ABSTRACT INTRODUCTION: The vehicle for propofol in 1 and 2% solutions is soybean oil emulsion 10%, which may cause pain on injection, instability of the solution and bacterial contamination. Formulations have been proposed aiming to change the vehicle and reduce these adverse reactions. OBJECTIVES: To compare the incidence of pain caused by the injection of propofol, with a hypothesis of reduction associated with nanoemulsion and the occurrence of local and systemic adverse effects with both formulations. METHOD: After approval by the CEP, patients undergoing gynecological procedures were included in this prospective study: control (n = 25) and nanoemulsion (n = 25) groups. Heart rate, noninvasive blood pressure and peripheral oxygen saturation were monitored. Demographics and physical condition were analyzed; surgical time and total volume used of propofol; local or systemic adverse effects; changes in variables monitored. A value of p < 0.05 was considered significant. RESULTS: There was no difference between groups regarding demographic data, surgical times, total volume of propofol used, arm withdrawal, pain during injection and variables monitored. There was a statistically significant difference in pain intensity at the time of induction of anesthesia, with less pain intensity in the nanoemulsion group. CONCLUSIONS: Both lipid and nanoemulsion formulations of propofol elicited pain on intravenous injection; however, the nanoemulsion solution elicited a less intense pain. Lipid and nanoemulsion propofol formulations showed neither hemodynamic changes nor adverse effects of clinical relevance.


RESUMO INTRODUÇÃO: O veículo do propofol em soluções a 1 e 2% é a emulsão de óleo de soja a 10%, que pode provocar dor à injeção, instabilidade da solução e contaminação bacteriana. Formulações foram propostas com o objetivo de alterar o veículo e reduzir essas reações adversas. OBJETIVOS: Comparar a incidência de dor à injeção do propofol com a hipótese de redução associada à nanoemulsão e a ocorrência de efeitos adversos locais e sistêmicos com as duas formulações. MÉTODO: Após aprovação pelo Conselho de Ética em Pesquisa, foram incluídos neste estudo prospectivo pacientes submetidas a procedimentos cirúrgicos ginecológicos: grupos controle (n = 25) e nanoemulsão (n = 25). Foram monitorados frequência cardíaca, pressão arterial não invasiva e saturação periférica de oxigênio. Foram analisados dados demográficos e estado físico; tempo cirúrgico e volume total usado de propofol; efeitos adversos locais ou sistêmicos; alterações nas variáveis de monitoramento. Considerou-se significativo valor de p < 0,05. RESULTADOS: Não houve diferença entre os grupos em relação a: dados demográficos, tempos cirúrgicos, volume total usado de propofol, retirada do braço, presença de dor durante a injeção e variáveis de monitoramento. Verificou-se diferença estatística significativa na intensidade da dor no momento da indução da anestesia, com menor intensidade no grupo nanoemulsão. CONCLUSÕES: Ambas as formulações de propofol, lipídica e em nanoemulsão, elicitaram dor à injeção venosa, porém a solução de nanoemulsão promoveu dor em menor intensidade. O propofol lipídico e o propofol em nanoemulsão não apresentaram alterações hemodinâmicas e efeitos adversos de relevância clínica.

Objectives To compare the effect of the consumption of buriti oil and soybean oil on the metabolism of rats under stress induced by iron overload.Materials and methods A total of 28 rats were randomized into control groups who consumed diet added of soybean (CS) or buriti oil (CB) and gavage with saline and two experimental groups who consumed diet added of soybean (ES) or buriti oil (EB) and daily gavage with iron II sulfate as stress inducer. The fatty acid profile of diets was analyzed. Body weight and diet consumption were evaluated every two days. The lipid profile and liver weight of animals were evaluated at the end of the experiment.Results Diet added of soybean oil showed higher percentage of polyunsaturated fatty acids (45.6%) and diet with buriti oil was rich in monounsaturated fatty acids (66.9%). There were no differences in food intake, total cholesterol, HDL-cholesterol and LDL-cholesterol among groups (p > 0.05). However, animals fed with diet supplemented with buriti oil showed intermediate triglyceride levels (CB: 65 mg/dL; EB: 68.7 mg/dL) compared to ES group (102.5 mg/dL). The liver of rats from the CS group had higher weight (2.06 ± 0.2 g) compared to the CB group (1.56 ± 0.1 g).Conclusion Buriti oil consumption was able to minimize some changes related to iron overload.

Pequi is the fruit of Caryocar brasiliense and its oil has a high concentration of monounsaturated and saturated fatty acids, which are anti- and pro-atherogenic agents, respectively, and of carotenoids, which give it antioxidant properties. Our objective was to study the effect of the intake of a cholesterol-rich diet supplemented with pequi oil, compared to the same diet containing soybean oil, on atherosclerosis development, and oxidative stress in atherosclerosis-susceptible LDL receptor-deficient mice (LDLr-/-, C57BL/6-background). Female mice were fed a cholesterol-rich diet containing 7% soybean oil (Soybean group, N = 12) or 7% pequi oil (Pequi group, N = 12) for 6 weeks. The Pequi group presented a more atherogenic lipid profile and more advanced atherosclerotic lesions in the aortic root compared to the Soybean group. However, the Pequi group presented a less advanced lesion in the aorta than the Soybean group and showed lower lipid peroxidation (Soybean group: 50.2 ± 7.1; Pequi group: 30.0 ± 4.8 µmol MDA/mg protein) and anti-oxidized LDL autoantibodies (Soybean group: 35.7 ± 9.4; Pequi group: 15.6 ± 3.7 arbitrary units). Peritoneal macrophages from the Pequi group stimulated with zymosan showed a reduction in the release of reactive oxygen species compared to the Soybean group. Our data suggest that a pequi oil-rich diet slows atherogenesis in the initial stages, possibly due to its antioxidant activity. However, the increase of serum cholesterol induces a more prominent LDL migration toward the intimae of arteries, increasing the advanced atherosclerotic plaque. In conclusion, pequi oil associated with an atherogenic diet worsens the lipid profile and accelerates the formation of advanced atherosclerotic lesions despite its antioxidant action.

Background & objectives Breast cancer is a leading cause of cancer death in women; dietary fat is the one of the factors that influences its incidence. In the present study we investigated the effect of feeding cow ghee versus soybean oil on 7,12-dimethylbenz(a)anthracene (DMBA) induced mammary cancer in rat and expression of cyclooxygenase-2 and peroxisome proliferators activated receptor- γ (PPAR-γ) in mammary gland. Methods Two groups of 21 day old female rats (30 each) were fed for 44 wk diet containing cow ghee or soybean oil (10%). The animals were given DMBA (30mg/kg body weight) through oral intubation after 5 wk feeding. Another two groups (8 each) fed similarly but not given DMBA served as control for the gene expression study. Results In DMBA treated groups, the animal fed soybean oil had higher tumour incidence (65.4%), tumour weight (6.18 g) and tumour volume (6285 mm3) compared to those fed cow ghee (26.6%, 1.67 g, 1925 mm3, respectively). Tumour latency period was 23 wk on soybean oil compared to 27 wk on cow ghee. Histological analysis of tumours showed that the progression of carcinogenesis was more rapid on soybean oil than on cow ghee. The expression of cyclooxygenase-2 was observed only in DMBA treated rats and it was significantly less on cow ghee than on soybean oil. The expression of PPAR-γ was significantly more on cow ghee than on soybean oil. Interpretation & conclusions Our results show that dietary cow ghee opposed to soybean oil attenuates mammary carcinogenesis induced by DMBA; and the effect is mediated by decreased expression of cyclooxygenase-2 and increased expression of PPAR-γ in the former group.

OBJECTIVES: The goal of this study was to determine the possible mechanism that is involved in the blood pressureraising effect of heated vegetable oils. METHODS: Adult male Sprague-Dawley rats were divided into 11 groups; the control group was fed with rat chow, and the other groups were fed with chow that was mixed with 15 percent weight/weight palm or soy oils, which were either in a fresh form or heated once, twice, five, or ten times. Blood pressures were measured at the baseline and throughout the 24-week study. Plasma nitric oxide levels were assessed prior to treatment and at the end of the study. Following 24 weeks, the rats were sacrificed to investigate their vascular reactivity using the thoracic aorta. RESULTS: Palm and soy oils had no detrimental effects on blood pressure, and they significantly elevated the nitric oxide contents and reduced the contractile responses to phenylephrine. However, trials using palm and soy oils that were repeatedly heated showed an increase in blood pressure, enhanced phenylephrine-induced contractions, reduced acetylcholine- and sodium nitroprusside-induced relaxations relative to the control and rats that were fed fresh vegetable oils. CONCLUSIONS: The blood pressure-raising effect of the heated vegetable cooking oils is associated with increased vascular reactivity and a reduction in nitric oxide levels. The chronic consumption of heated vegetable oils leads to disturbances in endogenous vascular regulatory substances, such as nitric oxide. The thermal oxidation of the cooking oils promotes the generation of free radicals and may play an important contributory role in the pathogenesis of hypertension in rats.

PURPOSE: Evaluate the effect of flaxseed, olive and fish oil on the lipid profile, preservation of villosities and lymphocyte migration in the intestinal mucosa of Wistar rats. METHODS: Thirty Wistar male rats were divided into four groups, which received the AIN-93M diet, with changes only to their lipid source: flaxseed, olive, fish, and soy oil (control group). The serum was separated for the biochemical parameter analysis. A histological evaluation was performed in the ileal portion. RESULTS: The group which was fed fish oil presented lower values when compared to the other treatments for Total Cholesterol, High-density Lipoprotein Cholesterol and Triacylglycerol (p<0.05). The animals treated with fish and olive oils presented better intestinal villosities preservation. Less deposition of lymphocytes was observed in the flaxseed group (p<0.001). CONCLUSIONS: This study demonstrated that flaxseed, olive and fish oils present different responses than soy oil for the intestinal mucosa preservation and lymphocyte proliferation in Wistar rats.


OBJETIVO: Avaliar o efeito dos óleos de linhaça, oliva e peixe no perfil lipídico, preservação das vilosidades e migração de linfócitos na mucosa intestinal de ratos Wistar. MÉTODOS: Trinta ratos Wistar foram divididos em quarto grupos e receberam dieta AIN-93M, modificando para cada grupo apenas a fonte lipídica: óleo de linhaça, oliva, peixe e soja ( grupo controle). O soro foi separado para análise dos parâmetros bioquímicos. A análise histológica foi realizada na porção ileal. RESULTADOS: O grupo que recebeu óleo de peixe apresentou menores valores de colesterol total, lipoproteína de alta densidade e triacilglicerol (p<0.05). Os animais tratados com óleo de peixe e oliva apresentaram melhor preservação das vilosidades intestinais. Menor deposição de linfócitos foi observado no grupo tratado com óleo de linhaça (p<0.001). CONCLUSÃO: Este estudo demonstrou que os óleos de linhaça, oliva e peixe apresentam diferentes respostas em relação ao óleo de soja na preservação da mucosa intestinal e proliferação de linfócitos em ratos Wistar.

The aim of this study was to investigate the effects of fish oil containing diet versus Iranian fat consumption pattern and standard diets on serum glucose and insulin sensitivity in Wistar rat. Thirty female Wistar rats were randomly allocated to three dietary groups: a standard diet [containing soy bean-oil], diet containing fish oil and diet containing mixed oil which was designed based on Iranian population fatty acid intake. Dams in each group were fed one of the diets during pregnancy and lactation and the pups were also weaned onto the same diet. Fasting serum glucose [Photometry] and insulin [ELISA] in pups were assessed and insulin sensitivity calculated on puberty. Fasting serum insulin in fish oil-fed group was significantly less than two other groups [P=0.018] and insulin sensitivity in fish oil-fed rats was significantly more than two other groups [P=0.002]. It seems a diet containing fish oil [rich in long chain omega-3 fatty acids] causes more insulin sensitivity comparing to diet containing soy bean oil [rich in omega-6 fatty acids] and diet with Iranian population fatty acid intake pattern [rich in saturated fatty acids]

OBJECTIVE: To observe the effects of consuming repeatedly heated soy oil on the aortic tissues of estrogen-deficient rats. METHODS: Thirty female Sprague Dawley rats (200- 250 g) were divided equally into five groups. One group served as the normal control (NC) group. The four treated groups were ovariectomized and were fed as follows: 2 percent cholesterol diet (OVXC); 2 percent cholesterol diet + fresh soy oil (FSO); 2 percent cholesterol diet + once-heated soy oil (1HSO); and 2 percent cholesterol diet + five-times-heated soy oil (5HSO). After four months, the rats were sacrificed, and the aortic tissues were obtained for histological studies. RESULTS: After four months of feeding, the NC, FSO and 1HSO groups had a lower body weight gain compared to the OVXC and 5HSO groups. The tunica intima/media ratio in the 5HSO group was significantly thicker (p < 0.05) compared to the NC, OVXC and FSO groups. Electron microscopy showed that endothelial cells were normally shaped in the FSO and NC groups but irregular in the 1HSO and 5HSO groups. A greater number of collagen fibers and vacuoles were observed in the 5HSO group compared to the other treatment groups. CONCLUSIONS: Fresh soy oil offered protection in the estrogen-deficient state, as these rats had similar features to those of the NC group. The damage to the tunica intima and the increase in the ratio of tunica intima/media thickness showed the deleterious effect of consuming repeatedly heated soy oil in castrated female rats.

Two groups of fatty acids are essential to the body, the omega6 (n6) series derived from linoleic acid (18:2, n-6) and the omega3 (n3) series derived from alpha-linolenic acid (18:3, n-3). Fatty acids provide energy, are an integral part of the cell membranes and are precursors of prostaglandins, thromboxanes and leukotrienes collectively known as eicosanoids. Eicosanoids participate in development and synthesis of immunological and inflammatory responses. The fixed oils (1, 2, 3 ml/kg) containing alpha-linolenic acid, obtained from the seeds of Linseed (Linum usitatissimum), Soyabean (Glycine max) and Holy basil (Ocimum sanctum) were screened for their antiinflammatory activity using carrageenan, leukotriene and arachidonic acid induced paw edema models in rats and the antiinflammatory effects were compared with the standard drug indomethacin. Significant inhibition of paw edema was produced by all the oils in the highest dose (3 ml/kg) in all the models. While O. sanctum oil produced the maximum percentage inhibition in leukotriene induced paw edema, L. usitatissimum oil produced maximum percentage inhibition in carrageenan and arachidonic acid induced paw edema models. The results show that oils with higher alpha-linolenic acid content (L. usitatissimum and O. sanctum) produced a greater inhibition of paw edema suggesting that modulation of the course of inflammatory disorders may be achieved by altering the eicosanoid precursor (i.e. poly unsaturated fatty acids: PUFA) availability through dietary manipulation.

The testicular parameters and sperm morphology were studied in twenty healthy Chinchilla breed bucks of rabbits fed with different planes of soybean meal. Their ages were between 6 to 10 months with their weights ranging from 1.4 to 1.9kg.The same quality of feeds was fed to each group throughout the study and at the rate of 0.5kg/head/day. The feeds contained graded percentage of soybean meal and growers mash. Semen used in this study was from the caudal epididymides while morphological aberrations were determined from a total count of 400 spermatozoa in smears obtained with Wells and Awa stain. Results of the spermiogram showed that the colour and consistency of the semen appreciated as the percentage of soybean meal increased. Group D with 90% soybean meal was significantly higher (P<0.05) over and above the values of other groups. Group D had a significant increase (P<0.05) over and above values of other groups in terms of percentage live spermatozoa. Semen concentration increased from 1.20 + 1.56 xl0(6)/ml in group B with 30% soymeal to 2.00+2.67 xl0(6)/ml in group D with 90% soymeal. Testicular parameters also showed that there was more weights increase as the percentage of soymeal in the feed increased. It is therefore concluded and recommended that soymeal can be part of normal diet of Chinchilla Rabbit without any adverse effect on the reproductive potential of the male rabbit.


Se estudiaron los parámetros testiculares y la morfología espermática de 20 chinchillas sanas, alimentadas con diferentes raciones de soya. Sus edades estuvieron entre 6 y 10 meses con pesos que fluctuaban entre 1,4 y 1,9 kg. Se dió la misma calidad de alimentación a cada grupo y recibieron 0,5 kg por día cada una de ellas. El alimento contenía porcentajes graduados de soya y de grano molido. El semen usado en este estudio provino de la parte caudal del epidídimo, mientras que las observaciones morfológicas fueron determinadas desde un conteo total de 400 espermatozoides teñidos a través de técnica de Wells & Awa. Los resultados del espermiograma mostraron que el color y la consistencia del semen se incrementó cuando hubo mayor concentración de soya en el alimento. El grupo D con 90% de concentración de soya fue estadísticamente significativo (p<0,05), sobre los valores de los otros grupos. El grupo D tuvo un incremento significativo (p<0,05) sobre los valores de los otros grupos en términos de porcentaje de vida de los espermios. En el grupo B con 30% de soya en su alimento. La concentración de semen fue de l,2+l,56xl0(6)/ml y en el grupo D con un 90% de soya fue de 2,0 + 2,67 x 10(6)/ml. Los parámetros testiculares también mostraron que hubo incremeento en el peso cuando se alimentaron con mayor porcentaje de soya. Concluimos y recomendamos que la soya se parte de la dieta normal de la chinchilla sin ningún efecto adverso sobre el potencial reproductivo de la hembra.

O consumo de cápsulas de óleo de peixe (OP) por humanos visa a atenuaçäo dos sintomas e prevençäo de várias patologias. As alteraçöes metabólicas e funcionais em células e órgäos do sistema imunológico causadas pelo OP pela administraçäo intragástrica (AIG) foram avaliadas. Ratos recém-desmamados (50-70 g) foram submetidos a AIG diária com óleo de peixe, óleo de soja ou manteiga de cacau (0,4 por cento do peso), por 28 dias. Os dados da AIG do OP foram também comparados com os da dieta enriquecida com OP.
