RESUMO

Immature hematopoietic progenitors are a constant source for renewal of hemocyte populations and the basic component of the tissue and cell repair apparatus. A unique property of these cells of internalizing extracellular double-stranded DNA has been previously shown. The leukostimulatory effect demonstrated in our pioneering studies was considered to be due to the feature of this cell. In the present research, we have analyzed the effects of DNA genome reconstructor preparation (DNAgr), DNAmix, and human recombinant angiogenin on both hematopoietic stem cells and multipotent progenitors. Treatment with bone marrow cells of experimental mice with these preparations stimulates colony formation by hematopoietic stem cells and proliferation of multipotent descendants. The main lineage responsible for this is the granulocyte-macrophage hematopoietic lineage. Using fluorescent microscopy as well as FACS assay, co-localization of primitive c-Kit- and Sca-1-positive progenitors and the TAMRA-labeled double-stranded DNA has been shown. Human recombinant angiogenin was used as a reference agent. Cells with specific markers were quantified in intact bone marrow and colonies grown in the presence of inducers. Quantitative analysis revealed that a total of 14,000 fragment copies of 500 bp, which is 0.2% of the haploid genome, can be delivered into early progenitors. Extracellular double-stranded DNA fragments stimulated the colony formation in early hematopoietic progenitors from the bone marrow, which assumed their effect on cells in G0. The observed number of Sca1+/c-Kit+ cells in colonies testifies to the possibility of both symmetrical and asymmetrical division of the initial hematopoietic stem cell and its progeny.

RESUMO

BACKGROUND:Pulmonary pericytes are located at the concavity where pulmonary vessels are interconnected, which is closely related to the formation and stability of pulmonary vascularization. However, there are few studies on how pulmonary pericytes affect the activity of pulmonary vascular endothelial cells in the pathogenesis of broncho-pulmonary dysplasia. OBJECTIVE:To analyze the relationship between the quantity of subgroups of pulmonary pericytes and endothelial cells in different stages of broncho-pulmonary dysplasia and to explore the effects of PDGFR-β+NG2+α-SMA+ pericytes on the early tube-forming activity of pulmonary vascular endothelial cells. METHODS:(1)Animal experiment:Twelve newborn C57BL/6 mice were randomly divided into normoxia and hyperoxia groups within 24 hours of birth, with six mice in each group.Mice in the hyperoxia group were exposed to an 85%O2 environment to build the mouse models of broncho-pulmonary dysplasia, while those in the normoxia group were fed in the same room air.The lung tissues of the mice in the two groups were taken at 7 and 14 days after birth. The pathological changes of the lung tissues were observed by hematoxylin-eosin staining.Three subgroups of pulmonary pericytes were measured by flow cytometry:PDGFR-β+NG2+α-SMA-,PDGFR-β+NG2+α-SMA+,and PDGFR-β+NG2-α-SMA+ cells.(2)Cellular experiment:Passage 3 PDGFR-β+NG2+α-SMA+ pericytes were co-cultured with mouse pulmonary vascular endothelial cells(experimental group)at a ratio of 1:4.Mouse pulmonary vascular endothelial cells cultured alone were used as controls. The tube-forming difference between two groups was analyzed after 15 hours of co-culture. RESULT AND CONCLUSION:(1)Animal experiment:Hematoxylin-eosin staining revealed that on day 7,the lung tissue of mice in the normoxia group had regular structure, obvious alveolar structure, and uniform size, while the number of alveoli in the lung tissue of mice in the hyperoxia group was less and the morphology of alveoli was irregular. On day 14,the alveoli of mice in the normoxia group gradually developed and matured, the alveolar structure gradually became regular and uniform in size, and the alveolar density gradually increased. The lung tissue structure of mice in the hyperoxia group was relatively disordered and the alveolar formation was delayed with the size gradually increasing and the alveolar structure being simplified.Flow cytometry results indicated that the number of PDGFR-β+NG2-α-SMA+ and PDGFR-β+NG2+α-SMA+ pericytes was increased in the hypoxia group compared with the normoxia group(P<0.01),while the number of PDGFR-β+NG2+α-SMA-pericytes and pulmonary vascular endothelial cells was decreased(P<0.01,P<0.04).(2)Cellular experiment:In the control group, the pulmonary vascular endothelial cells arranged in cords and extended around, and lumen-like structures formed in some areas. In the experimental group,PDGFR-β+NG2+α-SMA+ pericytes and their pseudopodia were not observed, the irregular grid structure of pulmonary vascular endothelial cells was significantly less than that of the control group, and the endothelial cells mainly clustered in clumps. To conclude,α-SMA+ pericyte subgroups are predominant in mice with broncho-pulmonary dysplasia.PDGFR-β+NG2+α-SMA+ pericytes can directly inhibit the tube-forming activity of pulmonary vascular endothelial cells, which may be involved in the process of abnormal vascularization in broncho-pulmonary dysplasia.

RESUMO

BACKGROUND:Exosomes are vesicle-like structures secreted by cells into extracellular compartments in the form of cytosol, which contain a large amount of microRNAs with important intercellular communication roles.MicroRNAs in exosomes rely on exosome transport and are able to enter target cells to exert important biological regulatory effects. In common bone and joint diseases, abnormal or damaged bone metabolism releases a large number of exosomes, while some exosome-derived microRNAs also promote the progression of osteoarthritis.Therefore,exosome-derived microRNAs are closely related to the skeletal system and are important for the development as well as diagnosis and treatment of many osteoarticular diseases. OBJECTIVE:To review the research progress of exosome-derived microRNAs in bone metabolism and bone and joint diseases. METHODS:Using"exosomes, extracellular vesicle, microRNA,miRNA,bone, bone diseases, bone formation, bone regeneration, bone resorption, bone destruction"as Chinese and English search terms, articles were searched on CNKI,Metasys, and PubMed databases.Finally,86 articles were included for summarization. RESULTS AND CONCLUSION:Exosome-derived microRNAs can regulate bone metabolism by affecting bone formation and bone resorption, and are closely related to the development of bone and joint diseases such as fracture healing, osteoporosis, osteoarthritis, rheumatoid arthritis, osteonecrosis of the femoral head, and osteosarcoma.Exosome-derived microRNAs will be an effective means of diagnosis and treatment of certain bone and joint diseases in the future. However, the current research on exosome-derived microRNAs in osteoarthritic diseases is limited, and more explorations and researches are still needed to diagnose and treat osteoarthritic diseases using exosome-derived microRNAs.

RESUMO

BACKGROUND:The application of orthodontic force triggers autophagy in the periodontal tissue via diverse signaling pathways, augmenting or attenuating the activity of relevant cell types such as periodontal ligament cells, osteocytes, osteoclasts, and osteoblasts, thus facilitating the process of periodontal remodeling. OBJECTIVE:To review the research progress in orthodontic force mediated autophagy in periodontal tissue and its impact on orthodontic tooth movement. METHODS:The PubMed,Web of Science,China Biology Medicine disc and CNKI were searched for literature published from 2010 to 2023 to summarize the progress in orthodontics-related autophagy. And 76 papers were finally included in the analysis and discussion. RESULTS AND CONCLUSION:Orthodontic force can trigger a series of biochemical signal changes through periodontal mechanical receptors and aseptic inflammation they cause, leading to autophagy in periodontal tissue.Subsequently, autophagy generates corresponding feedback through cascaded amplified signaling pathways such as Phosphoinositide 3-kinase/protein kinase B,Hippo, and mitogen-activated protein kinase pathways, promoting periodontal tissue remodeling and ultimately achieving tooth movement and stability.Orthodontic force-induced autophagy can differentially regulate bone resorption on the tooth pressure side and bone formation on the tension side.Related targets have good prospects in the clinical application of orthodontic treatment.Orthodontics and autophagy have complex mechanisms. However, existing research has only focused on exploring the role of autophagy in orthodontic tooth movement.Further exploration is needed to investigate the mutual regulatory effects between autophagy and orthodontic tooth movement,as well as the interactions between upstream mechanical receptors and signaling pathways involved in related pathways.

RESUMO

Ahmed glaucoma drainage valve (AGV) implantation is one of the main methods for the treatment of refractory glaucoma with a higher success rate than conventional filtration surgery. However, as a foreign body, the AGV often causes hyperplasia of scar tissue in the filtration area, wrapping around the drainage plate, thereby inhibiting aqueous fluid outflow and causing the intraocular pressure to rise again, leading to surgical failure.Although multiple injections of anti-metabolic drugs during and after AGV implantation can inhibit postoperative scarring, multiple postoperative subconjunctival injections will not only cause discomfort to patients, but also lead to complications. Therefore, it is necessary to improve the AGV to avoid repeated injection of the drug, achieve slow local release of the drug, and reduce the foreign body reaction of AGV at the same time.Recently, the development of new materials, such as Ologen collagen, poly (2-hydroxyethyl methacrylate), poly lactic-co-glycolic acid and opal shale and new techniques provides new methods to inhibit the scarring of filtration area after AGV implantation. This article reviews the methods and progress of inhibition of scar formation in filtration area from the aspects of development of AGV drainage plate materials, construction of drug delivery system of AGV combined with new materials, and improvement of AGV drainage plate structure.

RESUMO

[Objective]To explore the intervention effect of β-sitosterol,an effective monomer component of Eucommia ulmoides,on postmenopausal osteoporosis in mice and its potential mechanism.[Methods]Twenty-four female C57BL/6J mice of 12 weeks old were randomly divided into sham operation group, model group and β-sitosterol intervention group, with 8 mice in each group. The model of postmenopausal osteoporosis was established by bilateral ovariectomy.Mice in sham group and model group were given 0.9%sodium chloride solution and β-sitosterol intervention group was given β-sitosterol.Eight weeks after operation, all the mice were sacrificed and samples were collected.Micro-computed tomography(Micro-CT)was used to observe the protective effect of the β-sitosterol intervention on postmenopausal bone loss and trabecular bone breakage.Hematoxylin-eosin(HE)staining was used to analyze the bone histomorphology of the distal femur. The tartrate-resistant acid phosphatase(TRAP)staining and the alkaline phosphatase(ALP)staining were used to evaluate bone resorption and bone formation respectively. Then the expression level of β-catenin in each group was detected by immunohistochemistry.[Results]Micro-CT scanning showed that β-sitosterol could reduce bone loss and bone microstructure destruction in postmenopausal mice.HE staining also confirmed that β-sitosterol could effectively maintain the number and morphology of trabecular bone and inhibit abnormal fat accumulation caused by estrogen deficiency.TRAP staining also showed that β-sitosterol treatment did not affect osteoclast formation, while immunohistochemical results showed that β-sitosterol could significantly promote the expression of osteogenic marker ALP.Moreover,β-sitosterol can increase the expression of β-catenin in the distal femur to promote bone formation.[Conclusion]β-sitosterol treatment could enhance osteogenic differentiation and promote bone formation to relieve postmenopausal osteoporosis by upregulating β-catenin expression in vivo.

RESUMO

AIM:To investigate the effects of overexpressing α-Klotho(KL)in RAW264.7 cells stimulated by oxidative stress on the proliferation, migration, tube-formation and tight junction of human umbilical vein endothelial cells(HUVECs).METHODS:RAW264.7 cells were categorized into control, 4-hydroxynonenal(4HNE), and 4HNE+KL groups, with F4/80 expression assessed via immunofluorescence staining. Three groups of conditional media were prepared for HUVECs and culture divided into Mø-NC, Mø-4HNE, and Mø-4HNE+KL groups. Cell proliferation was evaluated using CCK8 assay, while scratch test and Transwell assays were employed to measure cell migration. Additionally, tube-formation assay was conducted to assess cell tubule formation, and Western blot assay was utilized to detect the protein expression levels of Claudin 5, Occludin and ZO 1.RESULTS:The results of immunofluorescence staining showed that the fluorescence intensity of F4/80 of RAW264.7 cells in the 4HNE group was significantly enhanced compared with the control group, while that of F4/80 in the 4HNE+KL group was significantly decreased compared with the 4HNE group(all P<0.05). The CCK8 assay results revealed a significant increase in the proliferation of HUVECs in the Mø-4HNE group compared with the Mø-NC group. Conversely, the proliferation of the Mø-4HNE+KL group exhibited a significant decrease compared with that in the Mø-4HNE group(all P<0.01). The results of scratch test and Transwell assays demonstrated a significant increase in the migration of HUVECs in the Mø-4HNE group compared with the Mø-NC group, while the migration of the Mø-4HNE+KL group exhibited a significant decrease compared with the Mø-4HNE group(all P<0.01). In the tube-formation assay, it was observed that the number of tubes formed by HUVECs in the Mø-4HNE group was significantly increased compared with the Mø-NC group, while that of tubes formed in the Mø-4HNE+KL group was significantly decreased compared with the Mø-4HNE group(all P<0.01). Additionally, the Western blot results revealed a significant decrease in the relative expression levels of Claudin 5, Occludin, and ZO 1 in the Mø-4HNE group compared with the Mø-NC group. Conversely, in the Mø-4HNE+KL group, there was a significant increase in the relative expression levels of Claudin 5, Occludin, and ZO 1 compared to the Mø-4HNE group(all P<0.01).CONCLUSIONS: KL inhibits the proliferation, migration, and tube-formation of HUVECs while enhancing the tight junction by changing the activation state of macrophages in the diabetic oxidative stress environment.

RESUMO

Coronary restenosis is an important cause of poor long-term prognosis in patients with coronary heart disease. Here, we show that lysine methyltransferase SMYD2 expression in the nucleus is significantly elevated in serum- and PDGF-BB-induced vascular smooth muscle cells (VSMCs), and in tissues of carotid artery injury-induced neointimal hyperplasia. Smyd2 overexpression in VSMCs (Smyd2-vTg) facilitates, but treatment with its specific inhibitor LLY-507 or SMYD2 knockdown significantly inhibits VSMC phenotypic switching and carotid artery injury-induced neointima formation in mice. Transcriptome sequencing revealed that SMYD2 knockdown represses the expression of serum response factor (SRF) target genes and that SRF overexpression largely reverses the inhibitory effect of SMYD2 knockdown on VSMC proliferation. HDAC3 directly interacts with and deacetylates SRF, which enhances SRF transcriptional activity in VSMCs. Moreover, SMYD2 promotes HDAC3 expression via tri-methylation of H3K36 at its promoter. RGFP966, a specific inhibitor of HDAC3, not only counteracts the pro-proliferation effect of SMYD2 overexpression on VSMCs, but also inhibits carotid artery injury-induced neointima formation in mice. HDAC3 partially abolishes the inhibitory effect of SMYD2 knockdown on VSMC proliferation in a deacetylase activity-dependent manner. Our results reveal that the SMYD2-HDAC3-SRF axis constitutes a novel and critical epigenetic mechanism that regulates VSMC phenotypic switching and neointimal hyperplasia.

RESUMO

ABSTRACT BACKGROUND: To the best of our knowledge, this is the first study to evaluate the effectiveness of specific concentrations of antibiofilm agents, such as N-acetyl cysteine (NAC), rifampicin, and ozone, for the treatment of pan-resistant Klebsiella pneumoniae (PRKp). OBJECTIVES: We evaluated the effectiveness of antibiofilm agents, such as NAC, rifampicin, and ozone, on biofilm formation in PRKp at 2, 6, 24, and 72 h. DESIGN AND SETTING: This single-center experimental study was conducted on June 15, 2017, and July 15, 2018, at Istanbul Faculty of Medicine, Istanbul University, Turkey. METHODS: Biofilm formation and the efficacy of these agents on the biofilm layer were demonstrated using colony counting and laser-screened confocal microscopy. RESULTS: NAC at a final concentration of 2 μg/mL was administered to bacteria that formed biofilms (24 h), and no significant decrease was detected in the bacterial counts of all isolates (all P > 0.05). Rifampicin with a final concentration of 0.1 μg/mL was administered to bacteria that formed biofilm (24 h), and no significant decrease was detected in bacterial count (all P > 0.05). Notably, ozonated water of even 4.78 mg/L concentration for 72 h decreased the bacterial count by ≥ 2 log10. CONCLUSION: Different approaches are needed for treating PRKp isolates. We demonstrate that PRKp isolates can be successfully treated with higher concentrations of ozone.

Abstract Introduction This paper presents an analytical approach to occupational therapy practice grounded in historical dialectical materialism. It understands occupational therapy professional practice as work within a capitalist system, and thus subject to analysis through the labor process theory. Objective To propose an analytical approach to occupational therapy practice aimed at supporting the development of an emancipatory practice framework based on the dialogue between Social Occupational Therapy and Latin American Collective Health. Method Utilizing emancipatory action research methodologies, this study conducted 10 workshops with 10 occupational therapists aimed at collective knowledge production. Results Human activity is identified as the object of occupational therapy working process, defined by the category work in its ontological dimension. Radical participation is set as the intended product of this process, which entails engaging in efforts to transform the social determination of collective living conditions. The individuals in this process include occupational therapists and the followed-up individuals, groups, and/or communities, all viewed as social beings collaborating toward social transformation. Four theoretical-methodological tools are identified: emancipatory analysis of human activities, emancipatory proposition of human activities, operationalization of radical participation, and interventions in the social fabric. Conclusion Occupational therapy, as a form of social practice, offers a specific contribution to the development of a collective revolutionary praxis. This praxis, aimed at realizing a concrete utopia, seeks to generate the means to create a new society.

Abstract: Organizational and Work Psychology (OWP) is permeated by a series of negative judgments and stigmas, mainly about its role as an intermediary of relations exploiting workers. Investigating the perceptions undergraduates have about it can bring access to what is central among the different ways of thinking about the subject. This study sought to analyze the perceptions of 100 undergraduates from two higher education institutions (HEIs), one public and private in Midwestern São Paulo about the importance of psychologists' role in OWP and their interest in working in the area in the future, comparing the results per HEI and between the first and fifth year of graduation. Graduates answered two structured questionnaires with open and closed questions. Results pointed to a greater appreciation of OWP in the private HEI and in the fifth year. Most first-year undergraduate students had little knowledge about OWP. Students from the private HEI took greater interest in working in the area than students from the public HEI. When asked if taking the courses changed their opinions, 49 students responded positively and only one negatively. Participants' responses were divided into positive, neutra,l and negative views about OWP. Results evaluated the perceptions on OWP in undergraduates the changes due to the impact of academic experiences.

Abstract: This article aims to discuss land rent as a basis for analyzing urban and agrarian issues. To this end, we carried out a thematic study whose analysis followed the historical-dialectic materialist method. We focused on the studies developed by Marx, Engels, as well as complementary bibliographical research. We sought to unveil the bond that unifies the essence of these expressions of the "social issue": private ownership of land and the process of its valorization based on the work incorporated into the land.
