RESUMO

ObjectiveTo explore the possible mechanism of dried fruiting bodies of Fomes officinalis (FOA) against Alzheimer's disease (AD) based on network pharmacology and experimental verification. MethodThe effective components of FOA were retrieved from a Bioinformatics Analysis Tool for Molecular mechANism of Traditional Chinese Medicine (BATMAN-TCM) and previous reports. The targets of the components were searched from PharmMapper and TargetNet, and the targets related to AD from Gene Expression Omnibus (GEO), DrugBank, among other databases. Thereby, the common targets of FOA and AD were obtained, and the protein-protein interaction (PPI) network and component-target network were established based on STRING and Cytoscape 3.7.1, followed by the topology analysis of the networks, and Gene Ontology (GO) term enrichment and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis of the common targets. The results were verified by the molecular docking and the in vitro cell experiment. ResultA total of 24 candidate components and 242 predicted targets of FOA, and 96 common targets of FOA and AD were screened out. The key components included [2-(1-carboxyhexadecylamino)-2-aminosuccinic acid], 3-keto-dehydrosulfurenic acid, and eburicoic acid, and the active targets were albumin (ALB), acetylcholinesterase (AChE), estrogen receptor 1 (ESR1), cysteine aspartate-specific protease-3 (Caspase-3), and beta-secretase1 (BACE1). The common targets were involved in 392 GO terms, and the key terms were the β-amyloid metabolic process and cholinesterase activity. A total of 77 KEGG pathways were obtained, which mainly included estrogen signaling pathway, cholinergic synapse, and AD. The results of molecular docking showed that 7 components of FOA had high binding affinity to amyloid precursor protein (APP), BACE1, AChE, and Caspase-3. The cell survival rate rose (P<0.01) and the mRNA and protein expression of APP, BACE1, AChE, and Caspase-3 reduced in FOA groups in a dose-dependent manner compared with those in the model group (P<0.05). ConclusionThis study reveals for the first time that FOA has multi-component, multi-target, and multi-pathway characteristics in the treatment of AD, which serves as a reference for further explaining the mechanism of FOA against AD.

Neolentinus ponderosus y N. lepideus son dos especies de hongos saprobios que poseen uso alimenticio tradicional en comunidades de los departamentos de Huehuetenango y Totonicapán, Guatemala. La capacidad degradadora de ambas especies les confiere potencial para el estudio de la producción de cuerpos fructíferos a nivel de sustrato, por lo que en esta investigación se evaluó el crecimiento de una cepa nativa de N. ponderosus y una de N. lepideus en el medio de cultivo agar extracto de malta (EMA) a diferente pH y también se estudió la degradación de la madera de dos especies de pino (Pinus tecunumanii y P. ayacahuite) en cámaras de podredumbre durante 12 meses. El mayor diámetro de crecimiento miceliar de la cepa N. ponderosus se observó a pH 7.0, en tanto que para N. lepideus se obtuvo a pH 5.0 y 5.6. Las colonias de ambas cepas evidenciaron textura afelpada, borde regular a irregular, color blanco, con o sin pigmento difusible, olor frutal, hifas de 1.0-5.0 µm de diámetro, con clamidosporas y fíbulas. La madera de P. tecunumanii degradada por la cepa de N. ponderosus y la madera de P. ayacahuite por la cepa N. lepideus, presentaron porcentajes de pérdida de peso de 8.76 (±5.58) y 12.07 (±5.66), respectivamente y en ambos casos se alcanzó solamente el estadio temprano de degradación. Los resultados de este estudio sientan las bases para investigaciones posteriores en los que se evalúe el cultivo en troncos de ambas especies para la producción de cuerpos fructíferos con fines alimenticios y comerciales


Neolentinus ponderosus and N. lepideus are two saprophytic fungi species used traditionally in Huehuetenango and Totonicapán, Guatemala. The degradative capacity of both species confers them potential for fruiting bodies production. This study evaluated the mycelial growth of two native strains of N. ponderosus and N. lepideus in malt extract agar (EMA) at different pH and the degradation of wood from two pine species in rot chambers during 12 months. pH 7.0 was the most appropriate for the mycelial growth of N. ponderosus and for N. lepideus were 5.0 and 5.6. The colonies of both strains showed fruity odor, velvety texture, regular to irregular edge, white color, with or without diffusible pigment, hyphae with 1-5 µm width, chlamydospores and clamp connections. Wood from Pinus tecunumanii and P. ayacahuite exhibit weight-loss percentages between 8.76 ± 5.58 and 12.07 ± 5.66, with N. ponderosus 145.2003 and N. lepideus 90.2002, respectively. In both cases reached the early stage of brown-rot decay. These results could be useful for future research that evaluate the fruiting bodies production in logs for food and commercial purposes

Objective To investigate the anti-tumor activity of triterpenoids from Ganoderma lucidum on different tumor cells . Methods The MTT assay was adopted to detect the in vitro inhibition effect on 5 kinds of tumor cells .The inhibiting curve was drawn ,IC50 was calculated for reflecting the compound′s cytotoxic activity .Results The in vitro experiments demonstrated that three kinds of triterpenoids compound monomer showed different degrees of inhibition effect ,in which the inhibitory effect of gano-derenic acid Y was stronger ,its IC50 on H460 lung cancer cells was 22 .4 μmol/L ,followed by 7-oxo-ganoderic acid Z2 ,its IC50 was 43 .1 μmol/L .Conclusion Ganoderenic acid Y shows a strong inhibitory activity on H 460 lung cancer cells ,7-oxo-ganoderic acid Z2 shows a certain inhibitory activity on H 460 lung cancer cells ,moreover the inhibitory activity is dose dependent .The three com-pounds of ganoderenic acid Y ,7-oxo-ganoderic acid Z2 and ganoderon B have no activity or very weak activity to the other detected cell lines .The anti-lung cancer activity of ganoderenic acid Y and 7-oxo-ganoderic acid Z2 needs to be further deeply studied .

It has reported that Ganoderma lucidum triterpenoids had anti-tumor activity. However, the anti-tumor target is still unclear. The present study was designed to investigate the anti-tumor activity of G. lucidum triterpenoids on different tumor cells, and predict their potential targets by virtual screening. In this experiment, molecular docking was used to simulate the interactions of 26 triterpenoids isolated from G. lucidum and 11 target proteins by LibDock module of Discovery Studio2016 software, then the anti-tumor targets of triterpenoids were predicted. In addition, the in vitro anti-tumor effects of triterpenoids were evaluated by MTT assay by determining the inhibition of proliferation in 5 tumor cell lines. The docking results showed that the poses were greater than five, and Libdock Scores higher than 100, which can be used to determine whether compounds were activity. Eight triterpenoids might have anti-tumor activity as a result of good docking, five of which had multiple targets. MTT experiments demonstrated that the ganoderic acid Y had a certain inhibitory activity on lung cancer cell H460, with IC₅₀ of 22.4 μmol•L ⁻¹, followed by 7-oxo-ganoderic acid Z2, with IC₅₀ of 43.1 μmol•L ⁻¹. However, the other triterpenoids had no anti-tumor activity in the detected tumor cell lines. Taking together, molecular docking approach established here can be used for preliminary screening of anti-tumor activity of G.lucidum ingredients. Through this screening method, combined with the MTT assay, we can conclude that ganoderic acid Y had antitumor activity, especially anti-lung cancer, and 7-oxo-ganoderic acid Z2 as well as ganoderon B, to a certain extent, had anti-tumor activity. These findings can provide basis for the development of anti-tumor drugs. However, the anti-tumor mechanisms need to be further studied.

We collected and identified 5,721 ectomycorrhizal fruiting bodies (EcM) from Naejangsan National Park from June 2004 to 2013, belonging to 1 phylum, 1 class, 6 orders, 19 families, 40 genera, and 196 species. Of these, 2,249 individuals were identified as 89 species belonging to 11 genera in 7 families in the Agaricales; 1,511 were identified as 43 species belonging to 2 genera in 1 family in the Russulales; 1,132 were identified as 50 species belonging to 21 genera in 6 families in the Boletales; 793 were identified as 8 species belonging to 3 genera in 2 families in the Cantharellales; 29 were identified as 3 species belonging to 2 genera in 2 families in the Thelephorales; and 7 were identified as 3 species belonging to 1 genus in 1 family in the Gomphales. Thus, most of the EcMs identified belonged to the following 3 orders: Agaricales, Russulales, and Boletales. Russulaceae were most common (43 species), followed by Boletaceae (39 species), and Amanitaceae (27 species); most individuals were Russulaceae (1,511), followed by Hydnagiaceae (1,071) and Boletaceae (804). The monthly distribution showed that the greatest number of individuals and species of EcM, including the dominant ones, occur around July~September at an elevation of 200~299 m, diminishing markedly above 600 m. The greatest number of individuals and species, including the dominant ones, were collected in the period with average temperatures 25.0~26.9degrees C, lows of 21.0~22.9degrees C, and highs of 30.0~31.9degrees C, relative humidity > 76%, and rainfall > 400 mm.

En el presente estudio se determinó la producción de cuerpos fructíferos de cinco cepas nativas de Agrocybe cylindracea sobre tres sustratos y dos tratamientos térmicos, a través del porcentaje de eficiencia biológica y la medición del diámetro de los píleos. Se encontró que el mayor porcentaje de eficiencia biológica de las cepas en los sustratos evaluados fue 115.84 %, que correspondió al sustrato constituido por 29% paja de trigo más 1% de harina de soya pasteurizado y obtenido por la cepa 58.01, la cual también produjo las mayores eficiencias biológicas en todos los sustratos evaluados. Al confrontar el porcentaje de eficiencia biológica en los diferentes sustratos, todas las cepas presentaron valores altos en el sustrato compuesto por 28% de paja de trigo, 1% harina de soya y CaCO3. Con respecto al diámetro de los cuerpos fructíferos, las cepas 58.01, 59.01, 60.01 y 638.08 produjeron píleos menores de 2 cm, entre 2-4 cm y mayores a 4.0 cm en los diferentes sustratos y tratamientos, excepto la cepa 59.01 que en el sustrato formulado con 29% de paja de trigo y 1% de harina de soya, solo produjo cuerpos fructíferos con píleos menores a 2 cm y entre 2-4 cm. En el análisis proximal de los basidiomas de las cepas evaluadas se obtuvo un alto porcentaje de proteínas, fibra cruda y carbohidratos, así como bajo porcentaje de grasas. Se recomienda que en futuras investigaciones o transferencia de tecnología a comunidades o entidades interesadas en el cultivo de este hongo, utilizar paja de trigo suplementada con harina de soya y como regulador de pH CaCO3, ya que en dicho sustrato se obtuvieron los mayores porcentajes de eficiencia biológica para la producción de cuerpos fructíferos de A. cylindracea.


This study determined the production of fruiting bodies of five native strains of Agrocybe cylindracea over three different substrates and two heat treatments, by the biological efficiency percentage and the measurement of diameters of pileus. The major percentage of biological efficiency found from the strains in the evaluated substrates was 115.84%, and corresponded to the substrate formulated by 29% of wheat straw and 1% of pasteurized soy flour, and obtained from the strain 58.01, which also produced the major biological efficiencies in all of the evaluated substrates. When confronting the percentage of biological efficiency in the different substrates, all the strains presented high values in the substrate comprising 28% of wheat straw, 1% of soy flour and CaCO3. In relation to the diameter of the fruiting bodies, the strains 58.01, 59.01, 60.01 and 638.08 produced pileus less than 2 cm, between 2-4 cm and greater than 4 cm in the different substrates and treatments, excepting the strain 59.01 which in the substrate formulated with 29% of wheat straw and 1% of soy flour, only produced fruiting bodies with pileus less than 2 cm and between 2-4 cm. In the chemical proximate analysis of the fruiting bodies of the tested strains, a high percentage of proteins, crude fiber and carbohydrates was obtained, and also a low percentage of fats. For future research or when transferring the technology to communities interested in mushroom cultivation, the utilization of wheat straw supplemented with soy flour and as a regulator of pH CaCO3 is recommended, as in this substrate the highest percentages of biological efficiency for the production of fruiting bodies of A. cylindracea where obtained.

To produce fruiting bodies of Oudemansiella mucida, porcelain fungus, on the oak sawdust medium, additives suitable for the mycelial growth and fruiting body formation were screened. In general, the mycelial growth of the three strains of O. mucida used in this study have been good on oak sawdust mixed rice bran of 20~30%. The mycelia incubated in potato dextrose broth for 7 days were inoculated on oak sawdust medium supplemented with various ratios of rice bran and incubated for 30 days at 25degrees C in the dark condition until the mycelia of O. mucida fully colonized the media from top to bottom. Then, top surface of the media in the bottles were horizontally scratched with a spatula and filled with tap water for 3 hours. To induce the primordial formation of O. mucida, the bottles were transferred to the mushroom cultivating room under 12 hrs of light (350 lux) and dark condition with relative humidity of 95% at 17degrees C. The primordia of O. mucida were formed on the surface of oak sawdust media after 7 days of incubation. The mature fruiting bodies were observed 5 days after primordial formation. The fruiting bodies O. mucida were formed on oak sawdust medium mixed with 5 to 30% rice bran. However, abundant fruiting-bodies of O. mucida were produced in oak sawdust medium supplemented with 20% rice bran. This is the first report associated with an artificial fruiting body production of O. mucida in Korea.

In order to breed a Cordyceps bassiana isolate that stably forms fruiting body in artificial cultivation, isolates derived from subculturing and single spores were tested through mating. From C. bassiana EFCC 783, three subcultured isolates EFCC 2830, EFCC 2831 and EFCC 2832 were obtained and fourteen single conidial isolates were obtained from these three subcultured isolates. Two different morphological types were found in the fourteen single conidial isolates. One type was able to form synnemata and another type was not able to form synnemata. Since switch of morphological type was not observed despite their continuous subculturing, cross was performed between the two types and the formation of fruiting body was examined. Ascospores were obtained from a selected fruiting body formed by hybrid of the cross. Self-cross and combinational cross of the ascospore-derived isolates generated hybrids that stably produce high quality fruiting body in artificial media.

To screen additives and their mixed ratio suitable for the mycelial growth and fruiting body formation of Oudemansiella radicata in the oak sawdust, additives such as rice bran, fermented soybean powder and wheat bran were used. Generally, the mycelial growth of O. radicata has been stable on oak sawdust mixed with rice bran of 5~20%. In case that O. radicata was cultured for about 30 days at 22 +/- 1degrees C under the illumination (350 lux) of 12 hours and moisture condition of 90 +/- 5%, the primordia have been formed gradually from red-brown crusts covering the surface of oak sawdust media. Based on the experimental results from 9 strains of O. radicata, fruiting bodies were produced widely on oak sawdust medium mixed with rice bran of 5 to 30%. Even though fruiting bodies of O. radicata have been produced well on oak sawdust media mixed with rice bran, fruiting bodies of O. radicata were produced intensively on oak sawdust media mixed with rice bran of 10%. Therefore, this result will provide a basic information for commercial production of fruiting body of wild O. radicata. This result is the first report associated with an artificial fruiting body formation of O. radicata in Korea.

To produce an artificial fruiting body of Armillaria mellea on the oak sawdust medium, seven strains of A. mellea were used. The top surface of oak sawdust medium covered with ground raw carrot was inoculated with each of 7 strains and cultured for 30 days at 25degrees C in the dark condition until the mycelia of A. mellea completely colonized the medium from top to bottom. Then, the mycelia which were fully covered on the top surface of the medium were scratched slightly with a spatula and filled with tap water for 3 hours. To induce the primordial formation, the 7 strains of A. mellea were transferred to the growth chamber under the illumination (350 lux) of 12 hours and relative humidity of 85 +/- 5% in a day and then cultured at 16 +/- 1degrees C. Only A. mellea IUM 949 could form primordia on the sawdust medium, but the other strains did not make primordia at the same condition. The primordia of A. mellea IUM 949 were formed 10 days after complete colonization of the medium and the fruiting bodies were produced 7 days after a primordial formation. The experimental results suggested that IUM 949 strain might be a good candidate for mass production of fruiting bodies of A. mellea.

Six strains of Sparassis crispa such as S. crispa DUM-01, DUM-02, DUM-03, DUM-04, DUM-05, and DUM-06 were tested for their mycelial growth on 6 different kinds of sawdust media and primordial formation on 10 different compositions of larch sawdust media. The highest mycelial growth was recorded on the larch sawdust. Of the 6 strains of S. crispa, S. crispa DUM-04 recorded the favorable formation of primordia. The primordial formation of S. crispa DUM-04 was more favorable on L-3 medium than 9 kinds of larch sawdust media. When S. crispa DUM-04 was cultured on the media of larch sawdust+pine sawdust, the formation of its fruiting bodies was more outstanding on the media of larch sawdust+pine sawdust than those of larch sawdust.

OBJECTIVE:To study the chemical constituents in the fruiting bodies of armillariella tabescens for searching active leading compounds.METHODS:Compounds were isolated from methanol extract of armillariella tabescens by chromatography column.RESULTS:Six compounds were isolated and identified as methyl oleate(1),stearic acid(2),3?-hydroxyergosta-5,7,22-triene(3),armillarisin B(4),5-hydroxy-uracil(5) and cerebroside B(6),respectively.CONCLUSION:Compounds 1,2,3,5,6 were firstly isolated from the fruiting bodies of armillariella tabescens.
