Aim To explore changes of gut microbiota in experimental model rats with hyperuricemia-induced mild cognitive impairment. Methods The experimental rats were provided with a diet containing 2% W/W uric acid (UA) and 2% W/W oxonic acid for period ranging from 1 day to 12 weeks. Morris water maze, blood biochemical indexes and other auxiliary models were used to evaluate the experimental animal model. Changes of gut microbiota were studied by 16S rDNA high-throughput sequencing. Results Compared with control group, the amount of UA of rats was significantly elevated. In addition, the rats showed significantly reduced spatial learning and memory in an escape latency and probe trial. The diversity of gut microbiota in model rats changed, and the relative abundances of 14 species of gut microbiota changed markedly. The abundance of Butyrivibrio, Erysipelotrichia, Erysipelotrichales, Erysipelotrichaceae, Erysipelatoclostridium, Lachnoclostridium 5, Anaeroplasmatales, Anaeroplasmataceae and Anaeroplasma in model mice increased, while that of Dorea, RuminococcaceaeUCG_005, Butyricicoccus, Tyzzerella 3 and Parasutterella decreased. Conclusions The continuous increase of UA can regulate gut microbiota, and the imbalance of gut microbiota is closely related to the development of mild cognitive impairment, which may be one of the mechanisms of hyperuricemia-induced mild cognitive impairment.

Background: The medicinal plant Murraya koenigii shown to have a wide variety of pharmacological activities (hypoglycemic and hypolipidemic). Objective of this study is the present study was designed to evaluate Antidiabetic and Hypolipidemic property of Murraya koenigii in experimentally induced diabetes in rats.Methods: Experimental diabetes was produced with single dose of Streptozotocin (STZ): 45 mg/kg IP. The rats were randomly allocated in various groups for 37 days. After the confirmation of diabetes on 7th day (>200 mg/dl), hydroalcoholic extract of Murraya koenigii (500 mg/kg) was administered orally to experimental rats from day 7th day and continued for 37 days thereafter. Various antidiabetic (Glucose, HbA1C), metabolic (Lipid profile), safety (pancreatic lipase, Creatinine, SGPT, Histopathology of Liver and Kidney) were evaluated in various group.Results: Efficacy of Murraya koenigii was observed on various parameter of diabetes. Administration of STZ resulted in a significant decrease in diabetic changes (increase in blood glucose, HbA1C), altered lipid profile (p<0.01) in the Control group rats as compared to sham group. Murraya koenigii treatment demonstrated significant antidiabetic indicated by restoration of blood glucose, HbA1C level (p<0.01) compared to Control group. In addition, Murraya koenigii also documented hypolipidemic property of test drug. As per biochemical assessment of Pancreatic lipase, Serum creatinine, SGPT and Histopathological report, the test drug reduce the pancreatic, liver and renal marker and also showed safe to pancreas, Liver and kidney. The histopathological assessment of the liver and kidney confirmed the biochemical findings.Conclusions: The study concluded that the Murraya koenigii possess antidiabetic efficacy.

Objective:To investigate the effect of bone marrow mesenchymal stem cell transplantation on immunoinflammatory response in experimental cerebral infarction rats,and to provide a method for the treatment of cerebral infarction.Methods:①Experimental rat model of ischemic cerebral infarction was established.Sixty SD rats in the lab from March 2015 to May 2017 were selected to complete the ischemic cerebral infarction model established by modified Zea-longa suture method.②Acquisition and culture of stem cells.In addition,5 rats were used to wash the bone marrow cavity with DMEM medium to collect bone marrow cells and the rat bone marrow mesenchymal stem cells could be isolated and prepared by adherent method.③Treatment of model rats.After successful modeling,60 rats were randomly divided into model group,control group and experimental group.After modeling,the model group only performed vascular ligation and did not insert the line plug;the control group injected equal volume of PBS at the lesion site;the experi-mental group injected 10 μl of third-generation bone marrow mesenchymal stem cells at the lesion site and measured the immune levels of the three groups of rats.To evaluate the effect of bone marrow mesenchymal stem cells in experimental rats with ischemic cerebral in-farction.Results:①In the inverted microscope,the volume of the first generation of MSCs became polygonal or fusiform on the third day,and the cells started to aggregate.The third generation of cells proliferated rapidly and arranged in a certain direction with swirling growth,and had multiple layers,the boundaries are unclear;②Fluorescence microscopy showed that:99% of cells obtained by fluorescence labeling,the obtained cells were bone marrow mesenchymal stem cells;③Control group and model group 1 month after treatment,there was no significant difference in neurological score between 2 months and 3 months(P>0.05).Neurological scores at 1, 2 and 3 months after treatment in the observation group were lower than those in the control group and the model group(P<0.05);④The levels of IL-6 and IFN-γ in the observation group were higher than those in the control group at 3 months after treatment(P<0.05).The levels of IL-10 and TGF-β in the observation group at 3 months after treatment were lower than those in the control group(P<0.05).⑤The percentage of CD4+CD25+Tregs in CD4+T cells at 1 month,2 months and 3 months after treatment in observation group was higher than that in control group and model group(P<0.05).Conclusion:Transplantation of bone marrow mesenchymal stem cells can help regulate rat immunity,reduce the level of inflammatory cytokines and promote the early recovery of neurological function.

ABSTRACT PURPOSE: To evaluate the effects of low intensity ultrasound on the healing process of third degree burn wounds in experimentally induced diabetic Wistar rats. METHODS: One hundred rats were divided into: control group; non-diabetic treated group; diabetic control group; diabetic treated group. The therapy was performed with a 3MHz ultrasound application, pulsed emission at 100Hz frequency, modulated at 20% with a dosage of 0.5W/cm2 during three minutes throughout 30 days. The surgical debridement of the wound was performed once at day 2. The wounds were morphometrically, macroscopically and microscopically evaluated at 3, 7, 14, 21 and 30 days. RESULTS: The wound contraction and collagen quantification were higher in all treated groups. Macroscopically, necrosis was higher in the diabetic control group. Granulation tissue was higher in treated groups during the proliferative and remodeling phase. Microscopically, there were greater mononuclear inflammatory infiltration, angiogenesis and fibroblast quantification in treated groups during the proliferative and remodeling phases. CONCLUSIONS: therapeutic ultrasound is beneficial in the inflammatory and proliferative phases of the healing process because it controlled the necrotic tissue, increased the granulation tissue and wound contraction. However in the remodeling phase it is not beneficial because of the continued angiogenesis and a mononuclear inflammatory infiltration.

RESUMO

Objective To analyze the mechanism of fentanyl combines midazolam,by comparison with some of the commonly used anesthetic method to determine in the ethical advantages of anesthesia.Methods 45 rats were randomly divided into three groups,Group A:1% anesthetized by intraperitoneal injection of sodium pentobarbital 40 mg/kg; Group B:5 % ketamine 120 mg/kg intraperitoneal injection; Group C:midazolam 5 mg/kg + fentanyl 0.05 mg/kg Last intravenous anesthesia.Observe the effects of anesthesia.Results The rats which are used of fentanyl conbines midazolam anesthesia,the onset of anesthesia time:0.02 ± 0.03 min,maintenance of anesthesia time:89.73 ± 22.59 min,the the narcotic operation time:1.51 ± 0.30 min,the success of anesthesia cases:15 cases.Conclusions Fentanyl combines midazolam compared with the currently used method of anesthesia,the advantages of the more prominent,it has significant improvement and protection about the security,utilization and recovery ethics in experimental rats.

RESUMO

The present paper studies and analyses the prevalence of epidemic hemorrhagic fever which occured in our university, The results showed that there was direct relationship between epidemic and infection of experimental rats. This paper also discusses the relation among the rat's positive detection rate, the feeding conditions and the rat's age.
