RESUMO

This study aimed to prepare silk fibroin nanoparticles (SF-NPs) and assess the physicochemical properties and biocompatibility of the formulation. An optimized and simplified solvent displacement method was employed to obtain SF-NPs. Single-factor prescription screening, such as silk fibroin (SF) solution concentration, the ratio of SF solution to organic solvent, ultrasonication power and time, and different types of organic phases, was used to optimize the formulation. The characterization of the optimal formulation included particle size, polydispersity index (PDI), zeta potential, morphology, and stability. The in vitro cell compatibility of the nanoparticles was evaluated using CCK-8 and Calcein-AM/PI cell viability staining. The results showed that when SF concentration was 20 mg·mL-1, volume ratio of aqueous phase to acetone was 1∶6, ultrasonic power was 80 W and ultrasonic time was 3 min, the best SF-NPs was obtained. The nanoparticles prepared in this study exhibit a near-spherical shape, with a uniform size distribution, having an average size of 144.8 nm, a PDI of 0.174, and a zeta potential of -27.35 mV. Results from in vitro cell experiments demonstrate excellent cell compatibility of SF-NPs, showing the ability to promote cell proliferation. The SF-NPs which were successfully prepared in this study exhibit uniform particle size and excellent biocompatibility.

The optimal prescription of tanshinone Ⅱ_A(TSN)-glycyrrhetinic acid(GA) solid lipid nanoparticles(GT-SLNs) was explored and evaluated in vivo and in vitro, and its effect on acne after oral administration was investigated. The preparation processing and prescription were optimized and verified by single factor and response surface methodology. The in vitro release of GA and TSN in GT-SLNs was determined by ultra-performance liquid chromatography(UPLC). The effect of GT-SLNs on acne was investigated by the levels of sex hormones in mice, ear swelling model, and tissue changes in sebaceous glands, and the pharmacokinetics was evaluated. The 24-hour cumulative release rates of GA and TSN in SLNs were 65.87%±5.63% and 36.13%±2.31% respectively. After oral administration of GT-SLNs and the mixture of GA and TSN(GT-Mix), the AUC_(0-t) and AUC_(0-∞) of TSN in GT-SLNs were 1.98 times and 4.77 times those in the GT-Mix group, respectively, and the peak concentration of TSN in the GT-SLNs group was 17.2 times that in the GT-Mix group. After intragastric administration of GT-SLNs, the serum levels of testosterone(T) and the ratio of testosterone to estradiol(T/E2) in the GT-SLNs group significantly declined, and the sebaceous glands of mice were atrophied to a certain extent. The results demonstrated that obtained GT-SLNs with good encapsulation efficiency and uniform particle size could promote the release of GA and TSN. GT-SLNs displayed therapeutic efficacy on acne manifested by androgen increase, abnormal sebaceous gland secretion, and inflammatory damage.

Based on the innovation of modern science and technology and concepts,the study of innovative Chinese medicine research has not been divorced from the traditional thinking of natural medicine research,in the theoretical system of traditional Chinese medicine,there was basically no inheritance and lack of innovation,which made the research of new medicine of traditional Chinese medicine fall into an awkward position.We sum up our experience in 1 1th Five-Year and 12th Five-Year,combined with the upcoming 13th Five-Year of the research and development idea of component Chinese Medicine,analyzed the impact of 3 aspects on the modernization of Chinese traditional medicine and put forward the solving methods.① The scientific screening of high-level candidate prescriptions has always been the bottleneck of the development of new Chinese medicine and the basic premise of modernization of Chinese Medicine;② It is an important outlet and measure to promote the modernization of traditional Chinese medicine to vigorously carry out the research on the transformation of traditional Chinese medicine and to formulate a scientific and rational mode of transformation of traditional Chinese medicine;③ From the point of view of the development and reform of component traditional Chinese medicine,it is necessary to follow the basic theory of traditional Chinese medicine,and comprehensively to consider the characteristics of the components of Chinese medicine,and to lay the foundation for the screening and research and development of Chinese herbal medicines

OBJECTIVE: To evaluate the effectiveness of using the Clinical Medication Decision Support (CMDS) software in a children's hospital, discuss the potential improvement methods and provide references for researches on rational drug use software applied in pediatric medications. METHODS: Problematic prescriptions screened by CMDS from 06/15/2014 to 06/30/2014 were collected. The data were processed and analyzed using microsoft excel. RESULTS: Among the 68834 issues, 51.2% of them were a-bout incorrect dosage and administration. Among the 294 drugs reported by CMDS with issues on dosage and administration, 37.1% had no information for dosage of pediatric use in the drug labels; 74.5% of the issues on dosage and administration corresponded to drugs with complicated pediatric dosage scenarios; while most of these issues had no clinical significance based on further clinical analysis. CONCLUSION: Application of rational drug use software for general purpose in children's hospitals may have specific problems such as difficulty in handling complicated drug dosage scenarios for children. These problems can be solved by leveraging big data technology.

RESUMO

OBJECTIVE:To reduce the incidence of the preventable adverse drug events by using information technology. METHODS:The intelligent prescription screening system was developed and the hospital information system-based link between "contraindications" and diagnosis of patients,their laboratory results and creatinine clearances were realized. RESULTS:Of the total 5 546 prescriptions selected randomly within one day,46 laboratory results-related,28 diagnosis-related and 32 creatinine clearance-related contraindications were detected by the system. CONCLUSION:This link mode is practical in that those prescriptions that violating the contraindications can be found out by the system; however,the clinical effect of the intelligent prescription screening system remains to be proved.

Objective To prepare dispersible compound melatonin tablets which can be quickly effective and used conveniently Methods The effective factors in the ingredients of the dispersible compound melatonin tablets were observed by orthogonal experiment design and the prescription was screened by disintegrating time limit Results The dispersible tablets could disintegrate completely in 50 seconds and the suspension could pass through 710 nm sifter in the dispersing test The dissolution rate could reach 92 9% in 5 minutes Conclusion The dispersible compound melatonin tablets can be prepared conveniently and easily disintegrate and dissolve
