RESUMO

Objective To optimize the extraction conditions for total flavonoids from Cibotium barometz by response surface meth-od(RSM). Methods According to the center combination of Box-Benhnken,using the RSM,the effects of ethanol concentration,the ratio of solid to liquid,the extraction time,and the extraction frequency were studied by central composite design. Results The opti-mal conditions of extraction were as follows:60%ethanol,the ratio of solid to liquid 1:40,refluxing and extracting twice,and 1.5 h for each time. Conclusion The actual extraction yield was 1.44%. The method of extraction has higher extraction efficiency than other methods and can provide a basis for the industrial production of the total flavonoids from S. barometz.

RESUMO

This study was aimed to find out the optimum parameters to decoction extraction process of Brassicae Radix cream . With methods of integrated score and uniform design , contents of the total extract and the total polysaccharide were set as the target to investigate the optimization process of extraction. The experiment showed that the optimum extraction condition was as follows: ten folds of water, four times decoction at boiling tempera-ture , one hour for each time . It was concluded that the optimum parameters to decoction extraction of Brassicae Radix were stable and feasible, which can be used for the preparation and reference for Brassicae Radix cream in Tibetan clinics .

Introduction Cryptosporidium is an important protozoan cause of waterborne disease worldwide of concern to public health authorities. To prevent outbreaks of cryptosporidiosis, the monitoring of this parasite in drinking water is necessary. In the present work, the polymerase chain reaction (PCR) and nested-PCR techniques were used to detect Cryptosporidium in raw water from catchment points of four water treatment plants (WTP) in Curitiba, Paraná, Brazil. Methods First, DNA extraction techniques were tested in samples containing decreasing amount of oocysts in reagent water, and PCR and nested-PCR with specific primers for 18SSU rDNA of Cryptosporidium were conducted to determine their sensitivity. In reagent water, a commercial extraction kit provided the best analytical sensitivity, and PCR and nested-PCR allowed the detection of five and two oocysts, respectively, with the primers XIAOR/XIAOF and XIAO1F/XIAO2R. Results In the spiking experiments, only the PCR with the primers AWA995F/AWA1206R was successful at detecting concentrations of 0.1 oocysts/mL. Two catchments samples of raw water and/or water sludge from four WTPs were contaminated with Cryptosporidium. Conclusions The application of the techniques to monitor Cryptosporidium in water and detect contamination in water catchments of WTPs in Curitiba are discussed in the present work. .

RESUMO

Objective To optimize the alcohol extraction technique of the medical herbs such as Rhizoma Corydalis and Radix Angelicae Dahuricae. Methods To optimize the alcohol extraction technique with L9(34) orthogonal test. The content of the tetrahydropalmatine in Rhizoma Corydalis,which was used as evaluating criteria,was determined by HPLC. Results The optimized extraction technique was as follows: the medicinal material was extracted for 3 times with 3 times amount of 75% alcohol,1.5 hours per time. Conclusions The optimized technique is reasonable and the extraction rate was high.

RESUMO

OBJECTIVE:To establish a method for content determination of baicalin in Scutellaria baicalensis Georgi extract and to optimize the extraction technique.METHODS:The content of baicalin was adopted as index and was deter?mined by HPLC;Single factor test and orthogonal test were employed to optimize the extraction technique.RESULTS:The detected concentration of baicalin showed a good linear relation in the range of0.05912~0.02956mg/ml(r=0.99974),the average recovery was99.5%,RSD=1.10%(n=5);The optimal technique was twice recirculation in water solvent14times the amount of Scutellaria baicalensis Georgi flour,each time for1hour.CONCLUSION:The established method is conve?nient,and the extraction technique is stable,reasonable and practicable.

RESUMO

OBJECTIVE:To optimize the extraction technique of tiannian capsules. METHODS:The extraction technique was selected by orthogonal method,with the contents of radix astragali and polycose,as well as the recovery of dry ointment as indicators,the influential factors for the extraction of effective ingredients that included the amount of adding water,length of decocting time and frequencies of decocting were studied.RESULTS:The optimal extraction technique is that medicinal herb is decocted in water 6 times as much as the herb for 3 times with 1 h per time.CONCLUSION:The optimized extraction technique is scientific,economic and practicable,which can be a guideline for manufacturing.

OBJECTIVE:To select an optimal extraction technique of Whitmania pigra whiman.METHODS:The af?fecting factors such as variety of solvent,temperature,time,the amount of solvent and the frequency of the extraction were studied on the basis of protein content in the hirudo extract and activated partial thromboplastin time(APTT).RESULTS:The optimum extraction result was obtained in4%NaCl solvent of10-fold as much as the medicine with3times of extraction at60℃for6h((3h+2h+1h).CONCLUSION:The present technique is simple,scientific and practicable.

RESUMO

OBJECTIVE: To study the optimum technique for extraction of mixture volatile oil from Olibanam and Sandalwood. METHODS: The orthogonal test was adopted to optimize the extraction process using the yield rate of volatile oil as indicator. RESULTS: The optimum extraction conditions were as follows: powder forming by 10mesh, 10- fold water added, soaking for 10h and extraction for 8h. CONCLUSIONS: This extraction method is simple, efficient and suitable for industria-lized production.

RESUMO

AIM: To confirm the optimum extraction of the total-saponins from Patrinia Villosa Juss. METHODS: Optimum conditions(ethanol concentration,extraction hours,ethanol solution quantity) were determined by the uniform design test,the extract was refined with macropore resin AB-8,and samples were measured colorimetrically at ?=560 nm compared with oleanolic acid as reference substance. RESULTS: The result was 95% ethanol as solvent, extraction time for 1 h adding 8 times amount of ethanol solution at 100 ℃ water bath for two times.under this condition,the total-saponins content was 0.668%,in accordance with the design forecast. CONCLUSION: The optimum extraction technique and purified methods could extract the total-saponins efficiently from Patrinia Villosa Juss.The technique is simple and adapts for production.
