manual colorimetric method were to conduct the detection analysis .Results The accuracy for detecting ABO blood group had no statistically significant difference between the two methods (P>0 .05) .In the ABO blood group screening , the detection rates of ABO subtype and antibody weakening in the Beckman Coulter Pk 7300 automatic blood group analyzer were higher than those in the micro plate method .3 cases of ABO blood group typing and reverse typing were consistent in the detection by the Beckman Coulter Pk7300 automatic blood group analyzer ,but was inconsistent in the detection by the microplate method .4 cases of ABO blood group typing and reverse typing were inconsistent in the the detection by the Beckman Coulter Pk 7300 automat-ic blood group analyzer ,but was consistent in the detection by the microplate method .Conclusion The Beckman Coulter Pk7300 automatic blood group analyzer can safely and effectively conduct the ABO blood group screening in blood donors .The samples of suspicious detection results still need to conduct the manual interpretation by combining with the test tube method .

RESUMO

Objective:To evaluate the performance of reticulocyte (Ret) detected by Sysmex XN-3000 automatic blood analyzer.Methods: 96 blood samples were collected randomly in clinical detection of reticulocytes, and respectively were detected by XN-3000 automatic blood analyzer and microscopy. The precision, stability, linearity and carryover of Ret in blood samples detected by XN-3000 automatic blood analyzer were evaluated by the correlation analysis of the results from the two methods and the correlation analysis would be completed by microscopy.Results: precision, linear range and sensitivity of Ret measured by XN-3000 were within the allowable range. The stability did not change significantly below 4℃ within 48h (CV<1%). The carryover rate was 0.14%. The results measured by XN-3000 were well compared with microscopy (r=0.979).Conclusion: Sysmex XN-3000 automatic blood analyzer is applied to the clinical detection of large quantities of specimens and it is ideal for measuring Ret.

Objective To compare the precision and correlation of test parameters betwee the two types of automatic blood ana-lyzer,Sysmex XT-2000i and Mindray BC-5800.Methods We selected 60 cases of EDTA-K2 anticoagulant whole blood samples, correlation analysis was carried out on the two instrument parameters,and high,medium and low value samples were chosen for 10 times′precision analysis in the two instrument s respectively.Results The variation coefficient(CV)values of blood count parame-ters of Sysmex XT-2000i were 0.02%-5.70%,while the CV values of white blood cell classification parameters were 0.06%-1. 66%.The CV values of blood count parameters of Mindray BC-5800 were 0.02%-5.3%,white the CV values of white blood cell classification parameters were 0.09%-1.17%.The determination coefficient(r2 )of each parameter of the two instruments were 0. 185-0.995.Conclusion the correlation between the two instruments is good,and the two instruments′precisions were consistent with the design range.

RESUMO

Objective To apply three automatic blood group testing instruments, WADiana®(Diana), Techno TwinStation and ORTHO AutoVue Innova/Ultra, in clinical cross match test and to assess their effect, so as to ensure safe clinical blood transfusion. Methods The three instruments were used for cross matching test of 8 073 blood recipients and blood specimens of Shanghai Blood Center. The incompatible positive samples were reexamined by experiment cross matching antiglobulin test tube method. Results The 3 instruments detected 589 positive blood samples. There was no false positive result in the WADiana®(Diana) system; Techno TwinStation and ORTHO AutoVue Innova/Ultra analyzer had 6 and 19 false positive results, respectively. Conclusion All the three instruments can accurately detect the positive samples in clinical cross match test, and WADiana® system has a more prominent performance.
