Oligoasthenozoospermia is the main cause of male infertility， with complex and diverse causes. Currently， there are still some unclear causes of oligoasthenozoospermia in clinical practice， known as idiopathic oligoasthenozoospermia. With the development of high-throughput sequencing technology， it has been found that intestinal microbiota disorder may be an important promoting factor for the onset of oligoasthenozoospermia. Traditional Chinese medicine believes that "deficiency of kidney essence" is the core pathogenesis of oligoasthenozoospermia. In clinical practice， the method of tonifying the kidney and strengthening the essence has a significant therapeutic effect on oligoasthenozoospermia， but its mechanism of action has not been fully elucidated. Based on the basic theories of traditional Chinese medicine and molecular biology research， it has been found that there is a similarity between "kidney essence" and intestinal microbiota. During the onset of oligoasthenozoospermia， the disorder of intestinal microbiota has similarities with the pathogenesis of "deficiency of kidney essence" in traditional Chinese medicine. Moreover， traditional Chinese medicine for tonifying the kidney and strengthening the essence can regulate the disorder of intestinal microbiota， which may be one of the effective mechanisms for the treatment of oligoasthenozoospermia with the Bushen Yijing method. Based on this， this article explored the mechanism of Bushen Yijing method of traditional Chinese medicine in treating oligoasthenozoospermia from the perspective of intestinal microbiota， so as to provide new ideas for the treatment of oligoasthenozoospermia with traditional Chinese medicine.

Objective:To observe the effect of Wuzi Yanzong Wan made of different processed products on the apoptosis of spermatogenic cells in rats with kidney essence deficiency， and explore its protective effect on spermatogenic cells. Method:SD rats were randomly divided into the blank group， model group， whole raw product group， pharmacopoeia group and salt-processed product group， with 8 rats in each group. The kidney essence deficiency model was replicated by giving tripterygium glycoside tablets （the dose of 20 mg·kg<sup>-1</sup>）. The flow cytometry （FCM） was used to analysis the apoptosis of spermatogenic cells in testis， the immunohistochemistry （IHC） and Western blot were used to detect the expression levels of B-cell lymphoma-2 （Bcl-2） and Bcl-2 associated X protein （Bax） in the testis. High performance liquid chromatography （HPLC） was used to compare the contents of eight components （chlorogenic acid， ellagic acid， hyperoside， isoquercitrin， verbascoside， astragalin， kaempferol and schisandrin） in Wuzi Yanzong Wan made of different processed products， the mobile phase was composed of acetonitrile （A）-0.4% phosphoric acid aqueous solution （B） for gradient elution （0-5 min， 5%-15%A； 5-10 min， 15%-17%A； 10-25 min， 17%A； 25-35 min， 17%-26%A； 35-60 min， 26%-56%A）， the detection wavelength was set at 254 nm. Result:Compared with the model group， the total apoptosis rate of spermatogenic cells， protein expression of Bax and Bcl-2 in each administration group were improved. Among them， the pharmacopoeia group and salt-processed product group had significant effects （<italic>P</italic><0.01）， and the improvement effect of the pharmacopoeia group and salt-processed product group was significantly better than that of the whole raw product group （<italic>P</italic><0.05）. The contents of chlorogenic acid， hyperoside， isoquercitrin and verbascoside in Wuzi Yanzong Wan were increased after the herbal medicines being processed with salt-water. The content of ellagic acid in the salt-processed product group increased， while it decreased in the pharmacopoeia group. The contents of verbascoside， astragalin， kaempferol and schisandrin in samples from the salt-processed product group were greater than those in samples from the pharmacopoeia group. Conclusion:Wuzi Yanzong Wan may reduce the apoptosis of spermatogenic cells in rat testis by inhibiting the expression of Bax and promoting the expression of Bcl-2， and exert its effect of nourishing kidney and enriching essence. The enhanced anti-spermatogenic effect of Wuzi Yanzong Wan after processing may be related to the changes in chemical composition content after processing.

Doctors of past dynasties already understood deficiency of kidney essence,phlegm and blood stasis were important for dementia.As the increasing recognition about Alzheimer disease and the offer of 'toxin damaging brain vessel' hypothesis,we thought ?-Amyloid,the pathology of Alzheimer disease,had the character of damaging brain and marrow,so it belonged to internal toxin,internal toxin damaging brain and marrow was also the pathology of the disease.Deficiency of kidney essence was the precondition,insufficient brain and marrow and internal toxin damaging brain and marrow were the two causes of brain and marrow consumption and spirit dysfunction,which were the pathology of Alzheimer disease in Chinese medicine.

This article put forward the ‘heart-kidney centric theory’ on basis of analyzing the physiologic function, pathomechanism, onset rules and measures of prevention and treatment of cardio-cerebrovascular disease and degenerative disease induced by kidney deficiency. According the new theory, a new TCM drug, the Tongmai Qiangshen Wine, was developed.In human body, kidney produces essential qi and produces energy source for human body; heart transports the essential qi and is the physiologic mechanism of transport of energy source. Deficiency of essential qi will result in stasis of blood vessel, on the other hand, stasis of blood vessel can cause defi ciency of essential qi. The opposition, mutual rooting, waxing and waning, mutual convertibility of them reflect the development rules of cardio-cerebrovascular disease and degenerative disease induced by kidney deficiency, and determin the vital process of birth, growth, adult, aging and death. The construction of heart-kidney centric theory and the development and application of Tongmai Qiangshen Wine not only provide new theory and model for prevention and treatment of cardio-cerebrovascular disease and degenerative disease induced by kidney deficiency, but also provide new thoughts for preparation of new TCM drugs from aspects of how to maintain the feature of TCM, how to inherit and create on basis of combing holism with disease, principle, method, medicines.
