RESUMO

Objective: The effect of Liuwei Dihuangwan on blood metabolism during growth and development of rats was investigated by taking the overall metabolic profile and biomarkers of metabolomics as indexes. Method: Ultra performance liquid chromatography-high definition mass spectrometry(UPLC-HDMS) was employed to establish a blood metabolomics study method for characterizing the blood metabolic profile of rats at different time before and after administration. The pattern recognition method was used to integrate and analyze the metabolic profiles, and the differential metabolic markers related to drug action were searched. Based on metabolomics pathway analysis(MetPA) and Kyoto encyclopedia of genes and genomes(KEGG) and other databases, the metabolic pathways related to differential markers were analyzed to study the effect of Liuwei Dihuangwan on blood metabolism during growth and development of rats. Result: Liuwei Dihuangwan significantly affect the blood metabolism profiles during growth and development of rats by regulating 30 blood metabolic markers and 12 related target metabolic pathways, and 8 key metabolic markers were delineated. Conclusion: Liuwei Dihuangwan can significantly regulate the blood metabolic network during the growth and development of rats, thereby affecting the growth and development of rats.

RESUMO

Objective To observe the changes of glycocholic acid (CG) and evaluate the diagnostic value of CG combined with total bile acid (TBA) and leucine aminopeptidase (LAP) in various liver diseases. Methods From October 2016 to March 2017,210 serum samples of healthy people, asymptomatic hepatitis B virus (HBV) infected, hepatitis, biliary obstruction, hepatocirrhosis and primary hepatocellular carcinoma patients were collected.CG and LAP were detected by corresponding kits, and liver function, coagulation function and other indicators of patients were collected and analyzed statistically. Results The serum level of CG were elevated in the 4 liver disease groups and differed statistically from the normal group or the asymptomatic HBV infected group.CG level was positively correlated with LAP (r =0.380,P < 0.01).In liver function indexes,CG was correlated with total bilirubin (TB),direct bilirubin (DB),TBA and alkaline phosphatase (AKP).At the same time,CG was correlated with fibrinogen(Fib),thrombin time(TT).LAP and TBA were introduced into regression equation Y =-0.835 + 0.157X1 +0.312X2 (X1:LAP,X2:TBA,R2 =0.685) as final variables in multivariate linear regression to analyse the influencing factors of CG.Receiver operator characteristic (ROC) curve analysis showed that CG had the strongest ability to diagnose liver diseases in combination with LAP.Conclusions The change of CG level is of great significance in all kinds of liver diseases. The combination of LAP has the strongest ability to diagnose liver diseases.

RESUMO

Objective To investigate the influence of total bile acid (TBA) ,cholyglycine(CG) ,biochemical parameters of liv‐er ,tumor necrosis factor‐α(TNF‐α) in the patients of intrahepatic cholestasis of pregnancy (ICP) on parturients and perinatal new‐borns .Methods The clinical data in 164 cases of ICP in our hospital from January 2012 to December 2013 were retrospectively ana‐lyzed .TBA ,CG and TNF‐αwere detected by the radioimmunoassay and TBIL ,DBIL ,ALT and AST were analyzed by the biochemi‐cal analyzer .Results TBA ,CG ,TBIL ,DBIL and ALT in the patients with ICP were increased than those in normal pregnant women ,i .e .the liver function was damaged ,serum TNF‐αexpression was increased ,the above indexes in the patients with severe ICP were increased compared with the mild ICP patients ,postpartum hemorrhage volume was increased and the Apgar scores were decreased (P<0 .05) .Conclusion ICP is high risk pregnancy .TNF‐α,CG and TBA reflect the severity degree of ICP .

RESUMO

Objective To investigate the relationship of vascular endothelial growth factor (VEGF) with bile acid in intrahepatic cholestasis of pregnancy patients (ICP). Methods Thirty three ICP patients and 30 normal pregnant women in late pregnancy were studied, maternal serum VEGF concentrations were examined by enzymelinked immunoadsorbent assay (ELISA). Capillaries density in placenta are detected by s ABC immunohistochemistry, radio immunity method is used to assay the maternal serum glycocholic acid. Results Serum VEGF in ICP (2 072? 17) ng/L has negative correlation with glycocholic acid (47 9?14 2) ng/L, however, positive correlation with capillaries density in placenta (68?5)/HP; The serum VEGF in normal pregnant woman (2 256?30) ng/L does not have relationship to glycocholic acid (1 6?0 7) ng/L but it has positive correlation with capillaries density in placenta (74?7)/HP.Conclusion Cytotoxicity of high concentration bile acids impairs vascular endothelial cells resulting in lower excretion of VEGF in vascular endothelial cells of ICP, which lead to placental blood capillaries dysplasia and lower permeability, it may be one of reasons of fetal distress and fetal death in uteri in ICP.

RESUMO

The serum level of cholygylycine acid was determined in 118 patients with liver cirrhosis and the patients were followed up for 1 year.37 patients out of the 118 died in the fol-lowup period. It was found that the serum level of cholygylycine acid was more closely correlated with the mortality rate of the patients with liver cirrhosis than other commonly used indices such as hepatic encephalopathy, ascites.nutritional status, hihrubin, albumin and gamma-globulin.The serum level of cholygylycine acid was compared with Child's grading concerning the prediction of the mortality rate. After Logistic regression analysis.it was confirmed that the serum level of cholygylycine acid was the most ideal index for the prognosis of the patients with liver cirrhosis.
