RESUMO

@#Androgen ablation therapy using gonadotropin-releasing hormone agonists is reported to be associated with metabolic abnormalities. Annatto tocotrienol (AnTT) is reported to reduce the expression of genes related to adipogenesis but the mechanism remains elusive. This study sought to determine the effects of annatto tocotrienol on body composition (lean and fat mass), serum adiponectin, leptin, and glucose levels in male rats treated with buserelin, a testosterone ablation agent. Three-month-old male Sprague Dawley rats (n=32) were randomly divided into four groups. The normal control (n=8) was given corn oil orally daily and normal saline subcutaneously daily. The remaining groups were injected with buserelin subcutaneously (75µg/kg/day). The buserelin group (n=8) was given corn oil orally, while the treatment groups were supplemented orally with AnTT at 60 or 100 mg/kg (n = 8/group). After treatment of 12 weeks, rats were euthanized. Dual-energy x-ray absorptiometry was performed to determine the lean and fat mass of the rats. Blood was collected for the evaluation of adiponectin, leptin and glucose levels. After 12 weeks, the lean mass, fat mass, adiponectin and leptin levels for all groups increased significantly compared to their respective baseline levels irrespective of their treatment (P<0.05). All groups, except rats receiving AnTT at 60 mg/kg, experienced a significant increase in glucose level after 3 months (P<0.05). Androgen ablation and AnTT do not influence body composition, adiponectin and leptin levels in male rats. However, annatto tocotrienol at 60 mg/kg may improve glucose metabolism.

The aim of this work was to study estrus synchronization and fixed time artificial insemination (FTAI) in dairy buffaloes during season anestrus. One hundred thirty-nine dairy buffaloes in seasonal anestrus were divided in two groups as G1(n=66) and G2(n=73). The protocols for both the groups were the same until day (D)14:D0 administration of 2.0 mg estradiol benzoate and implantation of progesterone device (P4) for 14 days; D14 removal of P4 plus 150 mg of cloprostenol and 400 IU of equine chorionic gonadotropin. On D16, G1 received 10 mg of buserelin and G2 100 mg deslorelin acetate. On D17, both the groups were submitted to FTAI. Ultrasonographic examinations of ovaries were performed on D0, D14, D16 and D17. Results showed that pregnancy rates in G1 and G2 were 20 and 41% (p<0.05) and the ovulation rates were 16.6 and 37%, respectively (p<0.05). The dominant follicle (DF) diameter on D16 was 7.9 mm in G1 and 8.9 mm in G2 (p>0.05). Thirty-five percent of the animals in G1 and 54.1% in G2 showed a diameter DF greater than 8.0 mm on D16 (p>0.05). Thus, it could be concluded that the protocols synchronized the estrus, leading the concentration of the parturitions in the period of low milk production. Deslorelin was more efficient than buserelin due the higher percentage of DF ovulation and higher pregnancy rates.

OBJECTIVE: To prepare Buserelin acetate nanoparticles(BA-NP) and to investigate its release property in vitro.METHODS: The BA-NP was prepared using double emulsion method.The content determination of BA-NP was performed using HPLC,and encapsulation efficiency and drug-loading rate of BA-NP were calculated.In vitro drug release property of nanoparticles was investigated by bag filter method.RESULTS: Prepared nanoparticles were even and regular in appearance.The linear range of buserelin acetate was 0.1~8.0 ?g?mL-1(r=0.999 9) with an average recovery of 105.38%.The RSD of intra-day and inter-day were lower than 1.78% and 0.93% respectively.The encapsulation efficiency of nanoparticles was(63.37?0.29)% and drug-loading rate of(1.03?0.09)%.The accumulative release rate of nanoparticles in phosphate buffer(pH=7.4) at 72 h was 62.35%.CONCLUSION: The preparation process of BA-NP is simple and particle with ideal release effect.
